Abstract

Water clover (Marsilea crenata) is one type of aquatic plants. Plants that fall into these ferns are found in the rice field, ponds, lakes, swamps and rivers. This plant has a very distinctive morphology of its leaves resemble an umbrella shape that is composed of four petals facing child leaves. In the area of young clover Java is widely used as food. Utilization of clover water not only as food, water clover leaves and stems can also be used as drug . A research about peculiar properties of water clover for human health is very important to do, like antioxidant activity assay and qualitative bioactive compounds assay. The objectives of this research were to determine content, chemical compounds (water, lipid, protein, ash and crude fiber), antioxidant activity and bioactive compounds of water clover. Methods that used in this research were proximate analysis, quantitative antioxidant activity assay using DPPH method, and phytochemical method. Water clover planted first in Laboratorim Characteristics of Raw Water Results, Aquatic Product Technology, FPIK, IPB. Crude extracts from water clover have antioxidant activity that measured as IC50 value. The IC50 value from chloroform extract, ethyl acetate extract, and methanol extract of water clover were 1285,39 ppm; 915,03 ppm dan 634,73 ppm, respectively. The crude extracts of water clover contained 6 bioactive compounds, i.e alkaloid, steroid/triterpenoid, flavonoid, carbohydrate, reducing sugar and free amino acid. These compounds suspected have positive physiology functions for human health, so that water clover can be used for raw material of nutraceutical. Abstract

Sea horse Hippocampus spp. is a group of unique marine biota and its distribution is spread all over the world. This study aims to determine the characteristics of seahorse Hippocampus comes from North Bintan waters by looking at the yield and degree of hydrolysis (DH) of the protein hydrolyzate sample. This samples obtained from the catch of fishermen in the North Waters of Bintan Island. The analysis is the measurement of the yield and the degree of hydrolysis. The results obtained from the study that the horse of sea H. comes from the North waters of Bintan shows that the highest yield in the sea horse body is bone that is equal to 43.69%. This proves that the seahorse is one of the ingredients that can be utilized as a further processed dry feed such as in the manufacture of flour and dried products (such as ginseng). As for the degree of hydrolysis generated from the substrate of H. comes a sea horse protein derived from meat and bone have a high value compared to the substrate of sea horse bone meal. Abstract

Histamine-producing bacteria to predict histamine level production can be identified by a molecular approach. The purpose of this study was to identify the types of histamine-producing bacteria on tuna, little tuna, and skipjack (TTC) meat, to analyze its bioinformatics through phylogenetic tree construction also to determine the levels of its histamine. The identification of histamine-producing bacteria was conducted using a molecular technique based on the hdc and 16S rRNA genes. Histamine levels were measured by a spectrofluorometer. The results showed types of histamine-producing bacteria had been successfully identified, both using specific hdc and 16S rRNA universal primers, including Morganella morganii, Enterobacter hormaechei, Klebsiella aerogenes, and Enterobacter bugandensis. The phylogenetic tree showed that the bacteria M. morganii and E. hormaechei were closely related to one cluster. Meanwhile, the other close relative cluster were Klebsiella aerogenes, Enterobacter bugandensis, and Escherichia fergusonii. In addition, histamine levels of frozen tuna, little tuna, and skipjack were 2.96±0.22 ppm, 2.14±0.23 ppm, and 1.02±0.97 ppm, respectively.

Abstract

Yellow velvetleaf (Limnocharis flava) is aquatic plant and known as vegetable and source of bioactive compound but it has aerenchyma system which affects to its proximate. The research purpose was to determine microscopic characteristics of yellow velvetleaf tissues such as leaf, stem, and root tissue. The proximate and bioactive compound from fresh and steam materials were also evaluated. The methods of the research consisted of plant dimension measurement, making of tissue slide by paraffin method in Johansen-TBA series, phytochemical analysis, proximate and carotenoid total analysis. The results showed that leaf tissue was composed by epidermis unilayer containing stoma, a palisades parenchyma layer, spongy parenchyma layer with some aerenchyma system, and vascular bundle. Stem tissue consisted of epidermis unilayer, cortex containing chlorophyll, starch, and some aerenchyma system, and vascular bundle was amphicribral type. Root tissue was composed by rhizodermis, cortex with aerenchyma system, endodermis multilayer, and vascular bundle. Fresh yellow velvetleaf contained 91,76% of moisture, 12,40% of ash, 7,95% of fat, 22,96% of protein, 11,93% of crude fibre, and 219,01 g/g of carotenoid total in its leaf. Its stem contained 95,33% of moisture, 16,38% of ash, 5,62% of fat, 13,23% of protein, 16,12% of crude fibre, and 92,99 g/g of carotenoid total. Steaming decreased crude fibre content, but increased ash, fat, and protein content. Steaming also increased leaf carotenoid total, but decreased stem carotenoid total. Leaf extract contained flavonoid, phenol, reducing sugar, and amino acid. Stem extract was composed by flavonoid, reducing sugar, amino acid. Flavonoid and reducing sugar were major bioactive compounds in this plant.

Abstract

Seahorse is marine biota that have commercial value and used as ornamental fish and traditional medicinal ingredients. Utilization of seahorses has still not been extensively studied, especially in the form of hydrolysates. Hydrolysates of seahorses are known to have efficacy in biological activity. This study was conducted to identify seahorse morphometrics, determine selected enzymes based on the degree of hydrolysis and determine the anti-inflammatory activity of hydrolysates. The results showed that morphometric identification showed morphological characteristics of Hippocampus kuda, namely the number of body rings 12 units, tail ring 35 units, eye protrusion and cheeks as much as 1 unit, average head length of 3.5 cm, number of body rings on dorsal fin 3 units. The best degree of hydrolysis of seahorses is protein hydrolysis using alkalase enzyme of 42.49%. The highest anti-inflammatory activity on sea horse hydrolyzate has %inhibition of 42.88%.

Abstract

Rumput laut Sargassum sp. berpotensi dikembangkan menjadi produk minuman serbuk, namun memiliki kekurangan yaitu aroma yang amis dan warna kehitaman. Kombinasi dengan ubi jalar ungu, garam rumput laut, dan jahe merah dilakukan untuk meningkatkan daya terima dan nilai fungsional produk. Penelitian ini bertujuan menentukan karakterisasi minuman serbuk berbasis Sargassum sp., ubi jalar ungu, dan jahe merah serta penambahan garam rumput laut terbaik. Tahapan penelitian ini yaitu pengeringan bahan baku menjadi serbuk, pembuatan formulasi, dan analisis. Formulasi 1 terdiri dari Sargassum sp. 25%, ubi jalar ungu 45%, jahe merah 15% dan gula rendah kalori 15%. Formulasi 2 terdiri dari Sargassum sp. 22,5%, ubi jalar ungu 42,5%, jahe merah 15%, gula rendah kalori 15%, dan garam rumput laut cokelat 5%. Hasil uji serat pangan larut air yaitu 5,06% (F1) dan 5,14% (F2), sedangkan serat pangan tidak larut airnya mencapai 22,62% (F1) dan 20,09% (F2). Mineral yang terkandung yaitu kalsium 6,53–7,35 mg/g, kalium 7,41–9,075 mg/g, magnesium 3,16–3,28 mg/g, natrium 1,95–6,32 mg/g, dan besi 0,07 mg/g. Minuman serbuk memiliki nilai aktivitas air sebesar 0,29. Kadar abu tidak larut asam pada produk hanya 0,15–0,20% dan kadar logam berat masih berada di bawah batas maksimum. Senyawa fitokimia yang terkandung yaitu alkaloid, flavonoid, triterpenoid, fenol, tanin dan glikosida. Nilai aktivitas antioksidannya sebesar 86,708 μmol troloks/g (F1) dan 87,499 μmol troloks/g (F2).

Abstract

Tilapia has white flesh, so it is very good to use in making surimi. In terms of texture, tilapia surimi has a slightly dense meat texture which makes the final result of the kamaboko a bit tough. Therefore, it is necessary to add an enzyme to the tilapia surimi so that it gets a softer kamaboko texture. This study aims to analyze the surimi characteristics of red tilapia with the addition of trypsin enzyme extracted from fish intestines. The method used is a simple experimental design with the addition of trypsin enzyme 0%, 1%, 2%, 3% and 4%. The parameters observed were proximate analysis, salt-soluble protein (PLG), pH, whiteness, particle size analysis (PSA), texture profile analysis (TPA), water holding capacity (WHC) and sensory analysis. Based on the analysis results, the addition of trypsin enzyme can improve the characteristics of red tilapia surimi. This can be seen from the results of the measured parameters, namely the hardness, chewiness, gummines, folding test and bite test which show that the texture of tilapia surimi is better and softer.

Abstract

Prebiotic food has become a food consumption trend because of its health benefits, especially for the prevention of stunting in toddlers and children. Mocaf flour combined with pea powder and seaweed salt can be processed into high-protein and high-mineral prebiotic crackers. This study aimed to determine the chemical composition, minerals, and vitamins of prebiotic crackers supplemented with Sargassum polycystum salt. This study consisted of making crackers without seaweed salt (F0) and with 2% seaweed salt (F1), followed by testing the chemical composition, minerals, and vitamins. The data were processed using Microsoft Office Excel 16 and descriptively analyzed. The results showed that prebiotic crackers with the addition of 2% S. polycystum salt increased the ash and lipid content but decreased the moisture, carbohydrate, and dietary fiber contents. Prebiotic crackers with and without the addition of S. polycystum salt had appropriate protein content appropriate SNI 2973:2018 about biscuit, namely 8.72% (F0) and 8.63% (F1). Prebiotic crackers with addition of S. polycystum salt has mineral content Na (389.72 mg/100 g), K (1,271.60 mg/100 g), Ca (224.61 mg/100 g), Mg (73.83 mg /100 g), Fe (4.22 mg/100 g), and vitamin B6 (7.02 mg/100 g). The addition of salt can increase the mineral and vitamin contents of prebiotic crackers.

Abstract

Healthy seaweed salt is low sodium salt from seaweed that offers health benefits for hypertension patients. Indonesian seaweed has the potential to produce healthy seaweed salt. Research to date still focuses on green and brown seaweed but there is still no report for red seaweed. Actinotrichia fragilis is one of red seaweed species that has been discovered in Indonesia’s seawater and has not yet been utilized. Thus, this study aimed to determine the chemical composition and antioxidant activity of A. fragilis flour and the optimum ratio for producing seaweed salt with a high yield, optimum %NaCl, Na/K ratio, and antioxidant activity. Seaweed salt production treatment was the ratio of seaweed flour and distilled water 1:3, 1:5, and 1:10 (w/v), extracted at 40°C for 10 minutes. The mixture was filtered, then dried at 60°C for 30 hours. Data analysis was performed by analysis of variance. The raw material for dried A. fragilis seaweed has a high ash and low-lipid content. Then the ethanol extract had a total phenolic content value of 84.34 mg GAE/g and an antioxidant activity value of 98.22 mg/L. Furthermore, the antioxidant capacity of the ethanol extract was 60.15 mmol ascorbic acid/g and 552.21 mmol Fe2+/g. The best treatment for producing A. fragilis salt is 1:10 with yield of 12.76±0.13%, %NaCl 47.22±1.38%, Na/K ratio 3.32±0.18, IC50 with DPPH and ABTS method 113 mg/L and 87.27 mg/L, total antioxidant capacity 38.21 mg/mL ascorbic acid/g, and 304.32 mmol Fe2+/g. Furthermore, A. fragilis can be used for the production of  healthy seaweed salt.

Abstract

Dinding sel Ulva lactuca menghasilkan ulvan yang merupakan hidrokoloid bersulfat dan berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk menentukan pengaruh lama pemanasan dalam autoklaf (15 dan 20 menit; 121℃) terhadap rendemen, kadar gula dan sulfat, serta aktivitas antioksidan ulvan. Rendemen ulvan, kandungan gula dan sulfat diukur masing-masing dengan metode gravimetri dan spektrofotometri. Ekstrak kering oven diuji gugus fungsi dan antioksidannya menggunakan FTIR dan metode DPPH. Ulvan hanya bisa dihasilkan melalui ekstraksi pada suhu 121℃ waktu 15 menit. Ekstrak kering ulvan memberikan rendemen 1,98%, kandungan gula total 46,06±0,16% dan kadar sulfat 21,53±1,16%. Spektrum FTIR menunjukkan adanya bilangan gelombang 850 dan 790 cm-1 yang merupakan ciri ulvan. Uji DPPH menghasilkan IC50 sebesar 469 ppm, yang tergolong sebagai antioksidan lemah.
