Abstract

ABSTRACT Hyperinsulinemia may precede insulin resistance in patients with Type 2 diabetes.  In this study, we tried to reveal an influence of long-term treatment of human  insulin to hyperglycemic and insulin resistance in male Wistar rats.  Before the experiment, blood-glucose  levels measured two hours after 2 g/Kg glucose imposition.  Rats receive human insulin (0.45-1.80) IU/KgBW/day subcutaneously every day for 14 days.   Furthermore, human insulin 1.8 IU/KgBW/day also provide a period of7,14and 21days. At theend of the experiment,we mademeasurements ofblood-glucoselevelstwo hoursafterthe imposition  glucose.There are, the influence oflong-termtreatment of human insulin determined bycomparing the blood-glucoselevelsbeforeandafter  insulin treatment.  Insulin resistance isdeterminedby comparing thehypoglycemiceffect of glibenclamide 10 mg/Kg  BW  to the control group.  The results concluded that the  humaninsulin1.80 IU/KgBW/day  treatment  for 14dayscan improveblood-glucoselevel'sWistarmale ratsup to126.369mg/dL(p<0.05)withpatternsdoes notdepend onthe length of timeof administration.The averageof % hypoglycemiceffect of glibenclamide10mg/kgBW  in ratswhoreceived treatment with human  insulin 1.80IU/kg BW/dayswereat 18,31%,smaller thancontrols (62.20%) (p<0.05). Decrease in hypoglycemic effects of glibenclamide 10 mg/kg BW caused by mice has insulin resistance might be due to a result of long-term insulin administration. Abstract

ABSTRACT Soursop leaves can be used as a drug hypercholesterolemia. Simvastatin is one of the drugs that can be used to lower total cholesterol and LDL. This study aims to determine the effectiveness of combination therapy EEDS and simvastatin in inhibiting elevated levels of total cholesterol and LDL induced rats a high-fat feed when compared with simvastatin alone or single EEDS. This study is experimental with post-test only control group design. Using the test animals were 30 male rats Wistar strain were divided into 6 groups. Test animals fed a high-fat twice daily for 14 days except for the normal control group, the test preparation administration once a day for 5 days. Measurement of total cholesterol using  the reagent kit CHOD-PAP and LDL measurement method  Direct Enzymatic Colorimetric Test with a spectrophotometer UV-Visibel. Data were statistically analyzed by One Way Anova followed by Tukey test. The results showed that administration of EEDS can inhibit LDL levels induced rats a high-fat feed. eeds combination and simvastatin can inhibit the increase in total and LDL cholesterol levels did not differ significan (p> 0.05), which means it has the same ability in lowering total cholesterol and LDL induced rats a high-fat feed simvastatin compared with administration of single and single EEDS. Abstract

ABSTRACT Hypertension is one of the degenerative disease of health problem  in among people. Bilimbi leaves (Averrhoa bilimbi L.) is one of plants which is have antihypertension effect and contains phytol, dietil phtalat, flavonoid.  This research is aimed  to know influence of ethanol extract of bilimbi leaves (EEBL)   through its on the increase of the antihypertensive effect of captopril in hypertensive rats. This research used a pre-test and post-test matched control group design.   Sample were 30 male Wistar rats  treated with MSG 100 mg/kgBW/day (po) for 14 days to induce hypertension. Hypertensive rats were divided into six treatment groups are  hypertension control group (0.5% CMC-Na 12.5 mL/kgBW/day) and positive control group (captopril 2.5 mg/kgBW/day), and treatment hypertensive rats EEBL (60 and 120) mg/kgBW/day, combination of EEBL-captopril (60+2.5)mg/kgBW/day and combination EEBL-captopril (120+2.5) mg/kgBW/day.  Suspension test were given for 14 days. Observed data is a decrease in blood pressure after giving of the suspension test. Data analysis was performed with the parametric t-test paired and nonparametric Wilcoxon with 95% level of confidence. The result shows that MSG can be as inducer of hypertension. EEBL (60 and 120) mg/kgBW/day has antihypertensive effect. The increase antihypertensive effect of captopril on systolic blood pressure is showed in the treatment EEBL 120 mg/kgBW/day.   Keywords: Hypertension, Monosodium glutamate, Averrhoa bilimbi L.

Abstract

ABSTRACT The absorbtion-based ointment of ethanolic extract of binahong leaves  (Anredera cordifolia (Ten.) Steenis) on 10% concentrate has antibacterial activity towards Staphylococcus aureus. In order to improve the activity of this ethanolic extract of binahong leaves,  it is necessarily to adjust the ointment’s base of formulation. The aims of study are to determine the ointment’s physical characteristics  which various base such as hydrocarbon, absorbtion and water solubility. The ethanolic extract of binahong leaves obtained from maceration method, using 70% of ethanol. The ointment was made based on the variation of its base which are hydrocarbon, absorbtion and water solubility. The obtained oinment then was examined on its physical  characteristics  properties including organoleptic test, pH, viscosity, stickiness, spreadability, and antibacterial activity toward Staphylococcus aureus on the agar plate medium. The organoleptic and pH test result data were analyzed descriptively. The data of stickiness, viscosity were analyzed using one-way ANOVA and Tukey with the level of confidence up to 95%, whilst the spreadability data was analyzed using Kruskal wallis and Man whitney. There were significant differences between the results of viscosity, stickiness, on each formula of ethanolic extract ointment of binahong leaves. However, there were no significant differences on the pshycal characteristic of the pH, organoleptic, homogenic, and spreadability.  Keywords: Ointment, Binahong leaves, Antibacterial activity, Staphylococcus aureus.

Abstract

ABSTRACT Recently, some research showed that there is an immunomodulator effect in the fermentation of Rosella kombucha mushroom  tea and it can be caused by flavonoid and organic acid. The purpose of this research was to show the activity of immunomodulator Rosella kombucha mushroom  tea to against the proliferation of lymphocyte cells of Balb/C mice in vitro and to show the dose and length of fermentation that induces highest immunomodulator activity.  Kombucha mushroom was fermented for 5 days, 10 days and also 15 days with boiled Rosella tea, water and sugar. The yield of fermentation in the concentration of 6,25; 12,5; 25; 50 and 100 µg/mL were tested on lymphocyte cells proliferation by the MTT Assay. The OD (Optical Density) data obtained was  statistically analyzed by nonparametric Friedman test and continuoused by Mann-Whitney test. The result of this research showed that the fermentation of Rosella kombucha mushroom tea  has an immunostimulatory activity on lymphocyte cells proliferation with the concentration 6,25; 12,5 and 25 µg/mL in the 10 days and the highest immunostimulatory activity  showed by the concentration 25 µg/mL and 10 days of fermentation time. Abstract

ABSTRACT Ethanol extract of Anacardium occidentale and rambutan bark  (Nephelium lappaceum) proved to have antifungal activity agains Candida albicans. Efforts to improve comfort and ease of use then made in the form of cream formulation.The aim of study is to know the antifungal activity of cream with various concentration of  Anacardium occidentaleleaf extract and rambutan bark againstCandida albicans. Extraction of ethanol extractAnacardium occidentale leaf (EAOL) and  rambutan bark (ERB) used maceration method. Solvents used ethanol 70% and ethanol 96%, respectively. Formulation cream of EAOL and ERB was maked 3 formulation.  Formulation cream EAOL with concentration of variations extract are FI (10,5%), FII (12,5%), FIII (14,5%). Formulation cream ERB with concentration of varions extract  are FI (10%), FII (15%), FIII (20%).  Antifungal activity was determined in the cream using agar diffusion method and  zone of inhibitionwere analyzed using descriptive. The results showed that  antifungal activity against  Candida albicans  of cream EAOL concentration 14,5% with zone of inhibition is 7,16 ± 0,28 mm and cream ERB concentration 20% with zone of inhibition is 9,5 ± 0,5 mm.  Kata Kunci : Antifungal,  Anacardium occidentale, Nephelium lappaceum,  Formulation ofcream

Abstract

ABSTRACTCandida albicans is a normal flora in the female genitalia which can cause vaginal discharge. White galangal rhizome contains essential oils of eugenol which have activities that inhibit the growth of Candida albicans. This study formulated the white galangal rhizome essential oil in the form of feminime hygiene to make it easier and more comfortable to use and to determine the antiseptic feminime hygiene activity of the white galangal rhizome oil of Candida albicans. The essential oil of white galangal rhizome is formulated in three variations of concentration namely 10%, 15% and 20%. Feminime hygiene of essential white galangal rhizome oil was evaluated such as organoleptic, homogeneity, specific gravity, foam power test, viscosity, pH and stability. Antiseptic activity testing was tested with SDA (Sabouraud Dextrose Agar) media and using agar diffusion method. The results of the feminime hygiene evaluation formulated showed results similar to the results of comparative evaluations (BN). The parameters of antiseptic activity are indicated by the formation of inhibitory zone diameters where F0 (negative control), F1 (10%), F2 (15%), and F3 (20%) respectively have an average diameter value of 11 mm, 14.65 mm 17.58 mm and 25.41 mm. The classification category of the response to microbial growth according to CLSI, F0 and F1 includes the weak group, the F2 is the moderate group and the F3 is the strong group. Based on the results of one-way ANOVA statistical analysis there were significant differences in liquid soap preparations F0, F1, F2 with F3 at (P <0.05). White galangal rhizome essential oil can be formulated into feminine hygiene and has antiseptic activity against the fungus Candida albicans.Keywords: white galangal, antiseptic, candida albicans

Abstract

ABSTRACT Getting old and environmental factors can deteriorate to hair problems. Celery (Apium graveolens L.) is a plant that is easily found in Indonesia and has been shown to have activity to increase hair growth. One of the pharmaceutical preparations that are applied in hair growth preparations is a gel. Gels generally use synthetic polymers, but in this study a natural polymer from chitosan and alginate was used as a base. Apart from their suitable rheological properties, chitosan and alginate are also known to have benefits in hair health. The purpose of this study was to maximize the use of celery leaves as hair growth in the form of a gel dosage form based on chitosan-alginate using the Box-Behnken response surface design method. This research was started from extracting celery leaves with ethanol, phytochemical screening, and determination of total flavonoid levels, making gels, and testing the hair growth activity. The results showed that the celery extract positively contained flavonoids (47.635 ppm / 0.5%), saponins, tannins, alkaloids, and steroids. The optimal formula is the middle formula with extract content of 0.5%, the ratio of chitosan: alginate (Chi/Alg) 1:1 and 4% base concentration. The FTIR spectrum showed that gel formation was due to the charge interaction between alginate and chitosan. The activity test results showed that the Chi/Alg base had an effect on hair growth but the concentration of celery extract 0.5% on that basis had no benefit compared to the base alone because the concentration was too low. Abstract

ABSTRACT Diltiazem hydrochloride (diltiazem HCl), a calcium channel blocker is widely used in the management of angina pectoris and hypertension. Because of its short biological half-life (3.5 h) and low oral bioavailability (40%) due to extensive hepatic metabolism leading to high frequency drug dosing. Therefore, diltiazem HCl is a suitable drug for transdermal formulation to improve bioavailability of the drug, patient’s convenience and compliance due to less frequent administration. The objective was to characterized the physicochemical properties and permeation of matrix film transdermal diltiazem HCl that prepared with polymers (PVA & EC) and PEG 400 as penetration enhancer. In this study, six formulation were prepared in different ratio of film forming polymers (5:5 & 7:3),  and 15%, 20%, 25% of PEG 400. Developed transdermal film  were characterized for  weight uniformity, thickness, folding endurance, moisture uptake, drug content and in vitro permeation study using commercial semi permeable membrane. The crystall of drug excipient interaction  were evaluated using X-ray diffraction technique. All the formulations were found to be suitable for formulating in terms of physicochemical characteristics and there was no significant differences. The in vitro skin permeation profiles showed increased flux values with increase  of PEG 400  as penetration enhancer. The X-ray diffraction studies revealed that the crystallinity of the drug decreased as the proporsion of PVA in the film increased.  Keywords : diltiazem HCl, PEG 400, PVA, patch transdermal, permeation

Abstract

ABSTRACTThe tannins contained in areca nut extract can inhibit the growth of Streptococcus mutans bacteria. To facilitate the use of betel seed extract as an antibacterial, it was formulated into an edible film with a concentration of 2.5%, 3% and 3.5%. Making edible films using solvent casting method and testing antibacterial activity using blood agar diffusion method. The edible film of areca nut extract made an evaluation included organoleptic, thickness, brittleness, water content, pH, and swelling test. The evaluation results of betel nut edible fim extract showed almost the same characteristics as the edible fim that had been circulating in the market which was used as a comparison. The presence of antibacterial activity from edible pinang seed extract is marked by the formation of clear zones. The test results of antibacterial edible activity of areca nut extract films 2.5%, 3% and 3.5% showed the average diameter of clear zone was 14.96 mm, 15.49 mm and 17.05 mm which were included in the strong category.Keywords: Areca catechu Linn, antibacterial, edible film, tannin, streptococcus
