Abstract

Sperma adalah bahan biologis yang sering digunakan sebagai bukti untuk kasus pemerkosaan. Penelitian ekstraksi DNA dari sperma dilakukan untuk mengetahui apakah DNA dapat diekstraksi dari sperma pada kondom yang tersimpan selama 15, 20, 25, 30, dan 35 hari serta untuk mengetahui keberhasilan amplifikasinya. Sampel sperma dari seorang probandus diteteskan ke dalam kondom 2000 µL, kemudian disimpan selama 15, 20, 25, 30, dan 35 hari. Ekstraksi DNA dilakukan dengan menggunakan metode fenol-klorofom yang sudah dimodifikasi dan amplifikasi DNA dengan menggunakan PCR Mastermix. Hasil penelitian menunjukkan bahwa DNA masih dapat diekstraksi dan diamplifikasi dari sperma dalam kondom yang tersimpan hingga 35 hari.

Abstract

Artificial diet for insect rearing is necessary at the time a lot of insects required periodically and continuously such as for pesticide efficacy research, biodiversity conservation, and for the activity of fruit fly management with the sterile male technique for instance where it needs millions fruit flies weekly to be sterilized and then subsequently released. Insect artificial diet will also increase the efficiency of time, energy and fund in the procurement of insects compared to insect rearing with its natural diet. Various artificial diet classifications and its compositions have been developed so far in varying degrees of rearing success.

Abstract

Study of genetic variation on three microsatellite DNA loci D2S1338, D13S317 and D16S539 from 76 unrelated individuals of Batak ethnic who live in Denpasar city and Badung regency were analyzed by using PCR method (SuperMix Kit, Invitrogen). DNA samples were extracted from blood cell using phenol-chloroform method and ethanol precipitation. The result of this study found 14 alleles on D2S1338 locus, 10 alleles on D13S317 locus and eight alleles on D16S539 locus. The finding showed high genetic diversity on three loci as well as on three sub-ethnic Batak of Batak Toba, Karo and Simalungun which were 0.8637; 0.7314 and 0.7692 respectively.

Abstract

Aplikasi teknologi transfer gen melalui perantaraan sperma dimungkinkan untuk memperbaiki kualitas genetik ikan. Keberhasilan transfer gen hormon pertumbuhan (Growth Hormone =GH) ikan budidaya melalui elektroporasi sperma dapat menghasilkan ikan transgenik. Pada penelitian ini dilakukan transfer konstruksi vektor ekspresi gen GH lele dumbo (pTarget-CMV-CgGH) ke sperma lele dumbo (strain mutiara) menggunakan kejut listrik 125 Vcm-1 dengan jumlah pulsa 3 dan 5 serta 50 Vcm-1 dengan jumlah pulsa 3 dan 5. Konsentrasi vektor yang digunakan 60 ng/?L, lama kejutan 30 milidetik dan interval kejut 0,1 detik. Dari hasil elektroporasi sperma lele mutiara sebesar 125 Vcm-1 (jumlah pulsa 3 dan 5) serta 50 Vcm-1 (jumlah pulsa 3 dan 5) ini, terdeteksi fragmen berukuran 600 bp (gen GH lele dumbo) pada sperma dan fragmen 1000 bp (96 % identik dengan GH Clarias gariepinus, no. aksesi AF416488.1) pada ikan umur 4 bulan menunjukkan keberhasilan transfer gen GH lele dumbo (GH eksogen) ke sperma lele mutiara. Rata-rata kelipatan tumbuh ikan lele mutiara transgenik umur 4 bulan (125 Vcm-1 dan 50 Vcm-1 dengan jumlah pulsa 3 dan 5) berturut- turut 3,53 kali; 3,05 kali; 2,75 kali dan 2,76 kali dari ukuran ikan non transgenik (over-ekspresi GH eksogen). GH eksogen terdeteksi pada ikan lele mutiara transgenik dan tidak terdeteksi pada ikan lele mutiara non transgenik (non elektroporasi). Ikan lele mutiara transgenik berhasildirakit dengan menyisipkan gen GH lele dumbo menggunakan teknik elektroporasi sperma.

Abstract

The aim of the research was to increase the production of seed potato size 31 - 60 grams by applying gibberellic acid (GA3). Seeds that passed their dormancy period were dipped in gibberellic acid solutions (10, 15 and 20 mg/L) for 15 minutes a day before planting. Control seeds were dipped in water. Seeds were then air dried for 24 hours. Seeds were planted in plots with length, width and height of 150 x 100 x 30 cm respectively with planting space 20 cm and planting depth 15 cm. The design was randomized block with four replications. The results showed that gibberellic acid enhanced shoot emergence. Gibberellic acid increased stem number, tuber number and yield per plant. Tuber number and yield of size M (31 ? 60 g) increased, whereas tuber number and yield of size LL decreased drastically. The optimum concentration of gibberellin for these increases was 15 mg/L. Plant height was not influenced by gibberellic acid.

Abstract

This experiment was performed to examine the effects of Andrographis paniculata Nees leaves extract on foetus morphology if given during organogenesis period. Thirty pregnant mice were randomly divided into 5 groups for teratogenic effects. Extract was given orally by gavage with 0 (as control); 0,004 (equal to one time dose to human); 7,5; 15; and 22,5 g/g body weight/ day. Treatment was given from day 6 to day 13 of gestation (organogenesis period). Foetus were removed on day 18 of gestation by caesarean section. Quantitative data were analyzed with Anova and Duncan's Multiple Range Test. The results showed that teratogenic effects increased in dose-related manner by causing foetus death and resorption, hemorrhage and clubfoot.

Abstract

Penelitian ini bertujuan untuk mengetahui keanekaragaman moluska di  Pantai Serangan, Bali. Lokasi penelitian terdiri dari dua lokasi yaitu lokasi pertama berada di depan SMP N 11 Denpasar, sedangkan lokasi kedua berada di Pantai III. Pengambilan sampel menggunakan metode transek yang dimodifikasi. Penelitian dilaksanakan dari bulan Oktober–Desember 2013. Hasil penelitian menunjukkan jumlah spesies moluska yang dijumpai di Pantai Serangan adalah 88 spesies. Tiga spesies diantaranya dijumpai di kedua lokasi penelitian. Pada lokasi di depan SMP N 11 Denpasar, Perna sp. merupakan spesies yang memiliki sebaran terluas dan kepadatan tertinggi. Pada lokasi di Pantai III, Conus planorbis merupakan spesies moluska yang memiliki sebaran terluas dan kepadatan tertinggi. Keanekaragaman moluska di Pantai Serangan khususnya lokasi di depan SMP N 11 Denpasar dan di Pantai III tergolong baik (H>3) dan keseragamannya tergolong baik (E=0,6 dan 0,8).

Abstract

This study aimed to investigate the presence of insects on mice carcasses in mangrove forest, the chronology of the insect presence and growth phase of the insects on estimating the time of death of the animal for the beneficial of forensic study. Collected insects were identified at The Laboratory of Animal Taxonomy and The Laboratory of Ecology, Department of Biology, Udayana University. The samples were divided into two based on the study areas: dry and watered areas. Five mice carcasses in three different spot were laid on each dry area. The data collected were analyzed descriptively. The differences of insect growths from egg to pupae and also the ex-pupae found on carcasses could be used to estimated the time of death of an animal.

Abstract

Pandanus plant is one of the important plant in Papua and Papua New Guinea. Traditionally pandanus plant is used by local people for lots of daily activities and daily needs, start from food flavoring, medicine, to religious ceremony needs. Pandan kelapa hutan fruits (P. jiulianettii Martelli.) is one of a kind of pandanus that been used from generation to generation the highlands of Papua and Papua New Guinea for the food ingredients. This research aims to determine the physicochemical properties, total phenol, total carotene,vitamin E and antioxidant activities of the forest coconut pandan oil (P. jiulianettii Martelli.) extracted using an aluminum pan. Physicochemical character that been analyzed is specific gravity, iod value, saponification value, free fatty acid content, acid value and peroxide value. Total phenol, total carotene and vitamin E measured respectively using Folin-Ciocalteu method, spectrophotometry method and colorimetric methods. Antioxidant activities is measured using a method that called DPPH. The result shows the pandan coconut oil has 1,00059 of specific gravity, 65.87 g/100gr of iod, 201,28 mg/g of saponification, 4.26% of acid and 5.64 Meq/kg of peroxide. The content of total phenols, total carotene and vitamin E respectively are 48.55 ppm, 2.75 µg/g and 5.0303 mg/100g. The antioxidant IC50 measuring result shows the value is 45.83 mg/mL.

Abstract

Fish cultures in rice fields or areas that use water from a pesticide-polluted irrigation or river, are particularly vulnerable to the negative impact of pesticides. This study aims to assess the effect of organophosphates on growth, ovarian histopathology and fecundity of red tilapia (Oreochromis niloticus). After acclimatization, 160 fishes (fingerlink stage) were kept in four ponds sized 2x1,5x0,8 m each. This study used a complete randomized block design and organophosphate administration through the feed. The study used four groups: control, and variations in dose of 2 ml (D1), 6 ml (D2), and 10 ml (D3) of pesticide per 100 grams of pellet, which is given in the morning (treatment) and in the evening (regular feed). Data were taken at week 3, 6, and 9, then the organophosphate treatments were stopped, and the last data were taken at week-18. Statistical analysis was performed by Anova and Duncan's Multiple Range Test. The results showed that the organophosphate pesticide inhibits the growth by reducing fish length and weight, reducing ovarian weight, increasing the number of oocyte atresia, damaging structure of the ovarian histology, and decreasing fecundity by reducing the number of egg production in the ovarian of red tilapia.
