Abstract

Substituting a part of soil on tea Camellia Sinensis (L.) O. Kuntze) nurseries by using organic matter could minimize the remove fertile soil from nature. Biofertilizing are a way to improve the biological quality of soil and subsequently tea seedling quality. The aim of the research was to determine the effect of planting media composition and biofertilizer doses on total fungal and bacterial population on planting media; as well as tea seedlings growth. The experiment was conducted using a randomized block design consisting of 9 treatments and 4 replications, each replication consisted of three plants. The treatment consisted of soil without biofertilizer (control), soil + 5% biofertilizer, soil + 10% biofertilizer, cocopeat without biofertilizer, cocopeat + 5% biofertilizer, cocopeat + 10% biofertilizer,  filter cake without biofertilizer, filter cake + 5% biofertilizer, and filter cake + 10% biofertilizer. Biofertilizer used in this experiment were Azotobacter vinelandii and Azotobacter chroococcum that was mixed with growing media before planting. Result of the experiment showed that planting media affect microbial populations differently. The population of total bacteria, total fungi and Azotobacter sp. in the filter cake media with 10% biofertilizer was significantly higher than those of other growing media. However, Azotobacter biofertilizer had no effect on seedling growth of tea (Camellia Sinensis (L.) O. Kuntze) clones Gambung-7

Abstract

Gold mine tailing in Buru island, Maluku Province, Indonesia contains high level of mercury but low in carbon and plant nutrients. Revegetation in such condition needs certain soil treatment which is suitable for plant growth. The objective of pot trials was to study the effect of indigenous Plant Growth Promoting Rhizobacteria Azotobacter and organic matter on bacterial survival as well as growth of groundnut grown in mine tailing. The experimental design was a Split Plot Design which tested three types of Azotobacter liquid inoculant and three soil total organic carbon (TOC) contents. Results showed that Azotobacter inoculation increased Azotobacter population in tailing at the end of vegetative growth of groundnut. Total nitrogen content in soil decreased when TOC level increased. However, nitrogen uptake and growth of groundnut did not change after Azotobacter inoculation or manure amendment. These experiments provided information that Azotobacter inoculation on groundnut maintain its proliferation in Hg-contaminated mine tailing.

Abstract

Corn cultivation on Ultisols is limited due to low nitrogen content of the soil. Efforts to increase the N content in the soil include the use of biofertilizers by symbiotic nitrogen-fixing endophytic bacteria. This study assesed various combination of nitrogen fertilizer doses and N2-fixing endophytic biofertilizer to increase the growth of maize in Ultisols Kentrong. The experiment was conducted at the Ciparanje Experimental Greenhouse, Faculty of Agriculture, Universitas Padjadjaran, Jatinangor. A randomized block design was used which consisted of 16 treatments with 3 replications, with a combination of doses of urea fertilizer and endophytic biofertilizer applied to seeds or planting holes. The experimental results showed that the combination of urea fertilizer and biofertilizer doses had a significant effect on the population of endophytic bacteria in the roots and leaves of corn plants, plant wet weight, root dry weight and shoot dry weight, plant height, stem diameter, plant nitrogen uptake and content of plants but was not significantly affected on the number of corn leaves. Combination of 80 kg ha-1 urea fertilizer and 3.6 kg ha-1 of biofertilizer was the best combination which increased the dry weight of the corn plant and reduced the dose of urea fertilizer

Abstract

The concept of balanced fertilization application is appropriate for tomato plant cultivation. However, most farmers still apply inorganic fertilizers excessively, which negatively impacts the environment and plant productivity. Alternative efforts can be made to reduce the usage for inorganic fertilizers through effective plant growth-promoting rhizobacteria (PGPR) application. This research studied the characteristics of the PGPR inoculants effectiveness (Azotobacter sp. and Pseudomonassp.) combined with NPK towards tomato in Lembang Andisol. The experimental design used was randomized block design, with three replications: A (Control), B (1 PGPR + 0 NPK), C (1 PGPR + NPK), D (1 PGPR + NPK), E (1 PGPR + NPK), F (1 PGPR + 1 NPK), G (1 PGPR + 1 NPK), H (2 PGPR + 1 NPK), I (0 PGPR + 1 NPK), J (½ PGPR +1 NPK). The results showed the treatments of C, F, and J resulted in higher total soil N content than the control treatment. Meanwhile, C and D treatment increased plant N uptake compared to control. Nitrogen uptake increased with the decrease of NPK dose application. All of the PGPR inoculation treatments with NPK combination did not increase Azotobacter sp. population and tomato yield. The yield of tomatoes per hectare showed that the PGPR inoculation at a dose of 3 l/ha (1 PGPR + NPK) increased yield by 28.7% compared to 1 NPK (150 kg/ha of Urea, 100 kg SP-36 and 50 kg/ha KCl) dose. Application of 3 l/ha of PGPR inoculant can save up to 75% of NPK usage

Abstract

The aim of this research is to find the characteristics of three PGPR isolates—Azospirillum sp., Bacillus sp., and Pseudomonas sp.: First, by profiling their characteristics in a liquid bioremediation system and secondly by measuring their performance as a bioagent in a soil phytoremediation system using ramie plant (Boehmia niviea L.). A Randomized Block Design in triplicate is used: (1) a Nitrogenfree medium with mineral media containing 1% (wt/v) petroleum hydrocarbons; and (2) 1% (wt/v) glucose medium as control. We tested their petroleum-degrading capacity, nitrogenase activity, phytohormones production, and ramie plant growth. The results showed that both Pseudomonas sp. (98.7%, 81.78% degradation efficiency) and Azospirillum sp. (93.80%, 83.70%) were the superior candidate in both systems. They both show reduced but adequate phytohormone production, managing to improve ramie plant growth. Both also showed reduced but sufficient nitrogen fixing capabilities to improve hydrocarbon degradation activity effectively. Meanwhile, Bacillus sp. has the lowest biodegradation capabilities (84.07%; 78.6%) and lowest nitrogenase activity, while failing to improve plant growth. Therefore Bacillus sp. would be more beneficial in a bacterial consortium where its characteristics (high IAA production) can be coupled with other isolates that can offset its lack of phytohormone or nitrogenase activity.

Abstract

Agricultural soil is possibly threatened by lead (Pb) contamination due to the intensive use of fertilizers. The rhizobacteria were recommended for the bioremediation of soils contaminated by low concentrations of Pb. The experiment was conducted to observe the Azotobacter's ability to proliferate in Pb-contaminated broth and to decrease the Pb availability in soil, Pb uptake by sweet potato roots, and sweet potato growth. The resistance test was performed by growing five Azotobacter isolates in N-free broth with various Pb levels. A pot experiment was conducted in a factorial randomized block design to test three levels of Pb in soil and two Azotobacter isolates. The results showed that Azotobacter Azv4 and A. choroococcum were resistant to 100 mg L-1 Pb in N-free broth. In the pot experiment, Azotobacter Azv4 Inoculation caused less Pb in soil and roots of sweet potatoes grown in Pb-contaminated soil than A. choroococcum. Either Azotobacter or Pb soil did not influence vine length. However, Azv4 was more prominent in increasing branch number, root volume and length; higher Pb in soil reduced branch number but did not affect root parameters. Azotobacter Azv4 increased more shoot and root dry weight compared to A. choroococcum, but both isolates did not change the shoot-to-root ratio (S/R). The Pb contamination only reduced root dry weight and reduced the S/R. This research considered utilizing rhizobacteria Azotobacter for reducing Pb levels in soil and roots; and increasing sweet potato biomass.

Abstract

Azotobacter sp. dan Pseudomonas sp. berperan sebagai binding agent yang dapat menghambat serapan hidrokarbon oleh akar sorgum.ABSTRACTThe synergism interaction between plant-bacteria can be used to improve performance of phytoremediation hydrocarbon (HC) contaminated soil. The study aimed to characterize  Azotobacter sp. and Pseudomonas sp. in petroleum phytoremediation performance using sorghum (Sorghum bicolor L). The research used a randomized block design with two factors: Azotobactersp. inoculation 0%, 1%, 2% and 3% per total petroleum hydrocarbon (TPH) concetration, and Pseudomonas sp inoculation:  0%, 1%, 2% and 3% per TPH concentration. The results showed there was no interaction between Azotobacter sp. and Pseudomonas sp. inoculation to each response variable. The independent effect of 3% Azotobacter sp. to biodegradation HC efficiency was significantly different from the control at phase 4 weeks after planting (WAP), but not significantly different at phase 14 WAP. Pseudomonas sp. effect did not show a significantly different between treatments on the hydrocarbon biodegradation efficiency in the two observation periods. Azotobacter sp. and Pseudomonas sp. population decreased at period 4 WAP, then increased at period 14 WAP. The Sorghum growth was retarded during 14 WAP. Hydrocarbon residues accumulated higher in the roots than in shoot. Azotobacter sp. and Pseudomonas sp. act as a binding agent that can inhibit the hydrocarbons uptake by sorghum roots.
