Abstract

Zona intertidal merupakan wilayah perairan laut yang masih dipengaruhi oleh pasang surut air laut. Salah satu keanekaragaman hayati yang terdapat di zona intertidal adalah lamun. Salah satu tanjung yang memiliki padang lamun adalah Tanjung Bilik Taman Nasional Baluran. Tujuan dari penelitian ini adalah mengetahui distribusi lamun di Zona Intertidal Tanjung Bilik Taman Nasional Baluran dengan menggunakan metode GIS (Geographic Information System). Metode pengambilan data yang digunakan yaitu metode jelajah. Data koordinat habitat lamun dipetakan menggunakan program arcGIS 10. Berdasarkan penelitian yang telah dilakukan ditemukan empat jenis lamun dengan urutan dari distribusi terluas, yaitu Thalassia hemprichii, Enhalus acoroides, Halophila minor, dan Halodule pinifolia. Thalassia hemprichii dan Enhalus acoroides memiliki pola distribusi acak. Halophila minor dan Halodule pinifolia memiliki pola distribusi mengelompok. Abstract

The tree ferns abundant in the ecotourism area of the geoforest erek-erek biosite is represented as ancient plants. However, species list and their composition as diversity richness data is still lacking the in the conservation area. The aims of of this study was to determine the tree ferns and provide the description of the species. Observations on the morphological characters of plants were carried out directly in the field of tree ferns habitat at the Ijen geopark, Banyuwangi, while other morphological and anatomical determinations were conducted in laboratory. The tree ferns identified in the erek-erek forest were two species i.e., Cyathea contaminans and Cyathea orientalis. The main distinguishing characteristics of these species are: stem height, stem surface, attachment of the remaining petiole, scale color, crozier size, indusium and spore shape. This article also provides descriptions of the species and the information regarding conspicuous characters that can be used for species determination in the field. We also propose further conservation efforts to preserve the tree ferns in their habitat.

Abstract

AbstrakBulbophyllum lobbii Lindl. merupakan anggrek dari famili Orchidaceae yang berpotensi sebagai bahan baku obat herbal. Identikasi morfologi anggrek B. lobii memiliki keterbatasan karena kemiripan spesies dengan anggrek lain. Alternatif identifikasi secara molekuler menggunakan sekuen matK dan ITS2 sebagai barcode dalam DNA barcoding diharapkan menjadi salah satu lokus pembeda spesies anggrek B. lobbii secara akurat dan efisien. Penelitian ini bertujuan untuk mengidentifikasi sekuen matK dan ITS2 sebagai penanda molekuler yang efektif untuk anggrek B. lobbii. DNA genom B. lobbii diisolasi dengan metode Cethyl Trimethyl Ammonium Bromide (CTAB) dan amplifikasi DNA dengan PCR. Hasil penelitian menunjukkan sekuen matK dari B. lobbii memiliki tingkat homologi tinggi dengan dua spesies B. lobbii (KY966747.1 dan KY966691.1) dari China dengan nilai Per. Ident sebesar 99,20%, sedangkan sekuen ITS2 memiliki homologi tertinggi dengan nilai Per. Ident sebesar 99,76% pada spesies B. lobbii (MG253848.1) dari Polandia. Hasil analisis menunjukkan sekuen ITS2 dapat mengidentifikasi spesies dari tingkatan subspesies atau diatasnya (ordo atau genus) dan juga meningkatkan resolusi filogenetik yang baik pada hasil BLAST, sedangkan sekuen matK memberikan sedikit kontribusi dalam pengelompokkan hubungan kekerabatan antara spesies B. lobbii dengan spesies pembanding lainnya. Sekuen ITS2 dapat direkomendasikan sebagai penanda molekuler yang paling baik untuk identifikasi sampel anggrek Bulbophyllum, khususnya Bulbophyllum lobbii.AbstractBulbophyllum lobbii Lindl. is an orchid from the Orchidaceae family which has potential as a raw material for herbal medicine. Morphological identification of the B. lobii orchid has limitations due to the species' similarity to other orchids. The alternative molecular identification using matK and ITS2 sequences as barcodes in DNA barcoding is expected to be one of the loci for distinguishing the B. lobbii orchid species accurately and efficiently. This study aims to identify matK and ITS2 sequences as effective molecular markers for the orchid B. lobbii. Bulbophyllum lobbii genomic DNA was isolated using the Cethyl Trimethyl Ammonium Bromide (CTAB) method and DNA amplification by PCR. The results showed that the matK sequence from B. lobbii has a high level of homology with two B. lobbii species (KY966747.1 and KY966691.1) from China with a value of Per. Ident is 99.20%, while the ITS2 sequence has the highest homology with a Per. Ident value of 99.76% with the species B. lobbii (MG253848.1) from Poland. The results of the analysis show that the ITS2 sequence can identify species from the subspecies level or above (ordo or genus) and also improves good phylogenetic resolution in BLAST results, while the matK sequence makes little contribution in grouping the relationship between the B. lobbii species and other comparison species. The ITS2 sequence can be recommended as the best molecular marker for identifying Bulbophyllum orchid samples, especially Bulbophyllum lobbii.

Abstract

Pembelajaran dengan pendekatan pengembangan ketrampilan merupakan strategi yang sering dipakai dalam pembentukan sikap ilmiah. Prakatikum merupakan metode pembelajaran yang diselenggarakan terhadapa mahasiswa STIKES dr. Soebandi Jember dalam pengenalan dan penggunaan Mikroskop, serta pendalaman materi pembelajaran bentuk dan struktur sel. Pelaksanaan praktikum selama pandemi Covid-19 di Jurusan biologi Universitas Jember dilakukan secara online. Tujuan penulisan artikel ini adalah untuk mengetahui efektifitas pembelajaran online terhadap capaian pembelajaran yang direkam dari kegiatan praktikum. Penelitian dilakukan dengan analisis data capaian pembelajaran dan menguji signifikansi skor rerata nilai laporan praktikum dan membandingkannya dengan tes akhir. Hasil penelitian diketahui bahwa capaian pembelajaran terendah adalah tes akhir yang dipengaruhi oleh beban materi pembelajaran yang diujikan. Pembelajaran berbasis aktivitas dan dilengkapi dengan kecukupan pengetahuan yang dimiliki oleh mahasiswa memberikan hasil optimal dalam penilaian capaian pembelajaran

Abstract

GRADSTEIN, R. et al. 2010. Bryophytes of Mount Patuha, West Java, Indonesia. Reinwardtia 13(2): 107-123. - This paper presents the results of a two-day survey of the bryophyte flora of Mt. Patuha and its surroundings near Bandung, West Java, carried out in the framework of the 5th regional training course on bryophyte and lichen diversity and conservation organized by SEAMEO BIOTROP, Bogor, in July 2009. A total of 159 bryophyte species were identified, including 98 mosses, 60 liverworts, and 1 hornwort, representing almost 1/6 of the total bryophyte flora of Java. Three moss species, Bryohumbertia subcomosa (Dix.) J.-P. Frahm, Fissidens gymnogynus Besch. and F. polypodioides Hedw., and one liverwort, Lejeunea pectinella Mizut., are new additions to the Javanese flora. The bryophyte diversity of Mt. Patuha is well representative of the Malesian flora and is rich in uncommon species. However, the relatively poor representation of shade epiphytes and commonness of sun epiphytes and generalists reflect disturbance of the forest by anthropogenic activities. Careful attention should be given to conservation of the remaining natural forest in order to prevent further losses of the rich bryophyte diversity of the area

Abstract

Dumortiera hirsuta, a thalloid liverwort, predominantly grows on the ground floor of coffee plantations in Gumitir, Jember District, East Java, and is known for its rich phytochemical content. This study aimed to comprehensively profile the volatile and non-volatile compounds present in the methanol extract of D. hirsuta and evaluate its antioxidant activity. The thallus was macerated using 96% methanol (1:10 ratio), followed by analysis using Gas Chromatography–Mass Spectrometry (GC–MS) and Liquid Chromatography–Mass Spectrometry Quadrupole Time-of-Flight (LC–MS QTOF). GC–MS identified 37 volatile compounds, with terpenes (29%), phenols (21%), and fatty acids (13%) as dominant classes. Several potent antioxidant sesquiterpenoids, including caryophyllene, guaiene, and aromadendrene derivatives, were notably abundant, along with unique compounds such as phytol, benzoic acid, pyrocatechol, and furanones. LC–MS analysis detected 15 non-volatile secondary metabolites, predominantly flavonoids (e.g., kaempferol-3-O-β-D-glucuronide, luteolin, leucocyanidin), phenolics (sesamol, euparin), and terpenoids (brefeldin A, E-p-coumaric acid), of which nine are well-documented for their antioxidant properties. These compounds were identified with high accuracy (mass error ±4 ppm) across positive and negative ion modes. Antioxidant potential was confirmed through the DPPH radical scavenging assay, which yielded a moderate IC₅₀ value of 101.13 ppm and a strong dose-response correlation (R² = 0.9526). The favourable microclimatic conditions of Mount Gumitir likely contributed to the phytochemical richness observed. Collectively, these findings highlight D. hirsuta as a chemically diverse bryophyte with promising antioxidant constituents, supporting its potential application in pharmacological development and natural antioxidant sourcing.

Abstract

Berkembang pesatnya teknologi dan industrialisasi di Indonesia menyebabkan terjadi pengikisan pengetahuan tradisional terutama pemanfaatan tanaman obat. Pemanfaatan tanaman obat dalam pengobatan tradisional memiliki manfaat yang sangat penting sebagai upaya pengobatan alami dengan meminimalisasi efek samping. Informasi tentang jenis-jenis tanaman obat sangat penting bagi masyarakat maupun para generasi muda, sejak mereka menempuh pendidikan dasar di sekolah. Tujuan dari kegiatan pengabdian ini untuk meningkatkan pengetahuan siswa tentang manfaat tanaman obat dan menumbuhkan rasa memiliki serta melestarikan warisan nenek moyang terkait pengobatan melalui pemanfaatan obat bahan alam. Metode kegiatan pengabdian melalui pemaparan materi tentang jenis tanaman obat dan manfaatnya, kegiatan tanya jawab dan diskusi, praktik menanam tanaman obat di lingkungan sekolah serta kegiatan education games dengan mencocokkan etiket pada pot tanaman obat yang sesuai. Kegiatan ini berhasil meningkatkan pemahaman siswa melalui kegiatan pemaparan materi dan education games. Kegiatan ini diharapkan dapat juga memupuk rasa memiliki dan melestarikan kearifan lokal Indonesia mengenai pemanfaatan tanaman obat sebagai pengobatan tradisional.

Abstract

Epiphytic orchids and ferns in urban forests simulating a lowland tropical forest ecology were studied. This study aims to provide information about the epiphytes' adaptation to living in a dynamic microclimate of the tree canopy. Five epiphytes from the two taxa (orchids and ferns) were observed in their stomatal structure and chlorophyll contents. The stomatal type, size, and density were analysed with microscopic observation. The chlorophyll content was measured using a spectrophotometer. The orchids developed different mechanisms for water conservation compared to the ferns. Orchis develop smaller sizes and denser stomata to support photosynthesis rate as they had water absorbers and storage organs. On the other hand, ferns tend to avoid drought stress by developing fewer numbers and sparse stomata. The higher chlorophyll content in ferns was for photosynthesis efficiency purposes

Abstract

matK is one of the universal loci which is commonly used as a barcode in the molecular identification of plants. Up to now, the problem of matK as a single locus is the use of the entire gene region as barcodes. The long amplicon size of matK raises several problems in DNA barcoding. The aim of this study is to uncover a small region of the matK gene as a potential mini-barcode region for alternative sites to get more efficient and effective results. In silico studies performed include the collection of matK sequence of medicinal orchid from GenBank, the amplicon size from matK primer sets that are commonly used in the Orchidaceae family, and PCR analysis. Data analysis was done by comparing the results of PCR amplification from several primer sets: matK 390F-1326R, 743F-R2, 712F-1154R and 712F-1216R. The results show that all primer pairs are able to amplify the DNA template isolated from Dendrobium purpureum. However, the shorter DNA band obtained from matK 743F-R2, 712F-1154R and 712F-1216R primers sets were visualized more thick than the longer amplicon band produced from matK primer 390F-1326R. This indicates that short matK sequence for DNA barcoding (mini-barcode) is more efficient in amplifying DNA templates compared to generally applied barcoding using full length sequences.
