Abstract

ABSTRACT The attack of Aeromonas hydrophila bacteria can cause mortality in goldfish approximately 100%. The controlling of this bacterial attack can be done through increased fish immunity. Honey is one of the natural ingredients that increases body immune system. This study aimed to determine the effective dose of honey supplemented in feed to increase goldfish fingerling resistance for disease prevention. Fish used in this study were goldfish fingerlings with 3.5 g average weight. This study was done using experimental complete randomized design method with five treatments and three replications. Treatments given were honey supplementation in feed with 0 ml/kg (A) as control treatment, 150 ml/kg (B), 200 ml/kg (C), 250 ml/kg (D), and 300 ml/kg (E). The result showed that honey supplementation in feed was effective to improve goldfish fingerlings resistance against Aeromonas hydrophila bacterial attack. The supplementation of honey in feed with 200 ml/kg was the best treatment for inducing goldfish fingerlings against A. hydrophila. This was proven by the increased white blood cells (leucocytes) (27.84 ± 5.07%) followed with no apparent clinical symptoms after attacked by A.hydrophila, such as hemorrhage, necrosis, exophthalmia or dropsy, besides showing the highest survival rate with 73.33 ± 11.5%. Keywords : Aeromonas hydophila, goldfish, honey, leucocyte, resistance ABSTRAK Serangan bakteri Aeromonas hydrophila dapat menyebabkan kematian ikan mas koki hingga mencapai 100%. Abstract

The male broodstock of Transgenic Mutiara catfish (P) (containing Clarias gariepinus Growth Hormone, CgGH) is a fish who show fast growth. Propagation of the Transgenic Mutiara catfish F1 and F2 progeny involves the crossing of broodstock P with Non Transgenic Mutiara catfish. New genetic variations (polymorphisms) can be generated from crosses broodstock P and F1 which lead to the fast growth inheritance of transgenic fish. Phylogenetic relationship analysis showed distance and genetic variation in broodstock P, F1, F2 and Sangkuriang catfish (as controls) using the RAPD-PCR method (Random Amplified Polymorphic DNA-Polymerase Chain Reaction) as the research purposes. The results showed that OPA-03 (5'-AGTCAGCCAC-3 ') primer was selective for determining phylogenetic among catfish samples. The closest genetic similarity index originates from the female broodstock of Non Transgenic Mutiara catfish (P) (93%), while the farthest originates from the male broodstock of Mutiara Transgenic catfish (P) (51%) with F1 dan F2. Closely related broodstock catfish is not recommended to be crossed.

Abstract

Mangrove forests, as coastal ecosystems rich in biodiversity, play an important role in ecology and the fisheries sector. Mangroves not only function as coastal protectors and habitats for aquatic organisms but also produce bioactive compounds that have potential as antibacterial, antioxidant, antifungal, and antiviral agents. Research on mangrove plants from the Rhizophoraceae family, such as Bruguiera, Kandelia, Ceriops, and Rhizophora, has shown that compounds like flavonoids, tannins, alkaloids, and saponins have significant biological activities. Flavonoids act as antimicrobials, while tannins inhibit bacterial growth by precipitating proteins. Alkaloids disrupt bacterial cell walls and have potential in combating cancer cells. Studies also reveal significant antioxidant activity, with some mangrove extracts showing great potential in combating free radicals. Moreover, these compounds offer potential in reducing reliance on synthetic antibiotics for the treatment of cultured fish diseases and provide opportunities for the development of natural-based health products. This review aims to integrate recent findings on the bioactivity of mangrove plants and propose innovative applications of bioactive compounds in fisheries and conservation with a focus on ecosystem sustainability and optimal use of natural resources.

Abstract

The prevention of fish infections caused by Aeromonas sp. bacteria can be achieved using Lactobacillus sp. bacteria. This study aims to identify and analyse the effect of Lactobacillus sp. using the microencapsulation method to treat catfish infected with Aeromonas sp. The identification of Lactobacillus sp. bacteria was conducted through microbiological and molecular tests. After obtaining the test results, the research continued with the application of microencapsulation using the emulsion technique. The impact of this microencapsulation was evaluated on catfish through an experimental approach employing a completely randomised design (CRD), with 10 catfish tested in each experimental aquarium. The study included five treatments, designated as A, B, C, D, and E. Treatment A served as the control, without the administration of Lactobacillus. Treatment B involved the application of Lactobacillus without microencapsulation. Treatments C, D, and E utilised varying doses of Lactobacillus at 0.003, 0.005, and 0.007 g/L, respectively. Each treatment was conducted in triplicate. Microbiological and molecular testing identified encapsulated Lactobacillus sp., specifically Lactobacillus helveticus and Lactobacillus johnsonii. The results indicated that treatment D, comprising a dose of 0.005 g/L of Lactobacillus helveticus and Lactobacillus johnsonii, was the most effective in promoting the healing of catfish infected with Aeromonas hydrophila bacteria. Keywords: Completely randomized design, bacterial infection mitigation, probiotic bacteria, emulsion technique.

Abstract

The Fish Auction Place (FAP) is a facility that supports fishermen in distributing their catches. Muara Angke FAP is one of the active auction places in Jakarta. Fish is a perishable commodity that requires proper handling to maintain its quality, including hygiene and sanitation facilities, especially to preserve the quality of little tuna (Auxis rochei). This study aims to analyze the sanitation and hygiene conditions at Muara Angke FAP based on Minister of Marine Affairs and Fisheries Decree No. 1 of 2007 (KEP. KP 01/MEN/2007), assess the organoleptic quality of little tuna, and analyze the relationship between sanitation-hygiene and tuna quality. The research was conducted from January to February 2025 using a case study method. Sanitation and hygiene data were obtained using purposive sampling, while little tuna samples for organoleptic and pH tests were taken using simple random sampling. Spearman correlation was used to analyze the relationship between FAP sanitation-hygiene and tuna quality. The results show that most sanitation and hygiene aspects meet the standards, although some deficiencies remain in facilities such as handwashing stations, toilets, environmental cleanliness, and warning signs. The organoleptic test yielded a final score of 7, indicating that frozen little tuna at Muara Angke FAP is still considered fresh. The pH ranged from 5.3 to 5.7, also indicating freshness. Spearman correlation analysis showed a weak positive relationship (r= 0.281) with no significant correlation (p= 0.231) between sanitation-hygiene and the quality of little tuna.
