Abstract

Freezing process in ram spermatozoa caused damage of the plasma membrane and acrosome thatlead to the decrease of spermatozoa fertility. Research was conducted to evaluate acrosomal damageduring freezing process using lectin histochemical technique. Semen was collected twice a week usingartificial vagina from 1-2 years old Garut ram. Immediately after collection, characteristic of semenquality was evaluated then diluted with Niwa and Sasaki Freezing (NSF) medium. Semen was loadedinto 0.25 mL mini straws and equilibrated at 4oC for two hours. Straws were then frozen and stored inliquid nitrogen. Evaluation of sperm characteristic (motility, viability and plasma membrane integrity)and acrosomal integrity were done during the freezing process. Detection of acrosomal integrity was observedusing Fluorescens isothiocyanate (FITC) and Avidin-Biotin-Complex (ABC) staining methods. Data ofcharacteristic spermatozoa and acrosomal integrity were analyzed using ANOVA. Result of the experimentsshowed that the percentage of motility, viability and plasma membrane integrity of spermatozoa before freezing (83 ± 2.7%; 88.8 ± 2.6%; 88.2 ± 3.7%) were significantly decreased (P<0.05) after equilibration (71± 4.2%; 84.2 ± 5.0%; 76.2 ± 1.3%) and after thawing (40 ± 3.5%; 61.08 ± 3.3%; 51.2 ± 10.4%). The percentageof intact acrosomal spermatozoa using FITC and ABC methods during freezing process were 93.63 ±2.73%; 88.04 ± 3.2% and 81.73 ± 4.77% VS 94.54 ± 0.26%; 88.17 ± 0.38% and 79.38 ± 2.06%, respectively.In conclusion, the characteristic of spermatozoa were significantly decrease (P<0.05) during freezing process.Furthermore, the integrity of acrosome spermatozoa can be well analyzed during freezing process by usinglectin histochemical staining methods.

Abstract

Urine crystal is a crystal nucleus which tend to form urine stone. The case of urine stone seems to beincreased every year. Crystallization could induce acute tubular necrosis which impact on renal dysfunction.The signs of this condition are high level of urea, creatinine and decrease glomerulus filtration rate. Theobjective of this research was to evaluate the effects of infusum Persea americana Mill as an inhibitorcrystallization which induced by ethylene glycol on white male rats. 20 male rats were divided into 4groups; K1 as negative group received only distilled water ad libitum, K2 as positive group receiveddistilled water containing ethylene glycol, K3 (dose 5%) and K4 (dose 10%) as treatment groups receivedwater containing ethylene glycol and avocado leaves infusion. Phytochemsitry screening of infusion avocadoleaves consisted of flavonoid, saponin, tanine and quinone. Result of analysis showed that the level ofureum and creatinine on K2 was higher than K3 and K4 group. The increased level could be inhibited byinfusion avocado leaves. The measurement of glomerular filtration rate in treatment groups wassignificantly different (p<0.05). Descriptive histopathology observation showed that renal lesio in grouptreatment (K3 and K4) were declined. Large crystal calcium oxalate on K2 group was observed by usingpolarized microscope, whereas small crystal calcium oxalate were seen in the infusion of avocado leavesgroups. These result showed the ability of infusion of avocado leaves as an inhibitor on the growth ofcrystallization calcium oxalate

Abstract

The research objectives were to analyze reproduction performance and productivity of swamp buffalofrom different altitudes in Cianjur and draw up a hierarchy of productivity strategy development usinganalysis of SWOT (Strength, Weakness, Opportunity, Threats) and Analytical Hierarchy Process (AHP)with four criteria: technology, costs, impact, and the response of farmers. Survey was conducted in Cianjurduring January-March 2014 by interview prepared questionnaires and direct observation of 63 buffalo farmers. Secondary data were also obtained from relevant agencies. Primary data were collected usingdirect observation of 139 reproductive female buffaloes then were further analyzed. A total of 58 buffaloesat their productive period were sampled and taken their morphometric data. Whilst 37 buffaloes weremeasured their frame size using Body Condition Score (BCS). The results showed that the reproductionperformance of buffaloes in the lowlands are not significantly different from those in the highland. The ageat first oestrus, first mating, first calving, gestation period were 25.6 months, 26.6 months, 38.7 months,11.8 months, respectively.. The oestrus period was 5.3 days, and post-partum mating interval was 54.6days. Differences in altitude and sex significantly affected (P <0.05) the morphometry assessment. Thebody weight of male buffaloes were found lower than the females both in highlands and lowlands (P<0.05).The body conditioning score of buffalo performance at highland was better compared to those in thelowland. Based on the SWOT analysis and AHP: (1) The main strategy is to improve the technology basedon the criteria of internal weakness by increasing scale holdings to seize opportunities buffalo meat selfsufficiency;(2) based on the criteria of cost and impact, the strategy was to cover threats over the professionout of the region by empowering farmers (facilitate increased productivity buffalo); (3) based on the responsecriteria, the primary strategy is to improve the quality of education of farmers by facilitating productivityimprovement opportunities to achieve self-sufficiency buffalo meat. The main development strategy basedon the four criteria: technology, cost, impact, and farmer response were increasing of buffalo ownershipscale, production facilities, and farmers education quality.

Abstract

Catharanthus roseus is one plant recognized as medical potential which can decrease cholesterol. Thepresent study was carried out to evaluate the effect of aqueous leaf extracts of C. roseus on plasma lipidprofil levels in rats cholesterol rich diet and atherogenic index. This study was carried out on 15 SpraqueDawley male rats randomly distributed into five groups (n=3). Rats hypercholesterolemic wereadministrated cholesterol rich diet containing 1% (w/w). Normal control group with normal diet whitoutextracts (K1), positive control hypercholesterolemic group (K2) with cholesterol rich diet whitout extracts,and others three groups (K3-K5) were feed high cholesterol and 1 mL of aqueous leaf extracts of C. roseuswith a dose of 20% (w/v), 40% (w/v) and 80% (w/v) respectively, administered twice daily 1 mL orally.Treatment was given for 28 days. After treatment, the plasma lipids profile such cholesterol total, highdensity lipoprotein (HDL), low density lipoprotein (LDL), triglyceride (TGA) level were measured. Theresults showed that during the period of treatment with cholesterol rich diet have produced under condition hypercholesterolemic of rats with average cholesterol levels of 124.33 ± 4.04 mg/dL (K2 group). Treatmentwith extract dose of 80% showed the best results among the other doses in reducing levels of total cholesterol,triglycerides, LDL and raise HDL levels. Extract treatment dose of 80% up to four weeks resulted incholesterol total (79.33±3.51 mg/dL), TGA (72.33±6.65 mg/dL), HDL (55.00±3.60 mg/dL), and LDL(9.87±5.34mg/dL). In addition, value ratio of cholesterol:HDL was 1.4: 1 and atherogenic index value was0.44. These results suggest that extracts of leaf C. roseus optimum doses 80% (w/v) has hipolipidemicactivity in hypercholesterolemic rats and it has low atherogenic index value.

Abstract

Lead is a heavy metal polluting the environment, and its accumulation in animal or human bodies canhave neurotoxic and nephrotoxic effects on animals and human. Lead-contaminated chicken meat can bethe source of lead to human. Lead exposure to human can be assessed by measuring its concentration andaccumulation in chicken body parts and analyzing chicken consumption patterns. This study was conductedto measure lead concentrations in chicken body parts and to estimate lead exposure caused by consumptionof chicken body parts (breast, legs, wings) and tissues (meat, skin, cartilage, spongy bones). Samples wereextracted by using aqua regia digestible method with a mixture of HCl: HNO3 (3:1; v/v) and leadconcentrations were measured by the Atomic Absorption Spectrophotometry (AAS) method. The leadconcentrations in chicken tissues varied from< 0.01 to 1.81?g.g-1dry weight.The average concentrations oflead in chicken tissues were lower than the recommended safety level of lead in chicken meat (1.0?g.g-1),except for the breast cartilage (1.03?g.g-1). The lowet accumulation level 2.6 ?g g-1 was found in domesticchicken wings while the highest of 32.9 ?g g-1 was found in broiler chicken breast (total of meat, skin,cartilage). Based on the data of lead accumulation in chicken tissues, a polynomial equation describing theprobability (P) to be exposed to certain amount of lead in chicken tissues (A, in ?g) was determined as P =-(1 x 10-3)A2 + (6,4 x 10-2)A.

Abstract

Neurotransmitter found in the enteric nervous system that plays an important role in a variety ofenteropathies, including inflammatory bowel disease. Alteration of nitrergic neurons has been reported to bedependent on the manner by which inflammation is caused. This study was performed to determine theprofile of neural nitrergic with Nicotinamide Adenine Dinucleotide Phosphate-diaphorase (NADPH-d)staining in the cecum of broiler infected by Eimeria tenella (E. tenella). Thirty six health free of coccidiosisbroiler one-day old chickens (DOC) used in this study. All of broiler adapted for 7 days with food and drinkgiven ad libitum then divided into three groups, each group consisted of 12 broilers. The 1st group wascontrol only given orally 1.0 ml of distilled water/ heads , while 2nd group was infected with a single dose of5x103 oocysts/ head orally, and 3rd group was infected with a single dose of 2x104 oocysts/ head orally. Onday 7 post infection, all of chickens were fasted for 12 hours then were euthanized and cecum was taken.Lesion score of cecum was assessed. Furthermore, tissue of the coecum was prepared for hisochemicalusing NADPH-d staining to determine morphology, and morhometric of nitrergic neurons. The resultshown that cecum lesion score of group I is 0, group II is +2, group III is +3. Neuron density in the cecum ofgroup I is 2794.96+4.65 neuron/cm2, group II is 4930.36+63.73 neuron/cm2 and group III is 7892.31+44.97 neuron/cm2. Neuron density of group II and III increased signifcantly (p<001) than group I. Nitrergic neurondesity of group III increased significantly (p<0.05) than that of group II. It was concluded that the infectionof E.tenella led to increase the number of neutrl mienteric nitrergic of the cecum. The higher lesion score ofcecum led to increase the number of mienteric nitrergic neuron.

Abstract

The objectives of this study to evaluate the effect of b-mercaptoethanol addition in maturation mediumon maturation and fertilization rate of sheep oocytes in vitro. Collected oocytes were then matured inmaturation medium without (control group) or with b-mercaptoethanol in concentration of 50 ?M or 100?M. There was a significant increase in the percentage of oocytes that reached the metaphase II (MII) stageafter cultured in maturation medium with b-mercaptoethanol either in concentration of 50 ?M or 100 ?M(p<0.05) (62.67%; 77.78%; 82.05% for control, 50 ?M, and 100 ?M group respectively). However no differencewas found in maturation rate of those oocytes in 50 ?M and 100 ?M groups (p>0.05). When maturedoocytes were fertilized, no difference was found on fertilization rate among the groups (P>0.05) (70.93%;77.50%; 69.14% for control, 50 ?M and 100 ?M group respectively). These data indicated that the additionof b-mercaptoethanol increased the percentage of sheep oocytes that reached the MII stage in vitro, but hadno effect on the fertilization rate.

Abstract

This research aim is to study the effect of E. longifolia Jack. roots methanol extract and its derivedfraction (n-hexane, chloroform, ethyl acetate, and methanol-water) on body weight of lactating mice. Thetreatment groups were administered 14 mg/20 g body weight of E. longifolia Jack. roots methanol extractand its derived fractions for 21 consecutive days. As a control are without treatment and placebo whichreceived aquadest at dose 0.056 ml/20 g body weight. The administration of E. longifolia Jack rootsmethanol extract showed the smallest change of body weight 29,63 g at the 3rd day and 29,31 g at the 21stday. Compared without treatment and placebo, oral administration of E. longifolia Jack. roots methanolextract and its derived fractions had no significant effects on body weight lactating mice at the 3rd and 21stdays in lactation period (p>0.05). It is concluded that the consumption of E. longifolia Jack. roots methanolextract and its derived fraction during lactation period not influence body weight of lactating mice.

Abstract

Bogor city is one of the greater Jabodetabek area which has a fairly high growth of the modern market.This should not shift the role of traditional market, if accompanied with an increase in the number andquality of traditional markets, among others by controlling infestations of flies on the market that can bea vector of various diseases.This research was conducted to identify the diversity and infestation of fliesspesies in five old Bogor traditional markets. The flies were collected using insect nets and then killed withchloroform to count and identification purposes. Measuring the prevalence of flies infestation in eachmarket were using sticky fly paper on block sale of meat, fish and outside market environment. Therewere ten fly spesies belong to four main families that Calliphoridae (C. megacephala, C. saffranea, C.rufifacies, and Lucilia sericata), Muscidae (M. domestica, M. conducens, and M. fasciata), Sarcophagidae (S.haemorroidalis, and S. fuscicauda), and Drosophilidae (Drosophila repleta). The others three families werePhoridae, Anthomyiidae, and Syrphidae. Fly diversity index on each markets were 1.203 (Bogor Market),1.038 (Sukasari Market), 2.678 (Anyar Market), 1.017 (Jambu Dua Market), and 1.618 (Gunung BatuMarket). Measurement of Calliphorid flies infestations as an indicator of the presence of litterdecomposition of organic material showed a high concentration in the market environment.These resultsillustrate the general environmental sanitation of traditional markets are bad.
