Abstract

The study aims to determine and analyze the relationship between breeders’ knowledge about feed formulations and the conversion value of duck feed. The subjects in this study were Alabio duck breeders in Hambuku Raya Village and Putat Atas Village, Hulu Sungai Utara, South Kalimantan Province, Indonesia with observed objects in the form of Alabio duck feed conversion and breeders’ knowledge of feed formulation knowledge and Alabio duck feed technology. The research method used was non-experimental explorative, the researcher did not give treatment to the group of Alabio ducks and conducted a survey by distributing questionnaires to farmers. Observation of feed conversion is carried out for 3 (three) months from February-April 2018 in each group. Observation of feed conversion is carried out starting from Alabio ducks to produce eggs, which are about 6 months old. Testing of feed conversion, normal data using analysis of variance (Anova), if significant continued with LSD. Survey research is the stage of analyzing using SWOT analysis to determine the level of weakness and strength in the framework of the development of Alabio ducks in the Hulu Sungai Utara Regency of South Kalimantan Province. The results of the analysis of feed conversion of the three groups were not significantly different, the group of Putat Atas Ducks for feed conversion an average of 4.51 ducks Hambuku Raya 1 group an average of feed conversion every month 5.30, the ducks of the Hambuku Raya 2 group the average conversion of feed every month 4.75. SWOT analysis value of weakness factor is 1,764, this factor includes one of them is knowledge of farmers about feed formulation. The results of this study are in line with the results of feed conversion in the first stage of research, the value of feed conversion obtained is still relatively high at 4.51-5.30. The conclusion is the SWOT analysis score of weakness factors one of which is the lack of knowledge of farmers about formulation and feed technology affect the high feed conversion.

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui daya anthelmintik dari ekstrak etanol daun kemangi (Ocimum americanum Linn.) terhadap mortalitas cacing Haemonchus contortus secara in vitro. Sediaan ekstrak dibuat dengan konsentrasi (berat/volume) 2,5%, 5%, 7,5%, dan 10%. Untuk kontrol positif digunakan PBS dan DMSO sedangkan untuk kontrol negatif digunakan Albendazole 10 mg/ml. Sebanyak 10 ekor cacing Haemonchus contortus betina direndam dengan berbagai perlakuan dalam cawan petri. Masing-masing perlakuan diulang sebanyak 4 kali. Pengamatan dilakukan setiap 1 jam sekali selama 6 jam untuk melihat apakah cacing masih hidup atau sudah mati. Hasil dari penelitian ini menunjukkan bahwa ekstrak daun kemangi memiliki efek anthelmintik terhadap cacing Haemonchus contortus secara in vitro. Makin tinggi konsentrasi ekstrak, maka daya anthelmintiknya juga semakin tinggi. Pada ekstrak konsentrasi 10% memiliki kemampuan anthelmintik yang hampir sama dengan Albendazole.

Abstract

Research of gastrointestinal protozoan in cats at Surabaya are still limited. Protozoa infection of the gastrointestinal tract can cause diarrhea and even zoonosis. This research aimed to identify and determine the prevalence of protozoan in cats in animal hostptal and animal clinic in Surabaya. Ninety  fecal samples were collected from 2 animal clinic and one animal hospital. Samples were  examined e wet mount (native, sedimentation, and floatation) and (Ziehl Nellsen) stain. Protozoa was identified by using a light microscope with 400x and 1000x magnification. The result showed 68,89% of samples were positively infected by gastrointestinal protozoa. The protozoa were Blastocystis sp, Cryptosporidium sp, Giardia sp, and Eimeria sp. By chi square test, there was  not significant differences the prevalence of gastrointestinal protozoan in cat between sex, age, breed, and diarrhea status

Abstract

Penelitian ini merupakan penelitian deskriptif dengan menggunakan 20 itik Alabio betina siap bertelur dengan rentang usia 6 bulan dan untuk pemeriksaan kualitas telur untuk masing-masing kelompok 10 sampel. Pengamatan tingkat produksi telur dilakukan selama tiga bulan dengan 10 pengulangan, kualitas telur (ketebalan kulit telur, persentase kulit telur, persentase kulit telur, persentase albumin, persentase kuning telur, huagh unit, warna kuning telur, kadar protein dan lemak kandungan). Data produksi telur dan kualitas telur diuji menggunakan uji Anova. Hasil pengamatan rata-rata produksi harian telur 15.45 ± 0.12, 14.72 ± 0.10, 14.91 ± 0.09; produksi berat harian (kg/hari) 925.27 ± 7.50, 875.40 ± 6.32, 884 ± 5.03; hasil dari ketebalan kulit telur kualitas 0.33 ± 0.01, 0.33 ± 0.12, 0.33 ± 0.12; persentase kulit telur 11.32 ± 0.93, 11.28 ± 0.10, 11.36 ± 0.05; persentase albumin 55.70 ± 1.08, 56.66 ± 1.22, 56.12 ± 1.00; persen kuning telur 32.07 ± 1.24, 31.55 ± 1.48, 31.64 ± 1.20; Haugh unit 88.70 ± 6.21, 91.41 ± 6.70, 94.51 ± 5.06. Warna kuning masing-masing memiliki skor 15. Dapat disimpulkan bahwa produksi dan kualitas telur itik Alabio baik.

Abstract

This study aimed to provide alternative information and solutions in an effort to increase reproductive productivity in etawah crossbreed goats (PE). The sample used in this study was 10 female PE with an average age of 2.5 - 3 years and primiparous at least . Ovulation induction was performed using 0.1 mg intra-muscular Gonadotropin-Releasing Hormone (GnRH), after seven days injection of Prostaglandin F2α (PGF2α) was given as much as 2.5 mg submucosa of the vulva, followed by a second injection of GnRH as much as 0,2 mg intramuscular in samples that have really experienced heat and selected selectively according to the signs of natural heat shown by the sample and followed by insemination as much as 2 doses or 0.50 ml of frozen semen. Blood sampling was performed at H0, H7th, H14th and H21th after insemination. All blood samples were collected and progesterone and estrogen hormone profiles were examined using the ELISA method. From the results of the ELISA test, the mean progesterone hormone profile H0 = 4.798 ng / ml, H7th = 4.887 ng / ml, H14th = 4.824 ng / ml, H21th = 5.148 ng / ml. The profile of the hormone estrogen at H0 = 19,461 pg / ml, H7th = 17,457 pg / ml, H14th = 18,248 pg / ml, H21th = 17,515 pg / ml. This study showed an increase in the levels of the progesterone hormone at H0 to H7th, then slightly decreased in H14th and a significant increase in H21th. For the estrogen hormone, there is a decrease from H0 to H7th then there is a slight increase in H14th and decreases again in H21th.

Abstract

This study aims to determine the effect of hepatitis B vaccine of spleen weight and white pulp diameter in rat (Rattus norvegicus). This study used 20 male wistar rats aged 8 weeks with an average weight of 200-250 grams, which was divided into four groups. Control group (P0) was given PBS solution, treatment group 1 (P1) was given type 1 hepatitis B vaccine, treatment group 2 was given type 2 hepatitis B vaccine, and treatment group 3 (P3) was given type 3 hepatitis B vaccine. Dose administered 0.4 ml and given repeatedly at days 0,30, and 60. Animals model to euthanize on day 75 for sampling. The spleen organ is weighed after the necropsing process and then the spleen is stored for the preparation of histopathologic with HE staining (Hematoxylin Eosin). The data obtained are weight data of spleen organ and spleen white pulp diameter. The results showed the highest weight of spleen organ 0.92 gram in group P0 and the largest white pulp diameter 34,34 μm in group P1. The results of data analyzed using one-way ANOVA (Analysis of Variance) and continued with Duncan alpha test. One Way Anova and Duncan alpha test showed that there were non significantly different between groups (p>0.05). The conclusion is we know that hepatitis B vaccine type 1,2, and 3 can't give the effect for spleen weight and white pulp diameter in rat (Rattus norvegicus).

Abstract

Monosodium glutamate (MSG) could cause increased production of reactive oxygen species (ROS), causing oxidative stress in the testicles, which adversely affected sperm quality. Dayak onion extract which is known for its high antioxidant content, could help alleviate oxidative stress caused by MSG. This research examined the effect of Dayak onion (Eleutherine palmifolia) on sperm abnormalities and plasma membrane integrity in mice (Mus musculus) exposed to MSG. Twenty-five male mice, 8 weeks old with a body weight approximately 20g, were divided into five groups, C– (received 0.5% Na-CMC), C+ (received 4 mg/g bw MSG), and three treatment groups (T1, T2, T3) which were given 4 mg/g bw MSG accompanied by Dayak onion extract at doses of 30, 60, and 120 mg/kg bw respectively. All treatments lasted for 52 days. Significant differences (p <0.05) in sperm abnormalities and plasma membrane integrity were observed among the groups. Sperm abnormalities found were (12.3 ± 1.92), (61.1 ± 3.10), (41.6 ± 2.87), (30.4± 1.91), and (18.2 ± 2.10) % respectively for C–, C+, T1, T2 and T3. Meanwhile, sperm plasma membrane integrity found were (33.0 ± 3.24), (69.3 ± 2.32), (41.8 ± 2.42), (55.4 ± 3.11), and (64.2 ± 1.27) % respectively for C–, C+, T1, T2 and T3. These results indicate that Dayak onion extract could help reduce sperm abnormalities and maintain plasma membrane integrity in mice exposed to MSG.

Abstract

BACKGROUND: During COVID-19 pandemic, effective preventive and therapeutic strategies was urgently necessary, especially with the emerging Delta variants. Previous existing interventions have some limitations, and passive immunization using immunoglobulin Y (IgY) is considered viable. However, the potential of IgY antibodies remains underexplored locally. Therefore, a feasible study was conducted to produce IgY against the SARS-CoV-2 Delta variant from chickens raised on a farm, instead of a laboratory environment, by using in-house recombinant protein, to offer a promising low-cost alternative for passive immunization.METHODS: In-house HEK293 cell line was produced by secreting recombinant S1 subunit protein of SARS-CoV-2 Delta variant. The protein was confirmed by western blot and used as an immunogen in two chickens. IgY was extracted from egg yolks. Titers and neutralization activity of IgY against SARS-CoV-2 Delta variant were confirmed by enzyme-linked immunosorbent assay (ELISA) and surrogate virus neutralization test (sVNT). The functionality of IgY was then tested on lysates from COVID-19 patients' nasopharyngeal swabs with dot blot method.RESULTS: The high anti-S1 IgY titers and neutralization activity was confirmed following immunization with 1 mg immunogen at 2–3-week intervals. IgY titers varied between chickens and time points. The mean titers showed significant increase after the fifth immunization (1.21, 1.72 and 1.48; with p<0.05 for all). Neutralization activity appeared after the second immunization and was significant after the third immunization (31.9%, 34.95%, 26.9%, 47.6%, 54.95% and 57.3%; with p<0.05 for all). The results of this study showed that the extracted IgY reacted to COVID-19 patients' nasopharyngeal swabs lysates.CONCLUSION: A high-yield and functional IgY anti-S1 SARS-CoV-2 Delta variant can be produced from chicken immunized with an in-house recombinant protein. Abstract

This study aimed to detect the presence of Salmonella sp. in broiler chickens raised in closed house systems using the Polymerase Chain Reaction (PCR) method. Liver and intestinal samples were collected from chickens showing clinical signs such as diarrhea, anorexia, and lethargy, along with pathological lesions observed during necropsy. Bacteriological identification involved isolation using selective media, Gram staining and biochemical tests. Two samples (3A and 3B) showed colony morphology and biochemical characteristics consistent with Salmonella sp. Confirmation using PCR targeting the invA gene (primers 139 and 141, expected amplicon size 284 bp) yielded negative results in both samples, while the positive control successfully amplified the target. The inconsistency between bacteriological and molecular results may be attributed to several factors: absence or mutation of the invA gene in the tested isolates, primer mismatch with local Salmonella strains, or inadequate DNA quality and concentration. These findings suggest that reliance on a single molecular marker may be insufficient for accurate detection and underscore the importance of optimizing PCR conditions. The study highlights the need for locally adapted primers and complementary diagnostic approaches to improve the reliability of Salmonella sp. detection in poultry, particularly in intensive production systems like closed house environments.
