Abstract

Endophytic bacteria are microbes that grow on plant tissue and can form a colony on plant tissue without producing a negative effect on the host. The endophytic bacteria have the potential to have the same activity and produce secondary metabolite as their host. The root of bitter melon (momordica charantia L.) has been known to have antibacterial activity, thus enabling the presence of endophytic bacteria, which also have antibacterial potential. This present research aims to isolate and characterize endophytic bacteria found in bitter melon (Momordica charantia L. The method used in the isolation stage is direct lanting so that the endophytic bacteria are obtained from plant tissue; macroscopic and microscopic characterization stage; inhibition test stage; inhibition test stage using disc method (Kirby-Bauer). In accordance with the results of macroscopic and microscopic characterization, the two bacteria indicate different characteristics.

Abstract

Off-label drugs can be interpreted as drugs that are used not in accordance with drug information, distribution permits and sales licenses from the Marketing Authorization (MA) related to indications, age, dosage, route of administration and contraindications. The use of off-label drugs in children occurs because of incomplete data on pharmacokinetics, pharmacodynamics and side effect of a drug. It is because clinical research in children is quite difficult and not in accordance with research ethics and morals, so it has the potential to cause errors in treatment and unwanted drug reactions. This study aims to describe the use of off-label drugs in hospitalized pediatric patients at RSUD of Dr. M.M Dunda Limboto, Gorontalo District, Gorontalo Province. This is an  observational study using cross sectional design and retrospective data collection. The sampling technique applies a purposive sampling technique with a total of 155 patients who meet the inclusion criteria during the January-June 2020 period. Abstract

In the developing materials for traditional medicines, it is necessary to standardize the quality of plant materials. Tuhus, quality, stability, and safety. This study aims to determine several standardization parameters and determine the flavonoid content of the 70% ethanol fraction of aur-aur leaves (Commelina longifolia L.) and bungango leaves (Flueggea virosa Royle) to ensure that the fractions have quality and measurable parameter values. The fraction was standardized with two parameters, thus specific and non-specific, and flavonoid content test using UV-Vis spectrophotometry method. The results of phytochemical screening, the fraction containing flavonoid compounds is 70% ethanol fraction. Standardization of specific parameters showed the organoleptic results of both aur-aur (Commelina longifolia L.) and bungango (Flueggea virosa Royle) leaf fractions in the form of a semi-solid viscous fraction, black in color, and characteristic odor. The content of water soluble extract and ethanol soluble extract of aur-aur (Commelina longifolia L.) leaf fraction were 22.84%, ethanol soluble extract 25.03%, while the fraction of Bungango (Flueggea virosa Royle) leaf was 30.70%. and 27.31%. Standardization of non-specific parameters showed the results of water content 21.40%, total ash content 8.92%, acid insoluble ash content 0.043%, drying shrinkage 10.48%. The results of non-specific standardization of bungango leaves (Flueggea virosa Royle) showed a moisture content of 14.45%, a total ash content of 9.62%, an acid insoluble ash content of 1.31%, a drying shrinkage of 10.64%. The results for the analysis of the flavonoid content of the 70% ethanol fraction of aur-aur leaves and bungango leaves were carried out at a wavelength of 438.36 nm. The total flavonoid content was calculated by the standard linear equation of quercetin, namely y= 0.0648x + (-0.0232) with a correlation coefficient (r2) = 0.9948 with the total flavonoid content of aur-aur leaf and bungango leaf each of 3.57. % and 9.578%.

Abstract

Hyperuricemia is a condition in which the kidneys fail to excrete uric acid, resulting in high levels of uric acid. (Kaneko et al., 2014). Uric acid will be excreted into the kidneys along with urine. The decrease of uric acid secretion into the kidney tubules due to interference of uric acid elimination to the kidneys which causes uric acid levels in the blood to increase (Ningtiyas Ramadhian, 2016). An adult male is suffering from hyperuricemia when his serum uric acid level is more than 7.0 mg/dl, while hyperuricemia in adult women occurs when the serum uric acid level is above 6.0 mg/dl. Hyperuricemia therapy management in the community is very diverse, ranging from traditional medicines and synthetic drugs. One of the traditional plants used as traditional medicine is pandan leaves. This study aimed to determine the anti-hyperuricemia effect of ethanol extract of Pandanus amaryllifolius leaves on mice (Mus musculus L.). The study involved 25 male mice which were divided into 5 groups; each group consisted of five male mice. Group I was a negative control with Na-CMC 1% w/v, while Group II was a positive control with Allopurinol 0.117 g w/v. Further, Groups III, IV, and V were given Ethanol Extract of 0.5 gr w/v, 1 g w/v, 1.5 g w/v of Pandan Leaves, respectively. The results showed that there was no significant decrease in uric acid levels in group I. Meanwhile, Groups II, III, IV, and V showed a significant reduction in uric acid levels with identical effectiveness. The results of the study were then tested using the One-Way ANOVA statistical test; the p-value was 0.01 (α 0.01). The best dose was 1.5 g of extract since there was a significant difference from the pandan leaves extract with 1.5 g (p 0.01). In conclusion, pandan leaves extract can reduce uric acid levels in the blood. This was evidenced by a decrease of 4.1 mg/dl in uric acid levels in mice during the experiment.
