Abstract

Orthodontic treatment aims to correct the improper arrangement of teeth. Tooth movement caused by orthodontic treatment causes tension and compression areas in the tooth's supporting tissues. The periodontal ligament will respond to this stimulus by triggering the inflammatory process. Flavonoids in Sauropus androgynus act as anti-inflammatories by inhibiting lipoxygenase and cyclooxygenase. This experimental research was conducted on 32 guinea pigs, which were grouped into 2 groups: the control group had a rubber separator applied for 14 days, and the experimental group had a rubber separator applied also given ethanol extract of Sauropus androgynus 78.3 mg/kg BW. Samples were taken on days 0, 3, 7 and 14 and statistically analyzed using an independent T-test and Mann-Whitney test. The results showed that the ethanol extract of Sauropus androgynus affected the number of inflammatory cells. Based on statistical results, there were significant differences between the control group and the treatment group in neutrophil cells (p=0.000), eosinophils (p=0.000), lymphocytes (p=0.002), and monocytes (p=0.000). In contrast to macrophage cells, statistical results showed no significant difference between the control and experimental groups with p=0.064. The observed effects of the ethanol extract of Sauropus androgynus on periodontal ligament inflammatory cells (neutrophils, eosinophils, lymphocytes, and monocytes) suggested its potential as an anti-inflammatory agent.

Abstract

Abstract

Dental caries and periodontal diseases are the most common diseases affecting the oral cavity. This can be prevented by maintaining oral hygiene. Toothpaste plays an important role in the reduction of plaque bacteria and inflammatory conditions. Rambutan honey has potential as a natural ingredient to make toothpaste with antibacterial and antioxidant properties owing to the presence of hydrogen peroxide and catalase enzymes, pH, osmolarity, and flavonoid content. The purpose of this study was to obtain a rambutan honey toothpaste formulation that effectively inhibits Streptococcus mutans growth. The laboratory experimental research method was used to combine CaCO3, glycerin, Na CMC, SLS, sodium benzoate, saccharin, menthol, and aquadest as the toothpaste base, which served as the control group. Subsequently, natural ingredients of rambutan honey at concentrations of 20%, 40%, and 60% were added to the toothpaste base to create rambutan honey toothpaste. The formulation of the rambutan honey toothpaste was followed by an antibacterial activity test using the Kirby Bauer method of disc diffusion, where the inhibition zones formed on Mueller Hinton Agar (MHA) medium were calculated. Data were analyzed using one-way and post hoc Tukey’s tests (p<0.05). The results showed that the formulation of rambutan honey toothpaste with a concentration of 60% had the greatest inhibition with 13.88±0.13 mm with a significant difference in each group (p = 0.000), so it can be concluded that rambutan honey toothpaste can inhibit the formation of Streptococcus mutans.  Keywords: antibacterial, formulation, rambutan honey, toothpaste, Streptococcus mutans

Abstract

Abstract

Oral health faces significant challenges due to increasing dental infections by pathogenic microbes such as Porphyromonas gingivalis, Streptococcus mutans, and Candida albicans. Topical antioxidants in toothpaste, gel, and mouthwash are often used to treat dental diseases. There is a growing interest in finding natural alternatives for oral hygiene without side effects compared to conventional toothpaste formulations. Nigella sativa (black cumin) has various pharmaceutical efficacy, making it a valuable plant-based source medicinal compound. This study evaluated black cumin ethanol extract's (BCSE) antimicrobial and antioxidant activities. Antibacterial effectiveness was evaluated through Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC), and disc diffusion assays against P. gingivalis, S. mutans, and C. albicans. Antioxidant activity was reported based on DPPH scavenging, ABTS reduction, H2O2 scavenging, and NO scavenging assays. Black cumin seed ethanolic extract (BCSE) showed antibacterial activity of 4.49 mm, 4.33 mm, and 7.12 mm for P. gingivalis, S. mutans, and C. albicans, respectively, with zones of inhibition increasing in a concentration-dependent manner (p<0.05). MIC and MBC evaluations also revealed that MIC for S. mutans was achieved at 12.5% BCSE concentration, while P. gingivalis and C. albicans required 25%, and MBC reached 100%. The IC50 for DPPH, ABTS, NO, and H2O2 were 25.41, 69.93, 87.18, and 95.16 μg/ml, respectively. The antioxidant activity increased in a concentration-dependent manner, especially at 100 µg/ml of BCSE. Based on these results, BCSE could be an optional ingredient in toothpaste.

Abstract

Morsicatio buccarum and labiorum are conditions generally caused by chronic habitual cheek and lip biting. Objective findings present thick, shaggy, irregular white lesions on the buccal and labial mucosa. Morsicatio is frequently associated with psychological conditions. This case report aims to assess the depression, anxiety, and stress levels in a patient with morsicatio buccarum and labiorum. A 22-year-old male patient visited the Integrated Polyclinic of Universitas Jenderal Achmad Yani Dental Hospital to check his oral status. Clinical examination revealed incidental findings. Extraoral examination showed clicking on both temporomandibular joint (TMJ) and lip desquamation sides. Intraoral mucosa examination revealed white lesions in the form of unsweepable plaque, irregular, slightly elevated from surrounding tissue, and interspersed with erythematous erosion areas on the buccal and labial mucosa. A pigmented lesion was found on the anterior attached gingiva mucosa on both the upper and lower jaws. The patient admitted to chronic habitual cheek biting since childhood. The Depression, Anxiety, and Stress-21 (DASS-21) assessment showed anxiety reaching a severe level. A psychological approach is needed as part of the treatment planning in morsicatio cases. Abstract

Early diagnosis of dengue hemorrhagic fever (DHF) is very important in determining further management. Diagnostic kits are intended as relatively faster, easier, and cheaper diagnostic tools. The objectives of this experimental laboratory study was to prepare a diagnostic kit candidate to detect dengue antibody in human serum using co -agglutination methods by utilizing protein A positive Staphylococcus aureus as a carrier to bind Fc immunoglobulin fractions without altering its ability to bind antigen. To enhance the sensitivity and specificity of the diagnostic kit, chicken IgY was used as one of the components. Rabbit immunoglobulin chicken anti-IgY was used as an intermediary connection between protein A positive and Staphylococcus aureus with chicken IgY. Solution A contains Staphylococcus aureus Cowan I and rabbit immunoglobulin chicken anti-IgY serum while Solution B contains anti-dengue chicken serum and dengue antigen. The laboratory experiments produced a formula of A: B = 1~3: 1~3 (v/v), which was then tested to human serum and compared with IgM and IgG ELISA tests as the gold standard. Data on the results of the test were statistically analyzed using chi - square test. Of the 65 qualified samples used, 47.69% resulted in true positive and 41.54% resulted in true negative. Meanwhile, the remaining samples demonstrated false negative (6.15%), positive predictive value (91.2%) and negative predictive value (87.1%). The sensitivity and specificity were 89% and 90%, respectively, with no significant diffe rences (p>0.05). Therefore, co-agglutination method using S. aureus with protein A positive as a carrier can be proposed as a diagnostic kit candidate to detect human serum dengue antibody to screen dengue hemorrhagic fever cases.
