Abstract

This study aimed to isolate and identify lactic acid bacteria (LAB) that could produce bacteriocins from urutan. Urutan is a Balinese sausage that is traditionally produced and naturally fermented. Urutans used in this study were obtained from several traders in the Baturiti, Mengwi, and Tegallalang region, Bali. More than 300 LAB colonies were isolated and underwent inhibition test against the growth of the indicator bacteria. Crude bacteriocins from two isolates, namely J2 and J6, showed inhibition zones against the growth of Lactobacillus plantarum E12.1 and Escherichia coli ATCC 8739. The J6 isolate also inhibited the growth of Enterococcus faecalis ATCC 29212. Both J2 and J6 were gram-positive, bacilli shaped, and showed negative catalase test. Based on the results of the 16S rDNA gene amplification, the two isolates were identified as Lactobacillus plantarum. Keywords: Lactic Acid Bacteria, Bacteriocin, Lactobacillus plantarum, Urutan

Abstract

The knowledge of farmers on pesticides and its association on health and the environment still remains an issue. Located in the rural area of Bali, the village of Songan, Kintamani, Bangli with its fertile soil accommodates many farm lands. The counseling and assistance included 20 farmers during their working hours to allow direct observation. The counseling and assistance targeted at issues regarding safe uses of pesticides and its association to health and the environment. The knowledge of participants was evaluated by pre- and post-test. Less than a half (40%) were able to identify an association between pesticide use and health, while almost all do not know the chronic effect of pesticides to health and/or the environment. Recommendations target health care facilitators are expected to continue counseling and assistance to the farmers and others in order to maintain a healthier and more sustainable future.

Abstract

Kebiasaan buang air besar sembarangan masih dilakukan oleh kurang lebih 45% penduduk Indonesia yang umumnya tinggal di pedesaan. Kebiasaan tersebut tidak hanya karena kurangnya kesadaran akan kebutuhan sanitasi, namun juga dikarenakan keterbatasan pilihan teknologi dan sarana sanitasi yang tersedia di tingkat lokal. Puskesmas Kubu II, Kecamatan Kubu telah berusaha menerapkan program Sanitasi Total Berbasis Masyarakat (STBM) untuk menggugah kesadaran masyarakat akan kebutuhan sanitasi dan memfasilitasi untuk terbangunnya aksi kolektif masyarakat untuk menyelesaikan masalah tersebut. Di sisi lain, isu terbuangnya air setelah menyiram closet menjadi isu penting dan berpotensi menjadi hambatan program sanitasi di daerah tersebut. Salah satu upaya yang dilakukan adalah dengan memperkenalkan teknologi Septik Tank Biofilter kepada wirausaha sanitasi di daerah tersebut. Para wirausaha diberikan pembekalan mengenai cara pengaplikasian Septik Tank Biofilter dengan semaksimal mungkin menggunakan media lokal. Selama proses pendampingan kendala yang dihadapi calon pengusaha sanitasi adalah pengetahuan tentang aplikasi teknologi Septik Tank Biofilter dan masih sedikitnya wirausaha yang mau menerapkan teknologi ini sebelum dipasarkan . Semua kendala ini diharapkan dapat diatasi dengan membangun komunikasi antara puskesmas, pemerintah desa, masyarkat pengguna mengenai solusi pendanaan dan dukungan untuk upaya peningkatan kebutuhan.

Abstract

Abstract Generally street vendor has little knowledge on hygiene and sanitation of food handling. Usually they trade in a night market or on street sides. There are two markets in Penatih Village that operate night market. Both markets located in a strategic location thus they have many visitors every day. In these two markets there are some vendors that need to be counseled and assisted in order to improve their knowledge on hygiene and sanitation of food handling. The counseling and assistance was conducted every week from 25 September to 10 October 2009, with 10 vendors as target participants. The venues were located in Penatih Market and Agung Market, Penatih Village, East Denpasar. The activities were conducted in the form of counseling and assisting during trading hours towards the participants in regards of hygiene and sanitation of food handling. In order to evaluate and measure the knowledge improvement of the participants, pre test and post test were carried out. The result shows that, most of participants have less knowledge on food additives in terms of usage, types and risk of usage. The participants have good knowledge on usage of clean water for utensil and food washing as well as knowledge on personal hygiene. It is recommended to conduct a continuous and regular counseling to street vendors with the involvement of local primary health care (puskesmas).

Abstract

Susu terfermentasi yang beredar di pasaran menggunakan bakteri probiotik yang bukan berasal dari strain lokal Indonesia. Beberapa strain yang diisolasi dari susu kuda sumbawa menunjukkan potensi untuk dikembangkan menjadi probiotik potensial. Namun, sebagian besar strain-strain belum diuji kemampuannya dalam proses karakterisasi susu untuk membuat susu terfermentasi. Penelitian ini bertujuan untuk menggali potensi Lactobacillus spp. isolat susu kuda Sumbawa sebagai starter susu terfermentasi. Sebanyak 32 isolat Lactobacillus spp. isolat susu kuda Sumbawa yang berpotensi sebagai probiotik diseleksi kemampuannya untuk dikembangkan sebagai starter susu fermentasi yang didasarkan pada produksi massa sel, pH dan karakteristik organoleptik susu terfermentasi. Dari 32 isolat Lactobacillus spp. yang dipergunakan diperoleh 2 isolat potensial yaitu Lactobacillus sp. SMM37 dan Lactobacillus sp. SMM42. Kedua isolat ini dapat memproduksi massa sel yang cepat, menghasilkan susu terfermentasi dengan pH yang baik yaitu Lactobacillus sp. SMM37 sebesar 3,68 dan Lactobacillus sp. SMM42 sebesar 4,19, serta karakteristik organoleptik yang baik yang meliputi rasa, kekentalan (tekstur), ada tidaknya butiran (curd).

Abstract

A study was conducted to isolate and identify of lactic acid bacteria originated from gastric fluid of bali cattle, and to determine their potential as the candidates of biopreservative. Lactic acid bacteria were isolated by culturing the gastric fluid of bali cattle in de Mann, Rogosa, Sharpe (MRS) medium; screening the bacteria, and identification of bacteria species by Analytical Profile Index (API) 50 CHL Kit. The results showed that, the new species of lactic acid bacteria were isolated and identified as Lactococcus lactis spp lactis 1 (SR21 isolate) and Lactobacillus brevis 1 (SR54 isolate) that have broad spectrum antimicrobial activities. It is clear from this study that a potential lactic acid bacteria producing antimicrobial agent can be isolated from the gastric fluid of bali cattle.

Abstract

This research aimed to isolate exopolysaccharides (EPS) producing Lactobacilli isolated fromsumbawa mare’s milk and its potential as prebiotics for modulating the growth of Bifidobacteriumbreve. Nine strains of Lactobacillus sp. were screened for their capabilities to produce EPS usingmodified MRS medium containing sucrose. Prebiotics potential of the EPS was verified by culturingB. breve JCM1273 in TOS medium containing EPS. The results showed that all strains ofLactobacillus sp. produced EPS on MRS sucrose medium and two strains (Lactobacillus SK3.1 andLactobacillus SK4) produced more EPS compared to the other strains tested. Bifidobacteriumbreve JCM1273 showed weak activity while in direct metabolism of EPS produced by Lactobacillussp. SK4 and its growth was enhanced on acid hydrolyzed EPS. Since this phenomenon mighthappened when the EPS exposed by the low pH during gastric passage, hence the EPS might be apotential source to be developed as prebiotics. Nevertheless, further investigation is necessary toevaluate the bifidogenic affects of EPS in Lactobacillus sp. SK4.

Abstract

This research aimed to study the effect of different forage composition in diet on nitrogen balance andmicrobial population of Bali cattle. Randomized Block Design consisted of four feed treatments with 3block of weight live as replicates were used in this study. Body weight of male bali cattle used rangedbetween 181-265 kg. These four treatments based on dry matter were: A (45% elephant grass + 0% ricestraw + 15% glyricidia + 10% calliandra + 30% concentrate); B (30% elephant grass +10% rice straw + 20%glyricidia + 10% calliandra+ 30% concentrate) ; C (15% elephant grass +20% rice straw + 25% glyricidia+10% calliandra + 30% concentrate) and D (0%elephant grass + 30% rice straw + 30% glyricidia + 10%calliandra+ 30% concentrate) . Variables measured were nitrogen balance and rumen microbial population.The collected data were analyzed by analysis of variance. The result showed that nitrogen intake in cattlefed with diet C was significantly (P<0.05) higher than these in other treatments and nitrogen retention(P<0.05) was significantly higher as compared to those fed with diet A. Amylolytic and cellulolytic bacterialpopulations were not significantly different (P>0.05) among all treatments, but the population of proteolyticbacteria was found the lowest (P<0.05) in cattle fed with diet A. It can be concluded that increasedglyricidia and rice straw in the diet could increased nitrogen intake, nitrogen retention and proteolyticbacterial population.

Abstract

A Shiga-like toxin producing Escherichia coli O157:H7 was detected in faecal samples of cattle, andhuman as well as in beef. The performance of agent indicated that it has been identified as harmful andoften life-threatening zoonotic agent. It is therefore important to analysed the genetic characteristic ofShiga toxin Escherichia coli (STEC) and to develop a diagnostic probe in order  to optimalized of diagnostictest  for the agent. The  study was started by amplifiying  stx2 gene, purifying of PCR product, sequencingof stx2 gene, analyzing  of phylogenetic tree, and finally  by analyzing   of  diagnostic  probe candidate.Homology study showed that the genetic sequence of the local isolate of  E. coli O157:H7 i.e SM25(1)isolated from cattle feces has  a genetic and fuctional similarity with  the control isolate i.e E. coli O157:H7ATCC 43894 originated from human.  Further study showed that a probe with  foreward primer  sequanceof 5’-AATTTATATGTGGCCGGGTTC-‘3 which were respectively designed as a PFS and PRS 176 bp product.Appeared to be potential candidate of diagnostic probe for the agent.
