Abstract

The aim of this study was to determine the estrus cycle and the length of estrus in Tragulus javanicuson the basis of its vaginal cytology. Vaginal swabs were collected daily at 7 am for two months. Smears ofthe swab were then prepared on glass slide and they were stained with Giemsa. Vaginal epithelial cells;parabasal, intermediet and superficial cells were counted and their percentages during proestrus, estrusand diestrus were determined. Diestrus was characterized by the absent of superficial cells in the vaginalepithel. Proestrus was characterized by the progressive increase in percentage of intermediet/superficialcells in vaginal epithel, whereas estrus was characterized by the presence of superficial/cornification cellsin most vaginal epithel. On the basis of its vaginal cytology, it was determined that the estrous period offemale Tragulus javanicus was 3 days (48-60 h) and the length was 11 days (10-14 day). The change in thecytology of vaginal epithelial cells appeared to a good marker to determine the estrus cycle phase of theTragulus javanicus

Abstract

The effectiveness of Streptococcus agalactiae vaccine in tilapia (Oreochromis niloticus) was evaluatedfor prevention of streptococcal disease. The vaccine was prepared using formalin-killed whole cell andconcentrated extracellular products/ECP (62.3 and 55.8 kDa) of â-haemolityc isolate and 62.3; 55,8 and51.8 kDa protein of  non-haemolityc ECP of S. agalactiae.  Vaccination and challenged (103 colony-formingunits (CFU)/fish of â-haemolityc and 105 CFU/fish of non-haemolityc S. agalactiae) trial was conducted byintraperitonial (IP) injection into fish with average body weight of 15 g.  Fish were vaccinated with wholecell, ECP and mixed (whole cell and ECP) vaccine.  Tilapia vaccinated with whole cell of â-haemolitycisolate had a relative percent survival (RPS) rates higher than those of ECP â-haemolityc vaccine. However,fish  vaccinated with mixed (whole cell and ECP) of â-haemolityc has a better protection rates as comparedto those of two type of S. agalactie infection. Whereas those vaccinated with mixed (whole cell non-haemolitycand ECP of â-haemolityc) vaccine has protection rate of 79% from â-haemolityc and 42% from non-haemolitycinfection.  Tilapia vaccinated with whole cell of non-haemolityc was only able to protect fish from non-haemolityc infection and was unable to protect fish from other types.  Tilapia vaccinated with ECP non-haemolityc had a worse RPS than others vaccines in which mix whole cell and ECP vaccine of non-haemolitychad a protection 50-56% from  S. agalactiae infection. Whereas vaccinated with mixed (whole cell â-haemolityc and ECP of non-haemolityc) vaccine showed a better to protect from â-haemolityc than non-haemolityc infection.  It showed thatvaccination with mixed (whole-cell and extracellular product)  vaccineof S. agalactiae â-haemolityc  was more effective to protect tilapia against Streptococcosis.

Abstract

The aims of this study were to determine both chemical composition and fraction of the proteincompounds of sumba mare’s milk. Determination of the chemical compositions of sumba mare’s milk havedone by analyzing protein content using the Kjeldahl method, fat content using Gerber method, lactosecontent and the total solids content. Identification of antimicrobial compounds of whey proteins in milkusing high performance liquid chromatography (HPLC) method. The results showed that the average ofsumba mare’s milk contained protein, fat, lactose and total solids were; 1.82%, 1.67%, 6.48% and 11.37%respectively. The average value of protein and fat in sumba mare’s milk was decrease significantly at fifthmonth of lactation period. Based on identification of antimicrobial compounds using HPLC method, thereare six main peaks with different polarities and retention times. In conclusion, sumba mare’s milk havea balance composition that can be used as a source of nutritious food and the milk likely also has six mainantimicrobial compounds in its whey protein.

Abstract

The use of AGP (Antibiotic Growth Promoter) in animal has been banned because of the risk of antibiotic resistance. As an alternative for growth promoter. Andaliman (Zanthoxylum acanthopodium DC.) were used as feed additives. Andaliman is an herbal plant which proven has an immunomodulatory effect. However, herbal plants have low bioavailability in the body, so we need to increase its bioavaibility by reducing particle size using nanoparticle technology. The aim of our study was to determine the effects of three forms of andaliman as extracts, nanoparticles and nanoencapsulation on leukocyte differential, leukocyte count and Heterophil (H) / Lymphocyte.(L) ratio index. In this study we used 24 quails, which is divided into 4 groups, consist of.4 birds each. Group K was control group, birds were given dexamethasone (immunosuppressant), group P1 was given dexamethasone and andaliman extract, group P2 was given dexamethasone and andaliman nanoparticles, whilst group P3 was given dexamethasone and andaliman nanoencapsulation. The results showed that andaliman consumption had a significant effect (P <0.05) on differential leukocytes, number of leukocytes and H / L ratio after induction with immunosuppressants. Total leucocytes count based on the given treatment were K (8080 cell/mm3), P1 (20040 cell/mm3), P2 (20440 cell/mm3), and P3 (22040 cell/mm3). Each mean lymphocytes were K (44,6%), P1 (53,6%), P2(57,4%), and P3 (65,8%). Each mean heterophile were K (50,6%), P1 (41,8%), P2 (37,8%), and P3 (28,6%). Each mean monocytes were K (2,8%), P1 (4,6%), P2(4,8%), and P3 (5,0%). Each mean ratio index Heterophile (H) / Lymphocyte.(L) were K (1,1), P1 (0,7), P2(0,6), and P3 (0,5). It could be concluded that administration of Andaliman extract, Andaliman nanoparticles and Andaliman nanoencapsulation can increase the number of leukocytes, differential leukocytes and decrease the H / L ratio.

Abstract

Sumbawa Island is an anthrax endemic area. One of the efforts for anthrax control is byvaccination, particularly in cattle and buffalo. The purpose of this research was to conduct costbenefitanalysis of the two scenarios of anthrax control in Sumbawa Island, namely Program A,anthrax control program has been conducted at this time on Sumbawa Island with vaccinationcoverage < 50% compared with Program B, anthrax control that are intended to reduce the prevalenceby vaccination coverage > 80%, and to analyze the anthrax control strategies in Sumbawa Island.The results of cost-benefit analysis of anthrax control in cattle and buffalo were : both programsprovided positive net present value (NPV) and internal rate of return (IRR) which were greaterthan the discount rate, but the criteria for B/C ratio of program A was > 1 on the discount rate of15% and 20%, while the B/C ratio > 1 of program B was only obtained in buffaloes with a discountrate of 15%. By considering the three criterias of cost-benefit analysis, it can be stated that theanthrax control in Sumbawa Island with vaccination coverage < 50% is a proper and more efficientcontrol compared to anthrax control with coverage > 80% or without control.

Abstract

An experimental study was conducted in order to evaluate the   of erythrocyte on bali cattle atVeterinary Physiology Laboratory, Faculty of Veterinary Medicine, Udayana University, Denpasar, duringone month.  Fifty blood samples were taken from bali cattle which slaughtered at Pesanggaran abattoir,Denpasar. The erythrocyte fragility was determined by using method Red Cell Fragility Procedure (2001).Results showed that the fragility (initial haemolysis) on red blood cell of bali cattle was observed, atranged between 0.45% - 0.55% NaCl , while for total  haemolysis range between  0.30 - 0,35 % NaCl.

Abstract

This research aimed to isolate exopolysaccharides (EPS) producing Lactobacilli isolated fromsumbawa mare’s milk and its potential as prebiotics for modulating the growth of Bifidobacteriumbreve. Nine strains of Lactobacillus sp. were screened for their capabilities to produce EPS usingmodified MRS medium containing sucrose. Prebiotics potential of the EPS was verified by culturingB. breve JCM1273 in TOS medium containing EPS. The results showed that all strains ofLactobacillus sp. produced EPS on MRS sucrose medium and two strains (Lactobacillus SK3.1 andLactobacillus SK4) produced more EPS compared to the other strains tested. Bifidobacteriumbreve JCM1273 showed weak activity while in direct metabolism of EPS produced by Lactobacillussp. SK4 and its growth was enhanced on acid hydrolyzed EPS. Since this phenomenon mighthappened when the EPS exposed by the low pH during gastric passage, hence the EPS might be apotential source to be developed as prebiotics. Nevertheless, further investigation is necessary toevaluate the bifidogenic affects of EPS in Lactobacillus sp. SK4.

Abstract

Koi herpesvirus (KHV)  is the cause of  a lethal disease that affects common carp (Cyprinus carpio)  andkoi (Cyprinus carpio koi). Although, it has been reported that common carps could act as carriers for KHV,their histopathologic findings, especially the gills  have not been identified up to now. In the present study,12  normal, healthy looking common carps including their gills were collected from the endemic area ofKHV in  Sleman, Yogyakarta. All fish were necropsied and the gills were collected and fixed in 10% bufferformalin. Then, the gills were processed histopathologically using routine hematoxyline-eosin stain andexamined under the microscope. Histopathologic examination of the gills exhibited an apparent infiltrationof inflammatory cells, especially lymphocytes. The large oval to polygonal basophilic cells containing largeintranuclear inclusion bodies were also identified. Gills epithelial cells show mass hyperplasia and adhesionwith necrotic changes. Thus, results of this study has led to a reasonable conclusion that KHV infection ispresent in the normal, healthy common carps. One possibility is those KHV  are live viruses (carriers forKHV) and might could act as a source of infection is being discussed.

Abstract

The aim of the present study was to investigate the characteristics of frozen-thawed Garut ramsemen when cryopreserved in extender supplemented with Orvus Es Paste (OEP) as surfactant. Ramsemen were collected and evaluated, then cryopreserved in extender supplemented with OEP at differentconcentrations: OEP-0 (without OEP); OEP-0.25%; OEP-0.50%; OEP-0.75%; and OEP-1.00%. Thecharacteristics observed were motility, viability, and Intact Plasma Membrane (IPM). The results showedthat semen which were cryopreserved and supplemented with OEP had a higher motility, viability, andIPM, respectively compared to those without OEP. No significant differences in the characteristics wereobserved within the different OEP concentrations. It can be concluded that supplementation of OEP in theextender could protect the quality of cryopreserved ram spermatozoa.
