Abstract

Penelitian ini dilaksanakan untuk menguji kapasitas dan stabilitas beberapa adsorben alami dalam mengikat aflatoksin B1 (AFB1) dengan metode in vitro yang melibatkan kondisi di dalam rumen. Percobaan in vitro dilakukan dengan menggunakan 3 macam adsorben alam (bentonit, zeolit dan karbon aktif) dan satu adsorben produk komersial (CAA). Pengujian dilakukan pada 3 macam medium in vitro (aquades steril, cairan rumen dan cairan rumen steril) dan 2 rasio AFB1:adsorben (1:1000 dan 1:10.000 yang setara dengan 1000 mg AFB1-contaminated feed:1 mg adsorben dan 100 mg AFB1-contaminated feed:1 mg adsorben). Inkubasi dilakukan selama 2 jam dengan penggojokan 70 rpm pada suhu 38,5oC menggunakan shaking incubator. Medium kemudian disentrifugasi selama 15 menit pada 3500 g. Supernatan diambil untuk dianalisis kadar AFB1 yang tidak terikat oleh adsorben, sehingga persentase AFB1 terikat dapat dihitung sebagai peubah kapasitas pengikatan. Presipitat selanjutnya diresuspensi dan diinkubasi kembali. Supernatan yang diperoleh setelah sentrifugasi 3500 g selama 15 menit dianalisis kandungan AFB1-nya untuk mendapatkan persentase AFB1 yang terlepas dari ikatan dengan adsorben untuk mendapatkan data stabilitas pengikatan. Data dianalisis variansi dengan prosedur general linear model rancangan acak lengkap pola faktorial menggunakan SPSS versi 17.0. Hasil memperlihatkan bahwa bentonit memiliki kapasitas pengikatan AFB1 tertinggi (77,54%) dengan medium aquades. Stabilitas pengikatan tertinggi ditunjukkan oleh CAA (99,78%) yang tidak berbeda dengan stabilitas bentonit (99,38%). Pengikatan AFB1 secara nyata (P<0,05) dipengaruhi oleh pH medium dengan kapasitas tertinggi diperoleh pada pH medium kurang dari 5,0. Berdasarkan peubah kapasitas dan stabilitas pengikatan serta pH optimum pengikatan dapat disimpulkan bahwa bentonit merupakan adsorben alami yang paling potensial untuk dipergunakan pada ternak ruminansia.(Kata kunci: Adsorben aflatoksin, Metode in vitro rumen)

Abstract

Penelitian ini bertujuan untuk mengetahui performa domba yang diberi complete feed kulit buah kakao (KBK) terfermentasi. Domba ekor tipis umur 1-1,5 tahun sebanyak 15 ekor digunakan dalam penelitian ini dengan pemberian pakan dan air minum secara ad libitum. Perlakuan yang dicobakan adalah K yang merupakan pakan kontrol terdiri dari hijauan (sumber serat) + konsentrat, NF merupakan complete feed dari kulit buah kakao (sumber serat) + konsentrat tanpa fermentasi dan F merupakan complete feed dari kulit buah kakao (sumber serat) + konsentrat yang melalui proses fermentasi (CFF) dengan perbandingan antara sumber serat dan konsentrat adalah 40% : 60%. Variabel yang diamati adalah konsumsi pakan, pertambahan bobot badan, konversi pakan, feed cost per gain dan income over feed cost. Hasil penelitian menunjukkan bahwa complete feed berpengaruh nyata terhadap konsumsi bahan kering (BK), bahan organik (BO), protein kasar (PK), serat kasar (SK), lemak kasar (LK) dan total digestible nutrients (TDN), sedangkan pertambahan bobot badan harian dan konversi pakan tidak berbeda nyata terhadap formulasi complete feed. Rerata konsumsi BK, BO, PK, SK, LK, dan TDN berturut-turut adalah 87,72; 78,21; 10,12; 12,21; 1,77, dan 59,52 g/kg BW0,75. Rerata pertambahan bobot badan harian adalah 128,67 g/hari atau 0,88 g/kg BW0,75, sedangkan rerata konversi pakan adalah 6,27. Disimpulkan bahwa pemberian ransum complete feed menghasilkan konsumsi nutrien yang tinggi tetapi tidak memberikan efek pertambahan bobot badan harian dan nilai konversi pakan yang lebih tinggi. Pemanfaatan fermentasi KBK menggunakan biofit menghasilkan PBBH 128,57 g/ekor/hari atau 0,88% g/kg BB0,75 efektif untuk digunakan dalam ransum penggemukan ternak domba.(Kata kunci: Domba, Complete feed, Kinerja)

Abstract

The aim of this study was to evaluate the effect of garlic extract (GE) in rations on the bloodcholesterol profiles of aflatoxin B-induced hepatotoxic in the early stage of broiler chickens. As many as140 one-day old chicks were used in this study. They were randomly divided into 16 treatment groups andeach group consists of 9 chickens. The experimental design adopted in this study was completely randomizedfactorial design consisting 4 concentrations of aflatoxin (A0 : 0 ppb, A1:500 ppb, A2: 1000 ppb and A3:1500 ppb) and 4 concentrations of garlic extract (B0: 0% B1:2% B2: 4% and B3 6%). The treatment wascarried out for 21 days. The serum levels of total cholesterol, low density lipoprotein (LDL) and highdensity lipoprotein (HDL), and HDL/LDL ratio were the examined. The result showed that Aflatoxin Bcaused hepatotoxicosis in broiler chickens indicated by the increase of total cholesterol and LDL, and thedecrease of HDL and HDL-LDL ratio. Treatment of chickens with 2% GE decreased total cholesterol andLDL level in serum, and increased HDL level and HDL-LDL ratio. No interaction effect was observedbetween aflatoxin B and GE extract treatments. It is evident that treatment of Garlic extract reduces thehepatotoxicosis effect of aflatoxin B treatment in broiler chickens.

Abstract

The objective of this study was to determine the effect of undegradable dietary protein (UDP) using protected soybean meal supplementation on blood metabolites of lactating dairy cows. Eighteen early lactating Friesian Holstein cows of 1 to 3 years old were used in this study and observed twelve weeks. All cows received forage and concentrate in a ratio of 62:38 (DM basis). Adaptation to the diet was conducted for 4 weeks. The  cows were divided into 3 groups with different dietary treatments: control diet without UDP and mineral mix (T0), control diet + UDP 40 g/L milk + mineral mix (T1), and  control diet + UDP 60 g/L milk + mineral mix (T2). Fresh water was also offered ad libitum in the pen. In the days 14 and 7 pre-treatment, and in the days 1, 7, 14 and 21 post-treatment, cows were weighted and their blood were collected for chemical analysis. Means between treatments were analyzed using one-way analysis of variance (ANOVA). The results obtained from this study revealed that blood metabolites pattern of lactating cows before and after UDP supplementation subjects to considerable varia-tions. The UDP supplementation had a significant effect on the concentration of Ca. For comprehensive assessment of the effect of UDP supplementation on blood metabolites at different stages of parturient period, further detailed studies should be performed

Abstract

               The content of AFB1 in feed with low levels and spend a long time, will cause primary damage or primary liver carcinoma. This study aims to reduce the toxicity of AFB1 with amino acid methionine-cystine supplementation in broiler chicken feed. This study used a 3 × 3 factorial design with methionine-cystine amino acid levels (M + C: 75, 100 and 125%) and AFB1 levels (0, 200, and 400 ppb). The variables collected were liver physiology, liver histopathology, SGPT levels, and SGOT levels. Observations of liver physiology showed that feed containing aflatoxin without methionine-cystine amino acid had a paler yellowish color (T4, T7 and T8). Pathological examination resulted that aflatoxicosis will attack the liver. Transition amino acid cystines in chicken feed contaminated with AFB1 did not occur in blood SGPT levels. Blood SGOT levels were highest at 21 days of age, namely T2 (M + C 100%) and T3 (M + C 125%) at AFB1 0 ppb which showed excess liver damage. The administration of methionine-cystine amino acids of 75, 100 and 125% in chicken feed contaminated with AFB1 0, 200 and 400 ppb consumed by broilers carries a risk of physiological and pathological damage to chicken liver.
