Abstract

Tarakan waters have geographical and landscape that supported a wealth of abundant and diverse fish. One of the natural resources on the Tarakan Island is Nomei fish. This fish is known by local names of Pepija or Lembe-Lembe. Nomei fish catch has decreased since 2009. There has been no real effort in determining the fish conservation strategies. Data on Nomei taxonomy is still lacking, so far limited research has been conducted only on bio ecology aspect. The purposed of this study was to determine the taxonomic status of Nomei fish from Tarakan based on 16S rRNA gene sequences and determine relationship with closely related species. Genetic data collection were done by taking two individual pectoral fin from each fish from Amal and Juata waters. Total DNA isolated using Roche KIT with some modifications. 16S rRNA gene amplification were done by using a universal primer. Alignment results showed that there were several automorfi nucleotide characters distinguishing of this genus. Based on 16S rRNA gene sequences with consistency of phylogenetic topology is generated, it can be concluded that Nomei fish from Amal and Juata included in Harpodon genus, species Harpodon nehereus. This is consistent with the description of morphological characters were Harpodon nehereus. It is also strengthened by divergent sequences of Harpodon sp. Amal 1 and Harpodon sp. Amal 2 (0%); while Harpodon sp. Juata 1 and Harpodon sp. Juata 2 (0%). Keywords : taxonomic status, Nomei fish, 16S rRNA gene, conservation

Abstract

Tarakan city has a potential local fishery resources which have not been considered and utilized optimally, one of which is fish Mudskipper / Tempakul (mudskipper). Mudskipper one of Gobiidae Family: subfamily Oxudercinae that resemble amphibians living and liked the muddy area scattered Mangrove coastal waters. One of the basic information that is needed is to know the variation of morphological characters mudskipper in Tarakan and characterization fenetic distance between populations. Morphological characters examined included morphometric studies, meristics and special characters of this fish. This research is a descriptive quantitative and qualitative descriptive, with a survey approach. This study founded four species of Mudskipper fish that live in the habitat settlements, open land, sea and mangrove and Juata areas. There are: Peripothalamus malaccensis, Periopthalmodon freycineti, Baleophthalmus boddarti and Periopthalamus barbarus. Dendogram of morphological analysis obtained by the two groups branch, that is Periophthalmus malaccensis and Periopthalmus barbarus clade with Periophthalmodon freycineti and Baleopthlamus boddarti form different branching. Grouping of mudskipper fish differentiated based on five main morphometric characters.  There are: SL: standard length, HL: length of the head, FDFB: first dorsal fin base length, SDFB: second dorsal fin base length, and VFL: long ventral fins.Keywords : morphology, Mudskipper, biodiversity, Oxudercinae, relationship

Abstract

Banyu Biru is a natural lake in Sumber Rejo Village, Winongan District, Pasuruan Regency. There are local fishes in the Banyu Biru called sengkaring and tambra. Biological studies on these fishes are not so much known. This study aims to identify the relationship of sengkaring and tambra compared with the related species ( Tor spp.: Cyprinidae) from Indonesia. Morphometric analysis can be used to clustering and understanding the relationship of taxa. Each species represents 17 specimens (Tambra and Sengkaring) and measured 22 characters (in mm) by using digital caliper and the additional characters (21 characters). The canonical, clustering, similarity and distances were used. Based on morphometric measurements, sengkaring and tambra had similarity with the four species found in Indonesia. Commonly, the population of Sengkaring and Tambra was critical; therefore, studies needed to be carried out to support conservation.

Abstract

Poeciliidae family consists of small live-bearer fishes with sexual dimorphic. This fishes are widely distributed across Indonesian archipelago, but biological studies on this fish are very limited. This research is aimed to identify the species of Peciliidae fishes which lives in Sari Lake Pasuruan based on gonopodium structures and morphometric analysis. A specific characteristic of the Poeciliidae family is its gonopodium structures which is a modification of 3rd, 4th, and 5th rays of the males anal fin. Each species represented 50 samples male and female from each type we found and measured 26 characters (in mm). Cladogram based on morphometric measurements showed that sample 1 and 2 had 100% similarity, sample 3 had 94% similarity with sample 1 and 2. Sample 4 had 54% similarity with sample 1, 2, and 3. Based on gonopodium structures sample 1 and sample 2 included Xiphophorus hellerii Heckel possess large claw on the terminus of ray 5 hook is small crescent-moon shaped on 3rd ray and lacking of gonopodium palp, sample 3 possesses distal platform with 11 retrosse serrae,and membranous hook so we were able to identify the sample 3 as Poecilia mexicana Steindachner, whilst sample 4 was Poecilia reticulata Peter had a long-pointed comb-like spines on ray 3rd and and has a small hook on the tip of 5p ray.

Abstract

Tor genus is a local Indonesian freshwater fish, and endangered species. The urgent conservation efforts should be made to determination of relationship based on phenetic approach. This is because the taxonomical position and classification of this fish were unclear. The data used in this study were meristic, general of morphology, and special character to determinate up to the species level. The special characters of this fish were the existence and size of the median lobe. Samples were taken from Banyu Biru lake (Lokal name were Sengkaring and Tambra), as well as the related species, They were consists of Tor duoronensis from Padang, Tor Tambraides from West Borneo, and Tor soro were obtained from North Sumatra. Fenogram formed based on UPGMA method. According to the phenogram, it was shown that 3 related groups of Tor Genus laid from near to far distance based on their relationship, 2 apomorphy and 4 automorphy group. The first consists of two subklad namely Tor duoronensis and Sengkaring with their similarity index 100%, and Tambra was sister clad of Tor duoronensis supported by the similarity index of 92.9 %. The second Subklad consists of Tor tambra and Tor soro (Apomorfi B) with a similarity index 59%, while the furthest distance Tor tambraides with similarity index of 45, 625%. Tambra and Sengkaring showed a grouping pattern that closely related to Tor duoronensis based on seven main distinguishing morphometric features. Seven main distinguishing features were consist of: size of median lobe, SL (standard length), MXBL (maxillary barbels lenght), CPL (caudal peduncle lenght), PDL (pre dorsal lenght), IW (interorbital width) and SNL (snout lenght). Furthermore, the results of morphometric analysis showed that Sengkaring and Tambra was indeed not a member of Tor Tambraides and Tor soro species due to significant difference of body size. Keywords: Tor, Relationship, Phenetic, Morphology, Special Character, Meristric, Conservation

Abstract

Anggota Famili Poeciliidae merupakan ikan introduksi dengan karakteristik berukuran kecil, live-bearers, dimorfi seksual dari segi ukuran tubuh, pola warna dan struktur gonopodium sebagai ciri diagnostik pada ikan jantan. Ikan ini berperan sebagai pengontrol perkembangan nyamuk malaria dan model pembelajaran biologi. Sejauh ini kajian taksonomi dan penelusuran hubungan kekerabatan didalam famili Poeciliidae belum ada. Tujuan penelitian ini adalah mengidentifikasi jenis, kekerabatan dan pengelompokkan berdasarkan karakter morfologi (morfometrik, meristrik, gonopodium dan ciri diagnostiknya). Jenis penelitian ini adalah deskriptif eksploratif, dengan pendekatan survey. Pada penelitian ini ditemukan tiga spesies ikan Poeciliidae yaitu Xiphoporus helleri, Poecilia mexicana dan Poecilia reticulata. Dendogram hasil analisis morfologi menggunakan metode UPGMA diperoleh dua kelompok percabangan apomorfi yaitu kelompok Genus Xiphoporus dan Genus Poecilia. Sub Clade automorfi antara Xiphoporus helerri A dan B dengan nilai kesamaan 92%.  Pengelompokkan ikan Poeciliidae dibedakan berdasarkan lima karakter utama terpilih yaitu: SL (standart length), Hdl (Panjang kepala), Panjang gonopodium, panjang sirip ekor dan PreDL (Panjang sebelum sirip dorsal).Kejelasan identifikasi ikan introduksi khas Telaga Sari menjadi modal utama dalam aquaculture, manajemen, konservasi, biodiversitas dan perkembangan ilmu pengetahuan.

Abstract

Foot-and-mouth disease is a severe problem that must be faced in the livestock sector. This disease has a negative impact on various aspects, especially the economy. One way to develop herbal medicinal compounds is through local Indonesian wild plants, meniran (Phyllanthus urinaria). This research aims to determine the potential of the wild plant P. urinaria as an antiviral agent for FMD using an in silico approach using molecular docking. The compounds used as ligands are flavonoid compounds in P. urinaria, namely, routine, quercetin 7-methyl ether, quercetin 3-O-β-D-glucoside, quercetin, rhamnocitrin, astragalin, and kaempferol. This study used the control drug ribavirin as a comparison. The research stage began with the preparation of the RdRp protein from the FMD virus with Discovery Studio, ligand preparation with the Lipinski druglikeness test and minimization using OpenBabel, followed by docking and visualization. The research results found that the six flavonoid compounds in P. urinaria have potential as antiviral FMD by inhibiting RdRp, with the most potent compound being quercetin (-7.9 kcal/mol). Further research is needed, including in vitro and in vivo testing, to provide confidence in the potential of this wild plant as an antiviral for FMD.

Monetaria annulus (Gastropoda: Cypraeidae) atau dikenal juga dengan siput cincin memiliki penyebaran di perairan Indo -Pasifik dan kerap dimanfaatkan oleh masyarakat dari masa sebelum Masehi sebagai alat pembayaran, perhiasan, dan bahan konsumsi. Penelitian Monetaria annulus ini bertujuan untuk mendapatkan karakteristik genetik spesies tersebut, yaitu dari nilai similaritas data BOLDsystems, komposisi basa nukleotida, jarak genetik, dan analisis filogenetik yang didasarkan pada sekuens gen COI. Sampel Monetaria annulus dari Pantai Nunuhu dilakukan tahapan ekstraksi, amplifikasi, elektroforesis, hingga sekuensing. Analisis genetik dilakukan dengan software bioinformatika, yaitu MEGA 7.0, BOLDsystems, Jupyter Notebook, Microsoft Excel, dan digunakan data pembanding dari Genbank. Hasil karakterisasi genetik dari tiga sampel segar, lima ingroup dan dua outgroup diperoleh sekuens dengan panjang 606 bp dan nilai similaritas 99,5-99,83%. Komposisi basa nukleotida menunjukkan kombinasi GC lebih rendah daripada AT dengan persentase berurutan 42,02% dan 57,98%. Rata-rata jarak genetik Monetaria annulus dari Pantai Nunuhu dengan ingroup adalah 2,69%. Hal ini menunjukkan bahwa sampel dari Pantai Nunuhu teridentifikasi satu spesies dengan Monetaria annulus ingroup. Dari pohon filogenetik dihasilkan satu kluster besar yang membagi Monetaria annulus Pantai Nunuhu dengan ingroupnya yang berasal dari China dan Papua. Hasil penelitian ini dapat dipergunakan sebagai dasar penelitian mengenai biogeografi Monetaria annulus terutama di kawasan Wallacea.

Abstract

Odonata merupakan ordo dengan tingkat keanekaragaman dan kelimpahan terbesar di antara filum Arthropoda lain tetapi banyak yang belum teridentifikasi. Capung menyukai habitat daerah terbuka hijau, utamanya rerumputan. Penelitian ini bertujuan menganalisis keanekaragaman dan kelimpahan insekta capung (Odonata: Anisoptera) di Lapangan Watu Gajah Kecamatan Semanding Kabupaten Tuban. Lapangan Watu Gajah termasuk ruang terbuka hijau yang mendukung kelangsungan hidup insekta, salah satunya capung. Kondisi iklim Lapangan Watu Gajah dengan rata-rata suhu 290C, ketinggian 40 mdpl, kecepatan angin 13 km/h, curah hujan 1,9 mm, kelembaban 70,33%, dan intensitas cahaya 4259,33 lux. Penelitian dengan metode jelajah (Cruising method) menggunakan jala insekta dan teknik hand sampling pada 3 titik area. Berdasarkan hasil penelitian diperoleh 6 spesies capung. Analisis data dilakukan dengan menggunakan rumus kelimpahan relatif, indeks keanekaragaman Shannon-Winner, dan indeks dominansi Shimpson. Hasil perhitungan indeks keanekaragaman capung tergolong rendah, yaitu 1,57 dan indeks dominansi 0,226 termasuk dalam kategori sedang. Lapangan Watu Gajah menjadi habitat ideal yang sesuai untuk kelangsungan hidup capung (Odonata: Anisoptera).
