Abstract

Acne is a skin condition characterized by the formation of skin bumps due to bacterial activity. Secondary metabolite compounds found in Gaharu leaves, such as flavonoids known for their antibacterial properties. This research aims to assess the physical and chemical stability of an anti-acne patch containing ethanol extract from Gaharu leaves and chitosan polymer. Additionally, this research aims to evaluate the antibacterial effectiveness of the anti-acne patch against Propionibacterium acnes. The experimental approach involves stability testing through cycling tests, observation of physical and chemical properties before and after stability tests, and antibacterial activity testing against P. acnes. The results from the stability test indicate overall stability in organoleptic aspects, thickness, folding ability, weight consistency, moisture content, vapor transmission, and moisture absorption of the anti-acne patch. However, instability is noted in the pH of the preparation. The antibacterial activity test reveals that the entire formulation exhibits antibacterial properties against P. acnes. Furthermore, there is a positive correlation between the increased concentration of ethanol extract from Gaharu leaves or chitosan polymer and the heightened barrier zone value.

Abstract

Gel is a semisolid dosage form that utilises a gelling agent, such as Carbopol 940, providing ease of application and efficient skin absorption. Ethanol extract of awar-awar (Ficus septica Burm. F) leaves, which contain flavonoids and tannins, has demonstrated promising anti-inflammatory potential. This study aimed to formulate and evaluate the stability of a gel containing the ethanol extract of awar-awar leaves, as well as assess its anti-inflammatory effects. The methods comprised phytochemical screening, gel formulation and evaluation, stability testing, and an in vitro anti-inflammatory assay. Four gel formulations were created: F0 (0%), F1 (1.5%), F2 (3%), and F3 (4.5%). Evaluation results showed that F0, F1, and F3 were homogeneous, with viscosities between 5,000-50,000 mPa·s, spreadability of 5-7 cm, and adhesion time exceeding 2 seconds, meeting Indonesian National Standard (SNI) requirements. However, pH testing during the cycling tests revealed instability in formulations F1 through F3 during storage. The anti-inflammatory test, conducted using the protein denaturation inhibition method, showed all three formulations had anti-inflammatory activity, with F3 being the most effective (92.20% inhibition). Therefore, F3 is considered the most promising formulation due to its high anti-inflammatory activity and acceptable pharmaceutical characteristics for gel preparations.

Abstract

Burns are skin tissue damage caused by hot objects or hot liquids. One of the potentially healing plants for burns is the bay leaves (Syzygium polyanthum (Wight.) Walp.). Bay leaves ethanol extract contains flavonoids known for their anti-inflammatory properties, which can aid in wound healing. This study aimed to determine the formulation and effectiveness of a gel made from ethanol bay leaves extract. Bay leaves were extracted using the maceration method with a 70% ethanol solvent; then, its extract was formulated into gel preparations. Following the formulation, the researchers conducted physicochemical evaluations and burn effectiveness trials using New Zealand albino rabbits. The research results indicated that the burn gel formulations with 3% and 6% concentration variations exhibited favourable physical properties, including organoleptic, homogeneity, and pH from 5.33 to 5.8, spreadability from 4.77 to 5.55 cm, adhesion strength from 5.47 to 11.6 seconds, and viscosity ranged from 12.507 to 16.080 cPs. The 15-day burn effectiveness test revealed no significant differences, indicating that the reduction in wound diameter was relatively consistent across all test groups. The percentages of wound healing in the negative control, bioplacentone, F1, and F2 were 42.48%, 54.73%, 43.84%, and 65.89%, respectively. Therefore, F2 demonstrated the best wound-healing performance.

Abstract

NAT2 serves as the key enzyme responsible for metabolizing the INH compound, with its expression and functional activity significantly contributing to the risk of hepatotoxicity. Due to the possible inhibitory role of curcumin on NAT2, it is important to assess its effect on the metabolic processing of INH and to examine the enzyme-related interactions that may occur between drugs. Molecular docking studies demonstrated that curcumin can localize in the hydrophobic pocket and form a strong bond with NAT2. The aim of this study was to predict the potential interaction of the isoniazid and curcumin compounds against NAT2 protein. In this study, NAT2 protein (PDB ID: 2PFR) was used as a receptor. The results obtained showed the binding energies of native ligand, isoniazid, and curcumin were -5.78, -4.47, -8.35 kcal/mol, respectively. The findings of this research suggest that curcumin is capable of suppressing NAT2 activity, thereby affecting the pharmacokinetics of INH. These results may offer insights into minimizing INH-related liver toxicity and enhancing its effectiveness through co-administration with curcumin.
