Abstract

Dimethylsulfoxide (DMSO) has been intensively studied the 19th century. Thissubstance is used as a medicine and solve. It can also be used as an absorptionenhancer of various drugs, either synthetical or herbal medicines. This study conductedwith DMSO as transdermal absorption enhancer of theophylline, which often used asan anti-cellulite dosage forms. The experiments studied in vitro transdermalabsorption of theophylline in gel dosage forms, containing 7% theophylline danvarious concentration of DMSO i.e. 0% , 3%, 5% and 7% DMSO respectively. Thesestudies used skin membrane of Wistar strain male rats in a vertical type diffusion Cell.The results indicated that DMSO was very potential as a permeation enhancertheophylline, the formula containing 7% DMSO increased the theophylline transportfrom 17,9 µg to 139,1 µg.

Abstract

Endophytic fungi used as a new resource to produce bioactive compounds such as antibacterial, antioxidant and anticancer. This study aimed to analyze the antibacterial, antioxidant, cytotoxic fraction of the separation ethyl acetate extract of mycelium of endophytic fungi genus Fusarium sp. Diethyl ether fraction showed high antibacterial activity in E.coli (20,75 µg/mL), S.typhi (35,08 µg/mL), and S.aureus (51,96 µg/mL). Test showed that the antioxidant activity of ethanol 96% fraction has highest antioxidant activity and total phenolic content (75,85 ± 0.87 mg GAE). Cytotoxic test on T47D breast cancer cells showed that the fraction of diethyl ether have highest activity with IC50 of 10,16 ± 0,88 µg /mL.

Abstract

Carotenoid compound, include licopene and lutein, has ability to increaseimmune system. One of organs included in immune system is spleen. The objective of thestudy is to observe the effect of licopene to the histopathologic appearance of ratspleen. Twenty female Sprague Dawley rat, 1 month old, 100 grams of weight devideinto 4 groups. The first group got licopene dosage 3 ug/kg of body weight, the secondgroup got licopene dosage 6 ug/kg of body weight, the third group got corn oil (vehiclecontrol) and the fourth group got aquadest. The treatment were given during 17 days,orally. At the day 14, the first, second and third groups were injected by aloxansubcutaneuslly. Three days after injection of aloxan, all of rats were sacrified, spleenwere taken in order to histopathological examination. Histopathologic appearances ofspleen were analized descriptivelly. The result showed that licopene dosage 6 ug/kg ofbody weigh able to increase centrum germinativum formation of spleen white pulp morethan licopene dosage 3 ug/kg of body weight. Base on the result can be concluded thatlicopene administration can increase centrum germinativum formation of spleen whitepulp.

Abstract

Faloak (Sterculia quadrifida R.Br) is used empirically by residents of East Nusa Tenggara for drug plant. The information of the active compound contained in the faloak bark yet specifically published. This study aims to determine the active compound contained in the faloak bark that have cytotoxic activity to T47D breast cancer cells. Extraction process using maceration method, isolation process using gradien isolation method, elusidation using combination of information of IR spectra, 1D-NMR, 2D-NMR and LC-MS, and cytotoxic activity test on T47D breast cancer cells using MTT method. The isolation result obtained that isolate of naphthoquinone derivative compound is 2,3-dihydro-6-hydroxy-2-methylenenaphtho [1,2-b] furan-4,5-dione with IC50 value on T47D breast cancer cell was 9,88 µg/mL and with an index selectivity value was 30,23.

Abstract

MeOH extract of Zingiber officinale Roscoe (ZO) were investigated for hepatoprotective effect at the doses (p.o) of 100 and 200 mg/kg body weight using CCl4 induction method. Male Swiss mouse (25±1,5 g) were treated with ZO doses 100 and 200 mg/kg bw and Sillymarin (100 mg/kg bw) for two week before treatment with a single dose of CCl4, 0.5 ml/kg bw, intraperitoneally. The mouse were sacrifactivity of SGPT-SGOT. The study showed the activity of SGPT of MeOH extract of ZO doses 100 mg/kg bw and 200 mg/kg bw, Sillymarin, control and normal groups are (34.51±5.34), (27.38±4.28), (27.26±1.06), (36.61±1.87), and (25.37±2.01) U/L, respectively. While the activity of SGOT of MeOH extract of ZO doses 100 mg/kg bw and 200 mg/kg bw, Sillymarin, control and normal groups are (88.88±0.94), (83.88±3.91), (83.99±7.24), (92.97±8.16), and (82.99±3.97) U/L, respectively. MeOH extract of ZO dose 200 mg/kg BW and Sillymarin significantly (p < 0,05) decrease the activities two enzymes (SGPT and SGOT). The results showed MeOH extract Zingiber officinale Roxb has hepatoprotective effect

Abstract

ABSTRACTKidney is one of the most important organs of the human body. The goal of diuretic therapy is to reduce edema in patients with impaired renal function. The therapy of diuretic is very important, because the therapy is effective to control of the volume of extracellular fluid, reduce protein exretion in urine and reduce on effects of hyperceluler complications. The purpose of this research is to provide information on the effect of giving diuretics with clinical outcome of patient with cronic kidney disease. The research was conducted by using longitudinal observational study design. Longitudinal observation is a method that perform data retrieval by a prospective census method with daily follow up until patient returns. The result of research showed that the most widely used diuretic was furosemide. Then the results also showed improvement of blood pressure, pulse, respiration rate, creatinine and urea although the results were not statistically significally (p>0.05). Mortality rate of the subjects in RSUP DR. M.Djamil Padang Hospital was very small. Patient with diuretic and without diuretic have equal mortality rate (p>0.05).Keyword: Kidney, CKD, Outcome, Diuretic

Abstract

Benzoic acid is one of the synthetic preservatives, work effectively at pH 2.5 to4.0, therefore it is widely used in acidic food or drink. This study aims to determinebenzoic acid content in some soft drink products, their conformity with Permenkes RINo.722/Menkes/Per/IX/88 on food additives, and to find out if there were any varianceof benzoid acid content in a different kinds of soft drink products.Determination of benzoid acid levels was performed by ultravioletspectrophotometry following solvent extraction of the benzoid acid with chloroform.Qualitatively, it was found that all of the sample contained benzoid acid. Quantitatively,the amount of benzoid acid in sample A was 227,73 mg/kg of materials; sample B was182,38 mg/kg of materials; sampel C was 259,52 mg/kg of materials; sample D was325,01 mg/kg of materials; sample E was 357,33 mg/kg of material. The resultindicated that there were variance of benzoid acid content in different soft drinkproducts. The use of benzoic acid in soft drinks was lower compaired to that ofPermenkes RI No.722/Menkes/Per/IX/88 (600 mg/kg of materials).

Abstract

Carvedilol is widely prescribed for long-term hypertension treatment. It is rapidly absorbable by oral administration, but its bioavailability is merely about 20% in humans. Drug delivery is therefore imperative to overcome this weakness. One form of transdermal drug delivery system is a patch. Transdermal patch is composed of various systems, for instance, a reservoir that uses a rate control layer to manage the rate of drug release. This research aimed to observe the effect of using resin gum as the control of drug release rate on the physical characteristics and release of carvedilol in a transdermal patch. The patches were prepared in 5 formulas with different quantities of resin gum, namely 50, 100, 150, 200, and 250 mg. Afterward, they were evaluated physically, and their dissolution and diffusion rates were analyzed. The results showed that resin gum with concentrations of 150 mg and 200 mg was physically qualified for rate control. Besides, the results of dissolution and diffusion tests revealed that transdermal patches with 150 mg of resin gum exhibited the best drug release and penetration.

Abstract

ARI is an acute respiratory infection requiring antibiotics. The use of antibiotics shouldbe routinely to prevent resistance. The design of this research was experimental by treatment ofthe cases. How to collect the data used prospective method. The subjects of research who fulfilthe inclusion criteria were 44 patients with acute respiratory infection were divided into twogroups: 22 patients who were given information as the treatment group and 22 patients whowere not given information as the control group. Data was collected through interview to knowthe rest of the drug and questionnaires filling compliance with Modification Morisky AdherenceScale (MMAS). The pattern for antibiotic used in patients of acute respiratory infection atcommunity health centre Kotagede I Yogyakarta were Amoxicillin (65.9%), Cotrimoksazol (25%), Erithromycin (6.8%), and Ciprofloxacin (2.3%). The level of patients compliance byquestionnaire MMAS was 59.09% high compliance, and 40.91 % low compliance. The level ofpatients compliance by the rest of drug was 86.36% and patients uncompliance was 11.36%.The results of questionnaire MMAS with the Chi-Square test p=0.220 (p>0.05), this shows thatgiving drug informations had no influence to level compliance for antibiotic used in patiens ofacute respiratory infection at Community Health Centre Kotagede I Yogyakarta.

Abstract

Potentially inappropriate medications (PIMs) use among elderly is a serious public health problem because it is intrinsically linked to increased morbidity, mortality, and healthcare costs. This study  aims to determine the association between number of drug prescribed, duration of hospital stay and PIMs use among elderly hospitalized patients at PKU Muhammadiyah Yogyakarta General Hospital. This study was analytic observational with cross-sectional study. Beers criteria 2012 were used to identify PIMs use among elderly hospitalized patients with osteoarthritis. Logistic regression analysis were used to determine association between number drugs of prescribed, duration of hospital stay and PIMs use. The results showed that 7 (38.9%) out of 18 hospitalized elderly patients were identified PIMs use and all of patients received atleast one PIM. The most common agents involved in PIMs were estazolam (11.1%), and metoclopramide (11.1%). Polypharmacy is a major predictor factor of PIMs use in this study.
