Abstract

Trypsin is a protease that breaks protein peptide bonds. Fish intestines canbe used as an alternative raw material for trypsin. Trypsin enzymes from theintestines of different fish species have different characteristics. This studyaimed to determine the characteristics and stability of trypsin enzyme inNaCl extracted from fish intestines based on differences in fish species. Trypsinactivity was optimal at 50 °C and pH 8, with specific activity values of 0.5993U/mg in rabbitfish, 0.3880 U/mg in sharks, and 0.6964 U/mg in flatfish. Themaximum reaction speed (Vmax) was the highest for trypsin from the intestineof rabbitfish (0.2585 mmol/s), followed by flatfish (0.1042 mmol/s), andshark (0.0599 mmol/s). The lowest Km value was obtained for trypsin fromsharks (0.4084 mM), followed by flatfish (1.0253 mM), and rabbitfish (4.5952mM). Trypsin from the intestines of rabbitfish and flatfish was stable in NaClsolution (concentration 5-30%), as it can maintain a relative activity of morethan 50%. In contrast, trypsin extracted from the intestines of milk fish hada relative activity below 21%. The average molecular weights of the threetrypsin enzymes were 26.8, 27.2; and 21.9 kDa, respectively. Differences inthe type of fish affected trypsin enzyme activity. Flatfish are omnivorous,and rabbitfish, as herbivores, have better enzyme activity values than sharks,as carnivores.

Abstract

Dinding sel Ulva lactuca menghasilkan ulvan yang merupakan hidrokoloid bersulfat dan berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk menentukan pengaruh lama pemanasan dalam autoklaf (15 dan 20 menit; 121℃) terhadap rendemen, kadar gula dan sulfat, serta aktivitas antioksidan ulvan. Rendemen ulvan, kandungan gula dan sulfat diukur masing-masing dengan metode gravimetri dan spektrofotometri. Ekstrak kering oven diuji gugus fungsi dan antioksidannya menggunakan FTIR dan metode DPPH. Ulvan hanya bisa dihasilkan melalui ekstraksi pada suhu 121℃ waktu 15 menit. Ekstrak kering ulvan memberikan rendemen 1,98%, kandungan gula total 46,06±0,16% dan kadar sulfat 21,53±1,16%. Spektrum FTIR menunjukkan adanya bilangan gelombang 850 dan 790 cm-1 yang merupakan ciri ulvan. Uji DPPH menghasilkan IC50 sebesar 469 ppm, yang tergolong sebagai antioksidan lemah.

Abstract

The sea sponge Stylissa carteri plays an important role because it is one of the constituent biota of coastal and marine ecosystems, especially in coral reef ecosystems. The increasing utilization of bioactive compounds in sea sponges is feared to significantly reduce the population of sea sponges. The prevention effort involves cultivating sea sponges for sustainable utilization. This study aims to determine the antioxidant activity of transplanted Stylissa carteri sponges and those that live naturally in nature. This study was conducted in Pramuka Island using the sponge explant combination method. The three types of transplants were number one (SO), number two (S. carteri and Aaptos suberitoides), and number three (S. carteri, A. suberitoides, and Acropora hard coral). Antioxidant activity was tested with three repetitions on three transplanted and naturally occurring S. carteri sponges (SA) using the DPPH (1,1-Diphenyl-2-Picrylhydrazyl) method. The results indicated that the four sample sources contained strong antioxidant activity, namely SO had an IC50 value of 86.22±23.6 μg/mL, SAP 86.58±30.7 μg/mL, SAC 86.10±23.4 μg/mL, and natural living SA 54.27±16.9 μg/mL. The transplanted S. carteri sea sponge has strong antioxidant activity and can be an alternative solution in terms of raw material availability for the development of potential pharmaceutical products.

Abstract

Jeroan ikan terkenal dengan konsentrasi enzim pencernaan yang tinggi, terutama proteinase misalnya tripsin. Karakteristik tripsin ikan dipengaruhi oleh jenis dan habitatnya. Jeroan ikan bandeng merupakan bagian dari saluran pencernaan ikan dan memiliki pH netral sehingga berpotensi menjadi sumber enzim khususnya tripsin. Jeroan ikan dapat menjadi alternatif pengganti enzim tripsin komersial yang berasal dari daging babi dan sapi. Tujuan penelitian ini adalah untuk menentukan karakteristik enzim tripsin dari jeroan bandeng dengan fraksi amonium sulfat. Proses penelitian diawali dengan ekstraksi enzim tripsin dari jeroan ikan bandeng, dilanjutkan dengan fraksinasi dengan amonium sulfat (0-80%). Suhu, pH, pengaruh ion logam, pengaruh NaCl terhadap aktivitas enzim, dan kinetika reaksi. Aktivitas enzim diukur menggunakan substrat N-α-benzoyl DL-arginine-p-nitroanilide (BAPNA). Hasil penelitian menunjukkan bahwa ekstrak kasar tripsin memiliki aktivitas sebesar 0,117 U/mL. Aktivitas enzim tripsin tertinggi pada fraksi amonium sulfat 20-30%, yaitu sebesar 0,295 U/mL. Enzim bekerja paling baik pada suhu 50°C dan pH 6. Aktivitas enzim tripsin pada ion NaCl, MnCl2, ZnCl2, CuCl2, dan CaCl2 mengalami penghambatan pada setiap jenis ion logam yang berbeda dan bersifat sebagai inhibitor. Aktivitas tripsin pada NaCl 5-30% meningkat pada rasio 10%, yaitu sebesar 0,249 U/mL. Nilai Vmax enzim ini adalah 0,285 mmol/s, dan nilai Km-nya adalah 0,374 mM.

Abstract

Ikan nila memiliki daging yang berwarna putih sehingga sangat baik digunakan dalam pembuatan surimi. Dari segi tekstur surimi ikan nila memiliki tekstur daging yang agak padat yang menjadikan hasil akhir kamabokonya agak keras. Oleh sebab itu, perlu dilakukan penambahan enzim pada surimi ikan nila sehingga mendapatkan tekstur kamaboko lebih lembut. Penelitian ini bertujuan untuk menganalisis karakteristik surimi ikan nila merah dengan penambahan enzim tripsin yang diekstrak dari usus ikan. Metode yang digunakan yaitu rancangan percobaan sederhana dengan penambahan enzim tripsin 0%, 1%, 2%, 3% dan 4%. Parameter yang diamati yaitu analisis proksimat, protein larut garam (PLG), pH, derajat putih, particle size analysis (PSA), texture profile analysis (TPA), water holding capacity (WHC) dan analisis sensori. Berdasarkan hasil analisis, penambahan enzim tripsin mampu meningkatkan karakteristik dari surimi ikan nila merah. Hal tersebut dapat dilihat dari hasil parameter yang diukur yaitu pada chewiness, hardness, gummines, uji lipat dan uji gigit yang menunjukkan bahwa tekstur dari surimi ikan nila menjadi lebih baik serta lebih lembut.

Abstract

High crab production encourages the implementation of strict standards for raw materials in canned crab products, particularly regarding labeling errors (i.e., mislabeling). DNA-based identification methods are accurate for species authentication; however, their application in the field remains limited because of laboratory equipment requirements. Therefore, the development of an on-site DNA-based analytical method is essential. This study aimed to design specific primers for Portunus pelagicus using the cytochrome c oxidase subunit I (COI) gene marker and to optimize the amplification time and temperature using the colorimetric loop-mediated isothermal amplification (LAMP) method with DNA isolates obtained from field-based extraction methods. The samples included fresh crabs, canned crab products, and non-target species (Charybdis feriata, Scylla serrata, and Podophthalmus vigil). The methods comprised target species confirmation, primer design and evaluation, and in vitro testing of the P. pelagicus primer set. Primer design resulted in a COI primer set within ideal parameter ranges and high specificity to P. pelagicus based on in-silico analysis. In vitro assays demonstrated that the primers successfully detected P. pelagicus DNA in both fresh and processed samples, with an optimal reaction time of 40–60 min at 65 °C. DNA isolates obtained using dipstick and direct lysis methods were also successfully amplified, indicating that these simple extraction techniques can be applied for on-site detection without loss of sensitivity. These findings demonstrate that the developed primer set is specific to P. pelagicus and that the optimized LAMP method has strong potential as a rapid and portable authentication system for crab raw materials in the field.
