Abstract

COVID-19 pandemic has no immediate ending in sight, and any significant increasing cases were observed worldwide. Currently, there are only two main strategies for developing COVID-19 drugs that predominantly use a proteomics-based approach, which are drug repurposing and herbal medicine strategies. However, a third strategy has existed, called small interfering RNA or siRNA, which is based on the transcriptomics approach. In the case of SARS-CoV-2 infection, it is expected to perform by silencing the viral gene, which brings the surface glycoprotein (S) gene responsible for SARS-CoV-2 viral attachment to the ACE2 receptor on the human host cell. This third approach applies a molecular simulation method comprising data retrieval, multiple sequence alignment, phylogenetic tree depiction, 2D/3D structure prediction, and RNA-RNA molecular docking. The expected results are the prediction of 2D and 3D structures of both siRNA and mRNA silenced S genes along with a complex as the result of a docking method formed by those silenced genes. An Insilco chemical interaction study was performed in testing siRNA and mRNA complex’s stability with the confirmation result of a stable complex which is expected to be formed before mRNA reaches the ribosome for the translation process. Thus, siRNA from the S gene could be considered a candidate for the COVID-19 therapeutic agent.

Abstract

Justicia gendarussa Burm.f., a medicinal plant, is Acanthaceae that has many functions. Furthermore, the compounds in gendarussa must be produced in high quantity and quality by applying callus culture method. Accordingly, it is important to study the effects of plant growth regulators (2,4-D, IBA, and BAP) on callus induction of gendarussa leaves. This research design utilized a factorial design with two factors (2,4-D and IBA: 0.5, 1, 1.5 mg/L and BAP: 0.5, 1, 1.5, 2 mg/L). The experiment consisted of 24 treatments, each of which was repeated 3 times. Observation was carried out in 6 weeks. Data on the time of callus formation, percentage of explants formed callus, and callus morphology were analyzed descriptively,while data on fresh and dry weight were analyzed by Two-Way ANOVA (α = 0.5). Interestingly, the results showed that various concentration of plant growth regulators (2,4-D, IBA, and BAP) affected callus induction from leaf explants of gendarussa. We concluded that the most optimal treatment combination of concentration of plant growth regulators in inducing callus from leaf explants of gendarussa is 1.5 mg/L 2,4-D and 2 mg/L BAP with a relatively long period of callus formation at the earliest, i.e. on day 5, 2.247 g of fresh weight, 0.108 gof dry weight, white callus translucent, and friable. Moreover, the optimum treatment will be used to produce secondary metabolite and seed synthetic by cell suspension culture.

Abstract

Penyakit yang disebabkan oleh kelompok Vibrio spp. selama program budidaya Litopenaeus vannamei berpotensi menyebar ke perairan melalui perpindahan larva, kontaminasi silang, hingga tingginya padat tebar. Penelitian ini bertujuan untuk menyelidiki kualitas air dan kelimpahan Vibrio spp. pada L. vannamei yang dibudidayakan menggunakan sistem super-intensif. Evaluasi fluktuasi kualitas air dan bakteri Vibrio spp. dilakukan dengan pengukuran parameter kualitas air (suhu, oksigen terlarut, salinitas, nilai pH, dan amoniak) pada media pemeliharaan L. vannamei. Pemeriksaan Vibrio spp. dilakukan pada air pemeliaraan dan tubuh udang L. vannamei dengan mengkultur pada media selektif CHROMAgar serta identifikasi pada kenampakan hepatopankreas. Pemantauan dilakukan mulai dari Days of Culture (DOC) 30 – 150. Hasil penelitian menunjukkan bahwa nilai DO pada pemeliharaan hari ke-150 cenderung mengalami peningkatan, yang diikuti dengan peningkatan nilai suhu dan kadar amoniak secara signifikan dibandingkan dengan usia pemeliharaan 30 hari (p≤0.05). Tidak terdapat perbedaan yang signifikan pada nilai pH dan salinitas pada DOC 30 dan 150. Cemaran Vibrio spp. pada media pemeliharaan cenderung tinggi pada DOC 30, sedangkan cemaran V. parahaemolyticus dan V. cholera cenderung tinggi pada DOC 90 dan 60 pada tubuh L. vannamei. Cemaran Vibrio spp., pada L. vannamei dapat dikaitkan dengan tingginya padat penebaran, berdampak pada penurunan DO dan peningkatan suhu dan amoniak pada system budidaya super-intensif.
