Abstract

Uji multilokasi merupakan merupakan tahapan penting dalam menyeleksi hibrida yang konsisten berpenampilan baik di semua lokasi pengujian ataupun yang berpenampilan baik spesifik lokasi. Dua puluh lima hibrida jagung manis SR diuji penampilannya dari bulan September 2008 sampai dengan Maret 2009 di tiga lokasi di Jawa Barat, yaitu: Jatinangor - Sumedang, Lembang – Bandung, Wanayasa - Purwakarta. Penelitian bertujuan untuk: mengidentifikasi adanya interaksi G x E pada karakter hasil; menyeleksi hibrida SR yang stabil di beberapa lokasi pengujian, dan atau yang adaptif pada lingkungan yang spesifik di Jawa Barat. Materi genetik tersebut ditanam di tiga lokasi berdasarkan rancangan tata ruang Rancangan acak kelompok dengan menggunakan 23 jagung manis hibrida SR Unpad dan dua kultivar cek sebagai perlakuan dengan setiap perlakuan diulang sebanyak tiga kali. Data dianalisis dengan analisis varians, analisis varians gabungan, analisis model AMMI (additive main effect and multiplicative  interaction). Berdasarkan hasil pengujian hibrida jagung manis SR di tiga lokasi di Jawa Barat terdapat interaksi G x E pada karakter hasil. Hibrida jagung manis SR 4, SR 27, SR15, dan SR 24 merupakan hibrida yang paling stabil, namun SR 4 dan SR 24 merupakan hibrida yang memiliki hasil di atas rata-rata yang stabil di beberapa lokasi pengujian. Hibrida jagung manis SR 9, SR 25, SR 22, SR 20, SR 30, SR 28, SR 32, SR 31, SR 43, SR 47, SR 52, SB beradaptasi baik di Jatinangor dan Purwakarta, namun kurang beradaptasi di Lembang. Disarankan melakukan pengujian pada lokasi yang lebih luas kisarannya serta musim yang berbeda untuk mengetahui stabilitas dan adaptabilitas hibrida jagung manis SR secara lebih akurat.

Abstract

Resistance to Philippine downy mildew (Peronosclerospora philippinensis) was observed in a BC1F2 population developed from a cross between the susceptible inbred line Pi 23 and the resistant inbred line P 345. Five components of resistance namely, disease incidence, disease severity, onset of systemic symptoms, the-area- underdisease-progress- curve (AUDPC), and rate of downy mildew development affected host resistance reactions. All components were significantly correlated. The adult plant resistant reaction was evident in resistant progenies showing prolonged onset of systemic symptoms. Transgressive segregation that exceeded the high and low parent values was also observed for all components of quantitative resistance to Philippine downy mildew.

Abstract

An experiment to locate QTL conferring resistance against Peronosclerospora philippinensis causing Philippine downy mildew was observed in a BC1F2 population developed from a cross between the susceptible inbred line Pi 23 and the resistant inbred line P 345. Phenotypic data were collected in Los Baños, UPLB for disease incidence. A total of four regions were associated with disease incidence. Some of QTL associated with disease incidence have been reported previously, and the remaining QTL are described here for the first time. The results on QTL analysis using molecular markers also agree with the previous study using classical generation mean analysis of the cross combination Pi 23 x P 345.

Abstract

Simple sequence repeats (SSRs) is an important DNA markers for mapping plant genomes particularly in mapping quantitative trait loci (QTL). To identify and map SSR markers for introgression resistance gene against Peronosclerospora philippinensis in maize, the parental materials (P 345 and Pi 23) together with 142 genotypes of BC1F1 of Pi 23 ×P 345 population were used. A total of 50 SSR markers were genotyped. However, only thirty-three markers were integrated into a molecular map together with RFLP and RGA markers. A molecular map of SSR was successfully constructed together with RFLP and RGA into 10 chromosomes. It covered a total map length of 1822 cM with an average distance of 18.22 cM between markers.

Abstract

Thirty-two TWC QPM hybrids were selected based on genotype × environment (G × E) interaction and stability. The experiment was conducted in three locations with differing conditions. TWC QPM hybrids were tested under nitrogen deficient and nitrogen sufficient fertilization at the Padjadjaran University Faculty of Agriculture Experimental Station, Jatinangor, Sumedang from March until July 2004. Another set of TWC QPM hybrids was tested under sufficient nitrogen fertilization at a high altitude location in Pangalengan from June until October 2004. Each experiment was arranged in a randomized block design with two replications with 32 TWC QPM hybrids and two check cultivars, i.e. Bisi-2 and Pioneer-7. Bartlett’s homogeneity test was applied to error derived from analyses of variances of every location. Combined analysis was done since error was homogenous for all observed characters. The combined analysis showed that there was G × E interaction for seed weight per 10 plants. Preliminary METs in West Java successfully selected newly developed TWC QPM hybrids based on stability and interaction between G × E. These hybrids are QPDMR-8-BCP1 and QPDMR-8-BCP2 for wide adaptive cultivars and QPDMR-35-BCP1, QPDMR-26-BCP1, QPDMR-8-BCP1, QPDMR-1-BCP2, QPDMR-8-BCP2, QPDMR-10-BCP2 and QPDMR-15-BCP2 specific adaptive cultivars.

Abstract

Proses pembentukan kultivar hibrida jagung dimulai dengan pembentukan galur-galur murni, uji daya hasilnya melalui uji daya gabung umum, dan akhirnya galur-galur murni yang memiliki potensi hasil terbaik digunakan sebagai tetua untuk persilangan membentuk kultivar hibrida.Penelitian bertujuan untuk mendapatkan galur-galur S4-A yang memiliki daya gabung umum yang baik dan mendapatkan hibrida testcross-nya yang memiliki daya hasil tinggi dibandingkan kultivar cek di dua musim tanam di Jatinangor. Percobaan dilakukan di kebun percobaan Universitas Padjadjaran, Jatinangor, pada musim I (kemarau) bulan Juli September 2003 dan musim II (hujan) bulan Maret–Mei 2004 berdasarkan Rancangan Perbesaran (Augemented Design) dengan 138 varietas hibrida testcross jagung S4-A dan kultivar cek Pioneer-7, Bisi-2 dan Cargil-7. Hasil Percobaan menunjukkan bahwa terdapat galur-galur S4-A yang mempunyai daya gabung umum yang baik di dua musim tanam, yaitu galur S4-A 280-2, 74-10, 24-11, 284-1, 240-2, 266-2, 289-2, 229-1, 203-1 dan 219-2. Sedangkan hibrida testcross-nya yang memiliki daya hasil melebihi kultivar cek pada dua musim tanam adalah pada TC-57, TC-73, TC-95, TC-115, TC-116, TC-120, dan TC-22.

Abstract

Informasi daya gabung umum dan daya gabung khusus komponen resistensi dari galur tetua diperlukan untuk pembentukan kultivar hibrida yang superior tahan hama gudang. Tujuan penelitian ini adalah untuk mengetahui nilai daya gabung umum dan daya gabung khusus komponen resistensi dari galur-galur Downy Mildew Resistance (DMR) dan Quality Protein Maize (QPM). Tujuh galur murni dan 12 hibrida silang tunggal jagung koleksi Laboratorium Pemuliaan Tanaman Fakultas Pertanian Universitas Padjadjaran, diuji menggunakan metode eksperimen ditata dalam rancangan acak kelompok diulang tiga kali. Percobaan dilaksanakan di Laboratorium Entomologi jurusan Hama dan Penyakit Tumbuhan Fakultas Pertanian Universitas Padjadjaran, dari bulan Agustus 2004 sampai November 2004. Galur DMR Ki 3 dan MR 10 merupakan penggabung umum yang baik untuk komponen resistensi persen kehilangan bobot dan persen kerusakan benih. Galur QPM CML 163 merupakan tester yang baik  untuk kehilangan bobot dan persen kerusakan benih. Sedangkan CML 161 merupakan tester yang baik hanya untuk karakter persen kehilangan bobot biji. Pasangan persilangan P 345 x CML 163 merupakan penggabung khusus yang baik untuk komponen resistensi kehilangan bobot biji dan persentase serangan. MR 10 x CML161 memiliki nilai tertinggi untuk heterosis baik MPH maupun HPH pada komponen resistensi kehilangan bobot biji dan persentase serangan.

Abstract

The information on germplasm diversity and genetic relatedness among elite breeding materials is an important element in maize breeding. Molecular characterization and genetic relationship of 11 QPM-DMR lines were analysed using thirty three SSRs markers. Genetic relationship was determined using Jaccard’s similarity coefficient, and dendogram was then constructed based on the unweighted pair-group method with arithmetical averages (UPGMA). Result showed that (i) all SSRs loci were informative for describing the genotypic variation as showed by their PIC, which ranged from 0.19 for umc1304 to 0.93 for phi112; (ii) the eleven maize inbred lines were clustered into one major group A and small groups B and C that corresponds well with the breeding programs adopted at different institutes of release, and (iii) thus, SSRs marker system is a valuable marker for varietals identification and for genetic diversity study of elite breeding materials.

Abstract

Sebagai bagian dari suatu penelitian kolaboratif AMBIONET (Asian Maize Biotechnology Network), Indonesia turut serta dalam kegiatan marker assisted selection (MAS) untuk pemuliaan jagung tahan bulai. Tujuan penelitian adalah untuk mengidentifikasi dan memetakan 146 marka RFLP (Restriction Fragment Length Polymorphism) yang berasosiasi dengan lokus karakter kuantitatif ketahanan dari 134 populasi genotip RIL (recombinant inbred lines) progeni CML 139 x Ki3. Penelitian dibagi atas dua tahap yaitu tahap analisis molekuler dan tahap pengujian di lapangan. Analisis molekuler memanfaatkan data dari Pusat Penelitian Internasional Jagung dan Terigu (CIMMYT), Mexico. Penelitian lapangan dilakukan di Bogor pada bulan Mei 2002 sampai Juli 2002. Data fenotipik (persentase serangan bulai) dianalisis dengan menggunakan program alpha lattice, sedangkan analisis QTL (Quantitative Trait Loci) menggunakan “software computer” program “Windows QTL Cartographer V.1.21”. Hasil analisis QTL menunjukkan bahwa dari 146 marka RFLP pada 134 populasi genotip RIL progeni CML 139 x Ki3, terdapat 14 marka yang terdistribusi pada enam kromosom dari 10 kromosom berdasarkan fenotaiping di Bogor. Efek aditif yang terdeteksi dalam analisis QTL pada umumnya bernilai negatif yang mengarah ke CML 139 (genotip peka).

Abstract

Exploration of heterotic patterns among maize inbreds has become important since it could provide information on which new germplasm could be used to improve base population in breeding program. This experiment was a preliminary study on the heterotic relationships between tropical quality protein maize (QPM) and downy mildew resistance (DMR) maize inbreds based on analysis on combining ability. Seven tropical maize inbreds were crossed in a line × tester. One commercial hybrid from Bisi Company, Bisi 2, was included for the evaluation of hybrids as check cultivar. The result revealed that two heterotic groups has been recognized, i.e., major group consisted of CML 161, CML 163, CML 172, Nei 9008, and P345. Whereas the small group contains MR 10 and Ki 3. The hybrids and their parental inbreds were evaluated in Jatinangor, Indonesia (753 m above sea level) in 2003 to 2004. The location of yield test has a humid climate with an annual rainfall of about 1925 mm. The field experiment was arranged in a randomize block design with two replications. The present study revealed two heterotic groups: major group involving CML 161, CML 163, CML 172, Nei 9008, and Ki 3; and small group containing MR 10 and P 345. Moreover, the major group can be divided into two subgroups involving all CML lines for the first subgroup and Nei 9008 and Ki 3 for the second subgroup.
