Pectinase is an enzyme that could hydrolyze pectin into galacturonic acid. Natural pectinase was produced by microbes such as bacteria, yeast, fungi and Actinomycetes. Application of pectinase in industry were mainly in juice industry, textile, pulp, tea, cocoa and coffee fermentation. In this research, we conducted purification and characterization of pectinase produced by Aspergillus ustus BL5 in submerged fermentation using commercial pectin. The result showed that the optimum of pectinase production was reached at 120 hours fermentation process with specific activity 0.59 U/mg. The crude extract of pectinase was then concentrated using PEG 6000 and purified by Sephadex G-75 gel filtration chromatography. There were 2 fractions contained pectinase which the activity was 4.15 U/mg (pectinase A) and 3.3 U/mg (pectinase B), respectively. Compare to crude extract, the yield product of pectinase A and B increased 6.94 and 5.53 times, respectively. The purified pectinase A have optimum temperature at 50 oC and optimun pH at 5.

Adanya variasi bahasa yang terdapat di Kecamatan Arosbaya dan Geger serta daerah tersebut memungkinkan untuk pengambilan data dikarenakan daerah tersebut termasuk daerah pesisir dan pegunungan menjadikan faktor adanya suatu dialek. Rumusan masalah yang muncul ialah bagaimana pemakaian Bahasa Madura dialek Bangkalan di Kecamatan Arosbaya dan Kecamatan Geger Kabupaten Bangkalan? Dengan tujuan menganalisis pemakaian Bahasa Madura dialek Bangkalan di Kecamatan Arosbaya dan Kecamatan Geger Kabupaten Bangkalan. Penelitian ini termasuk penelitian kualitatif deskriptif dengan data mentah berupa ujaran percakapan yang kemudian di transkripsikan ke bentuk tulisan yang berupa kalimat dan kata. Data tersebut diperoleh dari sumber penduduk asli daerah penelitian sesuai dengan criteria sumber data. informan dari penelitian ini peneliti itu sendiri yang di bantu oleh alat perekam suara berupa recorder/handphone. Metode pengumpulan data dalam penelitian ini yaitu metode simak dengan teknik dasarnya teknik sadap dan teknik lanjutannya menggunakan teknik simak bebas libat cakap, teknik catat, teknik rekam, teknik transkripsi data, dan teknik translate data. Selanjutnya, peneliti menganalisis data yang telah dikumpulkan dengan metode analisis data yakni metode padan. Metode ini memiliki teknik dasar sebagai teknik pilah unsur penentu (PUP), sedangkan teknik lanjutannya berupa teknik hubung banding membedakan (HBB). Peneliti juga menggunakan teknik keabsahan data berupa teknik pemeriksaan keabsahan data, yaitu 1) perpanjangan keikutsertaan, ketekunan, pengamatan; 2) triangulasi; 3) pengecekan sejawat, 4) kecukupan referensial; 5) kajian khusus negative; dan 6) pengecekan anggota.Kata kunci:Â Dialek Bangkalan, Dialektologi, dan Bahasa Madura

This research was conducted to increase children's sense of responsibility with cooking class activities for Group A children at the Dharma Wanita Persatuan Sedenganmijen Krian Kindergarten. Aspects of responsibility examined include: 1) children take care of public property, 2) children tidy up equipment/toys after use, 3) children carry out tasks that have been given by the teacher. This research method is included in classroom action research. This research was conducted in three cycles. The subjects of this study were Group A children with 15 children consisting of 5 boys and 10 girls. How to collect data by observation, interviews, and documentation. In the results of this study, it can be seen that there is an increase in children's sense of responsibility in the 3 cycles that have been carried out. Children's sense of responsibility has increased with cooking class activities because there are rules that must be followed and work to completion. So that it makes children more responsible which includes maintaining public property, tidying up equipment/toys after use and carrying out tasks that have been given by the teacher.

Indonesian hold presidential election in 2024. One of the most discussed topics by public is the presidential candidates. The discussion about the presidential candidate certainly reaped various kinds of responses from public, ranging from support to statements of disapproval. This research was limited to the candidacy of Anies Baswedan as a presidential candidate before a vice president candidate as his pair was selected. The purpose of this study is to conduct a sentiment analysis of public responses regarding Indonesia 2024 presidential candidate Anies Baswedan using tweets data from October 2022 to January 2023 using the naïve bayes classifier algorithm. This is expected to provide an overview of the public opinions on Twitter. Three test models were carried out with differences in the division of the amount of training data and test data, respectively 60%:40%, 70%:30% and 80%:20%. The test results showed the highest accuracy level was obtained by the 3rd model using training and testing data of 80%:20% with an accuracy value of 76.21%. Further research is recommended to conduct sentiment analysis on the pairs of Presidential and Vice-Presidential candidates who have been officially registered with the General Election Commission using various other classification algorithms.

ABSTRACT This research aims to ensure the safety of livestock-origin food products through the evaluation of microbial contamination potential in production-support facilities. The study was conducted by isolating and characterizing bacteria from poultry slaughter facilities in several traditional markets in Kendari City. Swab samples were taken from chicken holding pens in five traditional markets. The market locations were determined using purposive sampling, with the daily slaughter quantity as the main indicator. Isolation was performed by observing bacterial colony growth, and characterization was carried out by observing the morphology of bacterial colonies on specific differential media using Eosin Methylene Blue Agar (EMBA). Bacterial confirmation was done through microscopic observation based on the shape and morphology of bacteria in Gram staining. The results of this study indicate that bacteria can be isolated from all poultry slaughterhouses in traditional markets in Kendari City. Characterization results show that the bacteria were confirmed as Escherichia coli, Salmonella sp., and Pseudomonas sp. The low implementation of Good Hygiene Practices in production execution is a major supporting factor for the presence of bacteria as a source of contamination. Periodic supervision by relevant authorities is necessary as an effort to ensure the safety of livestock-origin food products.

The secondary metabolite compounds contained in clove leaves oil have the potential to be antibacterial Escherichia coli. The kinetic analysis is based on the inhibition zone formed as a result of the inhibition of clove leaves oil and the results of microencapsulation on E. coli bacteria. The determination of the inhibition zone uses the disk diffusion method by using amoxicillin as a positive control and tween oil as a negative control. The kinetic review is intended to find out the antibacterial inhibitory reaction process by determining the reaction order (n), rate constant (k) as well as the At-Ao-t relationship of clove leaves oil and the results of its microencapsulation. Based on the research results obtained by the reaction order (n) of clove oil as an antibacterial E. coli is n = 0.1913; with the rate constant k = 5.7679. The reaction order (n) results of microencapsulation as antibacterial E. coli are 0.5039; with the rate constant k = 4.7323.

This service program aims to increase the knowledge and skills of partner breeders regarding feed formulation techniques using local raw materials and superior local chicken breeding techniques using automatic hatching machines as well as seed production systems intensively and semi-intensively raised. The implementation of this service program uses group and individual counseling methods, which include non-physical and physical activities. Non-physical activities take the form of theoretical counseling using lecture and discussion methods. In contrast, physical activities are carried out using demonstration methods and field visits related to feed formulation techniques using local raw materials and superior local chicken rearing techniques using automatic hatching machines and intensive and semi-intensive seed production systems. The results of the implementation of this service program showed that the technical guidance activities for raising superior local chickens, which were carried out in Alebo Village, Konda District, South Konawe Regency, received a positive and enthusiastic response from the local village government and partner breeders. This technical guidance activity for raising superior local chickens that combined non-physical activities in the form of theoretical counseling with lecture and discussion methods with physical activities in the form of demonstrations and field visits can be a solution to problems experienced by partner breeders. Implementation of this service program is effective in increasing the knowledge and skills of partner breeders regarding feed formulation techniques using local raw materials and superior local chicken raising and breeding techniques using automatic hatching machines as well as seed production systems intensively and semi-intensively raised.

The goal of this research is to define the stages involved in applying the model of ProjectBased Learning (PjBL) in order to improve student learning results, particularly in scientific topics.The goal of this research is to define the stages involved in applying the model of Project BasedLearning (PjBL) in order to improve student learning results, particularly in scientific topics. Datacollection techniques used test and non-test techniques. The data analysis methods employed includedescriptive, qualitative, and quantitative. The findings revealed that the adoption model of PjBL withthe 4C, HOTS, and TPACK techniques improved learning outcomes in grade 4 pupils at SDN 4Kenanga. This is demonstrated by the rise in average student achievement observed in cycle 2. Incycle 1, the average individual value of 60.6 with 20 participants (67 percent) in the incompletesection of KKM calculated is 70, indicating that student achievement have not improved. While incycle 2, there is an improvement in student achievement from an average value of 78 to as many as23 or (76 percent) pupils finishing.

Marketers and retailers employ strategies to attract consumer attention and influence their purchasing decisions. Two increasingly popular strategies are flash sales and live streaming which utilize technology advancements to create a more engaging and dynamic shopping experience. The research aims to determine the influence of live streaming and flash sales on impulse buying among Shopee app users in Bengkulu city. The sample in this study consists of 100 respondents who use the Shopee app in Bengkulu. Data collection method involves the use of questionnaires. The analysis method employs the SmartPLS analysis tool version 4.0. The research results indicate that live streaming has a significant and positive influence on impulse buying among Shopee app users in Bengkulu, as the t-statistic value is 4.082 > 1.96 and the p-value is 0.000 < 0.05. The original sample value shows a positive value of 0.333, indicating that the relationship between the live streaming variable and impulse buying is positive. This means that the better the live streaming, the higher the impulse buying. Flash sales also have a significant and positive influence on impulse buying among Shopee app users in Bengkulu, with a t-statistic value of 7.045 > 1.96 and a p-value of 0.000 < 0.05. The original sample value shows a positive value of 0.517, indicating that the relationship between the flash sale variable and impulse buying is positive. This implies that the higher the flash sale provided by the seller, the higher the impulse buying.
