Abstract

The aim of this study was to find out DNA band pattern as a result of amplification using DNA primer from meat tenderness gene of Bali cattle. Sample used in this study was DNA isolated from blood Bali cattle. Blood samples was collected in heparin tubes from jugular vein. Leucocyte cells were isolated using RBCs (Red Blood Cells) lysis buffer. To get DNA fragmen, marbling and meat tenderness gene were amplified by using DNA primer for for meat tenderness (forward: 5’–CTACCGGGACGTCAACCT-3’; reverse: 5’-GGTTGTCGGGGTAGCTCA-3’). The size of DNA fragment were 210 bp. Key words: Bali cattle, meat tenderness gene

Abstract

BACKGROUND: Bone Morphogenetic Protein Receptor II (BMPR2) deficiency is associated with the pathologic development of pulmonary vascular changes in Pulmonary Arterial Hypertension (PAH). Fibroblast is the most abundant cell in vascular. However, there is only a little information regarding the effect of BMPR2 deficiency in fibroblast. This study aims to understand the effect of BMPR2 deficiency in fibroblasts.METHODS: This study applied the CRISPR/Cas9 technique to edit BMPPR2 in NIH-3T3 cells. The transfection of CRISPR/Cas9 for BMPR2 editing into NIH-3T3 cells was done by using chitosan nanoparticles. The evaluation of BMPR2 and Transforming Growth Factor (TGF)-β mRNA expression was done using Quantitative real-time polymerase chain reaction. The assessment of edited NIH-3T3 cells proliferation was done using a scratch test assay.RESULTS: The BMPR2 mRNA expression of CRISPR/Cas9-edited group was lower than the untreated group. The proliferation of the CRISPR/Cas9-edited group was higher than the untreated group. The TGF-β mRNA expression of CRISPR/Cas9-edited and untreated groups was similar.CONCLUSION: BMPR2 deficiency in fibroblast increase the fibroblast ability to proliferate.KEYWORDS: BMPR2, PAH, fibroblast NIH-3T3, CRISPR/Cas9, proliferation

Abstract

Penelitian ini bertujuan untuk  memilih calon pejantan sapi Aceh yang dipelihara di Balai Pembibitan Ternak Unggul – Hijauan Pakan Ternak (BPTU-HPT) Sapi Aceh Indrapuri berdasarkan berat badan ternak. Evaluasi yang dilakukan pada seleksi calon pejantan meliputi catatan berat lahir (BL), berat sapih (BS), berat setahunan (BY) dan berat dewasa (BD). Jumlah ternak yang digunakan untuk dievaluasi pada tahun 2014 adalah 34 ekor. Berdasarkan data catatan produksi diperoleh 10 calon pejantan sapi Aceh terbaik dengan kisaran nilai rasio berat dewasa (RBD) 1,03 sampai 1,65. Kesepuluh sapi Aceh ini akan direkomendasikan sebagai calon pejantan untuk progam uji zuriat pada tahap seleksi berikutnya.Kata kunci: sapi Aceh, catatan produksi, berat badan, rasio berat dewasa.

Abstract

This research was aims to determine the effect of formaldehyde using as a protective material in the soy groats and lemuru fish oil mixtured to escape of the protein and poly unsaturated fatty acid (PUFA) by in vitro. The rumen fluid was derived from donor of the Ongole crosbred fistulated as much as 3 heads. The experimental design used completely randomized design factorial 2x3 pattern consists of two factors:the first factor (soybean groats:lemuru fish oil in the ratio 2:1 and 4:1) and the second factor (the use of formaldehyde at levels of 0%, 2% and 4% base on the dry matter) and was repeated 5 times. The parameters are consist of the nutrient content, the substrates soybean groats and lemuru fish oil the rumen fluid fermented PUFA content by in vitro. Results of the study are levels of protein, fatty acids  oleic, linoleic and arachidonic much available on soybean groats:lemuru fish oil protected formaldehyde at levels of 2% and 4%. The difference between the ratio of soybean groats with lemuru fish oil had no significant (P≥0,05) but significantly different at different levels of formaldehyde (P≤0,05). The conclusion are formaldehyde content of 37% with a level of 2% can be used for protection the soybean groats and lemuru fish oil with a ratio of 4:1 and is able to provide protein as well PUFA. Keywords: soy groats, lemuru fish oil, in vitro, PUFA, formaldehyde protection

Abstract

(The genetic correlation of growth traits in Aceh cattle at indrapuri district Aceh Province)ABSTRACT. This research was conducted to estimate genetic correlation of growth traits on Aceh cattle including birth weight (BW), weaning weight (WW), yearling weight (YW), and Average Daily Gain (ADG). The research was done at Indrapuri Breeding and Forage Centre (IBFC) of Aceh cattle from March 2013 to April 2013. The materials of this research consist of growth records from 2010 to 2012. The genetic correlation was analyzed by variance and covariance. The results indicated that the lowest standard error (SE) value of heritability showed on birth weight 0.15+0.13. Most of genetic correlation value on growth traits was positive and high categorized ( 0.50). The lowest SE value showed on correlation between birth weight and pre-weaning ADG (0.55+0.54), birth weight and post-weaning ADG (0,63+0,62), pre-weaning ADG and post-weaning ADG (0.71+0.33), pos-weaning ADG and weight/age or W/A (0.72+0.33) then final weight and W/A (0.94+0.69). It could be concluded that most of genetic correlation value on Aceh cattle growth traits were positive and high therefore the selection based on growth traits can be done for increasing performance in Aceh cattle.
