Abstract

Koto Masjid Village is the largest production center in striped catfish fish farming activities in Riau Province. One of the inhibiting factors is the disease caused by bacteria, this study aims to determine the types of pathogenic bacteria that have the potential to infect striped catfish. This research was conducted in August - November 2019, samples of striped catfish used in the study amounted to 15 tails with an average length of 15-25 cm taken from 5 ponds in Kampung II, Koto Masjid Village, XIII Koto Kampar Subdistrict, and then identified bacteria by Conventional Laboratory in Fish Parasites and Diseases, Faculty of Fisheries and Marine, Riau University, Pekanbaru. This study uses a survey method that is by taking samples from the field in Pu and then the isolation of bacteria is carried out from the kidney organ in the TSB media then purified on the TSA media. The results of the study found 3 types of pathogenic bacteria namely Aeromonas sp., Edwardsiella sp., and Pseudomonas sp. Water quality during the study was pH 6.6-70, DO 4.05-4.35 mg/L, NH3 0.035-0.2 mg/L and Temperature 28-300C.

Abstract

Herbs Fermented consist of Kaempferia galanga L, Curcuma xanthorrhiza Roxb, and Curcuma domestica Val, which are environmentally friendly disease prevention alternatives that can be added to feed because they have several advantages, such as increasing appetite, digestion, growth, disease resistance, and reducing stress levels in fish. This research was carried out from August to December 2021 at the Marine Science Biotechnology Laboratory and Fish Parasites and Diseases Laboratory, Faculty of Fisheries and Marine, Universitas Riau. This study aimed to obtain the best dose from adding fermented herbs to increase the immune system of Oreochromis niloticus and prevent diseases caused by Aeromonas hydrophila bacteria in terms of profile erythrocytes. The method used was an experimental method using a Completely Randomized Design (CRD) with one factor, five levels of treatment, and three replications so that 15 experimental units were obtained. The challenge test was carried out on the 32nd day using A.hydrophila with a density of 108 CFU/mL. The best dose was found in P3 with a dose of 200 mL/kg of feed, which showed a total erythrocyte of 2.09x106 cells/mm3, hemoglobin level of 7.70 g/dL, hematocrit value of 37.70%, and a normal erythrocyte morphology. Water quality ranged from temperature 27-29oC, pH 6.9-7.2, DO 3.1-4.4 mg/L, and Ammonia 0.016-0.024 mg/L. Based on the data obtained, it can be concluded that fermented herbs can increase the immunity of O. niloticus to diseases caused by A.hydrophila.

Abstract

Multi-cell probiotic containing bacteria Nitrosomonas sp., Nitrobacter sp., and Bacillus sp. This research was conducted from March to May 2021 at the Fish Disease and Parasite Laboratory, Department of Aquaculture, Faculty of Fisheries and Marine, Universitas Riau. This study aimed to analyze the hematology of Asian redtail catfish reared in media that was added with probiotics periodically and to obtain the best time interval for giving probiotics to the rearing medium. The method used in this study is an experimental method by applying a Completely Randomized Design (CRD) with five treatment levels and three replications. The treatments used were the administration of probiotics at a dose of 0,01 mL/L of water with different time intervals, P0 (without treatment), P1 (5 days once), P2 (10 days once), P3: (15 days once), and P4 (20 days once). Fish were reared for 50 days. The results show that the Asian redtail catfish reared with the addition of probiotics in the rearing medium had a significant effect (P<0.05) with the best time interval being once every 5 days in the P1 treatment, seen from the total erythrocytes 2.43 x 106 cells/mm3, hematocrit level 28.67%, hemoglobin level 7.40 g/dL, total leukocytes 9.92 x 104 cells/mm3, leukocrit level 1.83%, and glucose 49.67 g /dL. Water quality during the study was still in the normal range that could support the growth of Asian redtail Catfish, namely temperature 26-27.1 C, pH 5.8-6.5, DO 6.0-7.4 mg/L, and ammonia 0.0011-6.5 0.0047 mg/L.

Abstract

Aeromonas hydrophila bacteria often attack internal organs such as the liver and intestines. This research was conducted in June – August 2021 at the Laboratory of Fish Parasites and Diseases, Faculty of Fisheries and Marine, Universitas Riau. Histology preparations were made at the Bukittinggi Veterinary Center. This research aimed to obtain the best dose addition of papaya leaf fermented solution which could reduce intestinal and liver tissue damage of striped catfish (P.hypophthalmus) which was challenged with A.hydrophila. The method used is an experimental method with a completely randomized design (CRD) with 4 treatment levels and 3 replications; Kp (Feed not given papaya leaf fermented solution + infected with A.hydrophila), P1 (16 ml/kg feed of papaya leaf fermented solution + infected with A.hydrophila), P2 (16 ml/kg feed of papaya leaf fermented solution) + infected with A.hydrophila), P3 (16 ml/kg of papaya leaf fermented solution feed + infected with A. hydrophila). The fish used was 8-10 cm with a density of 1 fish/3 L totaling 10 fish with a container measuring 40x30x30 cm. Infection by injection in the intramuscular section as much as 0.1 mL/head with a bacterial density of 108 CFU/mL. The parameters observed were clinical symptoms for 72 hours and histopathology of the intestine and liver of striped catfish. The results showed that the intestine saw the presence of hemorrhage, necrosis, and edema. While the liver shows abnormalities, such as hemorrhage, necrosis, hypertrophy, and fatty degeneration.

Abstract

Lactic acid bacteria can improve digestibility because they can be absorbed directly by the body. The purpose of this research was to obtain lactic acid bacteria types from the intestines of striped catfish (Pangasionodon hypophthalmus) that had been fed fermented herbal feed. This research was conducted from June 2019 to April 2020. The treatment used in this research was with different doses and stocking densities. The test was carried out using the disc diffusion method or the Kirby-Bauer method, which was done by measuring the inhibition zone around the paper disc. Identification of LAB was carried out by phenotypic and genotypic. The results showed that as many as four of the selected isolates were able to inhibit the growth activity of the pathogenic bacteria Aeromonas hydrophila. Isolates with code K had an inhibition zone of 6.7 mm against A.hydrophila, an inhibition zone of 7.5 mm for P1D2 isolates, P2D1 isolates had an inhibition zone of 7.0 mm, P2D2 isolates had an inhibition zone of 7.8 mm. Types of LAB that were identified phenotypically and genotypically include Lactobacillus casei, L.plantarum, and L. acidhopilus

Abstract

Desa Teluk Rhu merupakan salah satu desa yang terdapat di kecamatan Rupat Utara, yang memiliki potensi yang tinggi di bidang perikanan dan wisata bahari. Desa Teluk Rhu memiliki jumlah penduduk 1.885 orang yang terdiri dari 364 KK. Salah satu masalah saat ini ini adalah masih rendahnya pengetahuan keluarga nelayan terhadap pengolahan hasil tangkapan nelayan. Tujuan pengabdian adalah untuk meningkatkan pengetahuan dua kelompok istri nelayan Desa Teluk Rhu tentang pengolahan hasil perikanan menjadi produk olahan yaitu nugget, bakso, kemplang dan stik ikan dari ikan hasil tangkapan. Metode yang digunakan pada kegiatan ini adalah dengan metode ceramah, diskusi dan praktek pembuatan produk olehan ikan dan evaluasi. Materi penyuluhan yang disampaikan adalah: 1. Pentingnya packaging dan performance barang jualan, 2. Pembuatan keplak, 3. Pembuatan stik ikan. 4. Pembuatan otak-otak, 5. Pembuatan bakso bagi kelompok istri nelayan. Peserta diikuti dua kelompok keluarga nelayan. Dari hasil pengabdian dapat disimpulkan pelatihan sangat bermanfaat bagi peserta, Dapat meningkatan perolehan hasil tambahan, peserta menganggap bahwa 4 produk yang sudah dikenalkan bisa menjadi produk khas desa teluk Rhu Kecamatan Rupat utara, jika pembinaan terus menerus bisa dilaksanakan.

The method used is an experiment with a completely randomized design (CRD) one factor five levels of treatment. The doses used were negative control (without immersion cherry leaf solution and not infection with Aeromonas hydrophila), positive control (without immersion cherry leaf solution and infection with A.hydrophila), P1 (Immersion in cherry leaf solution at a dose of 2800 ppm), P2 (3,200 ppm), and P3 (3,600 ppm). Immersion was carried out 5 times for 5 minutes with an interval of 7 days. The results showed that immersion with cherry leaf solution at a dose of 3,600 ppm (P3) was the best dose (P<0.05) with a total leukocyte value of 10.66 x 104 cells/mm3, lymphocytes 84.67%, monocytes 5.67%, neutrophils 9.67%, phagocytic activity 36.67%, and survival rate 86.67%

Abstract

Lactic Acid Bacteria (LAB) are probiotic-forming bacteria that have characteristics, like be able to ferment sugar or carbohydrates and produce lactic acid as the final product of fermentation. The purpose of this study was to obtain isolates of lactic acid bacteria from the digestive tract of Asian Swamp Eel (Monopterus albus) and to determine the inhibition produced by these lactic acid bacteria in inhibiting pathogenic bacteria such as Aeromonas hydrophila and Pseudomonas sp. The method was used in this study is experimental method, by taking the digestive tract of Asian Swamp Eel and then identifying the lactic acid bacteria and characterized by morphological and biochemical characteristics and tested of the antagonist against the pathogenic bacteria A.hydrophila and Pseudomonas sp. using the Kirby Bauer disc method (disc method). From the research results obtained 5 types of candidate lactic acid bacteria with each code BL121, BL263, BL142, BL242 and BL342. The five isolates of lactic acid bacteria were able to inhibit A.hydrophila and Pseudomonas sp. with a Inhibition range of 8-16 mm. Based on the results of the study, there were three isolates that had inhibitory power against bacteria A. hydrophila (12-16 mm) and Pseudomonas sp (11-13 mm) with codes BL121, BL263 and BL142 and were classified as strong. The identification results obtained are bacteria from the genus Bacillus sp.

Abstract

Penelitian ini dilaksanakan pada bulan Desember 2019 s/d Februari 2020 bertempat di Laboratorium Parasit dan Penyakit Ikan Fakultas Perikanan dan Kelautan Universitas Riau. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan larutan daun kersen (Muntingia calabura L.) terhadap gambaran leukosit dan mendapatkan dosis terbaik penambahan larutan daun kersen ke dalam pakan. Metode yang digunakan adalah metode eksperimen dengan menggunakan Rancangan Acak Lengkap (RAL) dengan lima taraf perlakuan yaitu Kn= Kontrol negatif (pemberian pakan tanpa penambahan kersen), Kp= Kontrol positif (pemberian pakan tanpa penambahan kersen), P1= penambahan larutan daun kersen dengan dosis 1 mL/kg Pakan, P2= penambahan larutan daun kersen dengan dosis 2 mL/kg pakan dan P3= penambahan larutan daun kersen dengan dosis 3 mL/kg pakan. Hasil penelitian menunjukkan larutan daun kersen berpengaruh nyata terhadap gambaran leukosit ikan jambal siam (Pangasianodon. hypophthalmus) dilihat dari adanya peningkatan total leukosit, diferensiasi leukosit, dan aktivitas fagositosis (P<0,05). Dosis terbaik terdapat pada perlakuan P3 (3 mL/kg pakan) dengan rata-rata jumlah leukosit 9,38x104 sel/mm3, aktivitas fagositik sebesar 31,67%, dan tingkat kelulushidupan mencapai 86,67%. Sedangkan persentase limfosit 83,00%, monosit 9,00% dan neutrofil 8,00%.
