Abstract

Aim Lipoaspirate, a wasted by product from liposuction procedure recently has been shown to contain abundant mesenchymal stem cells (MSCs). MSCs have been studied in many research areas to regenerate many cell lineages including, myogenic, cardiomyogenic, and angiogenic lineages. The large quantity of MSCs in lipoaspirate, makes it an attractive source for stem cells used in research and clinical applications. A simplified method which is suitable to be performed in a basic laboratory will facilitate development of stem cell research in developing countries. Therefore the outcomes from this study are expected to encourage the progress of stem cell research in Indonesia. Methods Lipoaspirate was digested using collagenase type I, followed by a basic filtration method. Purification of MSCs was done by cell culture for 2-3 days followed by supernatant removal. To confirm the homogenous population of MSCs, an analysis using flowcytometry was performed based on the MSCs minimal criteria developed by Mesenchymal and Tissue Stem Cell Committee of the International Society of Cell Therapy.Resuts MSCs were able to be obtained at 16.41 ± 8.22 x 108 cells per 120 ml lipoaspirate. The cultured cells showed fibroblastic morphology which is characteristic for MSCs and were able to be purified from non-MSCs cells. This was confirmed by flowcytometry assay showing expression of CD105 and the absence of HLA-Class II, CD 45, CD 34, CD14, and CD19.Conclusions This study has shown that it was feasible to isolate messenchymal stem cell from human lipoaspirate. The procedure was practicable to be performed within a basic laboratory. (Med J Indones 2009; 18: 91-6)Keywords: mesenchymal stem cell, lipoaspirate, stem cell isolation technique

Abstract

Central obesity has become a public health problem which is currently rapidly increasing throughout the world, including in developing countries like Indonesia. The World Health Organization states that central obesity is a major risk factor for various cardiometabolic diseases, such as diabetes and hypertension. Inter-city bus drivers are high-risk occupations of cardiometabolic disease, this is due to unhealthy lifestyles and work patterns such as high consumption of fatty foods, smoking habits, lack of physical activity, long driving times and irregular sleep patterns. This study aims to determine the prevalence of central obesity based on waist circumference in inter-city bus drivers. This research is a descriptive cross sectional study. Data obtained from measurements of body weight, height and waist circumference, as well as calculation of body mass index (BMI) on 176 subjects obtained through consecutive sampling method. Obesity is said if BMI ≥25 according to WHO criteria for Asia-Pacific and central obesity if waist circumference> 90 cm for men and> 80 cm for women. In this study all subjects were men with an average age of 42.45  10.50 years. Based on the calculation of BMI there are 54.3% of subjects obese. Through measurement of waist circumference there are 50.6% of subjects with central obesity. Providing education is very important so that subjects can make lifestyle changes for the better so as to prevent cardiometabolic disease in inter-city bus drivers. AbstrakObesitas sentral telah menjadi masalah kesehatan masyarakat yang saat ini sangat meningkat pesat di seluruh dunia, termasuk di negara berkembang seperti Indonesia. Abstract

Sarcopenia is a medical condition characterized by a progressive decrease in skeletal muscle mass as well as muscle strength. Sarcopenia in the elderly causes an increased risk of falls, fractures and mortality. The main risk factors include advanced age, lack of physical activity, malnutrition, and chronic disease. Deficiency of albumin, a protein important for maintaining osmotic pressure and nutritional status, is often associated with malnutrition and sarkopenia. This community service activity was attended by 93 elderly people with an average age of 74 years. The results showed that the majority of participants had normal albumin levels, but there was a significant proportion who experienced hypoalbuminemia, more commonly found in men than women. Education and screening are carried out to increase awareness regarding the importance of albumin intake in preventing sarkopenia. A holistic and integrated nutritional approach is needed in the management of sarcopenia. Appropriate nutritional interventions, including increasing protein intake from foods such as snakehead fish, can increase serum albumin levels and prevent muscle mass loss. Collaboration between nutritionists, physicians, and other health professionals is essential to designing and implementing a comprehensive treatment plan. It is hoped that with appropriate monitoring and treatment, the quality of life of elderly people with sarkopenia can be improved. Keywords: albumin, education, hypoalbuminemia, elderly, sarcopenia

Abstract

The risk of sarcopenia and metabolic complications in the elderly can be caused by the aging process and decreased muscle mass in the elderly group. Sarcopenia and obesity in the elderly also increase the risk of cardiovascular disease, diabetes, and physical disability, which have the potential to reduce quality of life and increase morbidity and mortality. The purpose of this community service activity is to screen protein levels and lipid profiles in the elderly group in order to detect the risk of sarcopenia and dyslipidemia early. This activity is carried out using the Plan-Do-Check-Action (PDCA) method which includes planning, implementation, supervision, and follow-up. The community service activity was carried out at Panti Bina Bhakti, South Tangerang, involving 93 elderly participants aged >65 years. The results of the activity showed that 35 people (37.63%), 9 people (9.68%), 35 people (37.63%), and 7 people (7.53%) each had abnormal lipid profiles for HDL, Cholesterol, LDL, and Triglycerides. While 24 people (25.81%) had abnormal albumin levels. Therefore, it is necessary to monitor protein levels and lipid profiles regularly as well as early intervention through a healthy diet, regular exercise, and education for the elderly to maintain muscle mass and muscle strength.

Abstract

Aging leads to changes in body composition influenced by physical, psychological, and social factors. Understanding the relationship between fat distribution and nutritional status, particularly in older adults, is crucial for targeted interventions. This study examines the impact of body fat composition on nutritional assessment, particularly its influence on MNA scores in older adults. It aims to provide insights into how fat distribution and related metabolic changes affect nutritional status and inform targeted interventions for aging populations. The sample consisted of 31 elderly woman subjects selected through purposive sampling in a cross-sectional design from St. Francis of Assisi Catholic Church. Their nutritional status was determined using the MNA score, and the body composition measures were total body fat and skeletal muscle measured by the Omron Karada Scan HBF 375. SPSS analyzed statistical differences between body composition and MNA scores. This study found significant correlations between the Mini Nutritional Assessment scores and body composition parameters, including BMI (r = 0.473, p = 0.007), total subcutaneous fat (r = 0.468, p = 0.008), and visceral fat (r = 0.457, p = 0.010). Arm skeletal muscle showed a negative correlation (r = -0.486, p = 0.006). These results emphasize that fat composition is a critical determinant of nutritional status in the elderly. The study found a significant correlation between fat composition as a key determinant of nutritional status in the elderly, with significant correlations observed between MNA scores and various body fat parameters. Abstract

Imbalance of body lipid levels, especially total cholesterol and HDL, is a major risk factor for cardiovascular disorders. A simple lipid profile examination can be used to detect potential atherogenic dyslipidemia that has not shown clinical symptoms. Total cholesterol and HDL levels were examined in 104 participants using the Point-of-Care Testing (POCT) technique. Education was provided individually using leaflets, which contained information on the effects of saturated fat and trans fat consumption, and the importance of unsaturated fat and fiber intake on lipid profiles. Most participants had total cholesterol levels within normal limits. However, more than half of the population showed low to very low HDL levels, indicating hidden metabolic risks. Dietary education was provided in a targeted manner according to the results of each participant's examination. Total cholesterol and HDL examinations were used to assess the risk of lipid disorders early on. Nutrition education based on examination results is useful in raising awareness of the importance of regulating diet to reduce the risk of atherosclerosis and cardiovascular complications.
