Abstract

Tujuan penelitian ini untuk menentukan kualitas biologis ekstrak kasar dan isolat albumin ikan gabus yang optimal terhadap berat badan dan kadar serum albumin tikus mencit. Analisa data penelitian ini adalah uji kualitas biologis ekstrak kasar dan isolat albumin pada tikus mencit, perbedaan hasil tiap perlakuannya dianalisa dengan Rancangan Acak Kelompok (RAK) faktor tunggal. Hasil terbaik uji kualitas biologis ekstrak albumin kasar dan isolat albumin pada tikus Mencit (Mus musculus L.) adalah berat badan tertinggi 28,48 g, pada mencit yang diberi perlakuan ekstrak albumin kasar. Kadar albumin serum tertinggi 2,22 g/dl pada mencit yang diberi perlakuan ekstrak albumin kasar.

Abstract

Pre-ekstraksi alginat dari alga coklat dengan pelarut asam membutuhkan reaktan dan air dalam jumlah banyak, serta menghasilkan mengeluarkan banyak limbah. Penelitian ini bertujuan untuk mengoptimalkan penggunaan pelarut asam dengan melakukan penggunaan pelarut secara berulang dan mengetahui pengaruhnya terhadap mutu alginat, reduksi kebutuhan air dan reaktan, serta reduksi limbah pelarut. Penggunaan kembali pelarut asam pada proses pre-ekstraksi dilakukan dengan variasi penggunaan pelarut ke 0, 1, 2, 3, dan 4. Rancangan acak lengkap dengan tiga kali ulangan digunakan untuk menentukan pengaruh penggunaan pelarut berulang terhadap mutu alginat. Parameter yang diamati meliputi rendemen, viskositas, berat molekul alginat, kebutuhan air dan HCl, serta limbah pelarut asam. Hasil penelitian menunjukkan bahwa penggunaan pelarut berulang dapat menghasilkan alginat dengan rerata rendemen 26,7%. Penggunaan pelarut berulang ke 0-2 dapat menghasilkan alginat kualitas sedang dengan viskositas 270,9-360,0 mPa.s dan berat molekul alginat 122,04-175,44 kDa. Pre-ekstraksi makrolaga dengan pelarut berulang ke 0-2 dapat menurunkan kebutuhan air 22,2%, HCl 66,7%, dan limbah pelarut menurun 66,7%. Pre-ekstraksi makroalga dengan pelarut berulang mempunyai potensi efisiensi dan perlu dilakukan penelitian lebih lanjut pada skala yang lebih besar. AbstractPre-extraction of alginate from brown algae with acidic solvents requires a lot of reactants and water that produce a lot of solvent wastes. This study aimed to optimize the use of acidic solvents by using solvents repeatedly and to determine the effect on alginate quality, water consumption and reactant, and acid solvent waste. Repeated use of solvents on pre-extraction stage was carried out with variations of 0, 1, 2, 3, and 4 times. A Completely Randomized Design (CRD) with three replications was used to determine the effect of repeated use of solvents on the alginate quality. Parameters observed were yield, viscosity, molecular weight of alginate, water consumption, HCl requirement, and the amount of acid solvents waste. The result showed that the repeated use of solvents in the pre-extraction stage produced alginate with an average yield of 26.7%. Repeated use of solvents of 0 to 2 times produced medium quality of alginate with a viscosity of 270.9-360.0 mPa.s and alginate molecular weight of 122,04-175,44 kDa. Pre-extraction of macroalgae by repeated use of solvents can reduce water consumption 22,2%, HCl 66,7%, and waste solvents 66,7%. Pre-extraction of macroalgae by repeated use of acid solvents was potential for efficiency, and further research is needed for a larger scale extraction process.

Abstract

This Community Service Activity (PKM) has partners who are a group of goat breeders located in the Dayurejo Forest Village on the slopes of Mount Arjuno, Prigen District, Pasuruan Regency, East Java. Of the 14 problems faced by partners, cultivating a cage and five goats worth giving to the community is the main priority that has been mutually agreed to be carried out on an area of ​​approximately 14 m2. The purpose of doing community service by supporting goat breeders is to provide a view that in the future if it is carried out diligently, the welfare of the breeder will be achieved slowly but tends to be more certain. The presence of five goats supported by the Abdimas Team is a form of implementing community service after implementing ANSOS with partners with the aim of supporting the forest economy in the future, including for families of farmers.

Abstract

Snakehead fish was a fish that has a high albumin content. Snakehead fish was used in the health sector known as medicinal freshwater fish, to accelerate the process of healing wounds after surgery and childbirth. The purpose of this study was to analyze the relative mobility (Rf) of the snakehead fish (Ophiocephalus striatus) albumin isolates by steaming at different temperatures and times. The research method for analyze the relative mobility (Rf) of the snakehead fish (Ophiocephalus striatus) albumin isolates by using gel filtration column chromatography, was detected by the SDS-PAGE electrophoresis method. Electrophoresis was carried out at a constant voltage of 125 v/ slab. Gel staining was carried out with Commasie Brilliant Blue R-250 in methanol-acetic acid-water. A standard protein mixture consisting of: myosin (200.0 kDa), β-galactosidase (116.3 kDa), phosphorylase B (97.4 kDa), bovine serum albumin (66, 2 kDa), ovalbumin (45.0 kDa), carbonic anhydrose (31.0 kDa), trypsin inhibitor (21.5 kDa), lysozyme (14.4 kDa) and aprotinim (6.5 kDa). The data analysis of this research was descriptive analysis to see the photos of the electrophoresis results. Meanwhile, for the analysis, the measurement of albumin isolation by gel filtration Sephadex G-75 was carried out with a divided plot design. The results showed that SDS-PAGE electrophoresis with the most complex amount of protein was albumin isolate, the effect of the steaming temperature was 40 oC for 30 minutes, located in 5 ml of the 1st fraction, 5 ml of the 2nd fraction and 5 ml of the 3rd fraction, with values of relative mobility (Rf) 0.0833 to 0.6944.

Abstract

The brown alga has bioactive alginate and fucoidan, a potential for raw materials the biorefinery industry with core processing of fucoidan and alginate extraction. The research on sequential biorefinery of fucoidan and alginate was conducted in conventional and hydrothermal methods using a smart pressure-cooker, but the yield of fucoidan is low. The improvement was carried out by extracting fucoidan and alginate using microwave-assisted extraction (MAE). This study aimed to obtain the MAE processing conditions that produced the maximum yield of fucoidan and alginate. The factorial design of 2k was used to determine the effect of MAE processing conditions, i.e. temperature, time, and alga/water ratio on fucoidan and alginate yields. The parameters observed were fucoidan and alginate yield. Data analysis and prediction of first-order model and model accuracy was a regression modelling using a design expert program. The result showed that the elevate of temperature MAE processing to 80 oC,for 20 minutes, and alga/water ratio of 2:20 (w/v) increased fucoidan (4.5%) and alginate (40.6%) yield, but it decreased the yield at 100 oC, for 30 minute, and alga/water ratio of 3:30 (w/v). The equation of the first-order model was quadratic, by wich the model test in curvature was found significant at α=0.05. The MAE sequential biorefinery extraction improved fucoidan and alginate yield and had a potential efficiency for commercialization. The maximum response of fucoidan and alginate yield was obtained from a MAE sequential biorefinery at 80 oC,for 20 minutes,and alga/water ratio 2:20 (w/v); further research is needed to optimizing the process.

Abstract

Hutan di Lereng Gunung Arjuno di daerah Desa Dayurejo, Kecamatan Prigen,Pasuruan Jawa Timur sebagaimana hutan di Gunung lainnya perlu dilestarikan. Salah satu usaha pelestariannya setelah melaksanakan Analisis Sosial di lokasi setempat dan mendapatkan persetujuan dari Yayasan Cempaka Education dan pihak masyarakat Desa Hutan wilayah Desa Dayurejo yakni pengadaan bibit pohon pisang untuk ditanam di lereng Gunung Arjuna khususnya di lokasi yang dikelola oleh Yayasan Cempaka Education Center.

Abstract

Desa Bakalan berada di Kecamatan Purwosari Kabupaten Pasuruan, mempunyai potensi wilayah yang dilewati aliran sungai yang cukup melimpah airnya, sehingga dimanfaatkan untuk mengairi tanaman dan memelihara ikan antara lain nila, tombro ataupun lele. Salah satu permasalahan yang dihadapi adalah teknologi budidaya perikanan air tawar kurang berkembang, pengetahuan tentang perikanan masih cukup rendah, dan penggunaan alat produksi serta sarana prasaran masih sederhana. Rendahnya pengetahuan mengenai teknologi dalam budidaya menyebabkan kegiatan produksinya tidak berjalan secara kontinyu. Mitra kegiatan adalah Kelompok Swadaya Masyarakat (KSM) “Balesemi” desa Bakalan. Anggota kelompok berjumlah 20 orang, yang berpartisipasi aktif selama kegiatan berlangsung. Kegiatan ini bertujuan untuk meningkatkan pengetahuan dan keterampilan kelompok mitra guna menjaga kontinyuitas produksi dengan penerapan teknologi wadah kolam terpal sebagai media budidaya. Kegiatan dimulai dari tahap persiapan, sosialisasi, pelaksanaan sampai dengan pendampingan. Tahap persiapan dengan cara survei dan wawancara, melakukan pendekatan dan identifikasi masalah, diskusi bersama tentang pemecahan masalah. Adapun tahap pelaksanaan Bimtek meliputi: 1) Penyiapan kolam, 2) Pemilihan bibit, 3) Penebaran benih 4) Pemilihan pakan berkualitas; 5) Cara panen dan pasca panen. Solusi selanjutnya adalah pemberian bantuan kolam terpal yang dapat digunakan untuk budidaya ikan yang lebih baik. Evaluasi keberhasilan dilakukan melalui hasil survei dan kuisioner. Hasil survei didapat bahwa peningkatan pemahaman peserta sebelum Bimtek terhadap materi pada kisaran paling rendah 25% dan persentase pemahaman tertinggi 50%. Kemudian setelah dilakukan Bimtek mengalami peningkatan pemahaman tertinggi pada materi persiapan kolam mencapai 100% sedangkan terendah  pada materi mengenai pakan sebesar 80%. Hasil pengabdian ini penting artinya bagi perkembangan budidaya perikanan pada umumnya dan peningkatan pendapatan bagi kelompok mitra. Technical Guidance for Freshwater Fish Cultivation at the "Balesemi" Community Self-Help Group in Bakalan Village, Pasuruan Regency Abstract Bakalan Village is located in Purwosari District, Pasuruan Regency, has the potential of an area crossed by a river with abundant water, so it is used to irrigate plants and raise fish including tilapia, tombro or catfish. One of the problems faced is that freshwater fisheries cultivation technology is underdeveloped, knowledge about fisheries is still quite low, and the use of production tools and infrastructure is still simple. The low knowledge of technology in cultivation causes production activities not to run continuously. The activity partner is the Community Self-Help Group (KSM) "Balesemi" in Bakalan Village. This activity aims to improve the knowledge and skills of partner groups in order to maintain production continuity by implementing tarpaulin pond technology as a cultivation medium. Activities start from the preparation stage, socialization, implementation to mentoring. The preparation stage is by means of surveys and interviews, approaching and identifying problems, and discussing together about solving problems. The implementation stages of the Technical Guidance include: 1) Preparation of ponds, 2) Selection of seeds, 3) Spreading seeds, 4) Selection of quality feed; 5) How to harvest and post-harvest. The next solution is to provide assistance in the form of tarpaulin pools that can be used for better fish farming. Evaluation of success is carried out through survey results and questionnaires. The survey results showed that the increase in participants' understanding before the Technical Guidance on the material was in the range of the lowest 25% and the highest percentage of understanding was 50%. Then after the Technical Guidance was carried out, there was the highest increase in understanding in the material on pond preparation reaching 100% while the lowest was in the material on feed at 80%. The results of this service are important for the development of fisheries cultivation in general and increasing income for partner groups.

Abstract

Animal waste management is an environmental challenge that can be turned into an economic opportunity through the right approach. This service program aims to assist the Pokdarwis Pilang community of Probolinggo City in managing animal waste into organic compost to support the sustainability of urban farming. The Asset-Based Community Development (ABCD) approach is used to empower the community by utilizing existing local assets, such as animal waste, community skills, and infrastructure support. The service process includes five stages: Discovery, Dream, Design, Define, and Destiny. The Discovery stage focused on identifying local assets, where animal waste was found to be a valuable asset that was previously not managed properly. Through the Dream stage, the community formulated a shared vision to create a sustainable waste treatment system. The Design stage implemented technical training and procurement of composting infrastructure. At the Define stage, the community began to manage the waste treatment independently, followed by an evaluation of the results and social impact at the Destiny stage. The results showed an increase in the technical skills of Pokdarwis members, the establishment of a compost production unit, and changes in community behavior in waste management. In addition, the formation of partnerships with local markets helped improve the economic welfare of the community. The ABCD approach has proven effective in creating sustainable social change through community empowerment. Recommendations from this program are to expand the application of the ABCD approach to other communities and strengthen collaboration with stakeholders to support the sustainability of waste management and urban farming.

Abstract

This study is part of a community service project aimed at examining the role of human and social capital in the sustainable forest management of Mount Arjuna East Java. The objective of the community service initiative is to empower local communities around the forest to actively participate in forest conservation efforts. The study employs a descriptive method with a survey technique to gather factual data from the research area, which includes the spring areas around Mount Arjuna in East Java, Indonesia. The study population consists of communities from three villages: Leduk, Jatiarjo, and Dayurejo, located in Pasuruan, East Java. Data were collected from respondents through questionnaires and analyzed using Structural Equation Modeling (SEM) to understand the general responses regarding community participation in forest preservation. The results indicate that the human capital of the community, particularly education level and employment, contributes significantly to the success of sustainable forest development. Additionally, social characteristics such as mutual-cooperation and kinship within the community play a vital role in supporting long-term forest management. This community service initiative aims to strengthen both human and social capital to foster a sustainable forest management approach based on local wisdom and community-driven efforts.

Abstract

The research objectives was to determine the analysis of community empowerment participation for the forest revegetation in Arjuna Mount. This service uses a Participatory Rural Appraisal approach, with the study population covering communities around the forest of Arjuna Mount Pasuruan-East Java Indonesia. Data obtained from respondents with a questionnaire and analyzed using SEM (structural equation modeling), to find out an overview of respondents' responses about community participation in the preservation of the Arjuna Mount Forest. Community participation in the form of human capital, economy, social, local institutions, facilities and infrastructure has significant influence on community empowerment for the preservation of the Arjuna mount forest. Estimated regression weights for each indicator of community empowerment in Leduk, Jatiarjo and Dayurejo villages are human resources 0.99, economy 0.99, social 0.97, local institutions 1.00 and facilities and infrastructure 1.00. This explains that local institutional factors, facilities and infrastructure have a significant influence on community empowerment for the preservation of the Arjuna mount forest.
