Abstract

Abstract

Nystatin is the first line drug for oral candidiasis which is very difficult to dissolve in water. The Orabase Emulgel dosage form has good drug delivery capacity, as gel formulations generally provide faster drug release compared to ointments and creams. Gelling agent components play a role in determining the physical characteristics and permeability of emulgel. The type of gelling agent that will be used is carbomer 940 because it is a strong mucoadhesive. Nystatin is formulated in the form of orabase emulgel with 3 concentrations of carbomer 940, namely F1 (0.5%), F2 (1%), and F3 (2%). The physical characteristics of the preparations observed were organoleptic, homogeneity, pH, emulgel type, viscosity, adhesive power, spreadability, protective power and washability, and stability was tested. The results of the orabase emulgel nystatin preparation on physical characteristics using the MANOVA test (p<0.05) had an effect on pH, viscosity, spreadability and protective power.

Abstract

Mefenamic acid is a derivative of anthranilic acid that functions as a nonsteroidal anti-inflammatory drug (NSAID), antipyretic, and analgesic. Mefenamic acid is included in the Biopharmaceutical Classification System (BCS) class II which has low solubility and high permeability. The manufacture of SNEDDS requires a non-ionic surfactant with an HLB (Hydrophilic-Lipophilic Balance) value of more than 10. Mefenamic acid has high solubility in the surfactant Labrasol which has HLB 12 and Tween 80 which has HLB 15. This study aims to develop mefenamic acid into a Self Nano-Emulsifying Drug Delivery System (SNEDDS) with the optimization of labrasol and tween 80 as surfactants. The optimization of mefenamic acid SNEDDS is made using the simplex lattice design method. In the optimization, 6 running SNEDDS formulas were obtained with mefenamic acid. Chemical physics testing includes organoleptic evaluation, pH, viscosity, percent of transmitters, emulsification time, stability, particle size, potential zeta, and level determination. The results showed that the optimal combination of labrasol and tweenat SNEDDS mefenamic acid 80, namely 24% Labrasol and 1% Tween 80, with a desirability value of 0.984, had a clear, non-rancid, homogeneous, liquid-shaped color, pH of 4.69, viscosity 31.33 cP, transmitter percentage 96.80%, emulsification time 29.11 seconds, particle size 44.30 nm, zeta potential -28.87 mV, and a polydispersivity index of 0.563, with a percentage of levels of 100.08%, 100.70%, and 99.87%, respectively. Key words: Self Nano-Emulsifying Drug Delivery System (SNEDDS), Labrasol, Tween 80

Abstract

Background: Green coffee beans (Coffea liberica) are a rich source of bioactive compounds, including caffeine, chlorogenic acid, flavonoids, and polyphenols, which have potential health-promoting properties. Caffeine has been reported to enhance thermogenesis, stimulate fat oxidation, and increase energy expenditure, making it a promising natural compound for weight management. This study aimed to determine the caffeine content of green Coffea liberica beans using UV–Visible spectrophotometry and to evaluate the implementation of green coffee extract consumption on body weight reduction. Methods: An experimental study was conducted by extracting green coffee beans with 96% ethanol followed by phytochemical screening and quantitative determination of caffeine using UV–Visible spectrophotometry at a maximum wavelength of 272 nm. The analytical method was validated by assessing linearity, accuracy, and precision. A one-group pretest–posttest design was employed to evaluate the implementation of green coffee extract in overweight adults with a body mass index (BMI) ≥25 kg/m². Body weight was measured before and after the intervention, and statistical analysis was performed at a 95% confidence level. Results: The spectrophotometric method demonstrated satisfactory linearity, accuracy, and precision, indicating its suitability for caffeine determination. Green Coffea liberica beans were confirmed to contain caffeine and other bioactive compounds that contribute to antioxidant activity. Consumption of green coffee extract showed a tendency to reduce body weight following the intervention period. Conclusion: UV–Visible spectrophotometry is a reliable method for determining caffeine content in green Coffea liberica beans. The presence of caffeine and other bioactive compounds supports the potential application of green Liberica coffee as a functional food ingredient for weight management.

Abstract

Acne vulgaris is a common skin disorder, particularly among adolescents, often caused by the bacteria Cutibacterium acnes and Staphylococcus epidermidis. Antibiotic resistance in acne treatment necessitates the development of natural-based alternatives. This study aims to formulate and evaluate the antibacterial activity of acne patches based on kawista bark extract (Limonia acidissima L.) at varying concentrations of 25%, 50%, and 75%. Extraction was performed by maceration using 96% ethanol, and patches were formulated with a combination of HPMC and PVP polymers. Physical property tests included organoleptic evaluation, weight uniformity, drying shrinkage, thickness, folding endurance, and pH. Antibacterial activity was tested using the diffusion method against Cutibacterium acnes and Staphylococcus epidermidis. Results showed that all formulations met good physical patch standards, being : homogeneous, stable, thickness less than 1 mm, folding endurance greater than 300 times, and pH of 6. Antibacterial activity increased with higher extract concentration, with the largest inhibition zones observed at 75% concentration (15.16 mm for C. acnes and 18.05 mm for S. epidermidis), approaching the effectiveness of the positive control (clindamycin 10%). This study demonstrates that kawista bark extract acne patches have potential as an alternative topical acne therapy with strong antibacterial activity and physical characteristics that meet modern patch preparation standards.
