Abstract

Contamination of Aspergillus flavus can attacks food and feed ingredients, especially seeds during the storage. The using of biological agents, like microbes that have antifungal activity becomes a promising and important solution to be studied. This research aims to find out the antifungal activity of lactic acid bacteria (LAB) from digestive tract of “Kampung” chickens against the growing of A. flavus. Source of  the isolates are from colon, ventriculus, and crop. The inoculums of LAB which using were suspension of cell and cell free supernatant (CFS). There were 6 isolates of LAB (Bub1, Bub3, Bub4, V52, C51, and NNB) tested to obtain the potential of antifungal activity, by with well diffusion and and submerged culture method. The results showed that the 5 isolates suspension cell of LAB showed inhibitory zones, except Bub1, whereas all CFS LAB showed the inhibitory zones against the growth of A. flavus in MRSA medium.  Furthermore, antifungal activity of LAB against A. flavus to be observed by measuring the dry weight of the mycelium production, as well as the percentage of inhibition during the incubation period. All isolates of LAB, from suspension cell and CFS showing antifungal activity against growth of A. flavus. The Bub3 and NNB isolates, from suspension cell and CFS showed that the best antifungal activity compared to the others.

Abstract

Indigenous Trichoderma has widely used in late-blight epidemic areas in Magelang, Indonesia as biocontrol and biomodulator. But it application still lacks of scientific proof, even though it is potently to be developed. Therefore, this study aims to identify and evaluate indigenous Trichoderma application in eradicating late blight and increase productivity. This research was an experimental posttest-only using six experimental plots, with P0 plot with no treatment. The solution was made by dissolve indigenous Trichoderma solid-starter into water (m/v), then used for treatment. The treatment plots including: two groups of sprayed-only plots with doses of 100 g/ 10 L of solution for P1 and 150 g/ 10 L of solution for P2; and two group treated with sprayed-poured method using doses of 100 g/ 10 L of solution for P3 and 150 g/ 10 L of solution for P4. The positive control group was P5 group, treated using chemical pesticides and tuber booster. The microscopic identification revealed that the local bioagent was Trichoderma harzianum species. Application of T. harzianum in P4 was significantly increased the productivity. However, it was not effective in increasing growth, but able to reduce the intensity of late blight disease. The research revealed that native Trichoderma is able to be used as anti-infectious agent and potentially improve the quality of potato plants. In the future, this research may worthwhile for farmers to develop and produce trustworthy and proven Trichoderma-based biocontrol and help them increase the potato production economically.

Abstract

The enzyme amylase, cellulase and protease are an extracellular enzymes are many produced by microorganisms such as fungi Aspergillus section Nigri. This research aims to find out the potential biakanes of Aspergillus section Nigri DUCC collection (Diponegoro University Culture Collection) which is capable to produce some enzymes that consists of amylase, protease and celullase and also identification in molecular based. Activity enzymatic assay of Aspergillus section Nigri with  semi-quantitative method using selective medium, CMC for cellulolytic, Starch agar 1% for amylolytic and Skim Milk agar for proteolytic. Biakane potential is determined by looking at the Enzymatic Index (EI) is highest for all of three enzyme. Molecular identification is using the universal primer ITS4 and ITS5. The results showed that the culture of DUCC K207  has high activity for all of three enzyme. Index enzymatic of isolate DUCC K207 , 1.55 mm for amylolytic, 1.49 mm for cellulolytic and 1, 24 mm for proteolytic. Result of molecular identification DUCC K207  known as Aspergillus niger that has 100% similarity with Aspergillus niger MH 109325.1.

Abstract

Both eukaryotic and prokaryotic organisms can produce antimicrobial peptides (AMPs) which function as a self-defense mechanism against other harmful organisms in the same ecological niche. Pediococcus pentosaceus is a species of Lactic Acid Bacteria (LAB) capable of producing AMPs in the form of bacteriocins and bacterocin-like inhibitory substances. The compounds it produces have received Generally Regarded as Safe status by the Food and Drug Association (FDA) and have potential as biological preservatives in the food sector. The production of these compounds can be influenced by environmental factors and growth medium of producing bacteria, so that optimization studies for the production of bacteriocin have been developed, in order to obtain optimal its activity. Vitamin B Complex is one of the growth factors needed by living things including bacteria to support their metabolism. This study aims to determine whether the addition of vitamin B complex affects the production and activity of AMPs from P. pentosaceus. Vitamin B Complex concentrations of 0 ppm, 0.1 ppm, 1 ppm and 10 ppm were added to the production medium of P. pentosaceus. Cell-free supernatants were harvested by centrifugation, then their activity was tested against Bacillus cereus and Eschericia coli using the Kirby Bauer method and analyzed using one way ANOVA parametric statistical test with a significance level of 0.05 and Duncan's post hoc test. The results showed that the addition of vitamin B complex was not significantly different to the activity of AMPs compounds, but in higher concentrations could reduce the activity of these compounds.

Abstract

Ammonia emissions at the farm that was from the broiler faecal waste inflict restlessness and become the source of respiratory diseases to the local people because the farm is located adjacent to residential areas. Bacterial isolates were characterized as acid producer with BIS prebiotic mixture may be a bioremediation agent to reduce ammonia emissions in broiler feces. The purpose of this research is to create a biotechnology product which is ready to apply for reducing the ammonia at the poultry farm.  As the treatment used three isolates of bacteria such as L. acidophilus, L. bulgaricus and B. cereus were each used to ferment the BIS for 48 hours at a temperature of 38 ° C. Results of subsequent fermentation inoculated into the stool with a concentration of 8% (w / w), and incubated for 48 hours. Observations ammonia, pH and microflora in the stool, were done at 24 and 48 h of incubation. The results showed a decrease in the levels of ammonia and pH were significantly different between the control and all treatments, but there is no real difference between each treatment of bacterial isolates. Decreased levels of ammonia at 24 h incubation by L. acidophilus, L. bulgaricus and B. cereus, respectively for 92.89%, 98.78%, 98.56%, while the 48-hour incubation, respectively for 90.67 %, 89.60% and 96.80%.Keywords; Ammonia, Acid Producing Bacteria, Prebiotics, BIS, Broiler

Abstract

Fusarium sp. merupakan patogen tular tanah yang memiliki kemampuan bertahan dalam tanah selama bertahun-tahun, pada kondisi lingkungan yang tidak menguntungkan. Bakteri asam laktat (BAL) diketahui menghasilkan senyawa antikapang yang dapat digunakan sebagai salah satu alternatif pengendalian Fusarium sp. yang ramah lingkungan. BAL dapat ditemukan dalam produk fermentasi, antara lain pada tape ketan kemasan plastik. Tujuan penelitian ini adalah untuk mendapatkan isolat murni bakteri asam laktat (BAL) dari tape ketan kemasan plastik dan mengetahui aktivitas antikapang isolat BAL yang yang diperoleh  dalam  menghambat pertumbuhan kapang Fusarium sp. Penelitian ini menggunakan kultur BAL, supernatan BAL bebas sel (CFS) dan supernatan bebas sel BAL yang dinetralkan (CFSN). Uji aktifitas antikapang dilakukan dengan metode sumuran pada medium MRSA. Hasil penelitian menunjukkan bahwa dari tape ketan kemasan plastik diperoleh enam isolat BAL (BTP1, BTP2, BTP3, BTP4, BTP5, BTP6). Lima isolat BAL (BTP1, BTP2, BTP3, BTP4, BTP5) mampu menghambat pertumbuhan kapang Fusarium sp. dengan aktivitas antikapang fungistatik. Supernatan bebas sel BAL yang dinetralkan (CFSN) BTP1menunjukkan aktivitas antikapang yang paling besar dan berbeda nyata dengan isolat BAL lainnya.

Abstract

The production of fructose from inulin by inulinase would only take one stage enzimatis reaction and yielding 95% fructose. Inulin obtained from the dahlia tubers and inulinase produced by Pichia manshurica DUCC Y-015. The production of inulinase  (E.C 3.2.1.7) can be influenced by the presence of glucose as  additional carbon source. The purpose of this research is to analyze influence of  variation glucose concentration and incubation time to production inulinase P. manshurica DUCC Y-015 in dahlia tubers substrate. Measurement of production enzyme covering activity inulinase and activity invertase. Both analyzed by DNS method and determined based on 1 µmole fructose resulting. The design used in this research were Randomized Factorial Block Design ( RAFBD ). Factor I in the form of glucose concentration 0% (G0); 0.25% (G1); 0.5% (G2) and factor II were incubation time 6 hours (T6), 12 hours (T12), and 18 hours (T18) with repetition 3 times. The data obtained were analyzed by ANOVA method. The result showed variation of glucose concentration and incubation time difference had no significant effect on production of inulinase from P. manshurica DUCC Y-015. The highest production of the inulinase was demonstrated by treatment G1T12 with  0,25% glucose and incubation time 12 hours with value of the activity of inulinase as much as 0,574 IU/mL. Keywords : Dahlia tubers,  Glucose, Inulinase, Incubation time, Pichia manshurica DUCC Y-015,

Abstract

Natural dyes in food industry is more demand so its pushes eforts for developing the production. Monascus is one of fungi that is potentially used for anka production that used in industrial products such as meat, fish, soy sauce, and liquor. The aim of this study was to produce pigment from Monascus using corncob with 35 mesh (<0,5 mm), 18 mesh (0,6-1 mm), 10 mesh (1,1-2 mm) particle size substrate and 20%, 40%, 60% moisture content. The production of Monascus sp. pigments made of solid-state fermentation and  incubated for  14 days. The parameters observed were pigment production and biomass. Analysis of pigment using spektrophotometer with wavelength (λ)  of 500 nm as well as biomass analysis by measuring N-acetyl glucosamin. The results of analysis showed that particle with size 0,6-1 mm from the substrates and with 20% and 40% moisture content was the best condition in process of pigment production using corncob substrate. The production of pigment from fungi with substrate with size 18 mesh ((0,6-1 mm) in 20% moisture content reached 1,1 CVU (Color Value Unit)/ml, and  in 40% moisture content reached 1,07 CVU/ml. The highest biomass was reached on 10 mesh (1,1-2 mm)  particle size at 20% moisture content reached 0,83 A (absorbance)/gds (gram dry substrates), 40%  reached 0,72 A/gds, and 60% reached 0,81 A/gds.Keywords: Solid-state fermentation, corncob, Monascus sp., particle size, moisture content

Abstract

The rhizome of Curcuma aeruginosa has essential oils compounds and has been used traditionally to help for nourish the skin, asthma, appetite enhancer, and as anthelmintik. Bacillus subtilis and Staphylococcus aureus are gram positive bacteria that can cause skin disorder such allergic and acne. The aim of this research was to know antibacterial capacity of essential oils from C. aeruginosa in various concentrations against B.subtilis and S.aureus using disc diffusion method. This research was conducted using RAL design (Completely Randomized Design) with concentration of essential oils as a treatment, i.e. 25, 50, 75, and 100% (pure extract without dilution). The result showed that essential oils of C. aeruginosa has antibacterial capacity for both bacteria with different effect depending on the concentrations tested. Essential oils of C. aeruginosa with 100% concentration showed the highest antibacterial capacity against   S. aureus, whereas all concentrations of essential oils from  C. aeruginosa showed weak antibacterial capacity against B. subtilis. Keyword : Curcuma aeruginosa Roxb., distillation, essential oils, antibacterial, S.aureus, B.subtilis
