Abstract

Meniran (Phyllanthus niruri L.) is a plant that is reported to contain flavonoids and shows the activity as a type 2 anti-diabetic by a mechanism of inhibiting the absorption of glucose. Flavonoid compounds such as rutin, quercetin and hesperidin shows anti-diabetic activity. This research aims to determine the levels of total flavonoids and docking studies of flavonoids from meniran against α-amylase and α-glucosidase. Analysis of content of total flavonoids utilizes colorimetric method using AlCl3 and routine standard, the docking study uses Autodock Vina (version 1.1.2) program with the assistance of AutoDockTools (ADT) and visualization of docking results using Discovery Study. The results of the determination of total flavonoids level was 12.32 ± 0.53 mgRE/g extract whereas in ethyl acetate fraction and water each were 19.33±0,68 and 20.07±1.23 mgRE/g respectively. Results of all flavonoids dicking study shows the potential to bind to both receptors, rutin has better binding energy towards α-amylase whereas against α-glucosidase is demonstrated by 3-O -β-D-glucopyranosyl-(2-1)-O-β-D-xylopyranoside quercetin.

Abstract

Belimbing wuluh (Averrhoa bilimbi Linn.) has antihypercholesterolemic activity. This study aims to determine antihypercholesterolemic potential, optimal dosage and effect of water extracts of Belimbing wuluh (Averrhoa bilimbi Linn) on the index of rat organs. Test animals used were male Wistar rats, divided into 4 groups: normal control, negative control, positive conrol (simvastatin 0.18 mg/kg), and the aqueous extracts of fruit Belimbing wuluh dose of 63 mg/kg. The parameter which was measured in this study were total cholesterol levels which obtained using CHOD-PAP method, the weight of rats for 50 days, and organ indexes. Those parameters were further tested with One Way ANOVA. In parameter decrease blood cholesterol levels of rats between groups positive and group aqueous extract of the fruit starfruit  dose of 63 mg/kg did not experience a statistically significant difference. Meanwhile, the group aqueous extract of the fruit starfruit dose 63 mg/kg and a negative control experiencing significant difference (p<0.05). Conclusion, aqueous extract of the fruit starfruit dose of 63 mg/kg had the effect antihypercholesterolemic seen from the decrease in total cholesterol levels from day 30 and day 50, as well as statistically significant different from the negative control, and can affect the index spleen and pancreas but does not affect the index of the kidneys, liver, heart, and lungs.

Abstract

Ciplukan (Physalis angulata L.) empirically has been used to treat sore throat, inflammatory of the airways, gingivitis, and other diseases. This research was to determine the inflammatory activity of water herb extract of P. angulata L. on white male rats edema Wistar strain induced carragenan. P. angulata L. herbs were extracted using infundation method and were dried with frezee drying. Parameter that would be observed of this research was the inhibition foot edema of rat after induction of 0.1 mL of λ-carragenan 2% for 360 minutes. Measurement of edema volume was using pletismometer. The results of this research showed water herb extract of P. angulata L. in dose 100, 200, and 400 mg/kgBW has potensial as an antiinflammatory drug by percent respectively were 20.13; 28.93; and 34.70%. The three doses hasn’t showed any significant difference (p>0.05) with the positive control of diclofenac sodium dose 4.5 mg/kgBW was 33.90%. Conclusion of this research states water herb extract of P. angulata L. has antiinflammatory activity with effective dose of 400 mg/kgBW.

Abstract

Kratom (Mitragyna speciosa Korth.) has been known to have an analgesic opioid effect (antinociceptive). The major compound of kratom leaf is mitraginin, which has strong affinity on opioid receptor. The aim of this research is to prove antinociceptive effect of aqueous fraction of kratom leaf and its effective dose. The simplicia of kratom leaf was extracted with methanol 96%. Methanol extract fractioned with n-hexane, dichloromethane, ethyl acetate, and aquadest. This research used hot plate method on male Swiss Albino mice. The subject divided to 5 groups, negative control group, morphine 5,46 mg/kgBW, aqueous fraction 140, 240 and 560 mg/kgBW. Latency time was measured every 15 minutes over 2 hours period. Analytic statistical of latency time using One Way ANOVA shows that the aqueous fraction at the dose of 140, 280 and 560 mg/kgBW significantly differentiate with negative control group and positive control group. The antinociceptive effect increases with increasing doses. The three doses showed that the antinociceptive effect was no better than the positive control (Morphine)

Abstract

Extraction is an important step in separating bioactive compounds from the plant. The selection of extraction technique is also important in the standardization of herbal products, for if not observed it can remove the desired soluble constituents. One of Indonesia’s endemic plants which have been proven to have an antioxidant activity is Salam leaves (Syzygium polyanthum). This study aims to determine the effect of extraction method on total phenolic content and antioxidant activity of S. polyanthum leaves by counteracting free radicals mechanism using DPPH. The S. polyanthum leaves were extracted by maceration, soxletation, and infusa methods, and tested for the total phenolic content and the antioxidant activity. The results showed that the total phenolic content of each method extration is different. The total of the phenolic content extraction by maceration, soxhlet, and infusa method respectively were 338.62±21.3; 227.72±21.6; and 144.48±8.2 mgGAE/g. The best antioxidant activity was maceration method with IC50 17.53±0.11 µg/mL followed by soxhlet and infusa which were 18.73±0.31 and 40.26±0.18 µg/mL. The research conclusion is that the extraction method has an effect on the total phenol and antioxidant activity of S. polyanthum leaves.

Abstract

Phyllanthus niruri L. is known to promote hair growth. Menthol is one of the enhancers which promotes drug penetration into the skin. The aim of this research is to determine the potential of menthol in P. niruri Ethanol Extracts gel preparation as a hair growth promoter. The test animals were randomized into 4 groups: normal group, negative control (gel without extract and menthol), E1 group (Ethanol Extracts of P. niruri 5% gel without menthol) and E2 group (Ethanol Extracts of P. niruri 5% gel with menthol). Hair growth activity test was conducted by applying the gel on the rats’ back, and hair length was measured on the 7th, 14th and 21st days, while the hair was weighed on the 21st day. The results showed that the E2 group had an average hair length and hair weight of 12.45±3.457 mm and hair weight of 28.53±7.681 mg, greater than the E1 group of 10.67±2.455 mm and 19.87±9.552 mg respectively. Based on these results, it can be concluded that adding menthol in P. niruri Ethanol Extract gel preparation can influence hair growth and thickness.

Abstract

Buas-buas, meniran, secang, and rosella have biological and pharmacological activities as antioxidants. The combination of the four plants is expected to provide a more potent synergistic effect on antioxidant activity. The purpose of this study was to analyze the total phenol content, total flavonoids, and antioxidant effects before and after combination. The combination of extracts, buas-buas, meniran, secang, and rosella which are used in sequence is (1: 1: 1/2: 1/2), (1: 2: 1/2: 1/2), and (2: 1 : 1/2: 1/2). Plants used in the form of simplicia was extracted by maceration method. Radical capture activity uses DPPH and IC50 values are determined. Determination of total phenol is expressed equivalent to gallic acid. Total flavonoids are expressed as quercetin equivalents. The phenol and flavonoid content obtained are then correlated with antiradical activity. The results showed that the best IC50 values were in the combination of ratios (1: 1: 1/2: 1/2) that is (11.0 µg / mL), then (1: 2: 1/2: 1/2) which was 13.3 µg / mL, and (2: 1: 1/2: 1/2) which is 19.4 µg / mL. The highest total phenolic and flavonoid content in the ratio (1: 2: 1/2: 1/2) is 33.57% w/w EAG and 74.00% w/w EQ. Correlation analysis between IC50 values with total phenolic and flavonoid contents showed a positive correlation with R2 values of 0.8236 and 0.0102 with positive slope. Thus, it can be concluded that the total phenol content influences free radical scavenging activity by 82.36%, while the effect of total flavonoid content was only 1.02%.

Abstract

Antioxidant Activities Combination Of Brewing Buas-Buas (Premna serratifolia Linn.), Meniran (Phyllanthus niruri L.), Secang (Caesalpinia sappan Linn.) And Rosela (Hibiscus sabdarifa) with DPPH (2,2-Diphenyl-1-1) MethodBuas-buas leaves, meniran herbs, sappan wood, and roselle flowers are rich in phenols and flavonoids. The purpose of this study was to determine the IC50 value of the combination of wild-savage leaves, meniran herbs, sappan wood, and roselle flowers brewed in warm air using the DPPH method. The infusion is obtained from the simplicia of wild-savage leaves, meniran herbs, sappan wood, and roselle flowers which are powdered in a ratio of 1:1:0,5:0,5, 2:1: 0,5: 0,5, and 1:2: 0,5: 0,5. The results of the data show the IC50 values for each comparison, namely 119.4 µg/mL, 100.9 µg/mL, and 131 µg/mLKeywords:  DPPH; Antioxidant; Buas-Buas; Meniran; Secang; RoselaDaun buas-buas, herba meniran, kayu secang dan bunga rosela kaya akan fenol dan flavonoid. Daun buas-buas, meniran, kayu secang, dan bunga rosela sudah terkenal di masyarakat dan sudah banyak digunakan, akan tetapi belum banyak ditelusuri mengenai kombinasi  antara tanaman serta pemanfaatan tanaman sebagai antioksidan dan diolah menjadi minuman seduhan, oleh karena itu dilakukan penelitian ini. Tujuan dari penelitian adalah untuk mengetahui nilai IC50 dari kombinasi daun buas-buas, herba meniran, kayu secang dan bunga rosela yang diseduh dalam air hangat menggunakan metode DPPH. Kombinasi seduhan pada penilitan ini diharapkan dapat memberikan informasi kepada masyarakat mengenai seduhan yang kaya antioksidan. Seduhan diperoleh dari simplisia daun buas-buas, herba  meniran, kayu secang dan bunga rosela yang diserbukkan dengan perbandingan 1:1: 0,5: 0,5, 2:1: 0,5: 0,5 dan 1:2: 0,5: 0,5. Hasil data menunjukkan nilai IC50 masing-masing perbandingan yaitu 119,4 µg/mL, 100,9 µg/mL , dan 131,7 µg/mL.Kata kunci : DPPH; Antioksidan; Buas-Buas; Meniran; Secang; Rosela

Abstract

Standardization Of Buas Buas Leaf (Premna serrativeolia Linn.) Ethanol Extract And Secang Wood (Caesalpinia sappan Linn.).Extract standardization is a parameter used to ensure the safety and quality of extracts before being developed into pharmaceutical preparations. Meanwhile, Buas buas leaves (Premna serrativeolia Linn.) are useful for treating gastric disorders, carminatives, hemorrhoids, diuretics, fever, constipation, shortness of breath, and also as anti-inflammatory expectorants, antioxidants, antimicrobials, and anticoagulants. Furthermore,secang wood (Caesalpinia sappan Linn.) is widely known as a nutritious herbal drink. This research aims to determine extracts standardization including specific parameters (organoleptic and phytochemical screening) and non-specific (drying losses). The multilevel maceration with n-hexane and ethanol solvents extraction method was used. The results showed that the percentage of drying shrinkage in the ethanol extract of buas-buas leaves  and secang wood was 18.99% ± 0.60 and 15.27% ± 1.46 respectively. Moreover, the ethanol extract of buas-buas leaves has a thick shape, blackish-brown in color, has a distinctive aromatic odor, while secang wood has a thick, brownish-red color, as well as a distinctive aromatic odor. The Buas buas leaves extract contains flavonoids, phenolics, tannins, saponins, and terpenoids, while secang wood contains only flavonoids, tannins, and terpenoids.Keywords : Extract standardization, Graded maceration, Buas buas leaves, Secang wood.Standarisasi ekstrak merupakan parameter untuk menjamin keamanan dan kualitas ekstrak sebelum dikembangkan menjadi sediaan farmasi. Daun buas-buas (Premna serratifolia Linn.) berkhasiat sebagai mengobati gangguan lambung, karminatif, diuretik, demam, sesak nafas, ekspektoran antiinflamasi, antioksidan, antimikroba, sembelit, ambeien, dan antikoagulan. Kayu secang (Caesalpinia sappan Linn.) dikenal sebagai minuman herbal yang berkhasiat. Penelitian ini bertujuan untuk mengetahui standarisasi ekstrak meliputi parameter spesifik (organoleptis dan skrining fitokimia) dan non-spesifik (susut pengeringan). Metode ekstraksi pada penelitian ini menggunakan maserasi bertingkat dengan pelarut n-heksan dan etanol. Hasil penelitian menunjukkan bahwa persentase susut pengeringan ekstrak etanol daun buas-buas sebesar 18,99% ± 0,60  dan ekstrak etanol kayu secang sebesar 15,27% ± 1,46. Ekstrak etanol daun buas-buas memiliki bentuk yang kental, berwarna coklat kehitaman, berbau aromatik khas, sedangkan ekstrak etanol kayu secang memiliki bentuk yang kental, berwarna merah kecoklatan, berbau aromatik khas. Ekstrak etanol daun buas-buas mengandung senyawa flavonoid, fenolik, tanin, saponin, terpenoid. Sementara itu, ekstrak etanol kayu secang mengandung flavonoid, tanin, dan terpenoid.Kata kunci : Standarisasi ekstrak, Maserasi bertingkat, Daun buas-buas, Kayu secang.

Abstract

The leaf katuk (Sauropus androgynus L. Merr) extract at a dose of 400 mg / kg BW had antiinflammatory activity in rat by induction of carrageenan. Patch are adhesive plasters containing medicinal substances placed on the skin to delivery active substances. Patch can offer practicality, comfort and accuracy of dosage in the application of leaf S. androgynus extract.This research was a laboratory experimental research by making patch of leaf S. androgynus extract and inflammation effect used rats as its animal test. Inflammatory healing activity was seen from the percentage reduction in inflammation in rats after patch application. Patch of leaf S. androgynus extract dose 400 mg / kg BW has percentage of inflammatory inhibition ranged between 66,67-100%. Statistical test results using one-way anova patch leaf S. androgynus extract showed good effectiveness in curing inflammation. Effectiveness between leaf S. androgynus extract patch and diclofenac sodium patch relatively the same in curing inflammation.
