Abstract

Red Palm Oil (RPO) is the result of refining from Crude Palm Oil (CPO) which still maintains a high level of phytonutrients that are beneficial for health. The benefits of RPO can be used as a nutraceutical source, because RPO acts as a provitamin A and vitamin E. The addition of Lactic Acid Bacteria (LAB) in a product functions as a probiotic with metabolite activity that can inhibit pathogenic microorganism thereby increasing the durability of a product. The addition of LAB to RPO is expected to increase the nutraceutical value by adding probiotic properties to RPO products. Microencapsulation technology can protect the bioactive content of essential oils against unwanted chemical interactions with other components and provide increased stability during processing. This research is expected to be able to provide information about the potential raw material of fermented RPO as a nutraceutical source by showing the profile of RPO before and after the fermentation process, chemical and physical properties of the microencapsulant. The microencapsulation technique used is the extrusion method. Fermentation of the bioactive compound ingredients in RPO using L. casei has a significant effect. RPO after fermentation using L. casei (Red-PalmZym) produces major compounds in the form of 9-Octadecenoic acid, methyl ester, (E)- Fermentation of RPO with L. casei increases the components of chemical compounds contained therein. The stability of the RPO microencapsulated before fermentation provided an effective protection against oxidative damage to the oil and proved that the microencapsulation process can reduce the level of damage to the oil.

Abstract

Pilis, a traditional product containing various herbal ingredients, is currently less desir-able due to its impractical and unfashionable method of application. Pilis is used to treat dizziness, pain, eye fatigue, encephalitis in children, fever, and to aid recovery for mothers after childbirth. However, pilis also carries risks related to uncontrolled dosage and contamination, particularly from chemical or heavy metal pollutants, which may cause toxic reactions. To mitigate these risks, pilis should be used in recommended doses and produced under clean and safe conditions. This research aimed to provide the innovation of patches as an alternative treatment to increase attractiveness and control the active substances released and delivered for a long time. Transdermal patches are made from a combination of turmeric and black seed containing flavonoid active metabolite compounds, namely curcumin, carotenoids, quercetin, kaempferol, and apigenin. The combination has a synergistic effect that can increase total flavonoid levels and the therapeutic effect of pilis. The method used in this research was litera-ture review and data analysis. The results showed the effect of transdermal patch pilis from the combined extracts added to the formula in vitro and in vivo using animal tests. In conclusion, transdermal patch pilis has prospects as a topical treatment based on the combination of an active delivery system with a patch and metered dose system.

Abstract

Urinary tract infection (UTI) is one of the most common types of infection caused by the growth of microorganisms in the human urinary tract. Nitrite can be detected using spectroscopy, electrochemistry, chemiluminescence, chromatography, capillary electrophoresis, and flow injection analysis methods. However, this method takes quite a long time. So, this method is not suitable for rapid screening because it requires time for bacterial incubation and adequate sample preparation. So, there is a need for a fast-screening method that can diagnose UTIs through a nitrite colorimetric sensor. The colorimetric method is based on nanoparticles, which are relatively cheap and can provide visual and rapid detection. Nitrite is an indicator in diagnosing UTI. This research aims to determine the synthesis of Ag nanoparticles using avocado leaf bio-reductors (Persea americana Mill.) and chitosan as a stabilizer, which can be used as a colorimetric nitrite detector to minimize the use of dangerous and environmentally unfriendly inorganic materials. Characterization and analysis in this research used a UV-Vis spectrophotometer, FT-IR (Fourier Transform Infra-Red), and PSA (Particle Size Analyzer). The Ag nanoparticle synthesis process was carried out by varying the bioreductant volume, synthesis time, and chitosan concentration. It was found that the optimum synthesis time was 4 hours with an SPR value of 428 nm, an absorbance of 2.112 and a nanoparticle size. Apart from that, the optimum concentration of chitosan was obtained at a concentration of 2.5% with an SPR of 435 nm and an absorbance of 1.341 with a nanoparticle size. The results obtained from AgNPs/chitosan as a colorimetric nitrite detector showed a color change to purple with an LOD value of 0.1303 µM and an LOQ of 0.4345 µM. Abstract

One of the effective ways to stay healthy is maintaining cleanliness, including hand hygiene. Various diseases can be easily transmitted through the hands. However, modern developments have changed the habit of handwashing with water to using hand antiseptics. Hand sanitizer is an alternative to overcome this problem. Unfortunately, some hand sanitizer gels on the market still contain alcohol as an antibacterial ingredient. It is necessary to innovate in making hand sanitizers using non-alcoholic ingredients as antibacterials, for example, natural ingredients like garlic. Garlic contains allicin that functions as an antibacterial. Meanwhile, VCO contains high lauric acid with antibiotic properties that function as antiviral, antibacterial, and antiprotozoal. It also contains capric acid, with antiviral and antibacterial functions. This study aims to examine the effect of concentration variation of VCO on the physical stability of garlic extract emulgel preparation and antibacterial effectiveness against the growth of Staphylococcus aureus. The concentration variation of VCO in this study was 3%, 5%, and 7% for 3 types of formulas. The results showed that the concentration variation of VCO in emulgel preparations gave differences in the physical properties of the gel, including pH, viscosity, adhesive strength, and spreadability. It can be concluded that the higher the VCO concentration, the higher the pH, viscosity, and adhesive power, and the lower the spreadability and antibacterial inhibition power against Staphylococcus aureus. The best result from the evaluation of physicochemical properties and antibacterial tests was Formula 1, although it was still in the weak inhibitory power category.

Abstract

Parijoto (Medinilla speciosa Reinw. ex Bl.) is a traditional medicinal plant rich in polyphenolic compounds that are associated with antioxidant and cytoprotective activities. The present study aims to investigate the phytochemical profile, antioxidant capacity, cytotoxicity, and anti-senescence effects of parijoto methanolic extract. The methanolic extract of parijoto (MEP) was subjected to qualitative thin-layer chromatography (TLC) for the detection of flavonoids, quantitative high-performance liquid chromatography (HPLC) for the determination of quercetin and naringin, and antioxidant activity assessment using the DPPH method. The mobile phase for quercetin analysis was water: acetonitrile (45:55), while for naringin, it was acetonitrile:water: formic acid (21:78.8:0.2). Cytotoxicity was evaluated in Vero cells using the MTT assay, while senescence in normal kidney cells was assessed by senescence-associated β-galactosidase (SA-β-gal) staining. Results showed that the extract contained quercetin (5.76±0.11 ppm). The extract exhibited strong antioxidant activity with an IC₅₀ value of 40.46 ppm. Cytotoxicity testing of MEP on Vero cells revealed low toxicity (529±0.77 μM) at physiologically relevant concentrations, supporting its safety for normal kidney cells. Moreover, in normal kidney cells, the extract conferred cytoprotective effects by reducing oxidative stress–induced damage and attenuating senescence-associated β-galactosidase. Collectively, these findings suggest that MEP exhibits potent antioxidant, cytoprotective, and anti-senescence activity in vitro, warranting further in vivo studies to validate its potential effects on kidney health and age-related cellular damage.
