Abstract

Mulberry (Morus alba L.) is a plant from the Moraceae family which has a high flavonoid content. This research aims to determine the ability of NIR spectroscopy and chemometrics to classify mulberry leaf powder based on different planting heights and determine the total flavonoid content using the Uv-Vis spectrophotometric method. The classification of mulberry leaf powder originating from the lowlands, midlands, and highlands provides valid results using NIR spectra and chemometric data from the LDA and SIMCA models with an accuracy value of 100%. The average results of determining total flavonoid content using Uv-Vis spectrophotometry showed that the highest levels were in the highlands, followed by the medium and lowlands with total flavonoid levels respectively, namely 6.453 ± 1.194 mg QE/g powder; 5.889 ± 0.944 mg QE/g powder; and 5.177 ± 0.998 mg QE/g powder. The results of the One Way ANOVA test showed that the total flavonoid content of mulberry leaf powder from different altitude areas did not have a significant difference.

Abstract

Coffee contains bioactive compounds such as chlorogenic acid and caffeine. Chlorogenic acid is recognized as a natural antioxidant, while caffeine is widely used to reduce fatigue and drowsiness. This study aims to simultaneously determine the content of chlorogenic acid and caffeine in robusta coffee leaf powder (Coffea canephora) using Thin Layer Chromatography (TLC) densitometry. The study identified an optimal eluent composition of ethyl acetate:formic acid:distilled water (18:1:1), with maximum wavelengths of 330 nm for chlorogenic acid and 274 nm for caffeine. High linearity was demonstrated with correlation coefficients of 0.993 for chlorogenic acid and 0.994 for caffeine. Sensitivity was validated with LOD values of 100.664 ng for chlorogenic acid and 297.232 ng for caffeine, and LOQ values of 301.994 ng for chlorogenic acid and 891.697 ng for caffeine. Selectivity was confirmed as the method effectively separated chlorogenic acid, caffeine, and other compounds with a resolution (Rs) ≥ 1.5, while specificity was evidenced through purity and identity values exceeding 0.99. Precision was proven through repeatability (%RSD for chlorogenic acid: 2.94%; caffeine: 2.27%) and intermediate precision (%RSD for chlorogenic acid: 2.26%; caffeine: 2.18%). Accuracy was achieved with mean recovery rates of 102.22% for chlorogenic acid and 99.73% for caffeine. The determination of chlorogenic acid and caffeine concentrations in robusta coffee leaf powder revealed variations between samples: PTPN XII Renteng yielded 2.278% ± 0.209% for chlorogenic acid and 1.171% ± 0.821% for caffeine, while Puslitkoka Indonesia samples showed 1.238% ± 0.206% for chlorogenic acid and 0.917% ± 0.823% for caffeine. Based on validation parameter assessments, the method is deemed valid as it meets all acceptance criteria for each parameter. This research provides an efficient and accurate analytical approach for simultaneous determination of bioactive compounds in robusta coffee leaves.

Abstract

This study examines the potential antidiabetic activity of a combination of Mangifera foetida L. and Pandanus amaryllifolius Roxb. leaves. Mangifera leaves contain a compound called mangiferin, which acts as an antidiabetic and antioxidant. On the other hand, Pandanus leaves, are rich in terpenoids and steroids that also possess anti-diabetic effects. An Oral Glucose Tolerance Test (OGTT) was conducted on male mice of the Babl/c strain to assess the antidiabetic effects. The test involved administering a combination of ethanolic leaf extracts from Mangifera foetida L. and Pandanus amaryllifolius Roxb. in a 1:1 ratio. The study followed a pre-test design with a control group, comprising six treatment groups, each consisting of four male mice. The groups were described as follows: Group 1 was served as the normal group without any treatment. Groups II-IV received single extracts or combinations of extracts at doses of 62.5 mg/200g BW, 125 mg/200g BW, and 250 mg/200g BW. Group V acted as the positive control and was given oral glucose and glibenclamide at a dose of 0.09 mg/200g BW. Group VI was served as the negative control, receiving oral glucose and distilled water. The group that exhibited the highest percentage of decreased blood sugar levels was the one given oral glucose at a dose of 0.078g/20g BW of mice, along with the combination of Mangifera foetida L. and Pandanus amaryllifolius Roxb. leaves extract (1:1) at a dose of 35 mg/20g BW. This group demonstrated a decrease of 71.985 4.858 in blood glucose levels. The ANOVA analysis confirmed that the percentage decrease in blood sugar levels was significantly different from the positive control, indicating a higher effectiveness of the combination treatment in reducing blood glucose levels than the positive control.

Abstract

Abstract

Mesona palustris Bl. has considerable potential. Still, it has yet to be matched by its utilization in nanotechnology, especially nanoparticles (1-100 nm), which have superior physicochemical properties and activities. This laboratory study aims to determine the potential of of Mesona palustris Bl. leaf water extract as a bioreductant and capping agent in the biosynthesis of silver nanoparticles (AgNPs), as well as to evaluate the antioxidant activity of the synthesized AgNPs using the DPPH assay. The biosynthesis method produced optimum AgNPs with a ratio of 1% Mesona palustris Bl. leaf water extract, AgNO3, and dispersing medium 0.75:7:5 (v/v) synthesized for 60 minutes at 55C. The maximum wavelength of AgNPs produced was 433 nm with an absorbance of 0.677. FT-IR spectrophotometric characterization showed that phenolic compounds in the Mesona palustris Bl. leaf water extract is thought to act as a bioreductors and capping agent in the synthesis of AgNPs. SEM results showed that the AgNPs were spherical. Meanwhile, the PSA test results showed the average size of AgNPs was 81 nm, and the polydispersity index was 0.323 (moderately polydispersed). The IC50 value of AgNPs synthesized with Mesona palustris Bl. leaf water extract against DPPH free radicals was 71.501 1.347 g/mL, which is included in the potent antioxidant and better than Mesona palustris Bl. leaf water extract.
