Abstract

Piper betle and Piper crocatum leaf extract each has proven to have antimicrobe effect against Candida albicans, this is the reason why piper betle leaf often used as mixture of mouthwash or vagina cleanser. This research aim is to know the antimicbial activity of combination between stewed piper betle and piper crocatum leaf against Candida albicans. Experimental design that used is Completely Randomized Design (CRD) factorial design with 2 factor. Combination  of stewed green betel leaf and red betel leaf respectively 100% P.betle and 0% P. crocatum; 75% P.betle and 0% P. crocatum; 50% P.betle and 50% P. crocatum; 25%  P.betle and 75% P. crocatum; 0% P.betle and 100% P. crocatum. A negative control is steril aquadest, while second factor is contact time 0, 30 and 60 minute. The variables meansured were the growth of colonies in each treatment combination. The result showed that combination of 100% P.betle and 0% P. crocatum; 75% P.betle and 0% P. crocatum; 50% P.betle and 50% P. crocatum; 25%  P.betle and 75% P. crocatum have antimicrobial activity is  high. This result proven by absence of colony growth after treatment. The best antimicrobial activity found on treatment with only addition of red betel at 60 minutes of contact time. Keywords : Piper betle, Piper crocatum, antimicrobial, Candida albicans

Abstract

Advances in the field of fermentation technology, genetic engineering, and technology applications of enzymes lead to the use of enzymes in the industry is increasing. Enzymes used in the industry can be derived from microorganisms such as bacteria, molds, and yeasts. Protease is one of the important enzyme that has been used widely in various fields of industrial application and 65% of the sales of enzymes in the world. Alkaline protease is one of hydrolytic enzymes that can hydrolysis proteins which working at the pH range of 7-12. The aim of this study was  to investigate the influence of pH and incubation time on the protease production of A. niger PAM 18A. The experimental were done using Factor Block Random Design with 2 i.e pH of7, 8, 9, and incubation time of 5, 6, 7 days. The results showed that highest protease fromA. niger PAM18A were found in the treatment of pH 8 and of 5 days incubating with the protease activity and specific activity were 0,930 U/mL and 3,632U/mg respectively.Keywords: Production Enzyme, Protease, A. niger PAM18A, pH, incubation time.

Abstract

Diabetes mellitus (DM) is a hyperglycemia disease caused by insensitivity of cells to insulin that causes the presence of glucose in blood and urine. The α-amylase inhibitors is a compound that inhibit carbohydrates breakdown into glucose by amylase enzyme. Binahong (Anredera cordifolia) has been used by Indonesian people since long time ago to treat various diseases such as for anti-diabetic. The aims of study were to obtained the endophytic fungal isolated from Binahong plant, and find out of their ability of α-amylase inhibitor activity by treatment of carbon source and pH of production medium. The isolating endophytic fungal from binahong roots, stems, and leaves in medium PDA. The α-amylase inhibitory test was using starch substrate and the α-amylase inhibitory measure using spectrophotometric method. The study was used one factor CRD i.e. carbon source including sucrose, lactose, and maltose. The best carbon source then continued to test with variation pH 5,6, and 7. The data obtained were then analyzed by Analysis of Variance (ANOVA). Five endophytic fungal isolated from binahong were only one isolated i.e DT should high potensial in inhibiting α-amylase 91,43%. The best carbon source for inhibitory α-amylase i.e sucrose with the percentage of 58,91%, while pH 5 is the best pH medium production continous treatment carbon source sucrose with the percentage 37,83%. The result of statistical test showed that sucrose as the best carbon source just than lactose and maltose, and pH 5 as the best medium pH for the production α-amylase inhibitor. Key words : α-amylase inhibitor, endophyte fungi, Anredera cordifolia, diabetes mellitus

Abstract

East Kalimantan has a lot of processed food products traditionally, one of them is mandai which is fermented food made from fruit leather Cempedak (Arthocarphus champeden). Lactic acid bacteria involved in the fermentation mandai greatly affect the quality of the final product. This study aimed to perform the isolation and characterization of lactic acid bacteria in the mandai fermentation process. MRS medium was used in the isolation of bacteria for 14 days. Isolation of bacteria during the fermentation process mandai obtaining 17 isolates, nine isolates Gram negative isolates, and eight isolates Gram positive. Eight isolates showed positive results against some of the morphological and biochemical characterization. Including biochemical characterization, acid formation and gas production from glucose, motility test, catalase test, growth at 160C and 480C, hydrolysis of starch, fat hydrolysis, and hydrolysis of casein and acid formation test. Eight isolates bacteria made mandai into processed food products that had a sour taste. Keywords: lactic acid bacteria, BAL ,bacterial isolates, cempedak skin.

Abstract

Plastic waste is one of the environmental issues that still need to be handled properly. plastic  commonly  used  are  basically  formed  by  polyethylene  which  is  contains  chemical compound.  Less cavity of plastic can disturb soil aeration which is important for other aerobic organism. This problem lead to increasing ecological threat. The main objective of present study is to isolate the soil microorganism that have a role as an agent  through plastic degradation. Bacteria were isolated from the surface of the plastic which is buried in soil. The  isolation is been carried out through soil serial dilution method and inoculated into selective King’s B agar. Isolates were obtained and then tested the ability of degradation of the PE. Isolates with the highest degradation  activity isolates G then identified using molecular methods based on 16S rRNA. the results showed that the 16S rRNA sequencing isolates G measuring 1,500 bp with the highest homologous species derived from the genus Bacillus. Keywords: Polyethylene, bacteria, biodegradation

Abstract

 Produksi fruktosa secara langsung dari inulin oleh inulinase hanya memerlukan satu tahap reaksi enzimatis dan menghasilkan 95% fruktosa. Inulin diperoleh dari tepung umbi dahlia dan inulinase diproduksi Pichia manshurica DUCC Y-015. Produksi inulinase (E.C. 3.2.1.7) dapat dipengaruhi oleh keberadaan glukosa sebagai sumber karbon tambahan. Tujuan penelitian ini adalah untuk menganalisis pengaruh variasi konsentrasi glukosa dan waktu inkubasi terhadap produksi inulinase khamir P. manshurica DUCC-Y015 dalam substrat umbi dahlia. Pengukuran produksi enzim meliputi aktivitas inulinase dan aktivitas invertase. Aktivitas inulinase dan invertase dianalisis dengan metode DNS dan ditentukan berdasarkan 1 µmol fruktosa yang dihasilkan. Rancangan yang digunakan dalam penelitian ini adalah Rancangan Acak Kelompok (RAK) pola faktorial. Faktor I berupa konsentrasi glukosa 0% (G0) ; 0,25%  (G1) ;  0,5% (G2) dan faktor II berupa waktu inkubasi 6 jam (T6), 12 jam (T12), dan 18 jam(T18) dengan pengulangan sebanyak 3 kali. Data yang diperoleh dianalisis dengan metode ANOVA. Hasil penelitian menunjukkan bahwa variasi konsentrasi glukosa, perbedaan waktu inkubasi maupun  interaksi keduanya berpengaruh tidak signifikan  terhadap produksi enzim inulinase. Produksi enzim inulinase  tertinggi dari P. manshurica DUCC Y-015 ditunjukkan oleh perlakuan G1T12 dengan konsentrasi glukosa sebesar 0,25%  dan waktu inkubasi 12 jam dengan nilai aktivitas enzim sebesar 0,574 IU/mL.

Abstract

Sago processing waste is one of agro-industry waste, which contains cellulose 19,55% and lignin 25%. Genus Aspergillus has been widely known as cellulase-producing fungi. Isolation of Aspergillus from sago processing waste is expected to get a potential isolates indegenous cellulolytic fungi. Cellulase (EC 3.2.1.4) is an enzyme that can break down cellulose into glucose monomer form, with consists of three components that work in synergy are endoglucanase, eksoglucanase and β-glucoside. This study aims to obtain isolates of Aspergillus sp. cellulolitic potential from sago processing waste and cellulase production of the cellulolytic isolates in various concentrations of inoculum. The study was conducted using completely randomized design (CRD) 1 factor of 4 levels of inoculum concentration treatments were 0%, 2,5%; 5%; and 10% with three replications. The observed variables are biomass, CMCase activity and FPase activity. Data were analyzed by ANOVA with a significance level 95%. The result of isolation, two isolates of Aspergillus namely A. flavus and A. paradoxus with cellulolitic index respectively 2,63 and 2,06. Cellulase activity isolate of A. flavus either endoglucanase (CMCase) or total cellulase (FPase) are not affected by the concentration of inoculum.Keywords: cellulase, sago waste, Aspergillus, inoculum

Abstract

AbstrakPentana, dekana, pentadekana, dan parafin merupakan jenis kelompok senyawa alkana yang tidak dapat larut dalam air dan sulit terdegradasi. Sifat ini yang menjadikan senyawa alkana dapat mencemari lingkungan. Penelitian ini bertujuan untuk mengetahui konsentrasi optimum senyawa alkana bagi pertumbuhan isolat LBF-1-0025 dan mengidentifikasinya secara molekuler berdasarkan gen 16S rDNA. Dari skrining awal diketahui bahwa isolat LBF-1-0025 memiliki potensi tinggi dalam mendegradasi senyawa pentana, dekana, pentadekana, dan parafin. Optimasi pertumbuhan isolat LBF-1-0025 pada beberapa senyawa alkana dilakukan dengan menumbuhkannya dalam medium Artificial Sea Water (ASW) yang mengandung beberapa konsentrasi senyawa pentana, dekana, pentadekana, dan paraffin. Isolat LBF-1-0025 mampu tumbuh secara optimal pada pentana 150 ppm, dekana 200 ppm, pentadekana 150 ppm, dan parafin 200 ppm setelah 3 hari inkubasi. Hasil analisis sekuen gen 16S rDNA, bakteri LBF-1-0025 memiliki kemiripan sebesar 97% dengan Salinicola peritrichatus DY22.The Growth Optimization of Marine Bacteria  Salinicola  peritrichatus LBF-1-0025 in AlkanesAbstractPentane, decane, pentadecane, and paraffin are kind of groups of alkane compounds that are insoluble in water and difficult to degrade. This characteristic cause alkane compounds to pollute the environment. The aims of this research were to determine the optimum concentration of alkane compounds for the growth of isolate LBF-1-0025 and molecular identification based on 16S rDNA gene. From initial screening test, it was known that isolate LBF-1-0025 had high potential in degrading pentane, decane, pentadecane and paraffin compounds. Growth optimization of isolate LBF-1-0025 on several alkane compounds were carried out by culturing it in ASW medium containing various concentration of alkane (pentane, decane, pentadecane, and paraffin). Isolate LBF-1-0025 had optimum growth in pentane 150 ppm, decane 200 ppm, pentadecane 150 ppm and paraffin 200 ppm after 3 days incubation. Result analysis of 16S rDNA gene showed that isolate LBF-1-0025 had a similarity of 97% with  Salinicola  peritrichatus DY22.

Abstract

Broiler chicken meat is a a kind of food that is easily spoilage due to bacterial growth. The chicken meat sold in the market could potentially become a medium of microbial growth of foodborne diseases. The aims of this research was to  determined the microbiological quality of chicken meat sold at traditional market in  Banyumanik, Semarang City and  examined the source of bacterial contaminant especially E. coli on chicken meat. Samples were taken from two markets in Banyumanik, namely Dm Market and Jt Market of Semarang. The samples were purposive taken, from two different sellers in each market. Sampling was conducted at 08.00 AM and 10.00 AM. The parameters in this study are the total number of bacteria, the number of E. coli and the presence of Salmonella sp.  The results showed that all samples examined did not meet the standard of good quality microbiological requirements for chicken meat according to SNI 3924:2009, regarding total bacteria, E. coli content and the presence of Salmonella sp. The results of observations by contact plate technique found that 50% of tools used by chicken meat sellers were contaminated by E. coli.
