Abstract

Academic achievement is an indication level of learning effort by someone. One of them is grade point average (GPA). Some researches showed correlation between GPA and medical competency test. This medical competency test consists of two parts: multiple choice question computer-based test (CBT) and an objective structured clinical examination (OSCE). This cross sectional study aimed to determine the correlation of GPA with CBT scores for the period of November 2018 - August 2019 on students of Medical Faculty Riau University. The number of samples in this study were 149 students. GPA assessment were obtained from preclinical and clinical data of GPA. The majority GPA results are included in the value of 2.75-3.49 and have a percentage of 94% (preclinical), 96% (clinical) and 98.7% (combined). CBT score <66 were obtained from 12.8% student while 87,2% students get score ≥ 66. Spearman test showed significant correlation between GPA and CBT scores of students.

Abstract

Accumulation of triglycerides associated with  insulin resistance in type 2 diabetes mellitus (T2DM). Alpha lipoic acid (ALA) has hypotriglyceridemic effects by lowering triglyceride levels in blood and liver. This study aims to analyze effect of ALA on triglyceride levels in  type 2 diabetic rats. Fifteen Wistar male rats (±8-12 weeks, 200-250 gram) divided into 3 groups (control, T2DM  and  T2DM–ALA given for 3 weeks). Diabetes was induced by giving streptozotocin (50 mg/kg.bw) and nicotinamide (110 mg/kg.bw) after 15 minutes intraperitoneally. Diagnosis of T2DM was determined if the rats have perifer blood glucose >250 mg/dL after 3 days. The results showed that triglyceride levels in  ALA group (p=0,003) lower than T2DM group, T2DM group (p=0,031)  had higher triglyceride level than control and no difference of triglyceride level between  T2DM-ALA given group with control (p=0,696). This study concluded that ALA can affect triglyceride levels in T2DM thus prevents occurrence of hypertriglyceridemia.

Abstract

BACKGROUND: Diabetes mellitus (DM) is associated with an accelerated atherosclerotic macrovascular disease affecting medium-sized arteries. Several evidences support the role of oxidative stress in atherogenesis. However, the role of alpha lipoic acid (ALA) to prevent atherosclerosis is still debatable. This study was conducted to determine the effect of 60 mg/kg/day ALA for 21 days toward the expression of intercellular adhesion molecule-1 (ICAM-1) in rat model.METHODS: Eighteen male rats were divided into three groups labelled as control group, type 2 DM (T2DM) group, and T2DM+ALA group. The T2DM rat models were created by intraperitoneally injecting 50 mg/kg streptozotocin, followed by 110 mg/kg nicotinamide. Immunohistochemistry was used to evaluate the ICAM- 1 expression in rats. Quantitative image analysis of immunohistochemical stains was done on the abdominal aorta using Adobe Photoshop CS3 to find the area percentage and intensity. Kruskal-Wallis and Mann-Whitney tests were used to compare the mean value of area percentage and intensity.RESULTS: There was an increase in area percentage and intensity of ICAM-1 expression. The highest area percentage of ICAM-1 expression was found in the DM group, while the lowest was found in the control group. There were significant differences in the area percentage and intensity between DM+ALA group and DM group, where the area percentage and intensity of ICAM-I in DM group was higher than the DM+ALA group.CONCLUSION: In conclusion, our results demonstrate that ALA inhibits the expression of ICAM-1 in T2DM rat models.KEYWORDS: atherosclerosis, ICAM-1, alpha lipoic acid

Abstract

Background: The ongoing COVID-19 pandemic has led to the emergence of several variants of concern. To rapidly identify those variants, screening samples for whole-genome sequencing (WGS) prioritization could be performed. Objective: We optimized the polymerase chain reaction (PCR) screening method to identify the mutation in spike and ORF1a regions. Methods: We adopted primers targeting mutation in spike and ORF1a region from another study. We optimized the PCR screening method using kits readily available in Indonesia. Firstly, we compared N1 and N2 primers as internal positive control. We also compared GoTaq® 1-Step RT-qPCR System and Indonesia TFRIC-19 BioCOV-19 for the multiplex reaction. We used the optimized composition to screen SARS-CoV-2 positive samples from April – June 2021. Samples with spike and/or ORF1a target failure were subjected to whole genome sequencing (WGS). Results: The results demonstrated the N2 BioCOV-19 reaction as the optimized multiplex PCR composition for spike and ORF1a mutations screening. Whole-genome sequencing has shown that a sample with spike and ORF1a targets failure to be Alpha variant, while other samples with single target failure as non-variants of concern. Therefore, a multiplex RT-PCR composition has been optimized to detect mutation in spike and ORF1a regions. Conclusion: We have optimized a multiplex RT-PCR composition to detect mutation in spike and ORF1a regions.

Abstract

Various studies have been carried out to obtain proper management for atherosclerosis. Proteasome, a subcellular enzyme complex, is a potential therapeutic target for atherosclerosis. However, the effect of proteasome inhibitors on atherosclerosis still needs to be explored. It was an experimental study with a post-test-only control group design conducted at the Faculty of Medicine, Universitas Riau in Juni–November 2021. This study aimed to analyze the effects of proteasome inhibitors on catalase expression and intima-media thickness (IMT) in the thoracic aorta of atherosclerotic rats. Fifteen male Wistar rats were randomly divided into three groups (five rats per group), namely rats given standard feed (control, group I), rats induced atherosclerosis (group II), and rats induced atherosclerosis and given proteasome inhibitor (group III). The proteasome inhibitor, bortezomib, 50 µg/kgBW/day was given intraperitoneally on days one and three. After 4 days, rats were terminated, and the thoracic aorta was taken for the IMT analysis and catalase expression assessment using immunohistochemistry. Catalase expression was carried out quantitatively using Adobe Photoshop software. Analysis of variance test was used to compare the expression of catalase and IMT. A p value<0.05 was considered statistically significant. The results showed a significant decrease in IMT in group III compared to group II and an increase in catalase expression in group III compared to group II but not statistically significant. This study concludes that administration of bortezomib 50 µg/kgBW in atherosclerotic rats could inhibit thickening tunica intima-media in the thoracic aorta, although not significantly increasing the catalase expression.

Abstract

Ketidakseimbangan oksidan dan antioksidan dalam jaringan dapat menyebabkan kondisi stres oksidatif yang berakibat kerusakan dan kematian sel. Malondialdehid, sebuah produk dari peroksidasi lipid, sering digunakan sebagai salah satu indikator untuk mendeteksi kerusakan jaringan akibat stres oksidatif. Secara turun temurun, masyarakat telah meng­ gunakan bawang merah (Allium ascalonicum L.) setelah mengkonsumsi makanan tinggi lemak. Penelitian ini bertujuan untuk menganalisis pengaruh air perasan umbi bawang sebagai antioksidan pada tikus yang diberi diet tinggi lemak berdasarkan perubahan ka­ dar MDA plasma. Penelitian ini adalah eksperimental laboratoris dengan post test only design. Mencit dikelompokkan menjadi 3 yaitu kelompok diet normal, kelompok diet tinggi lemak, dan kelompok diet tinggi lemak ditambah perasan air bawang dengan konsentrasi20%, 40% dan 80%. Hasil penelitian menunjukkan bahwa terdapat perbedaan kadar MDA plasma yang bermakna (p<0,05) antara kelompok diet lemak tinggi yang ditambahkan perasan 80% bawang merah dibandingkan kelompok diet tinggi lemak saja. Penamba­ han air perasan bawang 80% dalam kelompok pakan tinggi lemak dapat menurunkan kadar MDA plasma ke kadar yang tidak berbeda bermakna dengan kelompok pakan normal (p>O.OS). Namun  , tidak ada perbedaan yang signifikan antara penambahan air bawang di 20% dan 40%'s pemerasan. Dari penelitian ini dapat disimpulkan bahwa air perasan dapat menurunkan kadar plasma MDA tikus dengan konsentrasi 80% sebagai konsentrasi yang terbaik.

Abstract

Type 2 diabetes is a chronic disease that is increasingly prevalent due to changes in lifestyle and economic factors. Early detection is crucial to prevent serious complications that can lead to high morbidity and mortality rates. An educational seminar and blood glucose screening were conducted in Mukti Sari Village, Tapung District, Riau Province, aimed at enhancing community knowledge about diabetes and its management. The activities included participant preparation, an interactive seminar, and blood glucose testing. Out of 76 participants aged 18-55, 57 individuals (73%) were tested; results indicated that 3 people (5.4%) had diabetes, 8 people (14.2%) were suspected of being prediabetic, and 46 people (82.1%) were normal. The prevalence of diabetes in this activity was higher than national data from 2018 (10.2%) and 2023 (11.7%). Discussions revealed that the community has good knowledge about diabetes and its complications but lacks appropriate attitudes and behaviors toward prevention, including dietary management, physical activity, and regular screenings. It can be concluded that this detection and education activity for type 2 diabetes can identify prediabetic groups in the community and inform local medical teams of the ongoing need to improve community attitudes and behaviors regarding type 2 diabetes.

Abstract

Cats are known to be potential reservoirs for a variety of zoonotic pathogens. However, the overall prevalence of zoonotic pathogens in the cat population remains unclear amid growing concerns. This study aimed to measure the combined prevalence of zoonotic pathogens in cats through a systematic review and meta-analysis. Relevant literature reporting the prevalence of zoonotic pathogens in cats, published from 2015 to 2025, was collected from databases. A total of 49 studies met the inclusion criteria, encompassing a total sample size of 18,206 cats. A meta-analysis was performed using a random-effects model. Toxoplasma gondii, Bartonella henselae, and Campylobacter spp. were the most frequently reported pathogens, with pooled prevalence estimates presented with 95% confidence intervals (CIs). The I² statistic was used to assess heterogeneity. The combined prevalence of zoonotic pathogens in cats was estimated at 24% (95% CI: 17–32%). Considerable heterogeneity was observed among the studies (I² = 98.4%, p < 0.001), and this likely reflects differences in geographic region, diagnostic methods, and pathogen type. Individual study prevalences ranged from 0.02 to 0.97, with varying weights according to sample size and variance. This meta-analysis highlights the relatively high prevalence of zoonotic pathogens among cats. The relevance of applying a One Health perspective is emphasized by these findings for the development of evidence-based public health policies to reduce zoonotic risks at the community and global levels.
