Microbiological Quality Analysis and Identification of Lactic Acid Bacteria from the Product of Fermentation of Mackerel Sukmawati Sukmawati

Abstract

Mackerel is a type of fish that has high nutritional value, this fish is a source of protein and is in great demand by many people. Mackerel can be processed into various types of processed food, one of which is through the fermentation process. The purpose of this study was to determine the microbiological quality, to determine the characteristics and the number of lactic acid bacteria , and to identify the species of lactic acid bacteria found in the fermented mackerel product. The results showed that the microbiological test of mackerel fermented products had the highest total plate number (ALT) in the Fit.L sample, namely 4.2x105 cfu/g, followed by the Fit.C sample with a value of 2.4x104 cfu/g, and the lowest value was in the Fit. A sample  4.3x103 cfu/g. The total plate number (ALT) in each test sample stated that the number did not exceed the maximum limit of the Indonesian national standard. Microbiological test results through the E.coli estimator test of the three samples showed a value of <3 APM/g, then the reinforcement test results were declared not to contain E.coli bacteria.The number of lactic acid bacteria (LAB) present in the mackerel fermented product sample showed the highest value in the Fit.L sample, namely 1.8x104 cfu/g, then in the Fit.C sample with a value of 2.6x103 cfu/g, while the lowest was in the Fit. A sample whose value is 3.6x102 cfu/g. The morphological characteristics of the LAB colonies from the three samples showed the same characteristics, namely round shape, smooth edges and convex elevation. The results of the identification of the two isolates from the two samples stated that the Fit.A isolate was identified as Staphylococcus condimenti strain DSM 11674 and the Fit.C isolate was identified as Lactobacillus reuteri strain 1444

Abstract

Kelurahan Jamaimo, Distrik Mariat, Kabupaten Sorong memiliki potensi lahan pertanian seluas 20 ha dengan topografi beragam, dari datar hingga berbukit, yang rentan terhadap erosi dan penurunan kualitas tanah. Masyarakat setempat, dengan berbagai latar belakang profesi, sebagian besar adalah petani yang masih mengandalkan sistem monokultur. Sistem agroforestry diperkenalkan sebagai solusi untuk meningkatkan keberlanjutan lahan melalui integrasi tanaman dan pohon guna mencegah erosi, meningkatkan kesuburan tanah, dan mengurangi ketergantungan pada bahan kimia. Kegiatan sosialisasi dilakukan dengan metode ceramah dan tanya jawab, melibatkan 22 petani untuk memberikan pemahaman mengenai teknik dan manfaat agroforestry. Hasil kegiatan menunjukkan bahwa tingkat pengetahuan petani tentang agroforestry masih rendah, sehingga diperlukan edukasi berkelanjutan untuk mendorong praktik pertanian yang lebih ramah lingkungan, produktif, dan berkelanjutan. Sosialisasi yang diberikan dapat meningkatkan pemahaman tentang agroforestry, dengan beberapa hal yang terkait pemanfaatan lahan secara optimal dan berkelanjutan, pengurangan ketergantungan pada pupuk kimia, dampak positif dari agroforestry dan kesadaran akan keberlanjutan lahan. Sehingga kedepannya kesadaran kelompok tani akan menerapkan system agroforestry dan keberlanjutan lahan dapat dilaksanakan lebih baik lagi dengan pendampingan secara kontinyu pada kelompok tani.

Abstract

Cellulose can be found in whole parts of plants. Cellulose can be degraded by cellulose enzyme. Cellulose enzyme is produced by cellulolytic bacteria which has ability to degrade the cellulose and fiber. Cellulose enzyme is extremely important in industry sector. This research aims to isolate the cellulosic bacteria of peel waste of Banana in Sorong, West Papua. This research used isolation and purification method by using 1 gram of Banana peel. It was isolated in CMC (Carboxymethyl cellulose) selective media and morphology characteristic test using 400X-1000X microscope and zone index test. The result showed that the cellulosic ability was found in isolate B with pure zone index 3mm and the smallest pure zone index was in isolate H with pure zone index 0,8 mm. while, the isolate C had zone pure index 1,5 mm, isolate D 1,625 mm, isolate E 1 mm, isolate F 0,714mm, and isolate G 2 mm.
