Abstract

The problem that often arises in tilapia cultivation is the low growth of fish caused by the provision of feed that contains high energy but cannot be digested by fish. Utilization of commercial feed as feed for tilapia growth is less effective because the feed is still complex so it requires energy to be broken down into more simple. The method used in this study was a completely randomized design (CRD) which was then analyzed using one way ANOVA variance. The best results were obtained in treatment 4 with an absolute weight value of 2.97, absolute length of 1.19, specific growth rate of 2.75, survival of 83.33% and feed conversion of 0.95%. The conclusion of this study is the treatment of 4 absolute weight of 2.97 g and absolute length of 1.19 cm.

Abstract

The mechanism of energy allocation related to increased appetite and feed intake in fish involves several neuropeptides encoded by specific genes. Orexin (OX) is a peptide secreted in the pars tuberalis of the hypophysis that acts as a stimulator in appetite increase (orexigenic factor). However, the expression of different orexigenic factors varies among vertebrate species, reflecting their unique types and lineages. Therefore, this study aimed to identify genes encoding appetite in Tilapia. RNA isolation, complementary Deoxyribose Nucleic Acid (cDNA) cloning, and DNA amplification were performed from brain samples of gonadally mature Tilapia. The PCR products were subsequently sent to Macrogen.Inc for sequencing. The amplification results of Orexin with β-Actin (positive control) observed using agarose gel electrophoresis showed that the size of the nucleotide base product of each gene was 196 bp and 197 bp. Confirmation of sequencing results carried out using the Basic Local Alignment Search Tool (BLAST) method - National Center for Biotechnology Information (NCBI) for Orexin and β-Actin were 97% and 100%. Based on these results, it can be concluded that each target gene isolated from the tilapia brain showed homology/similarity with the sequence available in the NCBI database

Abstract

Tiger shrimp (Penaeus monodon) is one of the featured products in Kalimantan Utara that still apply organic or traditional pond methods. Currently, production tiger shrimp has decreased due to a disease caused by Vibrio harveyi bacteria, which causes luminescent Vibrio disease or Vibriosis. Antibiotics are commonly used to treat this disease, but prolonged administration can lead to bacterial resistance The use of natural ingredients is an alternative solution to replace antibiotics, one of which is seaweed (Kappaphycus alvarezii). This study aims to determine the in-vitro inhibitory effect of seaweed extract against the growth of V. harveyi. This study using quantitative descriptive wih Kappaphycus alvarezii seeds from traditional farmers in Tarakan, Indonesia. Methods that were used in this research consisted of K. alvarezii extract preparation, bacteria preparation, phytochemical assay, and antibacterial assay of the extract followed by data analysis. The phytochemical assay consisted of alkaloid, phenol, flavonoid, saponin, and steroid assay. The antibacterial activity was evaluated using the paper disc diffusion method with seaweed extract concentrations of 5%, 10%, 15%, and 20%, while tetracycline served as the positive control and a solvent-only disc as the negative control. The results of phytochemical assay showed that the K. alvarezii extract contained alkaloid, phenol, flavonoid, saponin, and steroid. The antibacterial test showed that the seaweed extract with a concentration of 5% had an inhibition zone of 7 mm, 10% of 7.1 mm, 15% of 7.3 mm and at a concentration of 20% had the largest zone of inhibition with an inhibiton diameter of 13.3 mm. Based on these results, it can be concluded that the provision of Kappaphycus alvarezii is potentially inhibit the growth of V. harveyi in vitro but not significantly different when compared to the positive control. This activity is likely attributed to the natural compounds present in K. alvarezii, such as alkaloids, phenols, flavonoid, saponin, and steroids, which are known to possess antibacterial properties.
