Abstract

The presence of opportunistic pathogen bacteria in shrimp pond water is caused by 3 factor. There is environmental factor, shrimp condition and the existence of bacteria. Vibrosis disesase in shrimp is harmfull to farmers because it makes reducing result and caused disease in humans. Bacteria often found among types of Vibrio. The toxR gene is known as the transmembrane regulator gene that control the expression of toxin gene. This study aims to determine the existence of toxR gene in Vibrio parahaemolyticus in pond water samples by PCR method. Bacterial isolation was performed using selective media CDVP Nissui. Enumeration of bacteria by TPC method to determine the number of suspected V. parahaemolyticus on CDVP. Extraction of bacterial DNA using QIAamp® DNA Mini Kit. A good qualitative result of the DNA extraction then amplified by toxR primer. Sequencing process sent to PT. Genetica Science. V. parahaemolyticus inoculated on CDVP media showed blue-green color because it can not ferment sucrose. Dry media CDVP Nissui can be used as a medium in the enumeration of Vibrio parahaemolyticus (1,6 x 102 CFU/ml in water sample) because it can be distinguished by the type of bacteria colony color that appears. The PCR protocol used was annealing at 62ºC with a cycle 30 times. Electrophoresis results showed that bacterial samples from pond water positively containing toxR gene with length 368 bp. From sequence base data of nucleotides, the sample has 99% equation with V. parahaemolyticus in the NCBI bank gene. Samples of V. parahaemolyticus bacteria in shrimp pond water in Rembang are known contain the toxR gene.Keyword : V. parahaemolyticus, toxR gene, enumeration, TCBS, CDVP

Abstract

Bacteria can grow in variety of environments including those associated with marine organisms such as seagrasses, sponge, algae and soft corals. Bacteria association has the ability to produce bioactive compounds that can be used as a defense against pathogens and their hosts by producing secondary metabolites such as antibacteria compounds. This study aimed to asses the potential of bacteria associated with seagrasses Enhalus acoroides and Syringodium isoetifolium. This study was conducted with bacterial isolation from seagrass E. acoroides and S. isoetifolium, characterization of bacterial isolatesin morphology, antibacterial test, and biochemcal activity test. The result obtained four bacterial isolates, two isolates of Syringodium isoetifolium and two isolates of Enhalus acoroides. Two isolates from Syringodium isoetifolium had the largest ability inhibitory zone of inhibtion against the bacteria P. Aeruginosa (5.7mm) and against the bacteria E. coli (6.65mm).Key word : Antibacterial Compound, Seagrass-Associated Bacteria, Vibrio, Enhalus acoroides,      Syringodium isoetifolium

Abstract

Biofouling is a settlement process from marine organisms initiated by biofilm (microbial layers). Corrosion on the vessels, pipelines under the sea, oil plants, etc are big problems in the industrial today cause adversely affects. Antifouling paints have been developed to prevent settlement of organisms, however antifouling paints contain heavy metals and biocides which have toxic effects on marine ecosystems. Antifouling compound which environmental friendly is a solution for one of the paint component. The aim of this research was to isolate bacteria associated with brown algae which had potential to produce antifouling compound from Karimunjawa islands, Jepara.Isolate KS1-1 showed antifouling activity against Pseudomonas aeruginosa as biofilm-forming bacteria.This isolate had been characterized in morphology, moleculer and biochemichal identification.Moleculer analyses of 16S rRNA sequence, KS1-1 showed similarity 94% from 287 bp with Vibrio sp. Abstract

Plastic waste is one of the environmental issues that still need to be handled properly. plastic  commonly  used  are  basically  formed  by  polyethylene  which  is  contains  chemical compound.  Less cavity of plastic can disturb soil aeration which is important for other aerobic organism. This problem lead to increasing ecological threat. The main objective of present study is to isolate the soil microorganism that have a role as an agent  through plastic degradation. Bacteria were isolated from the surface of the plastic which is buried in soil. The  isolation is been carried out through soil serial dilution method and inoculated into selective King’s B agar. Isolates were obtained and then tested the ability of degradation of the PE. Isolates with the highest degradation  activity isolates G then identified using molecular methods based on 16S rRNA. the results showed that the 16S rRNA sequencing isolates G measuring 1,500 bp with the highest homologous species derived from the genus Bacillus. Keywords: Polyethylene, bacteria, biodegradation

Abstract

Shrimp is one of the main commodities in the aquaculture industry because it has high economic value and high demand product. The Litopenaeus vannamei shrimp have the advantage of being able to grow as fast as a tiger shrimp (3 g / wk), can be grown on a wide salinity range (0.5-45 ppt), lower protein requirement (20-35%) than tiger shrimp and stylirostris shrimp. Vibrio parahaemolyticus is a normal flora in the brackish waters environment which is pathogenic to shrimp commodities as well as in humans. The use of Probiotics as additional feed in the form of microbial cells intact widely used in shrimp farming as one effort to improve the quality of the environment and supress the growth of pathogenic bacteria. One of the probiotic products used is super PS. This study aims to obtain bacterial isolate probiotic products that can suppress the growth of Vibrio parahaemolyticus bacteria. Isolation is also done on mangrove sediments as a comparable type of bacteria that can suppress the growth of pathogens. Methods used include isolation of bacterial probiotic and mangrove sediments, antagonistic test, and molecular identification with PCR methods (Polymerase Chain Reaction). The isolation result obtained by seven isolate bacteria of probiotic product and eleven isolate of mangrove sediment bacteria. One selected bacterial isolate from isolation of probiotic product that is IP 7 which able to supress the growth of Vibrio parahaemolyticus with a diameter of clear zone 10.84 mm. The results of identification using the 16S rRNA gene sequence showed that IP 7 isolate had a similarity index of 92% with Lysinibacilllus cresolivorans.Keyword : Litopenaeus vannamei, Vibrio parahaemolyticus, probiotics, antibacterial, gen 16S rRNA.

Abstract

AbstrakPentana, dekana, pentadekana, dan parafin merupakan jenis kelompok senyawa alkana yang tidak dapat larut dalam air dan sulit terdegradasi. Sifat ini yang menjadikan senyawa alkana dapat mencemari lingkungan. Penelitian ini bertujuan untuk mengetahui konsentrasi optimum senyawa alkana bagi pertumbuhan isolat LBF-1-0025 dan mengidentifikasinya secara molekuler berdasarkan gen 16S rDNA. Dari skrining awal diketahui bahwa isolat LBF-1-0025 memiliki potensi tinggi dalam mendegradasi senyawa pentana, dekana, pentadekana, dan parafin. Optimasi pertumbuhan isolat LBF-1-0025 pada beberapa senyawa alkana dilakukan dengan menumbuhkannya dalam medium Artificial Sea Water (ASW) yang mengandung beberapa konsentrasi senyawa pentana, dekana, pentadekana, dan paraffin. Isolat LBF-1-0025 mampu tumbuh secara optimal pada pentana 150 ppm, dekana 200 ppm, pentadekana 150 ppm, dan parafin 200 ppm setelah 3 hari inkubasi. Hasil analisis sekuen gen 16S rDNA, bakteri LBF-1-0025 memiliki kemiripan sebesar 97% dengan Salinicola peritrichatus DY22.The Growth Optimization of Marine Bacteria  Salinicola  peritrichatus LBF-1-0025 in AlkanesAbstractPentane, decane, pentadecane, and paraffin are kind of groups of alkane compounds that are insoluble in water and difficult to degrade. This characteristic cause alkane compounds to pollute the environment. The aims of this research were to determine the optimum concentration of alkane compounds for the growth of isolate LBF-1-0025 and molecular identification based on 16S rDNA gene. From initial screening test, it was known that isolate LBF-1-0025 had high potential in degrading pentane, decane, pentadecane and paraffin compounds. Growth optimization of isolate LBF-1-0025 on several alkane compounds were carried out by culturing it in ASW medium containing various concentration of alkane (pentane, decane, pentadecane, and paraffin). Isolate LBF-1-0025 had optimum growth in pentane 150 ppm, decane 200 ppm, pentadecane 150 ppm and paraffin 200 ppm after 3 days incubation. Result analysis of 16S rDNA gene showed that isolate LBF-1-0025 had a similarity of 97% with  Salinicola  peritrichatus DY22.

Abstract

The genus Bacillusis one of the wellknown group of bacteria as Plant Growth-Promoting Rhizobacteria (PGPR) that can support plant growth or otherwise control the plant pathogens. In this study, three strain of B. pumilusfrom BIOTEC culture collection (BCC 7507, TBRC 2887 and TBRC 2903) were investigated for their potential to be used as fungal pathogen control.  The growth characteristics of these B. pumilusstrains were determined by making the growth curve of each strain. Further, antagonistic activity of B. pumilusagainst the anthracnose pathogens C. capsici, C. gloeosporioidesand C. acutatum, the rice dirty panicle pathogen Curvularia lunata, the leaf spot pathogen Alternaria brassicicola, and the rice blast pathogen Magnoporthe griseawere evaluated by identified the inhibition zone from their activity. Then, the secondary metabolites produced by these Bacillusstrains were also examined using anti-fungal activity assays. All B. pumilusstrains showed very low biocontrol activity against M. grisea, but not against C. acutatum, C. Capsici, C. gloeosporioides, A. brassicicolaand C. lunata. Culture extracts derived from these strains did not show any activity against M. griseain the spore germination inhibition assay.  These results showed that the three B. pumilusstrains did not exhibit strong potential for application as biocontrol agent.

Abstract

Dengue Hemorrhagic Fever (DHF) is an infectious disease caused by dengue virus and transmitted by Aedes aegypti and Aedes albopictus. In 1968 the Incidence Rate (IR) of this disease was 0.05 per 100.000 population and it increased to 49.5 per 100.000 population in 2015. Data of DHF in Semarang City from 2012 to 2016 showed that the incidence trend of this disease increased. For five years from 2010 to 2016, Semarang City was in the top three rankings of DHF IR in Central Java. The case of dengue fever in Indonesia is closely related to environmental conditions such as temperature inside the house. This aim of this study was to describe temperature inside the house and dengue in the productive age (13-60 years) in Semarang City. The subjects were 54 people consisted of 27 cases and 27 controls and selected by consecutive sampling. The source of this study was the primary data from observation and measurement of temperature inside the house using a thermohygrometer. These measurements did after the subjects of dengue case were health. The result showed that the proportion of subjects who lived at house with ≥25°C – ≤30°C temperature (Aedes aegypti’s optimal temperature for breeding) were mostly in the control group (37%). While the proportion of subjects who lived at house with <25°C or >30°C temperature (not an optimal temperature for Aedes aegypti’s breeding) were mostly in the case group (81,5%).

Abstract

Dengue Hemorrhagic Fever (DHF) is an infectious disease caused by one of 4 different Dengue virus and transmitted through Aedes aegypti and Aedes albopictus mosquitoes. DHF is one of the major public health problems in Indonesia, one of which is in Semarang and its surroundings. Incidence Rate (IR) DHF of Semarang District from 2006 to 2016 is always much higher than IR DHF Central Java and IR DHF National. This study aims to analyze the practice of insecticide use with the incidence of DHF in Semarang. The type of research used is observational analytic done with case control approach. The population in this study were DHF in March-May 2017 at three hospitals in Semarang City and not DHF (neighbor patient) who live in Semarang and surrounding areas (Kendal and Demak). The sample was 48 cases and 48 controls using consecutive sampling technique. Data obtained in this study were analyzed by Chi-Square test and calculation of Odds Ratio with 95% confidence interval. The results showed that there was no correlation between the use of insecticide (p=0,289, OR=0,636 and 95% CI=0,275-1,471). Based on these results, the practice of insecticide use is not related to the incidence of DHF in Semarang. Therefore, people are expected to always apply healthy hygiene behavior and do 3M Plus in order to avoid the incidence of DHF.

Abstract

Dengue Hemorrhagic Fever is an infectious disease caused by dengue virus that is transmitted through Aedes mosquito bites, both Aedesaegypti and Aedesalbopictus. DHF is one of the major public health problem in Indonesia.The Incidence Rate (IR) of DHF in Semarang City from 2006 to 2016 are always much higher than IR DHF of Central Java and IR DHF of National Indonesia.The cases of Dengue Hemorrhagic Fever (DHF) in Semarang City has increased significantly and cause death.This study aims to see the Breeding Places description and its relation to the incidence of Dengue Hemorrhagic Fever in Semarang city and surrounding areas.The type of research was an observational analytic study using Case-control design. The cases sample in this study were hospitalized DHF patients from the period of March to May 2017 in three hospitals in Semarang City.The control samples were health people and live around the cases with the same characteristics. Total sample were 27 cases and 27 controls. The data was analyzed by Mann Whitney test.The result showed that insignificant factors were existence of daily necessities containers (p=0,383), existence of containerswhat are not daily necessities (p=0,307), existence of total containers (p=0,142), opened containers condition (p=0,199), closed containers condition (p=0,683), and containers infested with larvae (p=0,253).  In conclusion, there is no association between breeding places factors with DHF incidence in Semarang city and surrounding areas.
