Abstract

Seafood product is an important commodity in the global trade market, however, seafood fraud has been increasingly reported. Therefore, a method to specifically identify species of seafood products is needed. DNA methods gain many interests for detection of seafood fraud due to its robustness. This study was aimed to determine the effectiveness of several methods for isolation of DNA from fresh and processed seafood. The DNA isolation methods used in this study were the CTAB DNAzol, TianGen, Qiagen, and Fasmac.. DNA concentration and purity were determinedusing nanodrop spectrofotometry and electrophoresis. DNA isolation showed that the DNA concentration of fresh seafood products was higher than the processed seafood products. DNA isolation using CTAB method have higher levels of DNA purity (1.60-2.00) than using commercial kits in all samples. The commercial kits offer a simple procedure and minimize the possibility of environmental contaminants.

Abstract

Jeroan ikan dikenal sebagai salah satu sumber enzim pencernaan yang melimpah, terutama sebagai sumber proteinase pencernaan yang baik, salah satunya adalah tripsin. Jenis dan habitat ikan akan memengaruhi sifat tripsin ikan. Tujuan penelitian ini adalah untuk membandingkan komponen asam amino daging dan aktivitas ekstrak kasar enzim tripsin dan usus ikan tuna sirip kuning (Thunnus albacares) dan ikan kakap merah (Lutjanus campechanus). Kedua jenis ikan diamati morfometrik dan rasio bobot jeroan ikan. Selain itu komposisi kimia, komponen asam amino daging, dan aktivitas ekstrak kasar enzim tripsin dari usus kedua jenis ikan tersebut kemudian dianalisis... Secara umum ikan tuna sirip kuning memiliki komposisi asam amino yang lebih tinggi dibandingkan ikan kakap merah, terutama pada komponen asam amino histidina, leusina, asam aspartat, lisina dan asam glutamat. Komponen asam amino yang paling dominan terdapat pada kedua jenis ikan tersebut adalah asam glutamat. Enzim tripsin ekstrak kasar usus ikan tuna memiliki aktivitas yang lebih tinggi yaitu 4,908 U/mg dibandingkan dengan ekstrak kasar usus ikan kakap merah sebesar 0,076 U/mg.

Abstract

Cinnamon leaves contain active compounds such as polyphenols and flavonoids which have antioxidant properties and play a role in lipid metabolism processes. This active substance is known to reduce liver fat levels and has the potential to increase the immune response in catfish. This research aims to determine the effect of adding cinnamon leaf extract to feed on liver fat levels and the health of catfish. Cinnamon leaf extract was added to the feed at five different doses, namely 0 g/kg (control), 0.5 g/kg, 1 g/kg, 2 g/kg, and 4 g/kg feed. The results of the study showed that the addition of cinnamon leaf extract to the feed reduced the fat content in the liver of catfish as seen from the decrease in fat degeneration in the liver of catfish compared to the control. Total erythrocytes, hemoglobin levels and hematocrit levels of fish treated with the addition of cinnamon leaf extract to the feed were lower and significantly different (P<0.05) compared to controls, but total leukocytes were not significantly different between treatment and control.

Abstract

Fish is a fishery product that has high nutritional value. Fish can be processed into various processed products. Processed fish products generally have the characteristic of not lasting long when stored at room temperature so that in the food processing, additives are added. Hazardous food additives that are often used in processed fish products are formalin. Formalin is a preservative that can cross-link with protein so as to prevent damage to product and can extend its shelf life. This study aims to determine the formalin content in processed fish products, namely fish meatballs and salted fish in traditional markets in Jambi City. The research method used laboratory experiments with 4 treatments and 3 replications. The results showed that the samples of fish meatballs and salted fish as a whole were positive for formalin. This shows that the food safety of processed products in traditional markets in Jambi is still low. The knowledge and awareness of the fisheries processing community about hazardous food additives is still low.

Abstract

Porphyridium cruentum has advantages in pigments and exopolysaccharides, so it has the potential to be developed in the food, pharmaceutical, nutraceutical, and cosmetic industries. Porphyridium cruentum cultivation media uses synthetic and indoors, causing high costs. This study aims to determine the growth performance of Porphyridium cruentum in different mediums and method of cultivation. The research methods used cultivating Porphyridium cruentum for 7 days with four treatments, namely indoors Guillard media (IGM), outdoors Guillard media (OGM), indoors fertilizer media (IFM), and outdoors fertilizer media (OFM. Each treatment had three replications. Growth was identified using an optical density spectrophotometer uv-vis, gravimetrically dried weight of biomass and pH adjustment during 7 days of cultivation. The result shows the best growth performance in IFM, followed by IGM, OFM, and OGM. Fertilizer media is better for the growth of Porphyridium cruentum than Guillard media. pH growth Porphyridium cruentum during cultivation 7-7.5. Indoor cultivation is better than outdoor cultivation. The highest biomass was IFM, which had the best performance in IFM with OD 0.247, pH 7, and biomass 79.6%.

Abstract

Taman Nasional Perairan Natuna merupakan salah satu kawasan konservasi laut yang dilindungi karena memiliki keanekaragaman hayati yang sangat penting dalam menjaga keseimbangan ekosistem laut serta mendukung keberlanjutan sumber daya alam perairan Indonesia. Pengelolaan kawasan konservasi memerlukan berbagai pertimbangan dan pemahaman mengenai kondisi fisik seperti adalah parameter suhu dan salinitas. Penelitian ini bertujuan untuk memberikan gambaran yang lebih komprehensif mengenai distribusi suhu dan salinitas di TNPN, khususnya di Pulau Laut. Data pada penelitian ini menggunakan data Argo Float dan akan dianalisis secara spasial dan vertikal dengan analisis stratifikasi perairan. Distribusi horizontal nilai suhu berkisar 25.99-27.31°C dan salinitas berkisar antara 33.01-33.31‰. Distribusi vertikal suhu berkisar antara 25.5-26.5°C dan 33.25-33.4‰. pada awal musim Barat terdapat lapisan homogen pada kedalaman 0-5 m dan bervariasi didaerah pesisir. Variasi nilai suhu dan salinitas diduga karena percampuran vertikal yang diinduksi oleh angin musim barat yang bergerak dengan kecepatan rata-rata 6.69 m/s. Sehingga, Taman Nasional Perairan Natuna cocok untuk dikembangkan sebagai kawasan konservasi.

Abstract

This study aims to determine the spawning techniques of Leptobarbus hoevenii fish at the Sungai Gelam Freshwater Aquaculture Center (BPBAT). Activities include maintaining prospective broodstock, selecting mature broodstock, injecting ovulation hormones, and the spawning process, stripping, egg handling, water quality monitoring to highlight its importance, and embryo development. Ovaprim hormone injections were administered to the female broodstock four times and once to the male broodstock to stimulate ovulation. The stripping process was carried out after ovulation and was followed by dry mixing of eggs and sperm before being spread into the hatching tank. The results showed a fertilization rate of 63,08% and a hatching rate of 9,73%, which were influenced by gamete quality and the physiological condition of the broodstock. Observations of embryogenesis showed developmental stages ranging from morula, blastula, gastrula, to organogenesis before the eggs hatch into larvae. Water quality parameters during the study were in the optimal range for spawning, namely pH 7,24–7,77, temperature 27–30,5°C, and DO 3,53–5,58 mg/L. The results of this study explain that the success of Hoven’s carp fish seeding is highly dependent on the readiness of the broodstock, the application of artificial spawning techniques, and good water quality management.
