Abstract

Karakterisasi Enzim α-Amilase Penggerek Batang Padi Kuning (Scirpophaga Incertulas) Pada Tanaman Padi Di Jember Moh. Nizar Sahadati A. Program Studi Agroteknologi, Fakultas Pertanian, Universitas Jember Jl. Kalimantan 37, Kampus Tegal Boto, Jember 68121 muhammadamri59@gmail.com ABSTRAK Padi sebagai tanaman pangan nasional sangat memerlukan perhatian sebagai negara agraris sangatlah penting untuk selalu memikirkan dan melakukan inovasi demi tercapainya swasembada pangan nasional. Sebagai tanaman pangan utama di Indonesia, padi memiliki banyak hama dari jenis serangga salah satunya adalah Penggerek batang padi kuning (Scirpophaga incertulas). Enzim adalah sekelompok protein yang berperan sebagai pengkatalis dalam reaksi-reaksi biologis. Enzim dapat juga didefenisikan sebagai biokatalisator yang dihasilkan oleh jaringan yang berfungsi meningkatkan laju reaksi dalam jaringan itu sendiri. Enzim α-Amilase menghidrolisis ikatan α-1,4 glikosidik amilosa, amilopektin dan glikogen. Enzim ini bersifat sebagai endoamilase, yaitu enzim yang memecah pati secara acak dari tengah atau bagian dalam molekul. Berat molekul α-amilase rata-rata ± 50 KD. Enzim ini mempunyai rantai peptida tunggal pada gugusan proteinnya dan setiap molekul mengandung satu gram atom Ca. Kalsium yang berikatan dengan molekul protein enzim, membuat enzim α-amilase bersifat relatif tahan terhadap suhu, pH, dan senyawa seperti urea. Aktivitas α-amilase dapat diukur berdasarkan penurunan kadar pati yang larut, kadar dekstrin yang terbentuk, dan pengukuran viskositas atau jumlah gula pereduksi yang terbentuk. Adapaun metode penelitian yang akan dilaksanakan terdiri dari beberapa pengujian yaitu : isolasi dan pemurnian α-amilase dari penggerek batang kuning, perhitungan protein terlarut, aktifitas enzim α-amilase, pengaruh suhu terhadap enzim α-amilase, pengaruh konsentrasi substrat terhadap enzim α-amilase. Hasil analisa data akan dianalisis menggunakan analisa statistik deskriptif. Kata Kunci : Padi, Penggerek Batang Padi Kuning, Enzim α-amilase

Abstract

Vermicompost is a fertilizer produced by composting organic matter by utilizing the interaction between earthworms and microorganism. Vermicompost also contains a number of macro and micro nutrients needed by plants. Besides containing macro and micro of nutrients, vermicompost also containing growth hormone such as cytokinins and auxins which are produced by worms during the composting process. The benefits of vermicompost are that it can improve the physical, chemical and biological properties of soil, increase the ability of the soil to retain water, rebuild damaged soil structures and increase ability of the soil to exchange cations. The fermentation process lasted for 14 days using Lumbricus rubellus worms. The choice of this type worm is based on its character which has a high level of productivity, can eat all types of organic matter, is much more adaptive and move less actively. This research was held in Sumbersalak Village, Ledokombo District, Jember Regency and also involving the Kenconowungu farmer group. Goat manure become waste in this village so it has potential to be developed further as the main ingredient for vermicompost. The other additional materials such as vegetable and fruits waste, and leaf litter were used as factor (treatment). These three types of materials are used to find out which treatment gives the best vermicompost quality. Chemical analysis was carried out in this study to determine the organic N, P, K, C organic, pH, and water content. The result of the analysis will be compared with the standards of Minister of Agriculture No.70 of 2011.

Abstract

Tobacco plants are an important plantation commodity in Indonesia. However, problems often occur in cultivation. These obstacles include abiotic and biotic stress. To overcome this problem, it is necessary to improve plant varieties through molecular-based breeding programs. This biotechnology program needs to carry out optimization research on the DNA isolation process so that the results produced can be used for further analysis. The method used uses a comparison of extraction techniques using tools and grinding with chemicals. Meanwhile, tobacco leaf samples were used with a comparison of wet samples and dry samples. From the results obtained, the treatment of the grinding method with chemicals and wet samples was able to increase the quality and quantity of DNA concentration.

Abstract

Implementation of community service regarding plant breeding was carried out in Sumbersalak Village, Ledokombo District with the Kenconowungu farmer group. The activities in this service program are to provide knowledge and understanding related to rice plant breeding. Breeding is carried out by crossing rice plants with the aim of producing superior rice varieties in accordance with the problems of farmers in Sumbersalak village. To improve theoretical mastery of the material, the method used is through material presentation activities, discussion sessions and evaluation. To facilitate the transfer of mastery of plant crossing skills in the field to target farmers, the direct practice method is used. The results of the service evaluation show that all target audiences have experienced a change in mastery of knowledge from not knowing and not understanding to knowing and understanding how to cross rice plants

Abstract

AbstrakTanaman krisan varietas Suciono merupakan tanaman hias yang potensial dikembangkan dan diminati oleh penduduk dunia, yang ditunjukkan dengan permintaan pasar terhadap tanaman krisan meningkat sepanjang tahun. Hal ini menyebabkan perbanyakan krisan secara in vitro sangat penting untuk dilakukan. Penelitian ini bertujuan untuk mengetahui pengaruh Indole Acetic Acid (IAA) dan Benzly Amino Purine (BAP) terhadap multiplikasi tunas krisan varietas Suciono. Desain penelitian ini menggunakan Rancangan Acak Lengkap dengan satu faktor yakni kombinasi hormon IAA dan BAP yang 9 kombinasi, yakni 0 mg/L +  1,5 mg/L; 0 mg/L + 2 mg/L; 0 mg/L + 2,5 mg/L; 0,5 mg/L + 1,5 mg/L; 0,5 mg/L + 2 mg/L; 0,5 mg/L + 2,5 mg/L; 1 mg/L + 1,5 mg/L; 1 mg/L + 2 mg/L; dan 1 mg/L + 2,5 mg/L dimana setiap kombinasi diulang sebanyak 3 kali. Variabel yang diamati meliputi analisis histologis kalus, waktu muncul tunas, jumlah tunas, tinggi tunas, dan jumlah daun. Hasil analisis ragam atau ANOVA menunjukkan bahwa kombinasi IAA 0 mg/L dan BAP 2 mg/L merupakan kombinasi konsentrasi terbaik untuk multiplikasi tunas. Penambahan BAP 2 mg/L mampu menghasilkan persentase muncul tunas. 100%, jumlah tunas 3 per-eksplan, tinggi tunas 7 cm, dan jumlah daun sebanyak 16 helai.AbstractThe variety Suciono of chrysanthemum is a potential ornamental plant that is in demand by the world's population, as indicated by the increasing market demand for chrysanthemum plants throughout the year. This  has led to the importance of in vitro propagation of chrysanthemum. The aim of the this study is to determine the effect of Indole Acetic Acid (IAA) and Benzly Amino Purine (BAP) on the multiplication of chrysanthemum shoots of the Suciono variety. The design of this study used nodus explants, treated with a combination of IAA hormones and BAP. It is make 9 combinations, and then the combinations of hormones is 0 mg/L + 1,5 mg/L; 0 mg/L + 2 mg/L; 0 mg/L + 2,5 mg/L; 0,5 mg/L + 1,5 mg/L; 0,5 mg/L + 2 mg/L; 0,5 mg/L + 2,5 mg/L; 1 mg/L + 1,5 mg/L; 1 mg/L + 2 mg/L; and 1 mg/L + 2,5 mg/L.  The observed variables included histological analysis of callus, time of shoot emergence,  number of shoots, shoots height, and number of leaves. The results of the analysis showed that the treatment with 0 mg/L IAA and 2 mg/L BAP was the best concentration for shoots multiplication. The addition of 2 mg/L BAP was able to produce a percentage of shoots emergence of 100%, 3 shoots per explant, shoots height of 7 cm and 16 leaves.

Abstract

Strengthening the independence of local rice seeds is a strategic effort to increase agricultural productivity while preserving the sustainability of local varieties. Sumbersalak Village, Ledokombo Sub-district, Jember Regency, has the potential of adaptive local rice, yet the seed quality still needs improvement. This community service program aims to provide training and technical assistance to farmers in applying backcross technology for the improvement of local rice seeds. The implementation methods include socialization, hands-on training in backcrossing, field assistance, as well as evaluation through pretests and posttests. The results showed an increase in participants’ understanding from an average of 30% (pretest) to 94% (posttest) in terms of backcross concepts and techniques. Field practices successfully demonstrated the stages of crossing and line selection, although a small number of participants still required additional guidance in the controlled pollination stage. This activity demonstrates that the transfer of backcross technology can enhance farmers’ capacity to produce high-quality and adaptive local rice seeds, thereby supporting seed self-reliance in Sumbersalak Village.
