Abstract

Graphical Abstract  Highlight Research Compounds produced by Roseivirga PAP.19 bacteria have potential as antimicrobials. The OSMAC approach was used successfully to activate BGCs of Roseivirga PAP.19. The use of medium in bacterial culture affects the secondary metabolites produced by Roseivirga PAP.19 bacteria. Bacteria cultured in A1Bfe+C medium produced more secondary metabolites compared to the other medium. The different incubation times during culture affect the amount of secondary metabolite compounds produced.   Abstract Marine sponge-associated bacteria are a rich source of bioactive metabolites. The biochemical interaction between bacteria and sponges enables symbiotic bacteria to produce metabolites similar to those of their host. Metabolite extracts from symbiotic bacteria contain bioactive compounds with cytotoxic, antiviral, and antimicrobial properties. There is an urgent need for new antimicrobial agents due to the increasing resistance to existing drugs. This study aims to optimize the medium and incubation time of secondary metabolites produced by the symbiotic bacteria of the sponge Roseivirga sp. PAP.19. To stimulate biosynthesis of silent gene clusters, the One Strain Many Compounds (OSMAC) approach was used. Bacteria were cultured in various media and at different incubation times. This has the potential to increase the production of secondary metabolites. Secondary metabolites were analyzed using LC-HRMS. The potential of these metabolites as antimicrobial agents was subsequently evaluated. Results showed six compounds with antimicrobial potential. Notably, the A1BFe+C medium produced more bioactive compounds than the other medium, and the highest compound production occurred on day 4. These findings suggest that optimizing culture conditions using the OSMAC approach can enhance the yield of potentially valuable antimicrobial secondary metabolites from symbiotic bacteria, which may contribute to the discovery of new antimicrobial agents.

Abstract

Marine biofouling causes significant economic and environmental damage creating an urgent demand for eco-friendly antifoulants. This in silico study aimed to evaluate the antifouling efficacy and ecotoxicological profile of macrolactin A (1), previously isolated from Indonesian sponge-associated Bacillus spp., alongside its computationally generated derivatives (1a-1f, 2a–2k) using Biotransformer 3.0. Molecular docking was utilized to assess binding affinities against key protein targets—the bacterial BAM complex, GSK-3 , and acetylcholinesterase (AChE)—and used EPI Suite™ to predict environmental safety. All tested compounds exhibited robust binding affinities (  kcal/mol) against all targets likely binding to allosteric sites. ANOVA revealed significant differences ( ) in binding strength, with derivatives displaying a distinct preference for GSK-3  over AChE and BAM ( ). However, the broad-spectrum affinity across all three targets supports a potential multi-mechanism mode of action. Crucially, most derivatives showed low toxicity and bioaccumulation potential compared to commercial antifoulants such as Irgarol 1501, SeaNine 211, and Selektope®. Notably, analogues 1a-1c and 2e were predicted to be readily biodegradable. This study identified 1a–1c and 2e as leading candidates for eco-friendly antifoulants and provides a strong basis for future experimental development of novel, sustainable marine coating candidates. Keywords: ecofriendly antifouling; in silico; macrolactin; molecular docking; sponge-associated bacteria

Abstract

This study attempts to explore and uncover the meaning of determining the selling price of fish traders. This study uses phenomenology as a method with data collection through interviews and documentation. The study shows that fish traders tend to ignore the calculation of the Cost of Goods Sold (COGS). The values of gratitude and love expressed in the COGS are used as the basis for determining the selling price of fish. The low selling price of fish for informants is conceptualized as a "feeling" of mutual love for others. Several non-material aspects underlie the neglect of COGS, such as the attitude of fish traders and collectors who often express gratitude for the results of fish sales. The implied gratitude illustrates the neglect of the cost aspect in COGS in determining the selling price. This study reveals the phenomenon of determining COGS and fish selling prices for fish traders.

Abstract

This study aims to evaluate the impact of implementing virtual simulation in Biology learning on concept understanding by Agrotechnology students. Along with the development of technology, technology-based learning, especially virtual simulation, has begun to be applied to support the understanding of more complex concepts. Although a number of previous studies have shown the successful use of virtual simulations in various disciplines, there are still research gaps that need to be filled regarding the application of virtual simulations in the context of Biology education, especially in Agrotechnology study programs. This creates a phenomenon where students have difficulty in understanding these concepts. This research fills the gap by evaluating the application of virtual simulation as a learning method in overcoming this challenge. Using an experimental method, this study involved 30 students as samples, who were tested using pretest and posttest to measure the improvement of their understanding of Biology concepts. The results of the analysis showed that the application of virtual simulation resulted in a significant increase in students' understanding, which was reflected in the N-gain score with an average of 78.25. This value indicates a substantial increase in understanding of Biology concepts after using virtual simulation as a learning method. This finding supports the effectiveness of virtual simulation in improving the understanding of complex concepts, so it can be an effective alternative in learning in Biology. The results of this study provide positive implications for the use of technology in education, especially in improving the quality of learning in higher education
