Abstract

Abstract

The problem that often arises in tilapia cultivation is the low growth of fish caused by the provision of feed that contains high energy but cannot be digested by fish. Utilization of commercial feed as feed for tilapia growth is less effective because the feed is still complex so it requires energy to be broken down into more simple. The method used in this study was a completely randomized design (CRD) which was then analyzed using one way ANOVA variance. The best results were obtained in treatment 4 with an absolute weight value of 2.97, absolute length of 1.19, specific growth rate of 2.75, survival of 83.33% and feed conversion of 0.95%. The conclusion of this study is the treatment of 4 absolute weight of 2.97 g and absolute length of 1.19 cm.

Abstract

The mechanism of energy allocation related to increased appetite and feed intake in fish involves several neuropeptides encoded by specific genes. Orexin (OX) is a peptide secreted in the pars tuberalis of the hypophysis that acts as a stimulator in appetite increase (orexigenic factor). However, the expression of different orexigenic factors varies among vertebrate species, reflecting their unique types and lineages. Therefore, this study aimed to identify genes encoding appetite in Tilapia. RNA isolation, complementary Deoxyribose Nucleic Acid (cDNA) cloning, and DNA amplification were performed from brain samples of gonadally mature Tilapia. The PCR products were subsequently sent to Macrogen.Inc for sequencing. The amplification results of Orexin with β-Actin (positive control) observed using agarose gel electrophoresis showed that the size of the nucleotide base product of each gene was 196 bp and 197 bp. Confirmation of sequencing results carried out using the Basic Local Alignment Search Tool (BLAST) method - National Center for Biotechnology Information (NCBI) for Orexin and β-Actin were 97% and 100%. Based on these results, it can be concluded that each target gene isolated from the tilapia brain showed homology/similarity with the sequence available in the NCBI database

Abstract

The production and demand of tilapia (O. niloticus) in some countries continue to increase but are not matched by good growth quality. Several methods have been used to increase growth, such as the use of synthetic hormones and radiation, however, the methods require such a high cost. Thus it needs to be investigated the potential replacement with natural prooduct. Papaya leaf contains papain enzyme thought to be able to improve the growth performance of fish body weight through the conversion of proteins into amino acids. The purpose of this study was to investigate the growth performance of tilapia (O. niloticus) fish that were given papaya meal (C. papaya) treatments. The concentrations pellet with papaya meal respectively T1(feed with 0 grams of papaya leaf meal), T2 (administration of papaya leaf meal with 1.25 g/kg feed), T3 (administration of papaya leaf meal with 1.75 g/kg feed), T4 (administration of papaya leaf meal with 2 g/kg feed), T5 (administration of papaya leaf meal with 2.25 g/kg feed). Parameters analyzed included: absolute length growth, absolute weight, specific growth rate, FCR survival rate and, water quality. The results showed that the highest weight growth of tilapia fed with the administration of papaya leaf meal was found at T4 of 21.23 grams. In the specific weight, the optimal treatment was found in T4 with a percentage of 20.97%. In the length growth of tilapia, it was known that the T1, T4 and T5 had highest lengths when compared to other treatments and the highest survival rate of tilapia (O. niloticus) was in the T2, T3, T5 treatments of 73%. The optimal FCR value was found in the T4 treatment of 1.14. Based on the results of the study, it can be concluded that the administration of papaya leaf flour can increase the growth performance of tilapia.Al-Nemrawi, N. K., Alsharif, S. S. M. & Dave, R. H.
