Abstract

Penelitian ini bertujuan untuk mengisolasi amilase dari biji mangga (Mangifera indica L.), mengetahui kadar protein, mengetahui kondisi optimum (derajat keasaman, suhu dan konsentrasi substrat), serta aktivitas spesifik amilasenya. Penentuan kadar protein total dilakukan dengan menggunakan metode Bradford. Penentuan kondisi optimum dan aktivitas spesifik amilasenya dilakukan dengan menggunakan metode asam dinitrosalisilat (DNS). Hasil penelitian ini menunjukkan kadar protein total yang terdapat dalam ekstrak kasar enzim biji mangga (Mangifera indica L.) adalah 3.176,9 μg/mL, aktivitas amilase sebesar 0,068 U/mL dan aktivitas spesifiknya sebesar 0,858 U/mg. Kata Kunci: Kecambah Biji Nangka (Artocarpus heterophyllus Lam), Ekstrak Kasar Enzim, Protein Total dan Amilase.

Abstract

Keterbatasan sumber daya air bersih yang dihadapi oleh masyarakat sekitar tambang menjadi dilema tersendiri, kondisi ini membuat masyarakat mengunakan air void sebagai sumber air baku untuk kebutuhan sehari-hari. Pembukaan lapisan tanah dalam proses penambangan telah membuat mineral di dalam tanah terbuka. Hal ini akan menyebabkanterjadi oksidasi mineral sulfida (pirit-FeS2) yang membawa kandungan-kandungan logam berat berbahaya, seperti timah hitam (Pb), seng(Zn), arsen (As). Kondisi ini diduga akan menimbulkan gangguan kesehatan,jika dikonsumsi masyarakat. Berdasarkan hasil studi pendahuluan yang dilakukan di Pondok pesantren Darul Fatta Loa Buah, Kota Samarinda menunjukkan bahwa air bekas void batu bara sudah dimamfaatkan sejak tahun 2019 sampai dengan saat ini. Tujuan dari penelitian ini untuk mengevaluasi kesesuaiaan kualitas air dari lubang bekas tambang batu bara dan mengidentifikasi keluhan kesehatan santri serta upaya yang dilakukan untuk mengelola air kolam pasca tambang sehingga memenuhi sayarat air bersih. Metode penelitian yang dilakukan mengunakan teknik penelitian observasional deskriptif dengan desain penelitian Cross Sectional dan pengumpulan sampel mengunakan metode total sampling yang berjumlah 98 santri. Berdasarkan hasil uji didapatkan tiga parameter yang tidak memenuhi standar yaitu kekeruhan, pH dan COD, dengan status kesehatan santri 60,2 % mengalami keluhan ringan , 33,7 % mengalami keluhan sedang dan 6,1 % mengalami keluhan berat dan untuk upaya pengelolaan air dengan menambahkan kaporitKata Kunci : Parameter Air, Status Kesehatan

Abstract

Blood cholesterol levels wh ich were higher than normal is trigger of various diseases. The objective of the research was to know “The Effect of Mackerel (Scomberomorus co mmerson) in the Feed to the Decline in Blood Cholesterol Levels in Hypercholesterolemic Male Mice (Mus musculus). The study was conducted over 40 days using completely randomized design with treatments, the positive control, negative control, and the mice were given the addition of mackerel on a standard feed with 3 different levels so grouped in L1 (2:1); L2 (1:1) and L3 (1:2). The parameters measured are total cholesterol blood serum of mice with the CHOD-PAP method. Cholesterol levels are measured with a photometer 5010 at a wavelength of 500 nm. The result in the (-) 86 mg/d L; (+) 113 mg/d L; L1 102 mg/d L; L2 86 mg/dL; and L3 82 mg/dL.

Abstract

Isolation and determination of optimum amylase work condition has been conducted from Bogor taro (Colocasia esculenta (L.) shoot). The aim of this research is to understand the optimum work condition and specific activity of amylase from Bogor taro. Amylase was isolated using homogenization techniques and sentrifugation. The raw extract of enzyme that contain amylase produced by Bogor taro then used to determine the total protein concentration and tested its activity in pH condition, temperature and maximum substrate concentration in various ways. The determination of total protein concentration was done with Bradford method, while the optimum work condition and specifik activity of amylase was done with dinitrosalycylic acid (DNS) method. The result of this reseach show the total protein concentration in Bogor taro is 7,88 μg/mL. Optimum work condition in Bogor taro is on acidity level (pH) 6, 600C temperature and 6 mg/mL substrate concentration. Specific activity from Bogor taro is 9573,68 U/mL. Abstract

Fish is a source of omega-3 which is good for our health. This study used one type of fish that contains high omega-3 (Rastrelliger kanagurta L.). This fish as s feed against the decrease in total blood cholesterol levels in mice (Mus musculus) are hypercholesterolemia. There are 5 treatments used in this study, i.e positive control, negative control, and mice which given the addition of bloated fish on standard feed with 3 different treatments L1 (2:1), L2 (2:2), L3 (1:2). Parameters measurement of total cholesterol level of mice is using CHOD-PAP method. The principle used in determining the total cholesterol level of mice blood cholesterol obtained is (+) 142 ; (-) 86 ; (L1) 108 ; (L2) 87 ; (L3) 74 .The result of this study showed that bloated fish (Rastrelliger kanagurta L.) used to reduce the total blood cholesterol level of male hypercholesterolemia mice (Mus musculus). Abstract

The aim of this study was to determine the antio xidant and toxicity act ivities of n-hexane and ethyl acetate of Piper aduncum L. (family : Piperaceae). Antio xidant activities of the ext racts was assessed by means of DPPH free radical scavenging method used ascorbic acid as standard with IC 50 value was 4,841 μg/mL. Piper aduncum L. showed n-hexane extract demonstrated the good antioxidant activity with IC 50 value of 40,034 μg/ mL and the ethyl acetate extract is 59,838 μg/ mL. The Brine shrimp letality bioassay met od as sed to determine t e cytotoxicity activities. The LC 50 values of n-hexane and ethyl acetate extracts were 25,12 and 17,78 μg/ mL, respectively.

Abstract

Research on metal concentration of Pb, Mn and protein content in windu shrimp’ meat (penaeus sp.) analysis in Dondang river, kecamatan Muara Jawa have done. This research is aimed to determine the concentration of heavy metal Pb, Mn and concentration of protein inside windu shrimp based by size and different sampling location. Concentration of heavy metal Pb, Mn and protein are correlated using Least Square method. Concentration of heavy metal Pb and Mn are analyzed by using AAS (Atomic Absorption Spectrophotometry) and concentration of protein are analyzed by using Kjeldahl method. Result of this research is the concentration heavy metal Pb and Mn in sampling point 1 in average concentration are 0.556 mg/L and 0.463 mg/L, sampling point 2 are 0.790 mg/L and 0.738 mg/L, sampling point 3 are 0.876 mg/L and 0.905 mg/L. Concentration of protein in windu shrimp in sampling point 1 in average are 23.4928%, sampling point 2 are 28.1701% and sampling point 3 are 29.1622%. Correlation of concentration of heavy metal Pb and Mn to the concentration of protein in windu shrimp (penaeus sp.) with R = 0.791; r = 0.948. Abstract

Research on the validation of As determination methods on wellbore water by using atomic absorption spectrophotometer at PT. Geoservices Balikpapan has been calculated. The validation method used is APHA-3114 C, with research stages such as linearity, detection limit, quantitative limit, accuracy, precision. The results showed that the linearity test had good result with R2 ≥ 0.9997. The detection limit value was 0,000038 µg/Land the quantization limit value was 0,00129 µg/L. The %KV value obtained is smaller than the 2/3 KVHorwitz value, indicating that this method has a good precision. The determination of As in drilled well water samples is 0.9167 µg/L, then below the standard threshold of clean water quality quality based on Permenkes. 416/Menkes/PER/IX/1990. Based on the results of the research, APHA-3114 B/C method can be used to determine As in drilled well water samples. Keywords: Validation method, arsenic, wellbore water, APHA-3114B/C, AAS-VHGA.

Abstract

Amylase is an enzyme that can catalyze stach hydrlysis into its simplier molecule as glucose. Coconut’s buds (Coccos nucifera L.) have high enough of carbohydrate content which amylase could have existed. The research are to determine protein concentration, optimum catalyst function and specified activity of amylase in coconut’s buds (Coccos nucifera L.). Coconut’s buds (Coccos nucifera L.) are isolated by extraction method using phospate buffer solution 0,1 M pH 7. The determination of protein concentration are used Bradford method. The determination of optimum catalyst function are used dinitrosalicylate acid (DNS), optimum catalyst fuction which are determined include of optimum pH, optimum temperature and optimum substrate concentration. The determination of specified activity of amylase are tested in optimum condition. The result of research are obtained the protein content are 2,933 mg/mL. The specified activity which are obtained from coconut’s buds (Coccos nucifera L.) are 80,376 U/mg in pH 6, temperature 50 ºC and substrate concentration 6,5 mg/mL Keywords : amylase, coconut’s buds, dinitrosalicylate acid (DNS) and specified activity.

Abstract

Diabetes mellitus (DM) is a metabolic disorder and characterized by hyperglycemic circumstances. Hyperglycemic is a condition where the glucose levels ini the blood increased dramatically. One of the pharmacological therapy ia to use ofdrugs that can reduse glucose levels in the blood. Phytochemical test and antihyperglicemic activity from durian (Durio zibethunus Murr.) peel have been carried out. Durio zibethinus Murr. peel was macerated with ethanol 96% then concentrated with rotary evaporators. It obtained rendemen 13.33% . Based on the test result of phytochemistry of secondary metabolites compounds from durian peel showed that etahanol extract containing flavonoid, alkaloid, fenolik, triterpenoid and saponin. Antihyperglicemic activity test use animal test male mice that is injected aloksan. Measurement of blood sugar levels using glucometer and glucose test strip.
