Abstract

Media pembelajaran yang digunakan di SMAS Kemala Bhayangkari pada materi sel menarik tetapi masih tidak interaktif. Penelitian ini bertujuan untuk mengembangkan media pembelajaran interaktif berbasis flashmateri sel yang layak digunakan. Pengembangan media pembelajaran interaktif berbasis flash ini menggunakan metode penelitian dan pengembangan (R&D) model 4-D, yaitu tahap Define (Pendefinisian), Design (Perancangan), Develop (Pengembangan) dan Disseminate (Penyebaran). Pada tahap Disseminate (Penyebaran) tidak dilakukan. Hasil penelitian menunjukkan kevalidan pada aspek materi sebesar 83,33% (sangat valid), bahasa sebesar 85 % (sangat valid), dan aspek media sebesar 80,42 % (sangat valid). Berdasarkan uji coba skala kecil dan uji coba skala besar, respon siswa dan respon guru terhadap media pembelajaran interaktif berbasis flash baik, sehingga media dapat digunakan dalam proses pembelajaran. Keefektifan media pembelajaran diperoleh berdasarkan nilai postest. Ketuntasan klasikal kelas 91,42%, yang menunjukkan bahwa media pembelajaran interaktif berbasis flash materi sel dinyatakan efektif. Berdasarkan hasil yang diperoleh, menunjukkan bahwa media pembelajaran interaktif berbasis flash layak digunakan untuk siswa dalam pembelajaran biologi materi sel.

Abstract

Three-dimensional (3D) culture is a technique commonly utilized in bioprocessing and biomedical research. Exosomes have been investigated as carriers for medications in numerous studies employing 3D culture methodologies. The objective of this research is to employ 3D cell culture for the isolation and treatment of exosomes targeting colon cancer cells. The isolation of exosomes obtained from HEK293 cells was conducted through the ultracentrifugation technique. Subsequently, exosome treatment was administered to WiDr cells at concentrations of 3.5 µg/ml, 7 µg/ml, and 14 µg/ml.The validation of molecular markers of exosomes (CD9 and CD81), along with BAX, BCL-2, and CD133, was performed using qRT-PCR. The findings revealed the successful isolation of exosomes derived from HEK293 cells, which exhibited the expression of markers CD9 and CD81. Furthermore, the expression of BAX and BCL-2 indicated the potential of exosomes to induce apoptosis, while the expression level of CD133 decreased with treatment at varying concentrations. These results suggest that exosome treatment has the capability to impede the proliferation of WiDr cells and reduce the expression of CD133, thereby signifying the potential application of exosomes as an in-vitro model for investigating cancer therapy in the future.

Abstract

Plastic waste is a serious environmental problem that requires immediate handling, especially in educational environments. SD Negeri 22 Seladu faces the challenge of managing plastic waste, especially used bottles, which have not been utilized optimally. This condition shows the need to increase students' awareness of plastic waste management and the provision of environmentally based learning media. This service activity aims to minimize plastic waste, increase students' understanding of waste management, and create learning media in the form of aquariums from used bottles. The method used includes preparation stages, delivery of materials, and practice of making aquariums from used plastic bottles. Activities are carried out with a participatory approach to actively involve students. The results of the activity show that students are able to understand the concept of recycling, increase creativity, and have a higher awareness of the importance of protecting the environment. This program provides significant educational and environmental benefits. By using used bottles as a learning medium, this activity can be used as a model for environmental education in other schools to support sustainable plastic waste management.
