Abstract

Type 2 Diabetes Mellitus is an endocrine system disorder influenced by genetic factors and lifestyle choices. Mutations in the TCF7L2 gene, particularly at rs7903146, have been closely associated with T2DM due to their disruptive effects on insulin secretion. The primary objective of this study is to identify mutations in the TCF7L2 gene within the T2DM population of the Sidoarjo Region. A total of seven samples were utilized in this research, employing DNA isolation, PCR, and sequencing techniques. The DNA samples were specifically amplified to target the TCF7L2 gene over a stretch of 318 base pairs. The findings of this study indicate an absence of mutations at rs7903146.

Abstract

Urinary Tract Infection is an infection of the urinary tract, especially the kidneys, bladder, ureters and urethra which causes various microorganisms, generally bacteria. If urine is left for too long it will affect the validity of the examination results in supporting the diagnosis of an examination, especially UTI. The aim of this study was to determine the effect of delaying urine at room temperature and cold temperature on the number of bacteria in Urinary Tract Infection patients. The method used was Laboratory Experimental with Accidental Sampling sampling techniques in accordance with the inclusion criteria, totaling 5 samples. The results of the study at room temperature (20-25°C) (12.9 /ml). The data were analyzed statistically using the Friedman test, showing a significance value of P<0.05, namely 0.003, that there was an effect of delaying urine at room temperature and at cold temperatures on the number of bacteria in sufferers of Urinary Tract Infections (UTI).

Abstract

Medicinal plants are plants that contain ingredients that can be used for treatment and the active ingredients can be used as synthetic medicinal ingredients. One of the medicinal plants that is often used is turmeric (Curcuma longa). In addition, there is young coconut (Cocos nucifera) which is a plant native to the tropics, has water on the inside of young coconuts and can be consumed without going through processing. This study aims to determine the potential of the combination of turmeric juice with young coconut water in inhibiting Clostridium perfringens bacteria. This study used 5 concentrations, namely 25%, 50%, 75% and 100% and the combination of turmeric juice with young coconut water, namely 25%: 75%, 50%: 50% and 75%: 25%. The concentration of 100% extract of turmeric juice and young coconut water was the best concentration to form an inhibitory zone against the bacterium Clostridium perfringens of 32.66 mm and 14 mm.

Abstract

Tamarind and turmeric are used by the community to treat respiratory diseases and fever obtained at the Pandaan market. This study aims to determine the effectiveness of tamarind leaves and turmeric by maceration in inhibiting Klebsiella pneumoniae bacteria at various concentrations. Antibacterial activity test using Kirby-Bauer disc diffusion method with concentrations of 25%, 50%, 75%, and 100%. Tamarind and turmeric leaves used maceration extraction method by immersion using 96% ethanol as solvent. The positive control used was Penicillin while the negative control used was DMSO 10%. The concentrations of the samples used were: tamarind leaf extract with concentrations of 25%, 50%, 75%, and 100%; turmeric extract concentrations of 25%, 50%, 75%, and 100%; a combination of 25% tamarind leaf extract and 75% turmeric extract; a combination of 50% tamarind leaf extract and 50% turmeric extract; combination of 75% tamarind leaf extract and 25% turmeric extract. The total number of samples is 33 samples. The best inhibition was obtained from the results of the inhibition zone > 20 mm. Turmeric extract had the best antibacterial activity against the growth of Klebsiella pneumoniae bacteria at concentrations of 50%, 75%, and 100%. With each inhibition zone diameter of 21.5 mm; 29.83 mm ; and 33.16 mm. Based on the results of the Two Way ANOVA test with a 95% confidence level (p > 0.05), it was found that there was no significant effect between samples of tamarind leaf extract and turmeric extract on the growth of Klebsiella pneumoniae bacteria.

Abstract

Plants have many chemical constituents. Utilization of plants as an alternative treatment in overcoming food poisoning, one of which is Gambas (luffa acutangula L) which contains flavonoids so that it can kill bacteria that attack the digestive system. This research was conducted in February-May 2021 and included an experimental study that aimed to determine the potential of maceration of Gambas fruit and fresh extract in inhibiting Clostridium perfringens bacteria at various concentrations. The antibacterial activity test was carried out by maceration method and fresh extract. This antibacterial potential is indicated by the form of a clear zone around the paper disk. This study used 4 concentrations, namely 25%, 50%, 75%, 100% and Penicillin as a positive control and aquadest as a negative control. Based on the non-parametric test using the Friedman test with a sign value (α <0.05) that is 0.008 which indicates that there are significant differences at various concentrations.
