Abstract

The presence of fungi in nature is very abundant, but at this time it is still necessary to know and explore the various types that are able to live in various places. Fungi can live in plant tissue, known as endophytic fungi, and can also live on the leaf surface of a plant. Avicennia marina lives on the coast and is used to reduce abrasion, besides being able to live in tidal habitats and high salinity levels. This is supported by the plant's physiology and the help of its microbial symbionts, therefore it is necessary to explore fungi, especially fungi that are able to symbionting with these plants because they are thought to be resistant to high levels of salinity. The purpose of this study was to determine the types of molds that have been isolated and are able to live in the tissue and leaf surface of A. marina. Characterization method was reviewed through macroscopic and microscopic morphology using a microscope and compared with literature from various sources. The characterization results of the 12 isolates obtained several genera, including Trichoderma, Aspergillus, Penicillium, Curvularia, Paecilomyces, Talaromyces, Syncephalastrum, and 1 unidentified isolate. These isolates need to be continued with further research to see their ability to produce metabolites and so on.

Abstract

Kempong is a traditional fermented food that is found only in South Karangpucung    Linggapura Bumiayu village, Central Java. It is prepared from palm kernel cake (PKC).  This fermented food is consumed mostly everyday by the people in the village as a side dish or snack. Study on the mold important in the kempong fermentation was done by isolating molds from kempong, PKC, laru and the air of preparation room. Eleven species of molds were successfully isolated from different samples. Among the mold isolates three species came out from kempong product, i.e. R. oryzae, E. chevaliery and A. oryzae. The isolates showed the capability in producing amylase, protease, lipase, and cellulase. Both R. oryzae and A. oryzae indicated as the main fermentation mold in kempong production, because the inoculum laru only contained these two species.

Abstract

AbstractLipstick is a non-sterile pharmaceutical product that is used for make-up on the lips. Liquid lipstick used by makeup artists may have a high level of microbial contamination because they are applied to different clients in turn. This study aims to determine the microbiological quality of liquid lipstick used by makeup artists based on the result of the Bacterial Total Plate Count (TPC), Yeast and Mold Count (YMC), and the presence of pathogenic microbes. Three samples of liquid lipstick were taken from three different makeup artists and one sample was a new liquid lipstick product. The Bacterial TPC test was carried out on TSA media and the YMC test was carried out on SDA with the spread plate technique. The presence of microbial pathogens P. aeruginosa, S. aureus, and C. albicans were carried out on selective media. The results of bacterial TPC was 9.0 × 103 to 3.0 × 104 CFU/mL, while the YMC was 1.1 × 103 to 3.1 × 103 CFU/mL. The pathogenic microbes detected were S. aureus in 2 samples and C. albicans in 1 sample. The bacterial TPC and YMC values exceed the contamination limit based on BPOM Regulation No. 12/2019.

Abstract

Kapang merupakan cendawan berfilamen yang hidup pada berbagai subsrat dan memiliki habitat yang beragam, antara lain hidup pada tanah, air, serealia, endofit dalam tanaman, dan buah. Habitat kapang yang beragam juga memperbanyak diversitas kapang pada berbagai substrat, salah satunya pada buah kedondong. Salah satu kapang yang sering ditemukan adalah Aspergillus. Kapang Aspergillus terdiri dari beberapa grup salah satunya adalah Aspergillus seksi Nigri yang lebih dikenal dengan black mold. Tujuan dari penelitian ini adalah untuk mengidentifikasi kapang Aspergillus seksi Nigri pada buah kedondong yang telah rusak atau busuk. Isolasi kapang tersebut menggunakan metode secara langsung (purposive sampling) dengan mengambil kapang Aspergillus yang diduga berasal dari seksi Nigri, lalu diinokulasikan ke media PDA, MEA, dan CDA. Kapang kemudian diamati morfologinya secara makroskopis dan mikroskopis serta selanjutnya diidentifikasi untuk mengetahui spesiesnya. Pada penelitian ini diperoleh hasil sebanyak sembilan isolat yang berasal dari buah kedondong, diantaranya Aspergillus niger, Aspergillus foetidus, Aspergillus aculeatus, Aspergillus japonicus dan Aspergillus sp. Penelitian selanjutnya isolat tersebut dapat diidentifikasi secara molekuler dan diuji potensinya.

Abstract

Protease is the most important enzyme in many industries, including detergent. Aspergillus flavus DUCC-K225 is a thermotolerant indigenous molds isolated from Madura island which is potential in producing thermostable alkaline protease enzymes. The enzyme produced by submerged culture on modified Czapeks Dox liquid medium containing glucose as carbon source and 1% of casein. The aims of this study were to determine the activity and stability of thermostable alkaline protease produced by A. flavus DUCC-K225 at various temperatures, also the compatibility to 5 local detergents. Research were done using CRD, with temperature variation for protease activity as treatment. The results showed that the optimum activity of thermostable alkaline proteases was 214.503 U/mL, with retained activities up to 78% in 60 minutes at 55°C. The alkaline thermostable protease compatible with 5 local detergents tested, with the retained activity varied 55.8%-95.22% at 29oC and 62.36%-97.7% at 55oC. The compatibility to detergent confirmed by blood stained cleansed. The higher the protease activity, the higher the blood stain solubility. Abstract

Indonesia has an abundant biodiversity of natural resources, one of which is phylloplane fungi. This abundant phylloplane mold needs further investigation to utilize its potential as an antagonistic agent. This study aims to prove the existence and to determine the types of phylloplane fungi species found on the leaf surface of pacing tawar (sweet ginger or crepe-ginger) plants, to analyze and determine the level of diversity and dominance of phylloplane, and to determine the types of phylloplane fungi species that have the antagonistic ability of the phytopathogenic mold Fusarium oxysporum. This research method includes isolating crepe-ginger phylloplane fungi using a contact plate, isolates purification, macromorphological and micromorphological identification, diversity and dominance index calculation, and antagonist test using a dual culture method. This study used a descriptive analysis research design. The isolation results obtained 57 species of crepe-ginger plants leaf phylloplane molds consisting of eight genera (Alternaria, Aspergillus, Cladosporium, Curvularia, Fusarium, Mucor, Penicillium, and Trichoderma). The results of the diversity index calculation with a value of 2.02 showed that the diversity of phylloplane fungi was moderate. The result of the dominance index calculation, with a value of 0.1521, shows no tendency for phylloplane fungi species to dominate. The results of the antagonist test showed that 15 isolates could antagonize the F. oxysporum with an inhibitory proportion of up to 82% obtained from T. viride isolates with code FD5B3.2

Abstract

Staphylococcus aureus merupakan bakteri penyebab infeksi kulit dan pada ulkus diabetikum, sehingga dibutuhkan antibakteri untuk mencegahnya. Daun jambu biji (Psidium guajava L.)  mengandung antimikroba. Spray gel mencegah infeksi silang, praktis, dan meningkatkan penetrasi obat. Kombinasi Na CMC dan kopovidon menghasilkan spray gel yang stabil dan membentuk lapisan film elastis. Penelitian ini bertujuan untuk mengetahui pengaruh kombinasi konsentrasi Na CMC dan kopovidon sebagai gelling agent terhadap karakteristik fisik spray gel SMEDDS ekstrak etanol 96% daun jambu biji dan aktivitas antibakterinya. Simplisia daun jambu biji dimaserasi dengan etanol 96%, kemudian dibuat SMEDDS, dan diformulasikan dalam spray gel dengan memvariasikan Na CMC dan kopovidon 1:1 (F1=0,1%, F2=0,2%, F3=0,3%). Uji KHM terhadap Staphylococcus aureus dilakukan pada ekstrak. Karakterisasi fisik dilakukan terhadap spray gel. Aktivitas antibakteri spray gel dilakukan dengan metode Kirby Bauer. Hasil yang didapat, yaitu KHM ekstrak sebesar 0,5%. Konsentrasi Na CMC dan kopovidon tidak berpengaruh terhadap karakteristik fisik spray gel. F2 dan F3 memenuhi semua peryaratan karakteristik fisik, sedangkan F1 tidak memenuhi persyaratan viskositas. Spray gel F1, F2, dan F3 memiliki aktivitas antibakteri terhadap Staphylococcus aureus berturut turut sebesar 28,10; 22,35; dan 22,09 mm, tetapi berbeda tidak bermakna dengan kontrol negatif.

Abstract

 Ulkus diabetik dari diabetes mellitus berupa luka terbuka pada permukaan kulit dan adanya kematian jaringan tubuh. Bakteri patogen paling utama ditemukan pada ulkus diabetik, yaitu Staphylococcus aureus. Daun kersen (M. calabura L.) dilaporkan mengandung senyawa metabolit sekunder yang berpotensi sebagai agen antibakteri dan diketahui ekstrak etanol 70% daun kersen (M. calabura L.) memiliki aktivitas antibakteri terhadap S. aureus. Gel mengandung komponen air yang lebih tinggi dibandingkan sediaan topikal lainnya. CMC Na dan gliserin digunakan sebagai gelling agent dan humektan. Penelitian ini bertujuan untuk mengetahui pengaruh variasi konsentrasi CMC Na dan gliserin terhadap karakterisitik gel dan aktivitas antibakteri terhadap S. aureus, serta mengetahui formula optimum sediaan gel ekstrak etanol 70% daun kersen (M. calabura L.) dengan variasi konsentrasi CMC Na dan gliserin yang memiliki karakteristik fisik gel dan aktivitas antibakteri yang baik. Simplisia daun kersen dimaserasi dengan etanol 70%, kemudian maserat dikentalkan dengan rotary evaporator. Gel diformulasikan dengan memvariasikan konsentrasi CMC Na dan gliserin, lalu dikarakterisasi fisik dan uji aktivitas antibakteri. Penentuan formula optimum dilakukan menggunakan design expert. Hasil yang didapatkan yaitu variasi konsentrasi CMC Na dan gliserin berpengaruh signifikan pada pH, daya lekat, dan daya sebar, tetapi tidak berpengaruh signifikan pada aktivitas antibakteri terhadap S. aureus. Formula optimum gel ekstrak etanol daun kersen adalah kombinasi konsentrasi CMC Na 2,234% dan gliserin 11,766%.
