Abstract

The aim of the research was to determine effect of Padina sp. extract on shelf life of red nile fi llet during storage at low temperature. Red nile fi llet was soaked up into Padina sp. extract solution at various concentration of 0; 0.5%; 1%; 1.5%; and 2% for 30 minutes, then was stored in chilling room for 10 days. The observation was carried out every 2 days. The parameters observed were pH, Total Plate Count (TPC), Total Volatile Base-Nitrogen (TVB-N), and organoleptic still be tests. The results showed that the different concentration of Padina sp. extract yielded the signifi cant effect (P<0,05) on pH, TPC, TVB-N and organoleptic of the red nile fi llet. The treatment of Padina sp. extract on red nile fi llet can still be consumed up to 8 days of storage based on TPC, TVB, and organoleptic namely odor and texture. The treatment 1% of Padina sp. extract was the best treatment in maintaining shelf life of red nile fi llet stored at low temperature.ABSTRAKPenelitian ini bertujuan untuk mengetahui pengaruh penggunaan ekstrak Padina sp. terhadap daya simpan fi let nila merah selama penyimpanan pada suhu dingin. Filet nila merah direndam selama 30 menit dalam larutan ekstrak Padina sp. dengan konsentrasi: kontrol; 0,5%; 1%; 1,5%; dan 2%, kemudian disimpan pada chilling room selama 10 hari dengan selang waktu pengamatan setiap 2 hari. Parameter yang diamati meliputi: pH, Total Plate Count (TPC), Total Volatile Base-Nitrogen (TVB-N), dan organoleptik. Hasil penelitian menunjukkan bahwa penggunaan ekstrak Padina sp. yang berbeda memberikan pengaruh nyata (P<0,05) terhadap nilai pH, kandungan bakteri total, kandungan TVB-N, dan nilai organoleptik keseluruhan fi let nila merah selama 10 hari penyimpanan pada suhu dingin. Filet nila merah yang diberi ekstrak Padina sp. masih layak dikonsumsi sampai hari ke-8. Penggunaan ekstrak Padina sp. 1% merupakan perlakuan terbaik dalam mempertahankan kesegaran fi let nila merah yang disimpan pada suhu dingin.

Abstract

The objective of this study was to investigate the antifungal activity of the soluble matter (SM) and crude saponins (CS) extracted from Stichopus japonicus using pressurized solvent extraction (PSE) with water or aqueous ethanol as a solvent, in comparison with traditional heat reflux extraction (HRE). The extraction yields were also determined for the SM and CS and compared for each extraction process and solvent. The antifungal activity of the SM and CS, extracted from the body wall of Stichopus japonicus using PSE or HRE with water or 70% aqueous ethanol, were investigated. Both SM and CS exhibited their highest antifungal activity when extracted by HRE with 70% ethanol and by HRE with water, respectively, while their highest yields were obtained when extracted by PSE with water. SM has more antifungal than potassium sorbate but weaker than propyl paraben, while CS has more antifungal than the two antifungal agents.

Abstract

Indonesia has a lot of seaweed that have high potential as a producer of alginate, but the method of extraction has not been as expected. The objective of this study to develop a method of extraction of sodium alginate from seaweed Sargassum through the calcium alginate pathway. This study used different variations of the concentration of calcium chloride. The concentration of calcium chloride used varied 0.5, 0.75 and 1 M. As a control, the extraction of alginate performed through alginic acid pathway which was developed at the Center for Research Product Processing and Biotechnology of Marine and Fisheries, Jakarta. Quality parameters were observed including alginate yield, product appearance, viscosity and gel strength. The result showed that the yield of alginate produced successively for 32.67; 44.67 and 53.33 % and 31.67 % for controls. In appearance, the concentration of calcium chloride did not significantly affect the alginate product appearance, but darker when compared with the product extracted through the alginic acid. Viscosity alginate produced successively 149, 131 and 144 cP, while 304 cP for control. In general, the alginate gel strength extracted through of calcium alginate pathway is lower than the alginic acid pathway.ABSTRAKIndonesia  mempunyai  banyak  rumput  laut  yang  berpotensi  tinggi  sebagai penghasil  alginate,  namun  metode ekstraksinya belum sesuai yang diharapkan. Penelitian ini bertujuan untuk mengembangkan metode ekstraksi natrium alginat dari rumput laut Sargassum melalui jalur kalsium alginat. Dalam penelitian ini digunakan berbagai variasi konsentrasi kalsium klorida yang digunakan pada pemisahan alginat dari filtrat hasil ekstraksinya. Konsentrasi kalsium klorida yang digunakan divariasi 0,5; 0,75 dan 1 M. Sebagai kontrol dilakukan ekstraksi alginat melalui jalur asam alginat yang dikembangkan di Balai Besar Riset Pengolahan Produk dan Bioteknologi Kelautan dan Perikanan, Jakarta. Parameter kualitas alginat yang diamati meliputi rendemen alginat, kenampakan produk, viskositas dan kekuatan gel yang dihasilkan. Hasil pengamatan menunjukkan bahwa rendemen alginat yang dihasilkan berturut-turut sebesar 32,67; 44,67 dan 53,33 % dan untuk kontrol 31,67 %. Secara kenampakan, konsentrasi kalsium klorida tidak terlalu mempengaruhi kenampakan produk alginat yang dihasilkan, tetapi lebih gelap jika dibandingkan dengan produk hasil ekstraksi melalui jalur asam alginat. Viskositas alginat yang dihasilkan berturut-turut 149, 131 dan 144 cP, sementara untuk kontrol 304 cP. Secara umum kekuatan gel alginat yang dihasilkan dari jalur kalsium alginat lebih rendah dibandingkan dengan yang diekstrak melalui jalur asam alginat.

Abstract

The purpose of this study was to determine antioxidant activity of polyphenol and phlorotannin extracts from brown algae Sargassum polycystum. Polyphenol was extracted with ethanol 96% then tested total phenolic content, while phlorotannin was extracted with methanol, chloroform, aquabidest, and ethyl acetate then tested total phlorotannin content. After crude polyphenol and crude phlorotannin extracts were obtained, the studied compounds were extracted using column chromatography. The extracts were then analyzed with HPLC (High Performance Liquid Chromatograpys). Antioxidant activity was analyzed by radical scavenging activity (RSA) and ferrous ion chelating ability (FIC) for four samples, crude polyphenol, crude phlorotannin, polyphenol, and phlorotannin. The result of total phenolic content was 1.18±0.67 mg GAE/g dry sample and total phlorotannin content was 0.61±0.27 mg PGE/g dry sample. Based on RSA and FIC analysis, phlorotannin (IC50 1.12±0.02 mg/mL and 1.34±0.01 mg/mL) showed the highest activity followed by crude phlorotannin (IC50 1.20±0.01 mg/mL and 1.47±0.10 mg/mL), polyphenol (IC50 1.23±0.01 and 1.55±0.02 mg/mL), and crude polyphenol (IC50 1.27±0.01 mg/mL and 1.63±0.02 mg/mL). The results of chromatographic analysis showed that S. polycystum extract contains phenolic compounds similar to the phloroglucinol.ABSTRAKPenelitian ini bertujuan untuk menentukan aktivitas antioksidan ekstrak polifenol dan florotanin dari rumput laut cokelat Sargassum polycystum. Polifenol diekstrak menggunakan pelarut etanol 96% kemudian diuji kandungan total fenolik, sedangkan florotanin diekstrak dengan menggunakan pelarut metanol, kloroform, akuades, dan etil asetat kemudian diuji kandungan total florotanin. Setelah memperoleh ekstrak polifenol dan florotanin, kedua senyawa tersebut dipurifikasi dengan kromatografi kolom. Hasil kromatografi kolom dianalisis menggunakan HPLC (High Performance Liquid Chromatography). Uji aktivitas antioksidan dilakukan dengan metode radical scavenging activity (RSA) dan ferrous ion chelating ability (FIC) terhadap empat sampel, yaitu ekstrak polifenol, ekstrak florotanin, polifenol, dan florotanin. Kandungan total fenolik yang diperoleh adalah 1,18±0,67 mg GAE/g ekstrak kering, sedangkan kandungan total florotanin diperoleh 0,61±0,27 mg PGE/g ekstrak kering. Berdasarkan uji RSA dan FIC, florotanin (IC50 1,12±0,02 mg/ mL dan 1,34±0,01 mg/mL) memiliki aktivitas tertinggi diikuti ekstrak florotanin (IC50 1,20±0,01 mg/mL dan 1,47±0,10 mg/mL), polifenol (IC50 1,23±0,01 dan 1,55±0,02 mg/mL), dan ekstrak polifenol (IC50 1,27±0,01 mg/mL dan 1,63±0,02 mg/mL). Hasil analisis kromatografi menunjukkan ekstrak S. polycystum mengandung senyawa fenolik relatif seperti senyawa floroglusinol.

Abstract

Fresh fishery products are highly perishable and susceptible to spoilage caused by rapid growth and activity of bacteria. Psychotropic bacteria are the major microorganisms responsible for spoilage when these products stored in ice or under refrigeration. The purposes of this study were to isolate psychrotrophic bacteria from fresh fishery products and to evaluate its resistance to bacteriocins. Fresh fishery products (kembung fish, shrimp, shellfish) samples were obtained from Demangan and Kranggan traditional market. Colony count methods were used to enumerate bacteria, whereas isolation and identification of bacteria was performed by the identification method of Lahallec and Colin (1995). The identification conducted were covering Gram staining and cell morphological examintaion (cell form), motilitym presence or absence of an oxidise, mode of glucose utilization, and capacity for glucose or lactose fermentation, H2S production, and gas production. The resistance assyas by bacteriocin were carried out using agar diffusion method. Seventeen isolates were found abd they were classified as Pseudomonas, Xanthomonas, and Flavobacterium/ Cytophaga. The results show that psychrotropic bacteria resistance to supernatant of lactic acid bacteria and nisin.

Abstract

The aim of this study was to determine the effect of storage temperature on the antioxidant activity of Sargassum hystrix. Samples were collected from Sepanjang Beach Gunung Kidul, Yogyakarta, and dried, pulverized, and then stored for two months at room temperature (17-33 °C), refrigerator (4 °C) and freezer (-18°C). Furthermore, the samples were tested every two weeks to determine their water content, total phenol, DPPH, and FIC. Also, Gas Chromatography-Mass Spectrometry (GC-MS) analysis was performed for two months before and after storage. The results showed that the water content increased after treatments, along with storage time which ranges from 8.55 to 14.27 %. Total phenol before storage was 175.73 mg GAE/g extract, and freezing has the highest total phenol levels after storage which was 22.07 mg GAE/g extract. Also, DPPH testing showed S. hystrix has anIC50 value of 0.45 mg/mL, and refrigeration has the lowest IC50 value of 3.18 mg/mL. FIC test showed IC50 value of 1.44 mg/mL, and freezing has the lowest FIC IC50 value of 15.36 mg/mL. In addition, GC-MS sample analysis before storage detected 8 compounds with antioxidant activity of 86.96 %. After two months, they were detected at room temperature, refrigerator, and freezing which are 4, 3, and 8 compounds respectively.

Abstract

Sargassum hystrix juice is a functional beverage made from S. hystrix through the immersion process, crushing, filtering, and pasteurization. Therefore, this study aims to determine the effect of S. hystrix concentration on antioxidant activity, level of consumer acceptance, and chemical composition of the juice. The fresh S. hystrix used were washed, immersed for three days, and blended at a concentration of 10, 20, 30, 40, and 50%, respectively. Meanwhile, the qualities of juice that were observed include yield, antioxidant activity, total phenol, water, ash, protein, and fiber content, formol number, total sugar, total plate count, and sensory analysis. The results showed that the S. hystrix juice had a value of yield 43.00-85.67%, pH 7.5, antioxidant activity 20.50-44.57%, total phenolic 32.78-293.11 mg GAE/g, water 93.03-95.39%, ash 0.36-6.34%, protein 0.88-4.96%, crude fiber 0.41-5.04%, formol number 15-21.67 mL N NaOH/100 mL, total sugar 3.10-4.61%, total plate count 2.3x103-4.3x103CFU/mL, sensory of color 1.83-2.95, flavor 2.49-3.29, and taste 2.29-3.50.

Abstract

Thyroxine hormone (T4) was one of the hormones related to metabolism rate. Animal transportation could be stressor interfering metabolism. Sargassum crasifoium was one of brown algae species that contained some important nutrients and also flavonoids. The study aimed at finding out the effect of the Sargassum on the decrase in thyroxine hormone and body weight loss. It used 9 Javanese Randu goats that were classified into 3 treatment groups. Group I served as control, Group II was treated with 450 mg/kg BW of Sargassum estract and Group III was treated with vitamin B1. All of the goats were transported for 12 hours using pickup truck at the speed of 60 km/hour. Blood samples were drawn before the transportation, during loading, 6 hours after the transportation and at the end of the transportation, while the body weight of the goats was weighed before and after the transportation. The results of the study showed that the T4 hormone decreased in the Group I and increased in the Group III during the transportation, while it was stable in the Group II. The biggest percentage of the body weight loss took place to the Group III (0.13%), while the smallest percentage took place to the Group II (0.063%). Statistical analysis showed that the Sargassum extract did not have any significant effect (p>0.05) on both the thyroxine hormone and the body weight loss during the transportation. Based on the results of the study it could be concluded that the Sargassum extract could stabilize the thyroxine hormone during the transportation so that it could prevent the body weight loss

Abstract

Penelitian ini untuk mengetahui kemampuan karagenan dan polifenol dariEucheuma denticulatum terhadap karbohidrat enzim α-amilase secara in vitro. Ekstrasi karagenan menggunakan larutan kalium hidroksida 0,5%. Polifenol diekstraksi dengan 50% (v/v) metanol. Identifikasi karagenan ditentukan dengan Fourier Transform Infrared (FTIR). Kandungan total fenol ekstrak ditentukan menurut metode Folin-Ciocalteu. Kemudian hasil ekstraksi diuji daya hambat aktivitas α-amilase. Ekstrak karagenan dan polifenol mempunyai kemampuan menghambat aktivitas IC50  α-amilase yaitu 12,16 dan  11,64 mg/mL. Ekstrak Polifenol   memiliki daya hambat α-amilase lebih tinggi dari pada ekstrak karagenan.Kata Kunci: Eucheuma denticulatum, karagenan, polifenol, α-amilase.

Abstract

Kitosan diketahui memiliki aktivitas antibakteri, biodegradable, dan biokompatibel tetapi tidak larut dalam air. Modifikasi kitosan menjadi karboksimetil kitosan diharapkan mampu meningkatkan kelarutan dalam air tanpa mengubah bioaktivitas sehingga memungkinkan aplikasi sebagai antibakteri. Penelitian ini bertujuan mengkaji pengaruh pemberian karboksimetil kitosan sebagai bahan antibakteri terhadap daya simpan filet nila merah yang disimpan pada suhu dingin. Karboksimetil kitosan dibuat dengan cara mereaksikan senyawa kitosan dengan asam monokloroasetat pada suhu 900C selama 3 jam. Sampel filet nila merah segar direndam dalam larutan karboksimetil kitosan dengan konsentrasi 0%; 2%; 3% dan 4% selama 60 menit, kemudian disimpan pada suhu dingin 50C selama 12 hari.
