Abstract

INTRODUCTION: UV-B exposure to the skin may produce ROS, decreasing collagen density and increasing MMP-1 expression. Longan leaf extract cream contains antioxidant and is expected to prevent decrease in collagen density and MMP-1 expression increase.OBJECTIVE: The research aims at administering longan leaf extract cream to increase collagen density and decrease MMP-1 expression in BALB/c mice exposed to UV-B rays.METHODS: This experimental research employed a Post Test Only Controlled Group Design. The subjects were 24 BALB/c mice, which were randomly divided into 4 groups. The Control Group (Ctr-G)was only administered with basic cream and exposed to UV-B rays, while Groups DLS-3, DLS-6, and DLS-13were respectively administered with longan leaf extract cream at doses 3.25%, 6.5% and 13% and exposed to UV-B rays for 4 weeks at a dose of 1 MED. On day 29, a tissue excision biopsy was performed to determine the collagen density and MMP-1 expression. The collagen density preparations were made using Masson’s Trichome staining and MMP-1 preparations were made using immunohistochemical staining.RESULTS: Kruskalwallis analysis showedstatistically no significant differences in collagen density and MMP-1 expression between groups(p value> 0.05).CONCLUSION: Administration of longan leaf extract cream at doses of 3.25%, 6.5% and 13% had proven unable to increase collagen densitynor decrease the MMP-1 expression significantly after UVB exposure.

Abstract

BACKGROUND: A high fat diet, accompanied by high frequency eating, results in hyperlipidemia. One prevention for hyperlipidemia is administration of celery extract.OBJECTIVE: to explore the effect of celery extract on the level of 8-OHdG (8 -Hydroxy-2’-Deoxy Guanosine) and the number of foam cell.METHODS: Experimental studies with post-test only control group design used Wistar rats, administered with adrenaline and high-fat diet forhyperlipidemic induction.Total of 35 rats were divided into 5 groups: negative controls (Neg-G); positive control treated with simvastatin (Nor-G); ES25-G,ES50-G, and ES75-G group were treated with 25 mg/200 g BW, 50 mg/200 g BW, and 75 mg/200 g BW celery extract, respectively. The treatment was given for 30 days. The 8-OHdG level was determined by ELISA, whereas the number of foam cells was determined by histopathologic preparations with HE staining. Data analysis was conducted by Kruskal-Wallis and Mann-Whitney.RESULTS: Mann-Whitney analysis showed levels of 8-OHdG in ES-25-G 12.13±0.21, ES-50-G 7.23±0.25, ES-75-G 4.41±0.45 significantly lowerthan the Nor-G 14.30±0.66, respectively p<0.001. The number of foam cells in the ES-25-G was 7.57±0.53, ES-50-G 6.57±0.79, ES-75-G 3.43±0.53,significantly lower than the Neg-G 13.57±1.27, respectively p<0.001.CONCLUSION: Celery extract capable of decreasing levels of 8-OHdG and the number of foam cells in Wistar rats induced with adrenaline and high-fatdiet.

Abstract

Introduction: Continuous Multi Stage Countercurrent Extraction Process (CMCE) propolis extract containing Caffeic Acid Phenethyl Ester (CAPE) may inhibit the formation of lipid peroxidation. However, it is still unclear whether or not CMCE propolis is able to increase SOD activity and reduce acute hepatocytes damages induced by CCl4. Objective: To evaluate the influence of CMCE propolis extract administration in increasing the SOD activity and reducing the hepatocytes fibrosis of male Wistar rats induced with CCl4.Method: an experimental research with a post-test only control group design. Twenty eight male Wistar rats were divided into four groups. Group C-N was only induced with CCl4, while group CM-3, CM-7, and CM-14 were induced with CCl4 and received CMCE propolis extract respectively with 3.6 mg/200 g BW, 7.2 mg/200 g BW, and 14.4 mg/200 g BW. CMCE propolis was administered for 14 days and then CCl4 was administered on the last day. SOD activity was measured using the colorimetric method, while the hepatocytes fibrosis degree was assessed using Image J Software method with NASH score modification.Results: Post Hoc LSD analysis indicated that SOD activity in group CM-3 (51.67 ± 4.20), CM-7 (70.21 ± 6.26), CM-14 (80.85 ± 4.59) was lower than that in group C-N (23.70 ± 5.12) with P <0.05. Meanwhile, the Mann-Whitney U test showed that the hepatocytes fibrosis degree in group CM-3 (0.85 ± 0.69), CM-7 (1.14 ± 0.37), and CM-14 (0.71 ± 0.48) was significantly lower than that in group C-N (2.28 ± 1.25) with p<0.05.Conclusion: The CMCE propolis extract administration was able to increase the SOD activity and reduce the hepatocytes fibrosis degree of male Wistar rats induced with CCl4

Abstract

Introduction: Strenuous physical activities cause an increase in free radicals, physical stress, psychological stress and decreased immunity. Psychological stress affects the hypothalamic-pituitary-adrenal (HPA) axis leading to increased levels of cortisol and blood cell apoptosis. However, the studies on the effect of strenuous physical activities on cortisol levels and leukocyte count showed inconsistent findings. Objectives: To examine the effect of strenuous exercise on cortisol levels and leukocytes count.Methods: In this study with post test only control group design, 15 male BALB/c mice were randomly assinged to the following 3 groups: control (non-exercised) group (G-0), once strenouse physical activity (G-1), 3 times strenuous physical activity (G-2). Cortisol levels were evaluated by ELISA.White blood count was determined. Data were analyzed by one way ANOVA test followed by Post Hoc and Pearson, with a confidence level of 95%.Results: The mean cortisol level in group G-0, G-1, G-2 were 11.86 ± 2.10μg/dL, 32.00 ± 0.86μg/dL, 44.58 ± 1.74μg/dL respectively. The number of leukocytes in G-0, G-1, G-2 were 6180±540.37/mm3, 4650±217.94/mm3, 4180±130.38/mm3 respectively. One way ANOVA showed a significant difference between groups (p <0.001). Post Hoc Test showed that the levels cortisol in G-1 and G-2 were significantly higher than thatof G-0 (p <0.001). The number of leukocyte in G-1 and G-2 was lower than that of G-0 (p <0.001). There was a significant negative correlation between cortisol levels and the number of leukocytes (r = -20.14, p <0.001).Conclusions: Strenuous physical activity increasescortisol levels and decrease the number of leukocytes in BALB /c mice.

Abstract

INTRODUCTION: UVB radiation on skin may increase reactive oxygen species (ROS) level and cause hyperpigmentation. Polyphenol, an antioxidant contained in tomato, is able to quench ROS and inhibit tyrosinase activity.OBJECTIVE: This study aims to evaluate the effect of topical tomato lotion on Malondyaldehide (MDA) level and the amount of melanin in skin following UVB radiation.METHODS: In this experimental study, 28 female Balb/c mice are randomly divided into 4 groups: control group (C-G) are given with base lotion, and Lycopersicum groups 0.14 (L-014), 0.7 (L-07) and 1.4 (L-14) are given with tomato lotion of concentration of 0.14%, 0.7% and 1.4% daily, respectively. All of the mice are exposed to UVB at 1MED once every two days for two weeks before lotion application. On day 16, all mice are terminated, and their skin tissues are prepared for MDA level and the amount of melanin assessment.RESULTS: The ANOVA statistical analysis shows a significant difference in the MDA level and the number of melanin of the groups, (p<0.05). The post-hoc LSD analysis shows that the level of MDA in L-14 (1.13) is significantly lower than that in L-07 (2.21), L-014 (2.47), and C-G (5.61), p<0.005. The amount of melanin in L-14 (10.00) is significantly lower than that in L-014 (23.50) and C-G (33.67), p<0.05. The amount of melanin in L-07 (13.00) is not significantly higher than that in L-14, p>0.05. The correlation between MDA level and the amount of melanin shows a strongly positive correlation value (r2) = 0.648CONCLUSION: Topical administration of tomato extract lotion significantly decreases the MDA level and the number of melanin in the mice’s skin following UVB radiation.

Abstract

Background: During exercise, oxygen decreases due to acidosis leading the production acid urine by kidney. Oxygenated water having capability to absorb more oxygen compared to mineral water is used to meet the oxygen need during exercise. This study investigated the effect of oxygenated water and mineral water on the O2 saturation and urine pH.Design and method: In this crossover study, 46 subjects were randomly assigned to either oxygenated water or mineral water for 24 hours followed by a crossover to the other regimen for an additional 24 hours of treatment. After 100 m sprint running for 20 minutes, the oxygen saturation and urine pH of the subjects were assessed.Result:The mean oxygen saturation before and after the treatment of oxygenated water were 96.78±1.32 and 97.61±0.93 respectively. The mean oxygen saturation before and after the mineral water treatment was 97.35±0.85 and 97.01±1.04 respectively. The urine pH after the administration of oxygenated water and mineral water were 6.643±0.69 and 6.585±0,58 respectively. Wilcoxon test resulted in no significant difference in pH after the treatment of oxygenated water and mineral water (p=0.498). Urine pH after the treatment of oxygenated water was found to be higher compared to that of mineral water.Conclusion: the oxygenated water increases the O2 saturation and urine pH compared to mineral water (Sains Medika, 3(2):162-167).

Abstract

Background: The use of probiotics to treat colitis is unsatisfactory, leading to a need for the combination with purple sweet potatoes (Ipomoea Batatas) and zinc. Objectives: This study aims to determine the effect of the combination of probiotics, purple sweet potatoes, and zinc on epithelial thickness.Methods: This was a post-test-only randomized controlled group experimental study using 30 mice divided into six groups, including KN, K-, K+, P1, P2, and P3. All groups except KN were induced with 4% acetic acid on day 1, then each received treatment for five days. KN, K+, P1, and P2 groups were given distilled water, sulfasalazine 1.3 mg/20 g BW, probiotics 0.078 mg/20 gBW + purple sweet potatoes 0.00169 g/20 gBW, and probiotics 0.078 mg/20 gBW + zinc 0.052 mg/20 gBW, respectively. Meanwhile, P3 received a combination of probiotics 0.078 mg/20 gBW + purple sweet potatoes 0.00169 g/20 gBW + zinc 0.0052 mg/20 gBW. Histological preparations were stained with Hematoxylin-Eosin and examined under 400× magnification across 10 fields of view. Results: The average epithelial thicknesses were KN (75.48±5.09), K− (28.02±5.60), K+ (69.1±13.82), P1 (47.05±9.41), P2 (48.6±9.73), and P3 (71.57±14.31). One-way analysis of variance (ANOVA) showed a significant difference among the groups with a p-value <0.001. Post Hoc LSD test identified significant differences (p-value<0.05) among all group pairs except between K+ and P3 (p-value=0.134) as well as P1 and P2 (p-value=0.349), but P3 had the highest average thickness value (71.57±14.31).Conclusions: The combination of probiotics, purple sweet potatoes, and zinc had an effect on intestinal epithelial thickness in male BALB/c mice with ulcerative colitis.

Abstract

Kanker payudara merupakan penyebab kematian kedua setelah kanker leher rahim pada wanita. Pengobatan kanker sampai sekarang masih belum berhasil dengan baik. Daun sirsak merupakan obat tradisional yang secara empirik dipakai masyarakat Indonesia sebagai obat antiinflamasi dan antikanker. Kandungan aktif acetogenin pada daun sirsak diketahui dapat menginduksi apoptosis dengan meningkatkan aktivitas caspase-3 dan menghambat proliferasi pada kultur sel kanker. Dengan tujuan untuk mengetahui potensi antikanker ekstrak daun sirsak, penelitian ini dilakukan dengan rancangan post test randomized control group design, menggunakan 24 ekor mencit C3H yang telah diinokulasi bubur tumor jenis adenokasrsinoma mamma dan telah tumbuh tumor. Mencit kemudian dikelompokkan secara random menjadi kelompok 1 (kontrol), 2 dan 3, berturut-turut mendapatkan aquades, ekstrak daun sirsak dosis 10 mg/kg dan 20 mg/kg. Perlakuan diberikan peroral selama 30 hari. Selama perlakuan, volume tumor diamati tiap 3 hari untuk mengetahui penghambatan per-kembangan tumor payudara. Pada hari ke-31, jaringan tumor diambil dan dibuat preparat histopatologi untuk pengecatan imunohistokimia terhadap ekspresi protein Ki-67 dan pengecatan Tunnel, berturut-turut untuk mengetahui tingkat proliferasi serta jumlah sel kanker yang mengalami apoptosis. Data di uji dengan uji One Way Anova. Hasil penelitian menunjukkan bahwa ekstrak daun sirsak mampu memperlambat pertumbuhan tumor payudara tetapi tidak mengurangi besar tumor akhir. Disamping itu, ekstrak daun sirsak mampu meng-hambat proliferasi sel melalui hambatan ekspresi Ki-67 dan mampu meningkatkan apoptosis. Abstract

Perbedaan mekanisme kerja obat Karbamazepin, Gabapentin, dan Amitriptilin dapat mempengaruhi efektivitasnya dalam mengurangi nyeri pada polineuropati diabetik. Untuk membantu pemilihan terapi yang paling tepat untuk mengelola nyeri tersebut, maka perlu dilakukan penelitian ini.  Penelitian dilakukan secara eksperimental dengan  rancangan pretest-posttest group design.  Pengukuran respon nyeri menggunakan Visual Analogue Scale (VAS) dilakukan sebelum dan sesudah perlakuan dengan Gabapentin, Carbamazepine, dan Amitriptyline. Perbedaan respon nyeri sebelum-sesudah dan antar perlakuan dianalisis dengan uji Mann-Whitney dan uji T-independent. Hasil penelitian menunjukkan bahwa ketiga obat tersebut  efektif dalam mengurangi respon nyeri pada pasien polineuropati diabetik, namun Amitriptyline lebih efektif pada pasien nyeri ringan sampai sedang dibandingkan dengan Gabapentin. Temuan ini memberikan informasi berharga bagi penyedia layanan kesehatan dalam memilih pengobatan yang paling efektif untuk pasien dengan nyeri polineuropati diabetik.

Abstract

Background: Islam has clear rules about halal products. For a Muslim, consuming halal products is a form of devotion to Allah SWT. The concept of halal and haram is the main point in people's consumption patterns. Like consuming food, consuming medicine must also be halal. This study was designed to learn about the proportion of halal-labeled drugs in Apotek Enggal Saras Ungaran and the influence of halal labels on consumer decisions to choose medicines. Methods: The research method uses a direct survey. The questionnaire was addressed to buyers at the Apotek Enggal Saras Ungaran who bought halal-labeled drugs and those not labeled as halal to determine the respondent's interest in halal-labeled products. Data collection is done by grouping the data according to the criteria and calculating the percentage of each group. Results: The use of halal-labeled drugs and drugs not labeled halal are 60% and 40%, respectively. Conclusion: Muslim consumers prefer halal products to those that are not labeled halal. Factors that determine community decision-making in choosing halal-labeled drugs at Apotek Enggal Saras Ungaran include knowledge of halal labels, price affordability, product evaluation, fulfillment of halal product needs, and alternatives in decisions to use halal drugs.
