Abstract

Sungai Opak adalah salah satu sungai utama yang mengalir di Provinsi DIY dan merupakan habitat bagi reptil seperti kura-kura, kadal dan ular. Environmental DNA (eDNA) merupakan metode yang lebih efisien untuk identifikasi spesies dibandingkan metode konvensional yang memiliki beberapa kekurangan. Tujuan penelitian ini adalah mengetahui keanekaragaman reptil di Hulu Sungai Opak dengan menggunakan metode sampling eDNA. Sampel air diambil dari bagian hulu Sungai Opak, kemudian dilakukan filtrasi, ekstraksi dan preservasi DNA, kemudian dilanjutkan PCR dan elektroforesis, sequencing, serta analisis bioinformatik. Hasil analisis eDNA menunjukkan adanya 6 spesies kura-kura dan 33 spesies ular. Metode eDNA dapat menunjukan hasil yang presisi dalam identifikasi sampel ke tingkat spesies, walaupun masih terdapat false positive dan false negative. Penggunaan eDNA metabarcoding dalam identifikasi keanekaragaman jenis reptil di hulu Sungai Opak dapat dilakukan secara cepat namun kurang efektif. Hal tersebut disebabkan oleh kontaminasi eDNA, kurang spesifiknya target sekuens dan primer untuk monitoring reptil, serta database referensi yang belum lengkap untuk reptil yang habitatnya di dalam dan sekitar perairan sungai di Indonesia.

Abstract

Keanekaragaman flora dan fauna Indonesia sangat beragam. Jenis fauna yang terkenal di Indonesia adalah lebah tanpa sengat yang tersebar di seluruh wilayah di Indonesia. Informasi keragaman lebah tanpa sengat dan hubungan kekerabatan masing-masing kelompok lebah akan memberikan informasi terkait data populasi. Analisis molekuler berguna sebagai ilmu dan alat untuk melakukan identifikasi spesies. Analisis molekuler keragaman lebah tanpa sengat dapat menggunakan DNA mitokondria (mtDNA) pada lebah. Salah satu gen pengkode DNA mitokondria adalah gen 16S rRNA. Penelitian ini bertujuan untuk menganalisis sekuen sampel lebah tanpa sengat secara molekuler dan menyusun peta kekerabatan sehingga dapat menjadi data genetik dan informasi keanekaragaman spesie lebah tanpa sengat di Kabupaten Lampung Timur. Metode yang digunakan adalah  isolasi dan amplifikasi gen DNA, elektroforesis dan visualisasi, sekuensing dan analisis hasil sekuensing  menggunakan Basic Local Alignment Search Tool (BLAST), dan  konstruksi pohon filogenetik menggunakan Molecular Evolution Genetic Analysis (MEGA11). Berdasarkan penelitian ini 3 sampel lebah tanpa sengat yang didapatkan dari eksplorasi di Kabupaten Lampung Timur terdeteksi sebagai spesies Heterotrigona itama dan memiliki kekerabatan yang dekat dengan spesies sundatrigona sp dan paling jauh dengan Paratrigona lineatifrons. Hal ini sesuai dengan hasil uji BLAST dan analisis data molekuler menggunakan software Mega 11.

Abstract

Avocado seeds (Persea americana Mill.) are known to possess various pharmacological properties, including notable anticancer potential. While preliminary studies have reported the cytotoxic effects of avocado seed extracts on breast cancer cells, there is still a lack of comprehensive research exploring the underlying molecular mechanisms responsible for these effects. This study explores bioactive compounds found in avocado seeds as potential agents targeting estrogen receptor alpha (ERα), a key biomarker in breast and cervical cancers. The investigation employs a range of computational approaches, including the Lipinski Rule of Five, ADME/Tox predictions, pharmacophore screening, and molecular docking analysis. Of the ten tested compounds, seven passed the Lipinski Rule of Five. ADME/Tox analysis revealed that most compounds exhibited adequate human intestinal absorption (HIA), poor blood-brain barrier (BBB) penetration, moderate Caco-2 permeability, and good plasma protein binding (PPB), while some were predicted to be mutagenic or carcinogenic. Pharmacophore modeling yielded an AUC of 0.87, with procyanidin B scoring 45.09 as a hit compound. Molecular docking revealed catechin, hyoscyamine, and atropine had the lowest Gibbs free energy (-5.15, -0.10, -0.07 kcal/mol). Among the compounds, catechin in avocado seed shows the highest potential for development as an ER-targeted anticancer agent.

Abstract

Red chili (Capsicum annuum L.) is one of the essential vegetable commodities in Indonesia. Nevertheless, the production of red chili is affected by disease due to fungal infection. Previous research showed that the consortial formulation of bacterial and Trichoderma without a carrier from Black Soldier Fly (Hermetia illucens) larva gut enhanced the disease resistance of red chili. Regardless, the research has not overcome several diseases caused by fungal infection. This study aims to analyze the effect of adding kaolin, talc, and zeolite as carriers with bacterial and Trichoderma consortia from BSF larval gut on the disease resistance of red chili caused by fungal infection. This experimental study was carried out in a randomized block design. The treatments were consortial of bacterial and Trichoderma + carrier kaolin (BTrK), talc (BTrT), zeolite (BTrZ), positive control of consortial without a carrier (K+(1)), positive control of inorganic treatment (K+(2)), and negative control of plants without treatment (K(-)). The results were analyzed by calculating disease incidence (DI) and severity intensity (SI) scores to determine disease resistance. New findings prove that BTrK enhanced resistance to fungal infection, namely cercospora leaf spot, leaf rust, and powdery mildew. BTrT formulation enhanced resistance to symptom severity. The study concluded that carrier formulation enhanced the resistance of red chili.

Abstract

Cabbage waste is household and market industry waste that contains significant nutrients, such as protein, fat, fiber, vitamins, and minerals. This waste has the potential to be utilized as animal feed, for example, for local catfish (Clarias batrachus). This study aims to utilize cabbage waste as an alternative feed. The research was conducted from April to June 2024 using a quantitative experimental method and proximate analysis on the feed, with four treatments, three replications, and a positive control. The results showed that treatment 3, with a feed dosage of 12%, was the most effective in influencing all observed parameters. This treatment resulted in a length growth of 14.22 cm, weight growth of 20.59 g, and a feed conversion ratio (FCR) of 1.73, which was better than the other treatments. The proximate analysis results indicated that the feed in treatment 3 contained 32.14% protein, 5.24% fat, and 9.30% carbohydrates.

Abstract

Postmenopausal women often experience insulin resistance, a condition linked to declining estrogen levels. Structurally similar to endogenous estrogen, phytoestrogens derived from Humulus lupulus L. (hops) hold potential as natural agents in hormone replacement strategies. This research utilized a comprehensive in silico methodology to assess 20 bioactive hop-derived compounds for their ability to act as agonists of estrogen receptor alpha (ERα). The evaluation involved Lipinski’s Rule of Five, ADME-Tox profiling, pharmacophore-based screening, and molecular docking analyses. Seven compounds satisfied the pharmacophore model criteria. Among them, Isoxanthohumol and 8-prenylnaringenin displayed the most favorable binding affinities, while Desmethylxanthohumol and Naringenin showed interaction patterns closely aligned with those of the native ligand. Although the majority demonstrated favorable pharmacokinetic properties, a few compounds presentedpotential mutagenic or carcinogenic effects. Considering binding performance, interaction resemblance, and predicted safety, these four compounds emerge as strong phytoestrogen candidates for ERα modulation in insulin resistance. Experimental validation through biological studies is necessary to substantiate these computational findings.

Abstract

Penelitian jenis penyu, sarang alami dan semi-alaminya di Pantai Selatan Yogyakarta telah beberapa kali dilakukan. Hasilnya pantai di Bantul termasuk Pantai Pelangi memiliki lokasi pendaratan penyu untuk bertelur yang baik. Penelitian jenis dan sarang penyu di Pantai Pelangi perlu diperbaharui. Penelitian ini bertujuan untuk mengkaji awal jenis penyu yang bersarang di pantai Pelangi selama Juni hingga Agustus tahun 2023 dan mengetahui pengaruh karakter fisik sarang alami dan semi-alami terhadap keberhasilan penetasan telur. Penelitian dilakukan pada Juni hingga Agustus 2023 di Pantai Pelangi, Kabupaten Bantul, Daerah Istimewa Yogyakarta. Penelitian dilakukan dengan metode survei. Data yang didapatkan: Penyu yang mendarat dan bertelur di Pantai Pelangi adalah penyu lekang (Lepidochelys olivacea). Kegagalan penetasan telur penyu di Pantai Pelangi disebabkan oleh beberapa hal, yaitu: lebar sarang semi-alami yang lebih sempit dari sarang alami; kedalaman sarang semi-alami terlalu dalam dan tegak lurus dibandingkan sarang alami; dan suhu pasir sarang semi-alami cukup panas. Saran: sarang semi-alami tidak perlu menggunakan ember, agar lebar dan kedalaman sarang dapat disesuaikan, kedalaman sarang dibuat agak miring guna memudahkan tukik merangkak keluar; perlu naungan di area sarang semi-alami guna mengurangi suhu panas pasir sarang semi-alami.

Abstract

Peradangan adalah respons perlindungan sistem imun terhadap  kekebalan rangsangan berbahaya. Aterosklerosis, penyakit pembuluh darah progresif yang ditandai penumpukan plak lemak dinding arteri, disebabkan oleh peradangan. NLRP3 berperan dalam berbagai penyakit inflamasi, termasuk aterosklerosis. Alternatif berbasis bahan alami seperti propolis Homotrigona apicalis dapat mengurangi peradangan dan mencegah perkembangan penyakit. Sebaliknya, obat antiinflamasi seperti NSAID dan steroid sering memiliki efek samping. Untuk pemahaman dari interaksi ini, digunakan pendekatan molekuler docking. Pendekatan ini memungkinkan penggunaan senyawa propolis H.apicalis sebagai antiinflamasi yang aman dan efektif. Dengan menggunakan metode molekuler docking, penelitian ini mengkaji potensi senyawa propolis H. apicalis sebagai penghambat jalur peradangan NLRP3 pada aterosklerosis. Senyawa dalam propolis, seperti Alpha-Amyrin dan Beta-Amyrin, menunjukkan aktivitas anti-inflamasi melalui interaksi hidrofobik dengan residu kunci pada protein NLRP3. Nilai afinitas ikatan tinggi Alpha-Amyrin (-10,7) kcal/mol dan Beta-Amyrin (-10,6kcal/mol), menunjukkan potensi penghambatan inflamasi yang efektif. Maka dapat disimpulkan bahwa Senyawa Alpha-Amyrin dan Beta-Amyrin dari propolis H. apicalis memiliki potensi sebagai penghambatan jalur inflamasi NLRP3 dalam aterosklerosis. Hal ini mengindikasikan Alpha-Amyrin sebagai kandidat yang sangat kuat dengan Beta-Amyrin sebagai senyawa pendukung dengan potensi serupa.

Abstract

Incision wounds are common occurrences in daily life, and appropriate wound management is essential to promote and accelerate the healing process. Wound care can be performed using topical preparations such as gels containing bioactive compounds. SCOBY kombucha is believed to contain bioactive compounds, such as compounds from green tea (especially polyphenols) and metabolites produced by microbes (organic acids, alcohols, vitamins, etc.) The aim of this research is to determine the effectiveness of SCOBY kombucha gel on the healing of incision wounds in mice (Mus musculus). The method used in this research is a Completely Randomized Design (CRD) with 8 treatment groups, including 2 control groups and 6 test groups. The concentrations of SCOBY kombucha gel used are 0%, 2%, 4%, 8%, 10%, and 12%. The SCOBY kombucha gel is applied twice daily until the wound closes. Observation of the incision wound is carried out by measuring the length of the wound that has not yet closed. Based on the conducted research, it was found that SCOBY kombucha gel is effective in healing incision wounds as evidenced by the reduction in wound size due to the bioactive compounds in SCOBY. The most significant wound healing effect was observed with the 12% SCOBY kombucha gel, as there was a significant difference compared to the negative control group, 0%, 2%, 4%, and 8% SCOBY kombucha gels.

Abstract

Tembakau (Nicotiana tabacum L.) merupakan salah satu tanaman unggulan yang berkontribusi dalam perekonomian Indonesia. Namun, perubahan iklim dapat memicu berbagai cekaman abiotik, seperti kekeringan yang dapat menghambat pertumbuhan tanaman. Salah satu upaya yang dapat dilakukan untuk meningkatkan toleransi tanaman terhadap kekeringan adalah melalui pemberian pupuk nitrogen. Penelitian ini bertujuan untuk menganalisis pengaruh pemberian pupuk nitrogen terhadap ketahanan tembakau (Nicotiana tabacum L. ‘Manilo’) pada cekaman kekeringan. Penelitian dilakukan menggunakan metode Rancangan Blok Acak Lengkap dengan perlakuan kombinasi tiga tingkat ketersediaan air (100%, 75%, dan 50% kapasitas lapang) serta tiga dosis pupuk nitrogen (0,6; 1,2; dan 1,8 g/polybag). Parameter yang diamati meliputi respons tingkat kerusakan serta aktivitas antioksidan enzimatik. Analisis data dilakukan menggunakan ANOVA dan uji DMRT pada taraf kepercayaan 95%. Hasil menunjukkan aplikasi pupuk nitrogen mampu meningkatkan aktivitas enzim antioksidan, terutama APX. Peningkatan dosis pupuk nitrogen berkorelasi positif dengan ketahanan tanaman terhadap kekeringan yang ditunjukkan melalui penurunan kebocoran elektrolit dan MDA, serta peningkatan aktivitas APX. Penelitian ini diharapkan dapat memberikan informasi ketahanan tembakau dengan pemberian pupuk nitrogen terhadap kondisi kekeringan.
