Abstract

Produksi ikan asap di Kabupaten Tuban tumbuh secara signifikan karena lokasi penjualannya dekat lokasi wisata.Salah satu kawasan penghasil ikan asap ini beradadi Desa Karangsari, Tuban. Permasalahan keamanan pangan masih belum diperhatikan oleh para produsen ikan asap yang terlihat dari tampilan produk dalam keranjang secara terbuka dan pengemasan yang seadanya.Hal ini memengaruhi daya simpan produk yang hanya bertahan 1-2 hari. Oleh karena itu, kegiatan Doktor Mengabdi Universitas Brawijayabertujuan untuk meningkatkan kualitas produk ikan asap (mikrobiologi dan organoleptik) melalui induksi teknologi pengemasan vakum pada kelompok usaha pengasapan ikan. Metode pelaksanaan program ini adalah penyediaan teknologi pengemasan vakum, penyuluhan dan pelatihan pada mitra, serta pendampingan dan pengawasan. Adanya peningkatan status keamanan pangan produk ikan asap melalui penerapan teknologi pengemasan vakum yang disimpan dalam tiga kondisi (suhu ruang, dingin dan beku) ini dievaluasi melalui uji kualitasbaik secara mikrobiologimaupun organoleptik. Hasil uji total bakteri dan organoleptik menunjukkan pengemasan vakum dapat mempertahankan kualitas produk menjadi lebih baik dan lebih diterima konsumen. Kombinasi antara pengemasan vakum dan penyimpanan pada suhu dingin danbeku dapat menghambat pertumbuhan bakteri secara optimal. Teknologi pengemasan vakum untuk produk ikan asap dapat meningkatkan keamanan pangan dannilai jual.

Abstract

The aim of this research was to determine the effect of addition of pectin on characteristics of edible film from gelatin of red snapper (Lutjanus argentimacalatus) skin. Concentration of pectin used were 0%, 0.2%, 0.4%, 0.6%, 0.8% and 1%. Water content, thickness, tensile strength, elongation, and the transmition rate of the water vapor of edible film were analysed. Results from analysis of variance showed that different concentration of pectin significantly affected (P<0.05) on the water content, thickness, tensile strength, elongation, and transmition rate of water vapor of edible film. Therefore, it was suggested that addition of 0.6% of pectin showed a good quality of edible film and in accordance with the Japan International Standard (JIS).

Abstract

Doktor Mengabdi is a community service program at Universitas Brawijaya that is carried out to improve the welfare of the community. One of the efforts to help the community in completing its packaging is by providing assistance in the form of business equipment.  The purpose of this study is to investigate the difference in income of Small medium enterprises, Lele Sehat (IGF) which has been provided with the help of vacuum frying and spinners machines. Data retrieval was done through interviews and live observations. The data were tested for its normality and further analyzed using a paired t-test. The results showed that there was a difference in income of small-medium enterprises difference between before and after getting tool assistance. Prior income was IDR 3,007,000. It was increased to IDR. 4,097,000. This indicates that providing tools to small-medium enterprises will increase income and indirectly improve their welfare.

Abstract

Kemasan aktif memiliki fungsi yang lebih luas dalam menjaga kualitas suatu produk. Salah satu bahan baku kemasan aktif adalah gelatin dengan penambahan senyawa aktif. Penelitian ini dimaksudkan untuk mempelajari karakteristik dan bioaktivitas kemasan aktif dari gelatin ikan ayam-ayam (Abalistes stellaris) dengan penambahan ekstrak daun jeruju (Acanthus ilicifolius) sebesar 0%, 1%, 3%, 5% dan 7%. Kemasan aktif yang dihasilkan diamati karakteristik dan bioaktifitasnya yang meliputi ketebalan, kuat tarik, elongasi, transmisi uap air, kadar air, pH, aktivitas antioksidan dan aktivitas antibakteri. Hasil analisa sidik ragam menunjukkan penambahan ekstrak daun jeruju berpengaruh nyata (p<0,05) terhadap ketebalan, aktivitas antioksidan dan aktivitas antibakteri dari kemasan aktif berbahan gelatin yang dihasilkan, namun tidak berpengaruh nyata (p>0,05) terhadap kuat tarik, elongasi dan transmisi uap air. Kemasan aktif dari gelatin ikan ayam-ayam dengan penambahan ekstrak daun jeruju 7% memiliki ketebalan 143,01 µm, mampu menghambat 65,45% radikal bebas DPPH dan menghasilkan zona hambat bakteri berkisar antara 2,18-2,61 mm. Hasil penelitian ini menunjukkan gelatin yang terbuat dari kulit dan sisik ikan ayam-ayam dengan penambahan ekstrak daun jeruju memiliki potensi untuk dikembangkan menjadi kemasan aktif.  AbstractPackaging has an important function in maintaining the quality of product. Active packaging can be made from gelatin as basic material with addition of bioactive substances. The purpose of this research was to study the characteristics and bioactivity of active packaging made from staryy triggerfish (Abalistes stellaris) gelatin incorporated with 0, 1, 3, 5, and 7% of Acanthus ilicifolius leaf extract. The active pakaging was observed for its charateristics and bioactivity including thickness,  elongation, tensile strength, water vapor transmission, moisture content, pH, antioxidant activity, and antibacterial activity of active packaging. The statistical analyses showed that the addition of A. ilicifolius leaf extracts significantly affected (p<0.05) the thickness, antioxidant activity, and antibacterial activity of active packaging. Active packaging from fish gelatin containing 7% of A. ilicifolius extract with 143.01 µm thickness, was able to inhibit 65.45% DPPH free radical and resulted in 2.18-2.61 mm inhibition zone. It was suggested that starry triggerfish gelatin and A. ilicifolius leaf extract have a potential to be developed as active packaging.

Abstract

The aims of this study were to screen phytochemical and antibacterial potency of methanol extract of Excoecaria agallocha. The phytochemical screening used qualitative analysis and the antibacterial assay determined by diffusion method. The results showed that extracts of leaf, flower, bark, stem and root of Excoecaria agallocha, respectively, contained alkaloid, steroid, flavonoid, tannin, terpenoid and saponin. Methanol extract of all part of Excoecaria agallocha, except the root, inhibited the growth of gram positive bacteria (Staphylococcus aureus ATCC 9144), Meanwhile, the leaf of Excoecaria agallocha extract not only inhibited the growth of gram positive bacteria (Staphylococcus aureus ATCC 9144) but also the growth of gram negative bacteria (Escherichia coli ATCC 8739).

Abstract

Abstrak Enzim gelatinase merupakan enzim yang berperan penting pada sektor industri pangan dan non pangan. Sumber dari enzim gelatinase dapat berasal dari hewan, tumbuhan dan mikroorganisme. Enzim gelatinase termasuk enzim protease dan fungsi dari enzim gelatinase sendiri adalah sebagai pengurai gelatin. Penelitian ini bertujuan untuk mendapatkan bakteri penghasil enzim gelatinase dari endofit mangrove Sonneratia alba dan untuk mengetahui jenis spesies bakteri penghasil enzim gelatinase dari endofit mangrove Sonneratia alba dari pantai Sendang Biru. Metode penelitian yang digunakan dalam penelitian ini adalah deskriptif eksploratif yang dilakukan dalam dua tahapan. Tahap pertama yaitu isolasi dan skrining bakteri yang menghasilkan enzim gelatinase dari endofit mangrove Sonneratia alba. Tahap kedua yaitu identifikasi bakteri penghasil enzim gelatinase berdasarkan uji microbact system.  Hasil skrinning enzim menandakan bahwa isolat dari daun Sonneratia alba merupakan isolat bakteri yang mampu tumbuh dengan baik dan menghasilkan enzim gelatinase yang paling baik. Bakteri endofit daun pada uji microbact system diketahui sebagai jenis bakteri Paenibacillus alvei dengan hasil oksidase negatif sehingga termasuk bakteri Gram positif sehingga dilakukan dengan uji microbact 12A/E. Karakteristik bakteri mempunyai karakteristik berbentuk bulat dan berwarna ungu.. Kata kunci: gelatinase, enzim, Sonneratia alba, Malang, halal.

Abstract

The increase in koi fish cultivation can lead to increase risks and caused problems. The main problem is high disease attacks caused by the bacteria Edwardsiella tarda. One of the alternatives to control the disease is immunostimulant treatment with the use of maggot, Hermetia illucens Larvae, known as Black Soldier Fly (BSF). This study aims to report the effect of maggot as a substitute for koi fish feed formulations on the immune system of koi fish infected with E. tarda. This study used a descriptive method, by quantitative approach with the IL-6 and IFN-Ɣ expression through RT-PCR analysis and fish survival rate (%). Identification of E. tarda was carried out by the PCR test method at a molecular weight of 200 bp. The density of bacteria that caused the death of the 50% koi fish population (LD50) for 72 hours was 107 cells/ml. The highest IL-6 expression in the spleen was found in sample C of 28,844 and the lowest in-sample K (+) of 26,096. The highest IL-6 expression in the liver was in sample K (+) of 29,060 and the lowest was in sample D of 25,428. The highest IFN-Ɣ in the spleen was in treatment C, 28,844, and the lowest was in treatment D, 27,219. The highest IFN-Ɣ in liver was sample K (+) of 26,795, and the lowest in-sample C of 25,615. The survival rate of fish in this study reached 30%-100%. The optimal amount of substitution of maggot treatment is 75% (Treatment C) of fish feed.

Abstract

Hypercholesterolemia is a health disorder in which cholesterol in the blood increases beyond normal limits. This disorder chronically can cause atherosclerosis. Sargassum aquifolium contains bioactive that can prevent an increase in blood cholesterol levels. This study aimed to prevent the increasing cholesterol levels and changes in the blood vessel profile of rats fed an atherogenic diet by S. aquifolium extract. S. aquifolium was obtained from Ekas Bay. Rattus norvegicus 2-3 months ages used in this study. Normal rats were fed an atherogenic diet for four months to produce an animal hypercholesterolemia model. The phytochemical profile was screened based on the Harborne method. Identity of bioactive determined by HPLC-HRMS. The parameters observed were cholesterol levels and blood vessel profiles. The juice of S. aquifolium contained steroids, saponins and tannins and their bioactive showed anticholesterolemic. Rats given an atherogenic diet had increased cholesterol levels. Administration S. aquifolium juice twice a day can reduce cholesterol levels and prevent the thickening of the aorta and arteries. In conclusion, S. aquifolium juice contains bioactive compounds and administration of S. aquifolium juice twice a day prevents an increase in cholesterol and damage to the aortic and arterial profiles.

Abstract

Banyuurip Village has considerable aquaculture potential, one of which is the culture of green mussels. Abundant production and its low price encourage people to do green mussel products processing. In 2019, UMKM Berkah Mandiri was established to increase the selling value of green mussels by innovating several products. However, the production process is still experiencing ups and downs, coupled with inadequate equipment. In addition, the distribution permit (SPP-PIRT) for the products have not been obtained. Therefore, the aim of the Doctoral community service program was to increase the productivity of UMKM Berkah Mandiri through a series of marketing strategy training activities, registration of NIB and SPP-PIRT and procurement of new production and packaging equipment. At the end of the Doctoral community service program, UMKM Berkah Mandiri has successfully obtained the SPP-IRT certificate and has better understanding of good manufacturing practice of products from green mussels. In addition, the crackers have been tested its nutritional content using a proximate test. The results showed that the protein, moisture, fat, ash and carbohydrate contents of Kopang cracker was 5.30%, 13.83%, 0.69%, 2.99% and 77.19%, respectively. The nutritional content list is used to improve product packaging in accordance with BPOM packaging standards.

HighlightsThe halophilic bacteria were isolated from salt-pond soil.Isolate C2 was identified as the best protease producer.Isolate C2 was identified as B. amyloliquefaciens strain UBCAbstractProtease is an important enzyme for various industries, such as pharmaceuticals, leather production, meat processing, protein hydrolyzate, food products, and waste processing industries. This study aimed to isolate and identify protease-producing bacteria isolated from salt-pond soil at the Tuban Regency, East Java, Indonesia. Bacterial communities in the soil samples were firstly isolated from the soil samples by culture-dependent technique on Luria Bertani agar enriched with 5% NaCl. Thereafter, bacterial colonies that grew on the media were purified and screened for their protease production using a skim-milk agar. The bacterial colony which produced protease was further identified using phenotypic (gram staining) and genotypic assays (the 16S rDNA sequence). The result showed that one isolate out of six (isolate C2) obtained from the soil sample was observed to produce a protease enzyme. Based on its 16S rDNA sequence, the isolate was identified as Bacillus amyloliquefaciens strain UBC. These results suggest that B. Amyloliquefaciens strain UBC is a salt-tolerant bacterium (halophilic bacteria) which has the potential to be further developed for protease-producing biological agents.
