Abstract

?-mangostin (?-MG) is a metabolite of xanthones isolated from fruit of Garcinia mangostana Linn pericarps. Skin cancer is one of the most common malignancies affecting many people. The development of skin cancer is mainly due to intermittent or long-term exposure to ultraviolet (UV) radiation resulting in reduced cell-mediated immune response, production of reactive oxygen species (ROS), and DNA changes. Increased levels of ROS can induce the production of matrix metalloproteinases (MMPs). ?-MG showing potential as an anti-metastatic agent with reduced the expression of MMP-1. This study aims to determine the affinity and stability profiles of ?-MG as anti-skin cancer by applying the In-Silico method. Molecular simulation of ?-MG was successfully docked to the MMP-1. ?-MG showed stable results after 100 ns molecular dynamics simulation based on the root mean square deviation (RMSD) and the root mean squared fluctuation (RMSF). The binding energies of the xanthone derivatives were calculated using the MM/PBSA method.

Abstract

Peningkatan aktivitas fagositosis sel makrofag sangat dibutuhkan oleh penderita penyakit tubekulosis (TB) untuk melawan invasi patogenitas dari bakteri Mycobacterium tuberculosis (Mtb) dan mengatasi keparahan infeksi sehingga pengobatan akan lebih optimal. Pengoptimalan fagositosis sel makrofag dengan pemberian imunostimulator dari tumbuhan seperti buah Etingera rubroloba A.D Poulsen penting diungkap. Buah diyakini sebagai imunostimulator berdasarkan penggunaan empiris oleh masyarakat Sulawesi Tenggara untuk memelihara daya tahan tubuhnya serta menyembuhkan penyakit infeksi demam typhoid, dan secara ilmiah terbukti sebagai imunomodulator, antiinflamasi dan antioksidan. Tujuan penelitian ini adalah membuktikan aktivitas buah E. rubroloba meningkatkan aktivitas fagositosis sel makrofag pada penyakit TB. Jenis penelitian ini eksperimental menggunakan 30 ekor mencit jantan yang terdiri dari kelompok normal (tanpa infeksi Mtb), kelompok negatif (Na.CMC 0.5% + infeksi Mtb), kelompok positif (ekstrak meniran komersial + infeksi Mtb), kelompok ekstrak dosis 200 mg/kg BB + infeksi Mtb, kelompok ekstrak dosis 300 mg/kg BB + infeksi Mtb, kelompok ekstrak dosis 400 mg/kg BB + infeksi Mtb. Hewan uji diberikan perlakuan selama 7 hari dan pada hari ke-8 diinfeksi antigen Mtb (Esat-6)  secara intra perintonium. Peningkatan fagositosis sel makrofag dihitung dengan mikroskop, selanjutnya data yang diperoleh dianalisis statistik one way ANOVA dan uji post hoc tukey. Hasil penelitian menunjukan ekstrak etanol buah E.rubroloba terbukti meningkatkan aktivitas fagositosis sel makrofag pada mencit distimulasi Mtb yang berbeda signifikan dengan kontrol negatif (P<0.05). Penelitian ini berhasil mengungkap informasi ilmiah baru dari tumbuhan E.rubroloba sebagai imunostimulator pada infeksi TB, sehingga berpotensi dikembangkan sebagai agen imunostimulator baru pada penyakit TB.

Abstract

ABSTRACT. Etlingera is a genus known for its potential medicinal properties. Etlingera rubroloba A.D. Poulsen is a recently identified species within the Etlingera genus found in Southeast Sulawesi. This study aims to isolate and identify secondary metabolite compounds from the methanol extract of the E.rubroloba A.D. Poulsen rhizome and its activity as an antimicrobial and antioxidant. The compound was isolated by using Thin Layer Chromatography (TLC), Vacuum Liquid Chromatography (VLC), and Radial Chromatography (RC) techniques. Isolated compounds were identified by 1H and 13C NMR spectroscopy techniques and compared the data with reference. Antimicrobial tests were carried out on Escherichia coli, Streptococcus mutans, and Candida albicans by broth microdilution method. The antioxidant test was carried out using the 1,1-dipheny1-2-picrylhydrazyl (DPPH) method. Four isolated compounds were successfully isolated : stigmasterol (1); 1,7-diphenyl-6-hepten-3-on; (2); and 1,7-diphenyl-heptan-3-on (3); and sinapyl alcohol diacetate (4). Antimicrobial activity test results extract were active against all bacteria and C. albicans. The results of antioxidant activity test showed that extract, compound 2, 3, and 4 had strong and medium antioxidant activity with IC50 values at 75.05 μg/mL; 201.4 μg/mL; 207.88 μg/mL, and 190.59 μg/mL, respectively.
