Abstract

Program pengabdian ini ditujukan untuk memfasilitasi peternak kelompok ternak sapi potong “Kalimasodo” untuk meningkatkan kualitas kesehatannya melalui perilaku sehat dan meningkatkan tingkat reproduksi ternak sapi potongnya. Para pternak sapi potong di kelompok ternak “Kalimasodo” beberapa bulan ini terjangkit penyakit diare, selain itu, tingkat reproduksi dari ternak potong dari kelompok ternak “Kalimasodo” sangatlah rendah, hal ini disebabkan banyaknya anakan sapi yang mati sesaat setelah dilahirkan indukannya. Program pengabdian ini dilakukan untuk meningkatkan kualitas kesehatan dan tingkat reproduksi dari ternaknya. Usaha peningkatan tersebut dilakukan dengan mengadakan penyuluhan perilaku sehat pada peternak, edukasi, diskusi tentang pentingnya manajemen kesehatan reproduksi ternak potong dan praktek langsung kelapangan terkait peningkatan performa ternak dan deteksi dini kebuntingan pada ternak. Tahatapan pelaksaanaan kegiatan meliputi observasi awal (16 November 2020), Penyuluhan dan edukasi (23 November 2020) dan Praktek Langsung (24 November 2020). Walaupun jumlah anggota peternak tidak maksimal dikumpulkan untuk edukasi dan praktek dikarenakan mengikuti protokol Covid-19, namun antusias dan respon positif dari para peternak menghasilkan berjalannya dengan baik program pengabdian ini.

Abstract

Ovarian follicle dinamycs and estrogen concentration interrelated to cow reproduction traits. Some previous studies said that preovulatory follicle directly related to estrogen concentration which induce estrous quality, and increasing pregnancy rate when artificial insemination happened. Another important factor to optimizing reproduction performance is altitude. Here in Indonesia these factors has not been studied further in PO cows. A total of 24 Peranakan Ongole (PO) cows, 12 cows from highlands or mountains of Kalibawang, Kulon Progo, Yogyakarta, Indonesia and 12 cows from lowlands or lowland of Galur District, Kulon Progo, Yogyakarta, Indonesia selected based on criterias; these cows ever had calves, not pregnant, 3-8 years old, have a Body Condition Score (BCS) of 2.5-3.5, in the peak phase of estrus and not experienced reproductive problems. The diameter of the cow ovarian follicles were examined using ultrasound and sera were taken to calculated the estrogen concentration using ELISA, meanwhile the NSPC taken from recording and farmer interview. The result showed that no difference of follicle diameter and estrogen concentration between mountain and lowland (p>0,05), a significant difference found in NSPC between mountain and lowland (p<0,05), and a strong correlation found betweend follicle diameter, estrogen concentration and NSPC (p<0,05), proved that the larger the follicle diameter, the higher the estrogen level and will improve the reproductive trait of PO cows.

Abstract

Jembrana disease, caused by the Jembrana virus, leads to high mortality (30%) and abortion (49%) in cattle, making vaccination essential. In this study, a multiepitope vaccine was designed using immunogenic TM and CA proteins. Predicted cytotoxic T lymphocyte, helper T lymphocyte, and linear B-cell epitopes were linked with flexible linkers, and the 50S L7/L12 ribosomal protein was added as a TLR4 agonist. In silico analysis confirmed the construct as non-allergenic, antigenic, and thermostable (aliphatic index 85.44), with 94.2% of residues in favored Ramachandran regions. Docking analysis revealed strong binding to TLR4 (−66 kcal/mol), and molecular dynamics simulations validated the structural stability. Immune simulations revealed increased antigen and antibody levels (IgM early, IgG1/IgG2 after day 15), progressive CD4+ T-helper expansion, transient CD8+ T-cell peaks, elevated IFN-γ and IL-2, and strong dendritic cell activation through MHC I and II pathways. These findings indicate the vaccine effectively stimulates humoral and cellular responses, supporting its potential as a promising candidate against the Jembrana virus.
