Abstract

The process of freezing and thawing on semen can lead to physical stress, often called cold shock, and couses the structural and biochemical damage that affecting cell function and ultimately lead to the death of the cell The aim of this study was to know the effect of the addition of various concentrations of dimethylsulfoxide (DMSO) as the intracellular cryoprotectant in phosphate yolk diluent on the post thowing quality of the green jungle fowl semen. The study used eight green jungle fowl semens which were collected with massage techniques. Semen was evaluated macroscopically and microscopically. Good quality semen was diluted with phosphate yolk which was added four different concentration of DMSO, namely 4%, 6%, 8%, and 10%. Semen was then filled and sealed in a mini straw (0.25 mL) with the concentration of 150.106 cells, and equilibrated at 4oC for 4 hours. The semen freezing was processed using conventional method. Evaluation was performed on post thawing semen. The evaluation of semen quality included the progressive motility and plasma membrane intact. Data were analyzed by analysis of variance. If there were any significant differences, the data were futher analyzed by Duncan test. Abstract

The objective of this study was to evaluate the viability and integrity of canine semen diluted in egg yolk glucose-citrate and Tris-citrate-glucose-soya bean. Semen was collected by manual manipulation from an apparently healthy kintamani dog stud. The semen was subjected to gross and microscopic examination to determine its viability following dilution in different extenders. Only semen with motility rate of 60% or higher was used in this study. Semen samples were diluted in Tris-citrate-glucose-soya bean and egg yolk glucose-citrate extenders at two levels of dilution; sperm to extenders ratio of 1:2 and 1:3. Following semen dilution, evaluations were made on sperm motility, percentage of live sperm, and DNA integrity at 0 hour, 3 hours, and 6 hours under 5oC of storage. Data was analyzed by multivariate analysis of variance (MANOVA).The present results showed a high significant different (p<0.01) in sperm motility, percentage of live and DNA integrity between treatment with egg yolk glucose-citrate and Tris-citrate-glucose-soya bean.  The result also showed that percentage of live sperm and DNA integrity of semen at 1:2 dilution difference significantly (p<0.01) to that of 1:3 dilution. The sperm viability and integrity were significantly difference (p<0,01) between the duration of storage at 5°C. Thus, the current study indicated that Tris-citrate-glucose- soya bean extenders  was sufficient to maintain motility, viability, and DNA integrity of kintamani dog  spermatozoa during storage at 5 oC. Further research should be conducted to evaluate the fertility of spermatozoa following dilution with these extenders.

Abstract

Chicken is important part in Balinese Hindu ceremony. Chicken were used as ceremony are chicken with black feather (siap selem), red feather (siap biing), white feather (siap putih) that a white feather chicken with white colour feet, yellow feather (siap putih siungan), that a white feather chicken with yellow colour feet, and chicken with mixed colour of those four colour (siap brumbun). Limitations of technology owned by community makes the chicken’s breeding doesn’t provide economic added value. The purpose of this service is to provide innovation of artificial insemination technology of chicken production for ceremony. The Innovation is given through counseling and workshops . The results showed that people knowledge about disease and breeding management was increased. The farmer have been able to do artificial insemination and vaccination in chicken. The participant were very enthusiastic in following the activities and understand the disease and how to prevent diseases in chickens. These skills are need to trained constantly to find the comfortable thechnique. The conclusions is farmers have been able to perform artificial insemination in chicken.

Abstract

Background: Long-tailed macaques (Macaca fascicularis) frequently share their habitat with humans, leading to conflicts, especially when habitat disturbances force them to exploit human food sources. In response, humans sometimes use air rifles to deter these animals. Preventive measures such as sterilization programs, habitat restoration, and public education about responsible interactions with macaques are essential to reduce the occurrence of such incidents and promote coexistence. Purpose: This report describes the anesthesia and surgical management of gunshot wounds in long-tailed macaques. Case(s): Two adult free-ranging male long-tailed macaques in the Ubud Monkey Forest, Bali, Indonesia, were presented with gunshot wounds on the inguinal region and right caudal pelvic area, respectively. These macaques were captured and anesthetized for a scheduled vasectomy. A detailed clinical examination indicated that the projectiles were lodged in the subcutaneous tissue. Based on their shape and size, the projectiles were suspected to be 0.177 caliber (4.5 mm) air rifle pellets. Case Management: Surgical removal was performed to extract the air rifle pellets. Preoperatively, the macaques received amoxicillin (15 mg/kg BW, IM), ketorolac (2.5 mg/kg BW, IM), and Visine® eye lubricant. Propofol was used for anesthetic maintenance (1 mg/kg BW, IV). Following pellet removal, the wound was sutured with monofilament absorbable poliglecaprone 25, and the planned vasectomy was conducted using the bilateral incision method. Postoperatively, the macaques were administered atipamezole (0.02 mg/kg BW, IM) for anesthesia reversal, placed in recovery cages in lateral recumbency, and closely monitored. Conclusion: Once the macaques showed clear signs of recovery from anesthesia, they were transported back to the capture site and released into their original group.

Abstract

Berkshire pigs are a popular choice for small and large scale pig farming enterprises worldwide, including in Bali. Berkshire pigs are well known for their superior meat quality and relatively fast growth rate. This study aimed to determine the effect of storage duration on the quality of Berkshire boar spermatozoa diluted with Beltsville Thawing Solution (BTS®) supplemented with melatonin. Semen from Berkshire boars was diluted using BTS® supplemented with melatonin at a concentration of 1.0 mM and stored at a temperature of 15 –20°C. Semen quality was evaluated at different storage intervals: (P1) 48 hours, (P2) 60 hours, and (P3) 72 hours. The observed semen quality parameters included sperm motility, sperm abnormalities, sperm viability, and plasma membrane integrity of spermatozoa, which were examined using smear preparations observed under a microscope. In addition, sperm malondialdehyde (MDA) levels as an indicator of oxidative stress were measured using the Porcine MDA ELISA Kit. Duncan’s multiple range test analysis showed that the storage duration of Berkshire boar semen diluted with BTS® supplemented with melatonin was able to maintain sperm motility, abnormality, viability, plasma membrane integrity, and malondialdehyde levels up to 72 hours of storage, with sperm motility of 40.66%, sperm viability of 53.00%, and sperm abnormalities of 6.33%. These values met the minimum standards set by the Indonesian National Standard (SNI 8034:2023) for liquid boar semen. These results indicate that melatonin acts as an antioxidant capable of neutralizing free radicals, thereby inhibiting the occurrence of oxidative stress in spermatozoa cells.

Abstract

This study aimed to evaluate the effect of palmyra fruit water–egg yolk diluents supplemented with moringa (Moringa oleifera L.) leaf extract on the intact plasma membrane, intact acrosomal membrane, and motility of boar spermatozoa during liquid storage. A completely randomized design was used, with seven diluent formulations varying in concentrations of egg yolk and moringa leaf extract. Each treatment was replicated four times and stored at 15-18°C for up to 72 hours. Semen quality was evaluated at 0, 24, 48, and 72 hours of storage. Data were analyzed using analysis of variance followed by Duncan’s multiple range test. Both diluent formulation and storage duration significantly influenced all semen quality parameters (p < 0.05). The diluent containing palmyra fruit water supplemented with 20% egg yolk and 15% moringa leaf extract produced the highest membrane integrity, motility, and viability with the lowest abnormality rate. The semen maintained acceptable quality for artificial insemination for up to 48 hours of storage. The superior performance of this formulation is biologically associated with the synergistic interaction between carbohydrate-derived energy from palmyra fruit water, phospholipid-mediated membrane stabilization from egg yolk, and antioxidant protection from moringa leaf extract against oxidative stress.
