Abstract

Spesies asing invasive adalah salah satu isu strategis di Indonesia karena keberadaanya menjadi ancaman terhadap kelestarian keanekaragaman hayati dan keberlanjutan konservasi ekosistem. Termasuk spesies ikan nila (Oreochromis niloticus) di Danau Dampelas, komoditas ini bukan asli Sulawesi sehingga dikhawatirkan mempengaruhi populasi spesies asli, komposisi struktur dan proses ekosistem. Penelitian ini bertujuan untuk identifikasi spesies invasive di Danau Dampelas secara morfologi dan molekuler. Identifikasi molekuler dilakukan dengan menggunakan DNA mitokondria berdasarkan daerah Cytochrome Oxidase Sub Unit I (COI). Sampel ikan nila dikumpulkan dari tujuh stasiun pengamatan di danau Dampelas. Hasil identifikasi dengan metode morfologi dan molekuler menunjukan hasil yang sama, yaitu Oreochromis niloticus. Berdasarkan identifikasi morfologi Oreochromis niloticus memiliki bentuk pipih dan oval, kepala sedikit runcing, warna tubuh kehitaman atau keabuan. Sirip ekor memiliki garis vertical, letak mulut terminal, sirip dorsal memanjang dengan mengeras dan bentuk ekor truncate. Hasil penelitian molekuler menunjukan bahwa semua sekuen ikan berhasil diamplifikasi dengan gen COI (654 bp). Hasil BLASNT menunjukan persentase identitas 100%.

Abstract

Rice pond cultivation is an effort to maintain fish in rice fields before planting rice and after the rice is harvested, the type of pond paddy field cultivation in Lamongan Regency uses a rotating rotation type that has a time interval for fish maintenance during the rainy season (Sequential/Alternate farming). Liquid organic fertilizer is a type of fertilizer derived from the fermentation of organic matter. The fermentation of these organic materials can produce more than one nutrient. The use of organic waste such as rice straw, water hyacinth, and fish offal can be used as an alternative source of nutrients that can be used to make liquid organic fertilizer. The expected target of this activity is training in the use of local resources to overcome the termination of inorganic fertilizer subsidies. Prolonged use of inorganic fertilizers will have an impact on the environment because it can result in nutrient imbalances. An alternative that can be done is the application of organic fertilizer to continue to increase the productivity of rice pond cultivation in Lamongan Regency. The manufacture of organic fertilizer can utilize local natural resources including rice straw, water hyacinth and fish offal which can be used as Liquid Organic Fertilizer ingredients. Making probiotics and composted cow dung as organic fertilizer to increase natural feed production. This activity was attended by 50 participants consisting of farmer groups from Babatagung Village, Deket District, Lamongan Regency. Before and after the training, questionnaires were filled out to the trainees for activity evaluation materials. Based on the questionnaire that has been distributed, 98% of fish farmers have known about making organic fertilizers from local resources. As many as 94% of farmers are interested in applying organic fertilizer to fish farming ponds and 70% of farmers are interested in increasing their income through the sale of locally-based organic fertilizers. In addition, farmers are also provided with how to maintain and measure water quality in fish farming ponds. After the training on making Liquid organic fertilizer, Liquid organic fertilizer was applied to denfarm ponds to determine the advantages of POC application.

Abstract

Manyar Sub-district, located in Gresik, is home to many fish and shrimp pond farmers. Vannamei shrimp farming is a particularly popular aquaculture activity in Tanggul Rejo Village, Manyar Sub-district. However, the shrimp and fish farming industries face challenges in production, primarily due to diseases arising from declining water quality. Many pond farmers lack knowledge about the importance of water quality management to support successful shrimp farming, highlighting the need for appropriate technology to improve production. This study aimed to apply probiotics to manage the water quality of vannamei shrimp rearing media in Manyar Sub-district. The activity was carried out in several stages, including identifying and mapping the problems faced by farmers, delivering educational materials about the use of probiotics in shrimp farming, providing assistance for probiotic application, and monitoring and evaluating the results. The findings showed that the application of probiotics to the rearing water media every two days resulted in low ammonia and nitrite levels, as well as adequate oxygen levels and pH, which are conducive to vannamei shrimp cultivation.

Abstract

Textile waste consists of insoluble solid particles, salts, dyes and heavy metals and is very difficult to degrade. Chitin derived from shrimp waste has the potential to overcome textile waste in chitin that causes environmental pollution through more economical absorption and easily available raw materials. Factors that improve adsorption performance include pH and temperature. This study was conducted to determine the effect of temperature, pH, and a combination of temperature and pH treatments on the ability of chitin to adsorb textile dye waste. The parameters observed were temperature, pH, combination of temperature and pH, functional groups (FTIR), chitin structure (SEM), and heavy metal test (AAS). Data analysis of statistical test results using two-way Analysis of Variance (ANOVA) followed by Duncan's further test. The ANOVA test results showed that the incubation treatment of temperature, pH, and a combination of different temperature and pH treatments resulted in (p < 0.05). This shows that incubation temperature and pH have an effect (significantly different) on the adsorption of chitin as an adsorbent on dyes. The results of Duncan's test showed that the best chitin adsorption was obtained in temperature incubation at 50°C (A3), incubation at pH 3 (B1), and the best combination adsorption at 50°C and pH 3 of 1,091±0.007a ppm.

Abstract

Udang vaname merupakan salah satu jenis udang yang memiliki nilai ekonomis tinggi. Hal ini dikarenakan banyaknya permintaan konsumen terhadap udang vaname. Namun budidaya udang vaname tidak mudah, komoditas tersebut mudah terserang penyakit apabila padat tebar terlalu tinggi dan kualitas air yang buruk. Kualitas air yang buruk dapat mengakibatkan imunitas udang vaname menurun dan mengakibatkan mudah terserang penyakit salah satunya Acute Hepatopancreatic Necrosis Disease (AHPND). Penyakit tersebut dilaporkan salah satunya diakibatkan oleh adanya barkteri Vibrio parahaemolyticus yang menginfeksi udang. Tujuan penelitian ini adalah untuk mengetahui kerusakan jaringan udang vaname yang terinfeksi bakteri Vibrio parahaemolyticus di tiga tambak yang berbeda. Metode yang digunakan pada penelitian ini adalah purposive sampling dengan metode observasi. Teknik pengumpulan data dilakukan dengan cara observasi lapang, yakni mengumpulkan data-data primer dan sekunder di lapangan. Data diambil dari tiga lokasi, yaitu Tuban, Sidoarjo dan Pasuruan. Hasil histopatologi dilakukan skoring kerusakan kemudian dianalisis dengan IBM SPSS menggunakan Uji Kruskal Wallis dan dilanjutkan Uji Z. Berdasarkan hasil penelitian menunjukkan bahwa kerusakan jaringan pada insang dan hepatopankreas akibat infeksi dari bakteri V.parahaemolyticus memiliki kerusakan yang cukup parah. Kerusakan yang terjadi pada insang mulai dari fusi lamella dan edema. Sedangkan kerusakan yang terjadi pada hepatopankreas adalah peluruhan sel dan infiltrasi. Maka dapat disimpulkan bahwa ketiga tempat yang terinfeksi V.parahaemolyticus terdapat perbedaan tingkat kerusakan jaringan hepatopankreas dan insang pada udang vaname yang terjadi pada hepatopankreas dan insang banyak hingga sangat banyak. Kerusakan terbanyak terjadi pada sampel udang yang berasal dari Kabupaten Sidoarjo

Abstract

Grouper (Epinephelus sp.) is a marine fishery commodity that is widely cultivated in marine cages and ponds. The export market demand for grouper fish continues to increase every year by 30.75% every year, so there is a need for improvement, through various technological applications. The main obstacle in cultivating grouper fish in ponds is the decline in water quality and disease attacks, which can cause up to 100% crop failure, just 3 – 5 days after infection. One effort that can be implemented is the cultivation method with the Immuno-Probiocirculation System (SIPBR). Brondong sub-district is one of the areas in Lamongan which has large fisheries potential, especially for breakwater water pools, which are the top among other districts. There have been many cases of grouper deaths in ponds to date, which are caused by disease attacks and water quality. The aim of this community service activity is to apply the Immuno-Probioculation System fish aquaculture technology in traditional plus pattern ponds (SI-PBR), increasing grouper fish production in Lamongan Regency, East Java. Observational method which includes socialization / counseling, planning and guidance for implementing SI-PBR technology in one period (three months). These results show positive indications. There is an increase in farmer knowledge, the SI-PBR method can reduce ectoparasite infestation and increase grouper yields from 272.43 kg / ha to 87.66 kg / ha, meaning an increase of 334%. The conclusion from this activity is that the SI-PBR model can increase fish production and can be applied in wider areas in the Lamongan region.

Abstract

Caulerpa racemosa, a type of green algae, has been studied  for its potential health benefits, including antioxidant and an- tibacterial properties. The extraction of bioactive compounds  from marine algae using green solvents like Deep Eutectic Sol- vents (DES) has gained interest due to their efficiency and en- vironmental friendliness. This research aimed to determine  the potential antioxidant activity of C. racemosa extract using DES solvents and different sonication times as an antibacterial  against pathogenic bacteria. The research employed a Facto- rial Completely Randomized Design (CRD) with two variables:  sonication time (10, 20, and 30 minutes) and different DES solvents (ethanol and ethylene glycol (1:20) with 90% aquades, and methanol and glycerol (1:10) with 90% aquades. The highest antioxidant activity was found in DES  Methanol-Glycerol (36.39 mg TE/g dw), followed by DES Eth- anol-Ethylene glycol (28.30 mg TE/g dw), both significantly  different (p<0.05). The C. racemosa extract inhibited the growth of V. parahaemolyticus with an inhibition zone of 8.08 mm at 50 ppm and 13.13 mm at 1,000 ppm. For V. harveyi, inhibition started at 6.37 mm at 50 ppm, reaching 11.70 mm  at 1,000 ppm. DES Methanol-Glycerol with a 10-minute soni- cation provided the highest antioxidant activity and effectively  inhibited V. parahaemolyticus and V. harveyi. Extraction of C. racemosa with DES-MG solvent and varying sonication times influences its antioxidant activity and antibacterial properties.

Abstract

One alternative approach to facilitate the utilization of feed by tilapia is the addition of probiotic in feed. In this study, we compare the effects of commercial probiotic EM4 and homemade probiotics on tilapia growth. This research was conducted in PT. Sahaja Berkah Abadi at wonosalam, jawatimur, indonesia. The study was designed as a completely randomized design with 3 treatments and 4 replications, encompassing control group (C), commercial probiotics (G1), and homemade probiotic (G2). The commercial probiotics (G1) contained Rhodopseudomonas sp., Lactobacillus sp., Streptomyces sp., and Actinomycetes sp. while the homemade probiotic (G2) contained starter bacteria Lactobacillus casei and herbal plant such as ginger, curcuma, and tumeric. Parameters observed during the study included weight gain (WG), specific growth rate (SGR), and survival rate (SR). The results revealed that both G1 and G2 performed better than the control group. The weight gain for the control, G1, and G2 groups were 2.38±0.03 g, 3.09± 0.01 g, and 3.13 ± 0.03 g, respectively. Similarly, the specific growth rates for the control, G1, and G2 groups were 4.25±0.03%, 5.52± 0.02%, and 5.59 ± 0.03%, respectively, while the survival rate remained at 100% across all groups. The water quality in all treatments during maintenance met the standard requirements for tilapia, with temperatures ranging from 25.5 to 26.7 °C and pH levels between 6.7 and 6.9. These findings suggest that the probiotics incorporated in G2 present a cost-effective alternative to those in G1, offering a promising option for tilapia cultivation.

Abstract

The purpose of this study was to investigate the water quality and phytoplankton abundance in Lake Toba to determine its trophic status. This research was conducted from January to March 2024 at four stations in Lake Toba. Water samples were taken using a UWITEC water sampler and a plankton net and brought to the laboratory for analysis. The study measured physical and chemical water quality and phytoplankton abundance. The results showed a brightness ranging between 5.8 and 6.3 meters, chlorophyll-a content <10 µg/L, total nitrogen <750 µg/L, and phosphorus ranging from 20 µg/L to 30 µg/L. Based on these parameters, the nutrient quality of Lake Toba was classified as mesotrophic. Phytoplankton abundance ranged from 22,514 to 33,475 individuals/L, with 33 genera identified across nine classes: Bacillariophyceae, Chlorophyceae, Cyanophyceae, Coscinodiscophyceae, Dinophyceae, Euglenida, Fragilariophyceae, Mediophyceae, and Zygnemophyceae. The diversity index (H') was 2.631 - 2.723, the uniformity index (E) was 0.828 - 0.872, and the percentage of blue-green algae was 14.75 - 20.85. Zygnemophyceae was the most dominant phytoplankton class, with Staurastrum sp. being the most prevalent species. In conclusion, Lake Toba's waters are categorized as mesotrophic based on the chemical parameters and as eutrophic based on phytoplankton abundance, indicating potential nutrient enrichment issues that warrant further investigation.

Abstract

Pasuruan Regency is one of the regencies that has potential in the fisheries sector, especially freshwater fisheries cultivation. The potential area for cultivation activities is located in Sukorejo District with the cultivated commodity being catfish. One of the farmer groups in Sukorejo District is the Pondok Darul Mukmin Sangkuriang Catfish farmer. However, this catfish cultivation activity has not been running optimally, because the cultivation system used is still traditional. This community service activity aimed to provide assistance related to water quality management and probiotic application in catfish cultivation. This activity was carried out at Pondok Darul Mukmin from July to September 2024. The method of implementing the activity was in the form of training, assistance, and demonstrations of water quality management, probiotic application in feed, and probiotic culture for catfish cultivation. The results of this community service activity showed that Pondok Darul Mukmin fish farmer  were very enthusiastic in participating in the training activities. In addition, the community can also carry out catfish cultivation by monitoring water quality and applying probiotics to feed. Cultivation activities resulted in increased catfish growth every week.
