Abstract

The coastal areas of Aceh Besar have high fisheries potential, but traditional fishermen using bagan boats in Gampong Lhok Layeun are facing declining catches due to technological limitations. The conventional lights used have limited light coverage and high fuel consumption. This service activity aims to improve the knowledge and skills of fishermen through education on energy-efficient underwater Light Emitting Diode (LED) technology as an alternative solution. The implementation method includes four stages: initial survey, socialisation, discussion, and evaluation and monitoring. The activity involved 25 participants consisting of fishermen, Panglima Laot, academics, and village officials. The results showed an increase in participants' understanding of underwater LED technology, with an average pre-test score of 57 increasing to 84 in the post-test. A total of 100% of participants expressed interest in adopting this technology, and showed high enthusiasm during the socialisation and discussion. This activity proves that participatory and educative approaches can strengthen traditional fisheries innovation at the local level.

Abstract

CORRESPONDENCE ANALYSIS OF THE RELATIONSHIP PATTERN BETWEEN DISASTER TYPES AND VICTIM STATUS IN SUMATERA IN 2024 This study analyzes the relationship between provinces, types of disasters, and types of disaster victims in 10 provinces on Sumatra Island using correspondence analysis. The data includes seven disaster types and five victim categories from Aceh, North Sumatra, West Sumatra, Riau, Riau Islands, Jambi, Bengkulu, South Sumatra, Bangka Belitung Islands, and Lampung. Descriptive results show floods as the most frequent disaster, while the "suffering victims" category appears most often. The chi-square test yields a p-value < 2.2e-16, indicating significant relationships between all variable combinations. The correspondence plot reveals distinct patterns: Lampung often faces extreme weather and earthquakes; West Sumatra and Riau are prone to landslides; Riau Islands and Aceh to forest and land fires; and Bengkulu to drought. Victim distribution also varies, West Sumatra reports more displaced victims, while most other provinces have predominantly suffered victims. In terms of disaster impacts, landslides are linked to fatalities, forest and land fires to displacement, and floods to suffering and missing victims. These findings highlight the interconnectedness of regions, disaster types, and victim impacts, offering important insights for disaster risk management and mitigation efforts across Sumatra.

Abstract

Biotechnology frequently employs Escherichia coli to produce recombinant proteins. However, its efficiency is strongly influenced by induction conditions, particularly the quantity of isopropyl ?-D-1-thiogalactopyranoside (IPTG). This study aimed to evaluate the effect of different IPTG concentrations on the expression of recombinant Jembrana Superficial Unit (JSU) protein in E. coli BL21. The JSU gene cloned into the pET21a vector was induced at 37°C for 1 hour using three IPTG concentrations (0.3, 0.6, and 0.9 mM), each performed in triplicate. SDS-PAGE analysis confirmed successful expression of the recombinant JSU protein, indicated by a distinct band at approximately 37 kDa, consistent with its predicted molecular weight. Bacterial cells were lysed by freeze–thaw, and protein expression levels were quantified by spectrophotometry from optical density measurements and expressed in arbitrary units. Statistical analysis using one-way ANOVA, following confirmation of data normality and homogeneity of variance, revealed mean expression levels of 1.69 ± 0.15, 1.59 ± 0.11, and 1.76 ± 0.28 a.u. for 0.3, 0.6, and 0.9 mM IPTG, respectively, indicating a non-linear trend with a modest increase at the highest concentration. Nevertheless, there were no statistically significant differences between the treatments (p>0.05). These findings suggest that, within the tested range, variation in IPTG concentration does not significantly influence JSU protein expression under the applied conditions. Further optimisation, including a broader range of inducer concentrations and induction durations, is recommended to enhance the production of soluble, functional JSU protein.
